The combination of sucrose and starch in the presence of surface-adsorbed salivary Î±-amylase and bacterial glucosyltransferases increase the formation of a structurally and metabolically distinctive biofilm by Streptococcus mutans. This host-pathogen-diet interaction may modulate the formation of pathogenic biofilms related to dental caries disease. We conducted a comprehensive study to further investigate the influence of the dietary carbohydrates on S. mutans-transcriptome at distinct stages of biofilm development using whole genomic profiling with a new computational tool (MDV) for data mining. S. mutans UA159 biofilms were formed on amylase-active saliva coated hydroxyapatite discs in the presence of various concentrations of sucrose alone (ranging from 0.25 to 5% w/v) or in combination with starch (0.5 to 1% w/v). Overall, the presence of sucrose and starch (suc+st) influenced the dynamics of S. mutans transcriptome (vs. sucrose alone), which may be associated with gradual digestion of starch by surface-adsorbed amylase. At 21 h of biofilm formation, most of the differentially expressed genes were related to sugar metabolism, such as upregulation of genes involved in maltose/maltotriose uptake and glycogen synthesis. In addition, the groEL/groES chaperones were induced in the suc+st-biofilm, indicating that presence of starch hydrolysates may cause environmental stress. In contrast, at 30 h of biofilm development, multiple genes associated with sugar uptake/transport (e.g. maltose), two-component systems, fermentation/glycolysis and iron transport were differentially expressed in suc+st-biofilms (vs. sucrose-biofilms). Interestingly, lytT (bacteria autolysis) was upregulated, which was correlated with presence of extracellular DNA in the matrix of suc+st-biofilms. Specific genes related to carbohydrate uptake and glycogen metabolism were detected in suc+st-biofilms in more than one time point, indicating an association between presence of starch hydrolysates and intracellular polysaccharide storage. Our data show complex remodeling of S. mutans-transcriptome in response to changing environmental conditions in situ, which could modulate the dynamics of biofilm development and pathogenicity.

A 21 h la formation de biofilms, la plupart des g&#xE8;nes diff&#xE9;rentiellement exprim&#xE9;s ont &#xE9;t&#xE9; li&#xE9;es au m&#xE9;tabolisme du sucre, tels que la r&#xE9;gulation positive des g&#xE8;nes impliqu&#xE9;s dans le maltose / maltotriose absorption et la synth&#xE8;se de glycog&#xE8;ne. En outre, les chaperons groEL / groES ont &#xE9;t&#xE9; induites dans le succ&#xE8;s + st-biofilm, ce qui indique que la pr&#xE9;sence d&#x26;#39;hydrolysats d&#x26;#39;amidon peuvent causer du stress de l&#x26;#39;environnement. Diese Host-Pathogen-Di&auml;t-Interaktion kann die Bildung von pathogenen Biofilme mit Bezug zu Zahnkaries Erkrankung modulieren. Wir f&uuml;hrten eine umfassende Studie um weiter zu untersuchen, den Einfluss der Nahrungskohlenhydraten auf S. Mutans-Transkriptom in unterschiedlichen Stadien der Biofilm Entwicklung mit ganzen genomische profiling mit einem neuen computational Tool (MDV) f&uuml;r Datamining. S. Mutans UA159 Biofilme Amylase-aktive Speichel gebildet wurden beschichtet Hydroxylapatit Scheiben in verschiedenen Konzentrationen von Saccharose allein (zwischen 0,25 bis 5 % w/V) oder in Kombination mit St&auml;rke (0,5 bis 1 % w/V). Insgesamt beeinflusst das Vorhandensein von Saccharose und St&auml;rke (Suc + St) die Dynamik von S. Mutans Transkriptom (im Vergleich zu Saccharose allein), die schrittweise Verdauung von St&auml;rke durch Amylase Oberfl&auml;che adsorbierten zugeordnet werden kann. Um 21 h Biofilm Formation betrafen die meisten der differentiell exprimierten Genen Zucker-Stoffwechsel, z. B. Hochregulation von Genen, die in Maltose/Maltotriose-Aufnahme und Glykogen-Synthese. Dar&uuml;ber hinaus wurden die GroEL/GroES Chaperone induziert im Suc + St-Biofilm, darauf hinweist, dass St&auml;rke Hydrolysat Umweltbelastung verursachen kann. Dagegen bei 30 h der Biofilm-Entwicklung, mehrere Gene zugeordnet Zucker Aufnahme/Transport (z.B. Maltose), zwei-Komponenten-Systeme, G&auml;rung/Glykolyse und Eisen-Transport wurden differentiell Suc + St-Biofilmen (im Vergleich zu Saccharose-Biofilme) ausgedr&uuml;ckt. Interessanterweise war LytT (Bakterien Autolyse) herraufreguliert, die mit dem Vorhandensein von extrazellul&auml;rer DNA in der Matrix von Suc + St-Biofilmen korrelierte. Spezifischer Gene mit Bezug zu Kohlenhydrat-Aufnahme und Glykogen-Stoffwechsel gefunden in Suc + St-Biofilme in mehr als ein Zeitpunkt, Angabe einer Assoziation zwischen der Anwesenheit von St&auml;rke-Hydrolysate und intrazellul&auml;re Polysaccharid-Speicher. Unsere Daten zeigen komplexe Umgestaltung von S. Mutans-Transkriptom in Reaktion auf wechselnde Umweltbedingungen vor Ort, die die Dynamik der Biofilm-Entwicklung und Pathogenit&auml;t modulieren kann.

La combinazione di saccarosio e amido in presenza di superficie adsorbita &alpha;-amilasi salivare e batterica glucosyltransferases aumentare la formazione di un biofilm strutturalmente e metabolicamente distintivo di Streptococcus mutans. Questa interazione ospite-patogeno-dieta pu&ograve; modulare la formazione di biofilm patogeni correlati alla malattia di carie dentale. Abbiamo condotto uno studio completo per indagare ulteriormente l&#39;influenza dei carboidrati alimentari su S. mutans-transcriptome a distinte fasi di sviluppo di biofilm con tutta genomic profilatura con un nuovo strumento computazionale (MDV) per il data mining. S. mutans UA159 biofilm sono stati formati su saliva amilasi-attivo rivestito dischi di idrossiapatite in presenza di diverse concentrazioni di saccarosio da solo (che vanno da 0,25 a 5% p/v) o in combinazione con amido (0,5-1% w/v). Nel complesso, la presenza di saccarosio e amido (suc + st) influenzato le dinamiche di S. mutans transcriptome (vs saccarosio da solo), che possono essere associate con graduale digestione dell&#39;amido dalle amilasi superficie adsorbita. 21 H di formazione di biofilm, la maggior parte dei geni differenzialmente espressi erano legata al metabolismo, quali sovraregolazione di geni coinvolti nella sintesi di assorbimento e di glicogeno maltosio/maltotrioso di zucchero. Inoltre, i groEL/groES accompagnatori sono stati indotti nella suc + st-biofilm, che indica che la presenza di idrolizzati di amido pu&ograve; causare stress ambientali. Trans-trans farnesol (tt-farnesol) is a bioactive sesquiterpene alcohol commonly found in propolis (a beehive product) and citrus fruits, which disrupts the ability of Streptococcus mutans (S. mutans) to form virulent biofilms. In this study, we investigated whether tt-farnesol affects cell-membrane function, acid production and/or acid tolerance by planktonic cells and biofilms of S. mutans UA159. Furthermore, the influence of the agent on S. mutans gene expression and ability to form biofilms in the presence of other oral bacteria (Streptococcus oralis (S. oralis) 35037 and Actinomyces naeslundii (A. naeslundii) 12104) was also examined. In general, tt-farnesol (1 mmol x L(-1)) significantly increased the membrane proton permeability and reduced glycolytic activity of S. mutans in the planktonic state and in biofilms (P < 0.05). Moreover, topical applications of 1 mmol x L(-1) tt-farnesol twice daily (1 min exposure/treatment) reduced biomass accumulation and prevented ecological shifts towards S. mutans dominance within mixed-species biofilms after introduction of 1% sucrose. S. oralis (a non-cariogenic organism) became the major species after treatments with tt-farnesol, whereas vehicle-treated biofilms contained mostly S. mutans (>90% of total bacterial population). However, the agent did not affect significantly the expression of S. mutans genes involved in acidogenicity, acid tolerance or polysaccharide synthesis in the treated biofilms. Our data indicate that tt-farnesol may affect the competitiveness of S. mutans in a mixed-species environment by primarily disrupting the membrane function and physiology of this bacterium. This naturally occurring terpenoid could be a potentially useful adjunctive agent to the current anti-biofilm/anti-caries chemotherapeutic strategies.

Mutans), virulenten Biofilme bilden. In dieser Studie untersuchten wir, ob Tt-Farnesol Zellmembran Funktion, S&auml;ureproduktion und/oder saure Toleranz von Plankton Zellen und Biofilmen von S. Trans-trans-farnesol (TT-farnesol) es un alcohol sesquiterpeno bioactivo se encuentra com&#xFA;nmente en el prop&#xF3;leo (un producto de la colmena) y los c&#xED;tricos, que altera la capacidad de Streptococcus mutans (S. mutans) para formar biopel&#xED;culas virulentas. En este estudio, se investig&#xF3; si tt-farnesol afecta a la funci&#xF3;n de la membrana celular, la producci&#xF3;n de &#xE1;cido y / o tolerancia al &#xE1;cido por las c&#xE9;lulas planct&#xF3;nicas y biofilms de S. mutans UA159. Adem&#xE1;s, la influencia del agente sobre la expresi&#xF3;n de S. mutans gen y la capacidad para formar biopel&#xED;culas en la presencia de otras bacterias orales (Streptococcus oralis (S. oralis) 35037 y Actinomyces naeslundii (A. naeslundii) 12104) tambi&#xE9;n fue examinado. En general, TT-farnesol (1 mmol L x (-1)) aument&#xF3; significativamente la permeabilidad de protones de la membrana y reducci&#xF3;n de la actividad glucol&#xED;tica de S. mutans en el estado planct&#xF3;nico y en biopel&#xED;culas (P &#x26;lt;0,05). Por otra parte, la aplicaci&#xF3;n t&#xF3;pica de 1 mmol L x (-1) tt-farnesol. Dos veces al d&#xED;a (1 min de la exposici&#xF3;n / tratamiento) la acumulaci&#xF3;n de biomasa reducida y evitar cambios ecol&#xF3;gicos hacia el S. mutans en la dominaci&#xF3;n de especies mixtas biopel&#xED;culas despu&#xE9;s de la introducci&#xF3;n de 1% de sacarosa S. oralis (un organismo no cariog&#xE9;nico) se convirtieron en las principales especies despu&#xE9;s de los tratamientos con tt-farnesol, mientras que los tratados con el veh&#xED;culo que figura en su mayor&#xED;a las biopel&#xED;culas de S. mutans (&#x26;gt; 90% de la poblaci&#xF3;n total de bacterias). Sin embargo, el agente no afect&#xF3; significativamente la expresi&#xF3;n de S. mutans genes implicados en la tolerancia al &#xE1;cido acidogenicidad, o la s&#xED;ntesis de polisac&#xE1;ridos en las biopel&#xED;culas tratados. Nuestros datos indican que tt-farnesol puede afectar la competitividad de S. mutans en un entorno mixto de especies principalmente por alterar la funci&#xF3;n de la membrana y la fisiolog&#xED;a de esta bacteria. Este terpenoide natural podr&#xED;a ser un agente adyuvante potencialmente &#xFA;til para las estrategias anti-biofilm/anti-caries quimioterap&#xE9;uticos actuales.
