V (D) J recombinase m&#xE9;die r&#xE9;arrangements des loci immunitaires et cryptiques s&#xE9;quences signal de recombinaison (SIR), r&#xE9;sultant dans une vari&#xE9;t&#xE9; de r&#xE9;arrangements g&#xE9;nomiques dans les lymphocytes normaux et les cellules leuc&#xE9;miques des enfants et des adultes. La fr&#xE9;quence &#xE0; laquelle ces r&#xE9;arrangements se produisent et de leurs &#xE9;ventuelles cons&#xE9;quences pathologiques sont d&#xE9;veloppemental d&#xE9;pendante. Pour avoir un aper&#xE7;u V (D) J recombinase m&#xE9;diation des &#xE9;v&#xE9;nements au cours du d&#xE9;veloppement humain, nous avons &#xE9;tudi&#xE9; 265 jonctions de codage associ&#xE9;es aux sites SCT au hypoxanthine-guanine phosphoribosyltransf&#xE9;rase (HPRT) locus dans des cellules T p&#xE9;riph&#xE9;riques de 111 enfants au cours des derniers stades du d&#xE9;veloppement foetal &#xE0; l&#x26;#39;adolescence au d&#xE9;but. Nous avons observ&#xE9; un certain nombre de caract&#xE9;ristiques V (D) J recombinase de traitement qui &#xE9;taient &#xE0; la fois l&#x26;#39;&#xE2;ge et le sexe d&#xE9;pend. V (D) J Recombinase vermittelt die Umlagerungen immun Loci und kryptische Rekombination Signal Sequenzen (CRS), was zu einer Vielzahl von genomic Rearrangements im normalen Lymphozyten und leuk&auml;mischen Zellen von Kindern und Erwachsenen. Die H&auml;ufigkeit, mit der diese Umlagerungen auftreten und ihre m&ouml;glichen pathologischen folgen entwicklungspolitisch angewiesen sind. Um Einblick in V (D) J Recombinase-vermittelten Veranstaltungen w&auml;hrend der menschlichen Entwicklung zu gewinnen, untersuchten wir 265 Codierung Kreuzungen CRS Websites bei der Hypoxanthin-Guanin-Phosphoribosyltransferase (HPRT)-Locus in peripheren T-Zellen von 111 Kindern w&auml;hrend der sp&auml;ten Stadien der fetalen Entwicklung durch fr&uuml;he Adoleszenz zugeordnet. Wir beobachteten eine Reihe von spezifischen V (D) J Recombinase Verarbeitung-Features, die Alter und Geschlecht abh&auml;ngig waren. Insbesondere TdT-vermittelte Nukleotid Einf&uuml;gungen variiert je nach Alter und Geschlecht, einschlie&szlig;lich Prozentsatz der Kodierung Kreuzungen mit N-Nukleotid-Eins&auml;tze, Dominanz der GC Nukleotide, und Pr&auml;senz der invertierte Wiederholungen (Pr-Nukleotide) an verarbeiteten Codierung enden. V (D) J Recombinase-vermittelte Verarbeitung der Kodierung von Kreuzungen bei kryptischen Rekombination Signal Sequenzen in peripheren T-Zellen w&auml;hrend der menschlichen Entwicklung.

Ricombinasi V (D) J Media riarrangiamenti loci immuni e sequenze segnale di ricombinazione criptico (CRS), risultante in una variet&agrave; di riarrangiamenti genomici in linfociti normali e le cellule leucemiche da bambini e adulti. La frequenza con cui si verificano questi riarrangiamenti e loro potenziali conseguenze patologiche sono evolutivamente dipendente. V(D)J recombinase mediates rearrangements at immune loci and cryptic recombination signal sequences (cRSS), resulting in a variety of genomic rearrangements in normal lymphocytes and leukemic cells from children and adults. The frequency at which these rearrangements occur and their potential pathologic consequences are developmentally dependent. To gain insight into V(D)J recombinase-mediated events during human development, we investigated 265 coding junctions associated with cRSS sites at the hypoxanthine-guanine phosphoribosyltransferase (HPRT) locus in peripheral T cells from 111 children during the late stages of fetal development through early adolescence. We observed a number of specific V(D)J recombinase processing features that were both age and gender dependent. In particular, TdT-mediated nucleotide insertions varied depending on age and gender, including percentage of coding junctions containing N-nucleotide inserts, predominance of GC nucleotides, and presence of inverted repeats (Pr-nucleotides) at processed coding ends. In addition, the extent of exonucleolytic processing of coding ends was inversely related to age. We also observed a coding-partner-dependent difference in exonucleolytic processing and an age-specific difference in the subtypes of V(D)J-mediated events. We investigated these age- and gender-specific differences with recombination signal information content analysis of the cRSS sites in the human HPRT locus to gain insight into the mechanisms mediating these developmentally specific V(D)J recombinase-mediated rearrangements in humans.

Aimsâ€ƒ Research on drug dependence often involves the administration of drugs of abuse to experienced drug users under controlled laboratory conditions. The primary objective of this study was to assess whether participation in such research alters the frequency of heroin use by non-treatment-seeking opioid-dependent volunteers after study completion. Designâ€ƒ Data were examined from four in-patient studies involving controlled opioid administration. Settingâ€ƒ Substance Use Research Center at Columbia University, New York State Psychiatric Institute. Participantsâ€ƒ Sixty-nine heroin-dependent volunteers. Measurementsâ€ƒ Participants' self-reported heroin use prior to and 1 month after study participation was compared using a Wilcoxon test. Because a number of participants reported that they had stopped using heroin, a logistic regression was used to identify correlates of heroin cessation 1 month after study completion. Findingsâ€ƒ One hundred and one participants entered laboratory studies and 69 completed them. Self-reported heroin use significantly decreased 1 month after study participation [1.7 (Â±2.0) bags per day] compared to baseline [6.8 (Â±4.2) bags per day], Pâ€ƒ<â€ƒ0.001 among the 69 completers. In addition, 42% of the completers were heroin-abstinent 1 month after study completion. Being African American, having a history of opioid dependence treatment, reporting heavier heroin use at baseline and a longer history of heroin use were correlated with cessation of heroin use. Conclusionsâ€ƒ Participation in opioid administration studies does not increase subsequent heroin use and for some individuals leads to accessing opioid dependence treatment and cessation of heroin use in the short term.

Impact of in-patient research participation on subsequent heroin use patterns: implications for ethics and public health.

Objetivos de investigaci&#xF3;n en la dependencia de las drogas a menudo implica la administraci&#xF3;n de drogas de abuso a los usuarios de drogas con experiencia en condiciones controladas de laboratorio. El objetivo principal de este estudio fue evaluar si la participaci&#xF3;n en este tipo de investigaci&#xF3;n altera la frecuencia de consumo de hero&#xED;na por la falta de tratamiento en busca de voluntarios dependientes de opi&#xE1;ceos despu&#xE9;s de la finalizaci&#xF3;n del estudio. Datos de dise&#xF1;o fueron examinados a partir de cuatro pacientes en los estudios sobre la administraci&#xF3;n de opioides controlada. Establecer el uso de sustancias Centro de Investigaci&#xF3;n de la Universidad de Columbia, Nueva York Instituto Psiqui&#xE1;trico del Estado.
