Sobres vitelino (VES) de Bufo arenarum fueron aislados con el fin de estudiar su composici&#xF3;n y su papel en la fecundaci&#xF3;n. VEs se componen de cuatro glicoprote&#xED;nas, con masas moleculares de 120, 75, 41 y 38 kDa. Para la caracterizaci&#xF3;n de sus propiedades biol&#xF3;gicas, que determina cuantitativamente el esperma-VE vinculante y la inducci&#xF3;n de la reacci&#xF3;n del acrosoma. Heter&#xF3;logo uni&#xF3;n de B. arenarum esperma a los componentes de Xenopus laevis VE se observ&#xF3; con aproximadamente un tercio de la eficiencia de la uni&#xF3;n hom&#xF3;loga. Vinculante Equivalente de X. laevis espermatozoides al arenarum B. VE se observ&#xF3;. Cuando B. esperma arenarum fueron incubadas con fluoresce&#xED;na marcado con isotiocianato de VE, las glicoprote&#xED;nas marcada unida al extremo anterior de la cabeza de los espermatozoides, que muestra una distribuci&#xF3;n lateral. La inducci&#xF3;n de la reacci&#xF3;n acrosomal se evalu&#xF3; mediante la incubaci&#xF3;n de los espermatozoides en medios salinos hipot&#xF3;nicos con glicoprote&#xED;nas de VE. VEs indujo la reacci&#xF3;n del acrosoma en un tiempo y de manera dependiente de la concentraci&#xF3;n. La reacci&#xF3;n de acrosoma fue m&#xE1;xima a los 10 min. El efecto semi-m&#xE1;xima se obtuvo a una concentraci&#xF3;n glicoprote&#xED;na de 1 mcg / ml. La especificidad se determin&#xF3; a trav&#xE9;s de las glicoprote&#xED;nas de fertilizaci&#xF3;n sobre, que no han podido inducir a la reacci&#xF3;n del acrosoma. El VE B. arenarum es bioqu&#xED;micamente similar a otros sobres de huevo. Tambi&#xE9;n parece que sus propiedades biol&#xF3;gicas son similares a otras especies en lo que se refiere a la uni&#xF3;n de espermatozoides y la inducci&#xF3;n de la reacci&#xF3;n del acrosoma. Buste vitellina (VEs) di Bufo arenarum sono stati isolati per poter studiare la loro composizione e il loro ruolo nella fecondazione. VEs sono composte da quattro delle glicoproteine, con masse molecolari di 120, 75, 41 e 38 kDa. Per caratterizzare le sue propriet&agrave; biologiche, abbiamo determinato quantitativamente associazione sperma-VE e l&#39;induzione della reazione acrosomiale. Associazione eterologo di sperma arenarum b. ai componenti di Xenopus laevis VE &egrave; stata osservata con circa un terzo l&#39;efficienza dell&#39;associazione omologa. Associazione equivalente di X. laevis sperma per il b. arenarum VE &egrave; stata osservata. Quando b. arenarum sperma sono stato incubato con fluoresceina isotiocianato-etichetta VE, le glicoproteine etichette legate alla parte anteriore della testa degli spermatozoi, che mostrano una distribuzione laterale. Induzione della reazione acrosomiale &egrave; stata valutata incubando sperma in media salina ipotonica con glicoproteine VE. VEs indotto la reazione acrosomiale in maniera dipendente dal tempo e concentrazione. La reazione acrosomiale era massima dopo 10 min. L&#39;effetto di met&agrave;-massimo &egrave; stato ottenuto a una concentrazione di glicoproteina di 1 microg / ml. specificit&agrave; &egrave; stata determinata utilizzando delle glicoproteine busta di fertilizzazione, che non &egrave; riuscito a indurre la reazione acrosomiale. Il b. arenarum VE &egrave; biochimicamente simili ad altre buste di uovo. Sembra inoltre che le propriet&agrave; biologiche sono simili ad altre specie per quanto riguarda l&#39;associazione di sperma e induzione della reazione acrosomiale. Tuttavia, per quanto siamo consapevoli, questa &egrave; la prima osservazione di VE che inducono la reazione acrosomiale sperma negli anfibi. Le relativamente piccole differenze osservate in associazione sperma eterologo-VE in arenarum X. laevis e B. sono incompatibili con l&#39;attuale paradigma che specificit&agrave; specie nella fecondazione &egrave; regolata al passo associazione sperma-VE.

Vitelline envelopes (VEs) of Bufo arenarum were isolated in order to study their composition and their role in fertilization. VEs are composed of four glycoproteins, with molecular masses of 120, 75, 41, and 38 kDa. To characterize its biological properties, we quantitatively determined sperm-VE binding and the induction of the acrosome reaction. Heterologous binding of B. arenarum sperm to Xenopus laevis VE components was observed with about one-third the efficiency of homologous binding. Equivalent binding of X. laevis sperm to the B. arenarum VE was observed. When B. arenarum sperm were incubated with fluorescein isothiocyanate-labeled VE, the labeled glycoproteins bound to the anterior end of the sperm head, showing a lateral distribution. Induction of the acrosome reaction was evaluated by incubating sperm in hypotonic saline media with VE glycoproteins. VEs induced the acrosome reaction in a time- and concentration-dependent manner. The acrosome reaction was maximal after 10 min. The half-maximal effect was obtained at a glycoprotein concentration of 1 microg/ml. Specificity was determined using fertilization envelope glycoproteins, which failed to induce the acrosome reaction. The B. arenarum VE is biochemically similar to other egg envelopes. It also seems that its biological properties are similar to other species in regard to sperm binding and induction of the acrosome reaction. However, as far as we are aware, this is the first observation of the VE inducing the sperm acrosome reaction in amphibians. The relatively small differences observed in heterologous sperm-VE binding in X. laevis and B. arenarum are inconsistent with the current paradigm that species specificity in fertilization is regulated at the sperm-VE binding step.

A characterization of the Amphibian Bufo arenarum oocyte envelope is presented. It was made in different functional conditions of the oocyte: 1) when it has been released into the coelomic cavity during ovulation (surrounded by the coelomic envelope, (CE), 2) after it has passed through the oviduct and is deposed (surrounded by the viteline envelope, (VE), and 3) after oocyte activation (surrounded by the fertilization envelope, (FE). The characterization was made by SDS-PAGE followed by staining for protein and glycoproteins. Labeled lectins were used to identify glycosidic residues both in separated components on nitrocellulose membranes or in intact oocytes and embryos. Proteolytic properties of the content of the cortical granules were also analyzed. After SDS-PAGE of CE and VE, a different protein pattern was observed. This is probably due to the activity of a protease present in the pars recta of the oviduct. Comparison of the SDS-PAGE pattern of VE and FE showed a different mobility for one of the glycoproteins, gp75. VE and FE proved to have different sugar residues in their oligosaccharide chains. Mannose residues are only present in gp120 of the three envelopes. N-acetyl-galactosamine residues are present in all of the components, except for gp69 in the FE. Galactose residues are present mainly in gp120 of FE. Lectin-binding assays indicate the presence of glucosamine, galactose and N-acetyl galactosamine residues and the absence (or non-availability) of N-acetyl-glucosamine or fucose residues on the envelopes surface. The cortical granule product (CGP) shows proteolytic activity on gp75 of the VE.

Una caratterizzazione della busta anfibi Bufo arenarum ovocita &egrave; presentata. &Egrave; stato fatto in diverse condizioni funzionali dell&#39;ovocita: 1) quando &egrave; stato rilasciato nella cavit&agrave; celomatiche durante l&#39;ovulazione (circondato da busta celomatiche, (CE), 2) dopo che &egrave; passato attraverso l&#39;ovidotto e viene deposto (circondato dalla busta di viteline, (VE) e 3) dopo l&#39;attivazione degli ovociti (circondato dalla busta di fertilizzazione, (FE). La caratterizzazione &egrave; stata fatta da SDS-PAGE seguita da colorazione per proteine e glicoproteine. Etichettati lectine sono state utilizzate per identificare residui glicosidici sia in componenti separati su membrane di nitrocellulosa o di embrioni e ovociti intatti. Propriet&agrave; proteolitica del contenuto dei granuli corticali sono stati anche analizzati. Dopo SDS-PAGE di CE e VE, &egrave; stato osservato un pattern di proteine diverse. Ci&ograve; &egrave; probabilmente dovuto all&#39;attivit&agrave; di una proteasi presente nella recta pars dell&#39;ovidotto. Confronto tra il pattern di SDS-PAGE di VE e FE ha mostrato una diversa mobilit&agrave; per una delle glicoproteine, gp75. VE e FE dimostr&ograve; di avere residui di zucchero differenti nelle loro catene di oligosaccaridi. Sono presenti in gp120 delle tre buste solo residui di mannosio. N-acetil-galattosamina residui sono presenti in tutti i componenti, ad eccezione di gp69 alla FE. Residui di galattosio sono presenti principalmente in gp120 di FE. Lectina-associazione dosaggi indicano la presenza di glucosamina, galattosio e residui di N-acetil-galattosamina e l&#39;assenza (o non disponibilit&agrave;) dei residui di N-acetil-glucosamina o fucosio sulla superficie buste. Il prodotto di granulo corticale (CGP) Mostra attivit&agrave; proteolitica gp75 della VE.

The vitelline envelope (VE) participates in sperm-egg interactions during the first steps of fertilization. In Bufo arenarum, this envelope is composed of at least four glycoproteins, with molecular masses of 120, 75, 41, and 38 kDa and molar ratio of 1:1.3:7.4:4.8, respectively. These components were isolated and covalently coupled to silanized glass slides in order to study their sperm-binding capacity. When considering the molar ratio of the glycoproteins in the egg-envelope and assuming that each protein is monovalent for sperm, the assay showed that gp41 and gp38 possess 55 and 25% of total sperm-binding activity. We obtained a full-length cDNA of gp41 (ZPC), comprising a sequence for 486 amino acids, with 43.3% homology with Xenopus laevis ZPC. As in the case of mammalian ZP3 and Xenopus ZPC, Bufo ZPC presented a furin-like (convertase) and a C-terminal transmembrane domain (TMD) reflecting common biosynthetic and secretory pathways. As it was reported for some fishes, we obtained evidence that suggests the presence of more than one zpc gene in Bufo genome, based on different partial cDNA sequences of zpc, Southern blots and two-dimensional SDS-PAGE of deglycosylated egg-envelope components. As far as we are aware, this is the first observation of the presence of different zpc genes in an Amphibian species.

L&#x26;#39;enveloppe vitelline (VE) participe dans le sperme d&#x26;#39;&#x153;ufs interactions pendant les premi&#xE8;res &#xE9;tapes de la f&#xE9;condation. Dans Bufo arenarum, cette enveloppe est compos&#xE9;e d&#x26;#39;au moins quatre glycoprot&#xE9;ines, ayant des masses mol&#xE9;culaires de 120, 75, 41, et 38 kDa et le ratio molaire de 1:1.3:7.4:4.8, respectivement. Ces composants ont &#xE9;t&#xE9; isol&#xE9;s et coupl&#xE9; de fa&#xE7;on covalente &#xE0; des lames de verre silanis&#xE9;es afin d&#x26;#39;&#xE9;tudier leur sperme capacit&#xE9; de liaison. Lorsque l&#x26;#39;on consid&#xE8;re le rapport molaire des glycoprot&#xE9;ines dans l&#x26;#39;oeuf-enveloppe et en supposant que chaque prot&#xE9;ine est monovalent pour le sperme, le dosage a montr&#xE9; que la gp41 et GP38 poss&#xE8;dent de 55 &#xE0; 25% du total des spermatozo&#xEF;des activit&#xE9; de liaison. Nous avons obtenu un ADNc pleine longueur de la gp41 (ZPC), comprenant une s&#xE9;quence de 486 acides amin&#xE9;s, avec une homologie de 43,3% avec Xenopus laevis ZPC. Comme il a &#xE9;t&#xE9; signal&#xE9; pour certains poissons, nous avons obtenu des preuves qui sugg&#xE8;rent la pr&#xE9;sence de plus d&#x26;#39;un g&#xE8;ne dans zpc Bufo g&#xE9;nome, fond&#xE9;e sur diff&#xE9;rentes s&#xE9;quences partielles d&#x26;#39;ADNc de zpc, les transferts de Southern et &#xE0; deux dimensions de SDS-PAGE de d&#xE9;glycosyl&#xE9;es &#x153;uf enveloppe composants. Pour autant que nous sommes conscients, c&#x26;#39;est la premi&#xE8;re observation de la pr&#xE9;sence de diff&#xE9;rents g&#xE8;nes ZPC une esp&#xE8;ce d&#x26;#39;amphibiens.

In Bufo arenarum, questa busta &egrave; composto di almeno quattro delle glicoproteine, con masse molecolari di 120, 75, 41 e 38 kDa e rapporto molare di 1:1.3:7.4:4.8, rispettivamente. Questi componenti sono stati isolati e covalenza accoppiati alle diapositive di vetro silanizzata al fine di studiare la loro capacit&agrave; di sperma-associazione. Quando si considera il rapporto molare di glicoproteine nella busta di uovo e supponendo che ogni proteina &egrave; monovalente per lo sperma, il saggio ha mostrato che gp41 e gp38 possiedono 55 e il 25% del totale degli spermatozoi-associazione attivit&agrave;. Come &egrave; stato segnalato per alcuni pesci, abbiamo ottenuto la prova che suggerisce la presenza di pi&ugrave; di un gene zpc nel genoma di Bufo, basato su sequenze diverse parziali del cDNA di zpc, Southern Blot e bidimensionale SDS-PAGE delle componenti di uovo-busta deglycosylated. Per quanto siamo consapevoli, questa &egrave; la prima osservazione della presenza di geni differenti zpc in una specie di anfibi.

La envoltura vitelina (VE) participa en las interacciones de huevo de esperma en los primeros pasos de la fecundaci&#xF3;n. En Bufo arenarum, esta envoltura se compone de al menos cuatro glicoprote&#xED;nas, con masas moleculares de 120, 75, 41 y 38 kDa y la relaci&#xF3;n molar de 1:1.3:7.4:4.8, respectivamente. Estos componentes se aislaron y se acopla covalentemente a portaobjetos de vidrio silanizados con el fin de estudiar su capacidad de uni&#xF3;n de esperma. Al considerar la relaci&#xF3;n molar de las glicoprote&#xED;nas de la envoltura de huevo y suponiendo que cada prote&#xED;na es monovalente de los espermatozoides, el ensayo mostr&#xF3; que la gp41 y GP38 poseen 55 y 25% del total de esperma actividad de uni&#xF3;n. Se obtuvo un ADNc de longitud completa de la gp41 (PCC), que comprende una secuencia de 486 amino&#xE1;cidos, con 43,3% de homolog&#xED;a con Xenopus laevis PCC. Como en el caso de los mam&#xED;feros y ZP3 ZPC Xenopus, Bufo ZPC presenta un dominio tipo furina (convertasa) y un C-terminal de transmembrana (TMD) que refleja biosint&#xE9;tico com&#xFA;n y v&#xED;as de secreci&#xF3;n. Como se inform&#xF3; por algunos peces, se obtuvo evidencia que sugiere la presencia de m&#xE1;s de un gen ZPC en Bufo genoma, basado en diferentes secuencias parciales de ADNc de ZPC, transferencias Southern y de dos dimensiones de SDS-PAGE de desglicosiladas envoltura de huevo componentes. Nous avons r&#xE9;cemment identifi&#xE9; un nouveau r&#xF4;le pour l&#x26;#39;MAD3 membre de la famille Mad dans le PNB prolif&#xE9;ration et l&#x26;#39;expression NMYC. Fait int&#xE9;ressant, l&#x26;#39;expression est r&#xE9;gul&#xE9;e positivement MAD3 dans des mod&#xE8;les murins de m&#xE9;dulloblastome, la tumeur du cerveau la plus fr&#xE9;quente chez les enfants. During development, Sonic hedgehog (Shh) regulates the proliferation of cerebellar granule neuron precursors (GNPs) in part via expression of Nmyc. Mutations in the Shh signaling pathway lead to brain tumors in mice and humans. We have recently identified a novel role for the Mad family member Mad3 in GNP proliferation and Nmyc expression. Interestingly, Mad3 expression is upregulated in mouse models of medulloblastoma, the most common brain tumor in children. These results are surprising because current models suggest that Mad proteins should antagonize Myc proteins by competition for direct DNA binding via Max heterodimerization to inhibit cellular proliferation and potentially tumor progression. Here, we discuss our recent work in the context of candidate Mad3-interacting proteins and Mad3 expression in human brain tumors that together suggest interesting insights into the role of Mad3 in cellular proliferation and tumorigenesis.

During development of the central nervous system, precise synaptic connections between presynaptic and postsynaptic neurons are formed. While significant progress has been made in our understanding of AMPA receptor trafficking during synaptic plasticity, less is known about the molecules that recruit AMPA receptors to nascent synapses during synaptogenesis. Here we identify a type II transmembrane protein (SynDIG1) that regulates AMPA receptor content at developing synapses in dissociated rat hippocampal neurons. SynDIG1 colocalizes with AMPA receptors at synapses and at extrasynaptic sites and associates with AMPA receptors in heterologous cells and brain. Altered levels of SynDIG1 in cultured neurons result in striking changes in excitatory synapse number and function. SynDIG1-mediated synapse development is dependent on association with AMPA receptors via its extracellular C terminus. Intriguingly, SynDIG1 content in dendritic spines is regulated by neuronal activity. Altogether, we define SynDIG1 as an activity-regulated transmembrane protein that regulates excitatory synapse development.

Durante el desarrollo del sistema nervioso central, precisas conexiones sin&#xE1;pticas entre las neuronas presin&#xE1;pticos y postsin&#xE1;pticos se forman. Si bien los progresos se han realizado importantes en nuestra comprensi&#xF3;n de la trata de receptores AMPA en la plasticidad sin&#xE1;ptica, se sabe menos sobre las mol&#xE9;culas que reclutan a los receptores AMPA en las sinapsis incipientes durante la sinaptog&#xE9;nesis. Aqu&#xED; identificamos una prote&#xED;na transmembrana de tipo II (SynDIG1) que regula el contenido de los receptores AMPA en las sinapsis de las neuronas en desarrollo de hipocampo de rata disociados. SynDIG1 colocalizes con los receptores AMPA en las sinapsis y en los sitios extrasin&#xE1;pticos y asociados con los receptores AMPA en c&#xE9;lulas heter&#xF3;logas y el cerebro. Los niveles alterados de SynDIG1 resultado en las neuronas cultivadas en cambios notables en el n&#xFA;mero de sinapsis excitadoras y otras funciones. Bien que des progr&#xE8;s significatifs ont &#xE9;t&#xE9; r&#xE9;alis&#xE9;s dans notre compr&#xE9;hension de la traite des r&#xE9;cepteurs AMPA au cours plasticit&#xE9; synaptique, on en sait moins sur les mol&#xE9;cules qui recrutent r&#xE9;cepteurs AMPA aux synapses naissantes au cours de la synaptogen&#xE8;se. A subset of medulloblastomas, the most common brain tumor in children, is hypothesized to originate from granule neuron precursors (GNPs) in which the sonic hedgehog (SHH) pathway is over-activated. MXD3, a basic helix-look-helix zipper transcription factor of the MAD family, has been reported to be upregulated during postnatal cerebellar development and to promote GNP proliferation and MYCN expression. Mxd3 is upregulated in mouse models of medulloblastoma as well as in human medulloblastomas. Therefore, we hypothesize that MXD3 plays a role in the cellular events that lead to medulloblastoma biogenesis. In agreement with its proliferative role in GNPs, MXD3 knock-down in DAOY cells resulted in decreased proliferation. Sustained overexpression of MXD3 resulted in decreased cell numbers due to increased apoptosis and cell cycle arrest. Structure-function analysis revealed that the Sin3 interacting domain, the basic domain, and binding to E-boxes are essential for this activity. Microarray-based expression analysis indicated up-regulation of 84 genes and down-regulation of 47 genes. Potential direct MXD3 target genes were identified by ChIP-chip. Our results suggest that MXD3 is necessary for DAOY medulloblastoma cell proliferation. However, increased level and/or duration of MXD3 expression ultimately reduces cell numbers via increased cell death and cell cycle arrest.

Role of MXD3 in proliferation of DAOY human medulloblastoma cells.

Hyperamylinemia, a common pancreatic disorder in obese and insulin-resistant patients, is known to cause amylin oligomerization and cytotoxicity in pancreatic islets, leading to β-cell mass depletion and development of type 2 diabetes. Recent data has revealed that hyperamylinemia also affects the vascular system, heart, and kidneys. We therefore hypothesized that oligomerized amylin might accumulate in the cerebrovascular system and brain parenchyma of diabetic patients.

Amylin deposition in the brain: A second amyloid in Alzheimer disease?

MXD3 is a basic-helix-loop-helix-leucine-zipper transcription factor involved in cellular proliferation. In previous studies we demonstrated that knock-down of MXD3 in the human medulloblastoma cell line DAOY resulted in decreased proliferation. Surprisingly, overexpression of MXD3 in DAOY cells also decreased proliferation and increased cell death, suggesting that persistent expression of MXD3 triggers an apoptotic response, perhaps as a fail-safe mechanism. To investigate this apparent paradox in detail we developed a tamoxifen inducible system to analyze the temporal effects of MXD3 in the proliferation and transcriptional response of DAOY cells upon acute induction compared with long-term expression of MXD3.

MXD3 regulation of DAOY cell proliferation dictated by time course of activation.

Conventional chemotherapy for precursor B-cell (preB) acute lymphoblastic leukaemia (ALL) has limitations that could be overcome by targeted therapy. Previously, we discovered a potential therapeutic molecular target, MDX3 (MAX dimerization protein 3), in preB ALL. In this study, we hypothesize that an effective siRNA therapy for preB ALL can be developed using antiCD22 antibody (αCD22 Ab) and nanoparticles. We composed nanocomplexes with super paramagnetic iron oxide nanoparticles (SPIO NPs), αCD22 Abs and MXD3 siRNA molecules based on physical interactions between the molecules. We demonstrated that the MXD3 siRNA-αCD22 Ab-SPIO NP complexes entered leukaemia cells and knocked down MXD3, leading the cells to undergo apoptosis and resulting in decreased live cell counts in the cell line Reh and in primary preB ALL samples in vitro. Furthermore, the cytotoxic effects of the MXD3 siRNA-αCD22 Ab-SPIO NP complexes were significantly enhanced by addition of the chemotherapy drugs vincristine or doxorubicin. We also ruled out potential cytotoxic effects of the MXD3 siRNA-αCD22 Ab-SPIO NP complexes on normal primary haematopoietic cells. Normal B cells were affected while CD34-positive haematopoietic stem cells and non-B cells were not. These data suggest that MXD3 siRNA-αCD22 Ab-SPIO NP complexes have the potential to be a new targeted therapy for preB ALL.

Targeted therapy with MXD3 siRNA, anti-CD22 antibody and nanoparticles for precursor B-cell acute lymphoblastic leukaemia.

HB22.7, an anti-CD22 monoclonal antibody has shown consistent preclinical activity against non-Hodgkin lymphoma (NHL). Histone deacetylase inhibitors (HDACi) have demonstrated efficacy in lymphoma and can modulate cell surface receptor expression. To augment the lymphomacidal activity of HB22.7 we examined the combination of AR42 (an HDACi) and HB22.7 in vitro and in vivo. The combination resulted in 10-fold increased potency in 6 NHL cell lines when compared to either drug alone. Both drugs reduced tumor progression in xenografts, but the combination was significantly more efficacious and resulted in regression of established tumors, without toxicity. AR42 inhibited HB22.7-mediated CD22 internalization, suggesting that increased efficacy could be due to higher availability of CD22. Overall, the synergistic effects of HB22.7 and AR42 on in vitro cytotoxicity and in vivo anti-tumor activity make this combination an attractive option for further pre-clinical and clinical evaluation.
