The transcriptional mechanisms governing the development and plasticity of somatopic sensory maps in the cerebral cortex have not been extensively studied. In particular, no studies have addressed the role of epigenetic mechanisms in the development of sensory maps. DNA methylation is one the main epigenetic mechanisms available to mammalian cells to regulate gene transcription. As demethylation results in embryonic lethality, it has been very difficult to study the role of DNA methylation in brain development. We have used cre-lox technology to generate forebrain-specific deletion of DNA methyltransferase 1 (Dnmt1), the enzyme required for the maintenance of DNA methylation. We find that demethylation of neurons in the cerebral cortex results in the failure of development of somatosensory barrel cortex. We also find that in spite of functional thalamocortical neurotransmission, thalamocortical long-term potentiation cannot be induced in slices from Dnmt1 conditional mutants. These studies emphasize the importance of DNA methylation for the development of sensory maps and suggest epigenetic mechanisms may play a role in the development of synaptic plasticity.

Les m&eacute;canismes transcriptionnels r&eacute;gissant le d&eacute;veloppement et la plasticit&eacute; des somatopic cartes sensorielles du cortex c&eacute;r&eacute;bral n&#39;ont pas &eacute;t&eacute; &eacute;tudi&eacute;s intensivement. En particulier, aucune &eacute;tudes n&#39;ont &eacute;tudi&eacute; le r&ocirc;le des m&eacute;canismes &eacute;pig&eacute;n&eacute;tiques dans le d&eacute;veloppement de cartes sensorielles. M&eacute;thylation de l&#39;ADN est l&#39;un principaux m&eacute;canismes &eacute;pig&eacute;n&eacute;tiques disponibles pour les cellules de mammif&egrave;res pour r&eacute;guler la transcription du g&egrave;ne. Comme la d&eacute;m&eacute;thylation aboutit &agrave; la d&eacute;termination de la l&eacute;talit&eacute; embryonnaire, il a &eacute;t&eacute; tr&egrave;s difficile d&#39;&eacute;tudier le r&ocirc;le de la m&eacute;thylation de l&#39;ADN dans le d&eacute;veloppement du cerveau. Nous avons utilis&eacute; la technologie de cre-lox pour g&eacute;n&eacute;rer la suppression du cerveau-sp&eacute;cifique d&#39;ADN m&eacute;thyltransf&eacute;rase 1 (Dnmt1), l&#39;enzyme n&eacute;cessaire pour le maintien de la m&eacute;thylation de l&#39;ADN. Nous trouvons que la d&eacute;m&eacute;thylation de neurones dans les r&eacute;sultats du cortex c&eacute;r&eacute;bral dans l&#39;&eacute;chec du d&eacute;veloppement du cortex somatosensoriel baril. Keine Studien haben insbesondere die Rolle der epigenetischen Mechanismen in der Entwicklung der sensorischen Karten angesprochen. DNA-Methylierung ist eine der wichtigsten epigenetischen Mechanismen zur S&auml;ugetier-Zellen, Gen-Transkription zu regulieren. Denn Demethylierung embryonalen T&ouml;dlichkeit f&uuml;hrt, war es sehr schwierig, die Rolle der DNA-Methylierung in der Entwicklung des Gehirns zu studieren. Wir haben Cre-Lox-Technologie verwendet, um Stirnhirn-spezifischen Streichung von DNA Methyltransferase 1 (Dnmt1), das Enzym ben&ouml;tigt, die f&uuml;r die Wartung der DNA-Methylierung zu generieren. Wir finden diese Demethylierung von Neuronen der Hirnrinde ergibt das Scheitern der Entwicklung der Fass somatosensorischen Kortex. Wir finden auch, dass trotz der funktionalen Thalamocortical Neurotransmission, Thalamocortical langfristige Erm&ouml;glichung in Scheiben von Dnmt1 bedingte Mutanten induziert werden kann nicht. I meccanismi trascrizionali che regolano lo sviluppo e la plasticit&agrave; delle mappe sensoriali somatopic nella corteccia cerebrale non sono state ampiamente studiati. In particolare, studi hanno affrontato il ruolo dei meccanismi epigenetici nello sviluppo di mappe sensoriali. La metilazione del DNA &egrave; uno dei meccanismi epigenetici principali disponibili alle cellule mammifere per regolare la trascrizione genica. Come demetilazione risultati nella letalit&agrave; embrionale, &egrave; stato molto difficile studiare il ruolo di metilazione del DNA nello sviluppo del cervello. Abbiamo utilizzato la tecnologia cre-lox per generare specifiche del proencefalo eliminazione delle DNA metiltransferasi 1 (Dnmt1), l&#39;enzima necessario per il mantenimento della metilazione del DNA. Troviamo che demetilazione di neuroni nei risultati della corteccia cerebrale nel fallimento dello sviluppo della corteccia somatosensoriale barile. Troviamo anche che nonostante la neurotrasmissione talamocorticale funzionale, potenziamento a lungo termine talamocorticale non pu&ograve; essere indotta in fette da Dnmt1 mutanti condizionali. Questi studi sottolineano l&#39;importanza della metilazione del DNA per lo sviluppo di mappe sensoriali e suggeriscono meccanismi epigenetici possono giocare un ruolo nello sviluppo della plasticit&agrave; sinaptica.

No se han estudiado ampliamente los mecanismos transcripcionales que regulan el desarrollo y la plasticidad de los mapas sensoriales de somatopic en la corteza cerebral. En particular, no hay estudios han abordado el papel de mecanismos epigen&eacute;ticos en el desarrollo de mapas sensoriales. Metilaci&oacute;n del ADN es uno de los principales mecanismos epigen&eacute;ticos disponibles a las c&eacute;lulas mam&iacute;feras para regular la transcripci&oacute;n gen&eacute;tica. Como desmetilaci&oacute;n da lugar a mortalidad embrionaria, ha sido muy dif&iacute;cil estudiar el papel de la metilaci&oacute;n del ADN en el desarrollo cerebral. Hemos usado tecnolog&iacute;a cre-lox para generar eliminaci&oacute;n prosenc&eacute;falo espec&iacute;ficas de DNA metiltransferasa 1 (Dnmt1), la enzima necesaria para el mantenimiento de la metilaci&oacute;n del ADN. ALS8 is caused by a dominant mutation in an evolutionarily conserved protein, VAPB (vesicle-associated membrane protein (VAMP)-associated membrane protein B)/ALS8). We have established a fly model of ALS8 using the corresponding mutation in Drosophila VAPB (dVAP33A) and examined the effects of this mutation on VAP function using genetic and morphological analyses. By simultaneously assessing the effects of VAP(wt) and VAP(P58S) on synaptic morphology and structure, we demonstrate that the phenotypes produced by neuronal expression of VAP(P58S) resemble VAP loss of function mutants and are opposite those of VAP overexpression, suggesting that VAP(P58S) may function as a dominant negative. This is brought about by aggregation of VAP(P58S) and recruitment of wild type VAP into these aggregates. Importantly, we also demonstrate that the ALS8 mutation in dVAP33A interferes with BMP signaling pathways at the neuromuscular junction, identifying a new mechanism underlying pathogenesis of ALS8. Furthermore, we show that mutant dVAP33A can serve as a powerful tool to identify genetic modifiers of VAPB. This new fly model of ALS, with its robust pathological phenotypes, should for the first time allow the power of unbiased screens in Drosophila to be applied to study of motor neuron diseases.

A Drosophila model of ALS: human ALS-associated mutation in VAP33A suggests a dominant negative mechanism.

ALS8 es causada por una mutaci&oacute;n dominante en una prote&iacute;na conservada evolutivamente, VAPB (prote&iacute;na de membrana asociada a ves&iacute;culas (VAMP)-asociados a la prote&iacute;na de membrana B) / ALS8). DNA methylation is a major epigenetic factor regulating genome reprogramming, cell differentiation and developmental gene expression. To understand the role of DNA methylation in central nervous system (CNS) neurons, we generated conditional Dnmt1 mutant mice that possess approximately 90% hypomethylated cortical and hippocampal cells in the dorsal forebrain from E13.5 on. The mutant mice were viable with a normal lifespan, but displayed severe neuronal cell death between E14.5 and three weeks postnatally. Accompanied with the striking cortical and hippocampal degeneration, adult mutant mice exhibited neurobehavioral defects in learning and memory in adulthood. Unexpectedly, a fraction of Dnmt1(-/-) cortical neurons survived throughout postnatal development, so that the residual cortex in mutant mice contained 20-30% of hypomethylated neurons across the lifespan. Hypomethylated excitatory neurons exhibited multiple defects in postnatal maturation including abnormal dendritic arborization and impaired neuronal excitability. The mutant phenotypes are coupled with deregulation of those genes involved in neuronal layer-specification, cell death and the function of ion channels. Our results suggest that DNA methylation, through its role in modulating neuronal gene expression, plays multiple roles in regulating cell survival and neuronal maturation in the CNS.

Metilaci&oacute;n del ADN es un importante factor epigen&eacute;tico regulan la reprogramaci&oacute;n del genoma, la diferenciaci&oacute;n celular y la expresi&oacute;n g&eacute;nica del desarrollo. Para entender el papel de la metilaci&oacute;n del ADN en las neuronas del sistema nervioso central (SNC), generamos condicionales ratones mutantes Dnmt1 que poseen aproximadamente el 90% hypomethylated corticales y hippocampal c&eacute;lulas en el cerebro dorsal anterior de E13.5 en. Los ratones mutantes eran viables con una vida normal, pero aparecen muerte neuronal severa de la c&eacute;lula entre E14.5 y tres semanas postnatal. Acompa&ntilde;ado con la degeneraci&oacute;n cortical y hippocampal llamativa, ratones mutantes adultos exhibieron neurobehavioral defectos en el aprendizaje y la memoria en la edad adulta. Inesperadamente, una fracci&oacute;n de las neuronas corticales Dnmt1(-/-) sobrevivi&oacute; a lo largo del desarrollo postnatal, por lo que la corteza residual en ratones mutantes contiene 20-30% de las neuronas hypomethylated en toda la vida. During neocortical development, neurons exhibit highly synchronized patterns of spontaneous activity, with correlated bursts of action potential firing dominating network activity. This early activity is eventually replaced by more sparse and decorrelated firing of cortical neurons, which modeling studies predict is a network state that is better suited for efficient neural coding. The precise time course and mechanisms of this crucial transition in cortical network activity have not been characterized in vivo. We used in vivo two-photon calcium imaging in combination with whole-cell recordings in both unanesthetized and anesthetized mice to monitor how spontaneous activity patterns in ensembles of layer 2/3 neurons of barrel cortex mature during postnatal development. We find that, as early as postnatal day 4, activity is highly synchronous within local clusters of neurons. At the end of the second postnatal week, neocortical networks undergo a transition to a much more desynchronized state that lacks a clear spatial structure. Strikingly, deprivation of sensory input from the periphery had no effect on the time course of this transition. Therefore, developmental desynchronization of spontaneous neuronal activity is a fundamental network transition in the neocortex that appears to be intrinsically generated.

Using spatiotemporal multiplexing we circumvented light-scattering ambiguity inherent to deep-tissue multifocal two-photon microscopy. We demonstrate calcium imaging at multiple axial planes in the intact mouse brain to monitor network activity of ensembles of cortical neurons in three spatial dimensions.

Although many genes predisposing to autism spectrum disorders (ASD) have been identified, the biological mechanism(s) remain unclear. Mouse models based on human disease-causing mutations provide the potential for understanding gene function and novel treatment development. Here, we characterize a mouse knockout of the Cntnap2 gene, which is strongly associated with ASD and allied neurodevelopmental disorders. Cntnap2(-/-) mice show deficits in the three core ASD behavioral domains, as well as hyperactivity and epileptic seizures, as have been reported in humans with CNTNAP2 mutations. Neuropathological and physiological analyses of these mice before the onset of seizures reveal neuronal migration abnormalities, reduced number of interneurons, and abnormal neuronal network activity. In addition, treatment with the FDA-approved drug risperidone ameliorates the targeted repetitive behaviors in the mutant mice. These data demonstrate a functional role for CNTNAP2 in brain development and provide a new tool for mechanistic and therapeutic research in ASD.

Absence of CNTNAP2 leads to epilepsy, neuronal migration abnormalities, and core autism-related deficits.
