Hepatitis B virus (HBV) X protein (pX) is implicated in hepatocarcinogenesis of chronically infected HBV patients. To understand mechanism(s) of pX-mediated cellular transformation, we employed two tetracycline-regulated, pX-expressing cell lines, constructed in AML12 immortalized hepatocytes: one a differentiated (3pX-1) and the other a de-differentiated (4pX-1) hepatocyte cell line. Only 3pX-1 cells undergo pX-mediated transformation, via sustained Ras-Raf-mitogen-activated protein kinase pathway activation. pX-nontransforming 4pX-1 cells display sustained, pX-dependent JNK pathway activation. To understand how pX mediates different growth characteristics in 3pX-1 and 4pX-1 cells, we report, herein, comparative cell cycle analyses. pX-transforming 3pX-1 cells display pX-dependent G(1), S, and G(2)/M progression evidenced by cyclin D(1), A, and B(1) induction, and Cdc2 kinase activation. pX-nontransforming 4pX-1 cells display pX-dependent G(1) and S phase entry, followed by S phase pause and absence of Cdc2 kinase activation. Interestingly, 4pX-1 cells exhibit selective pX-induced expression of cyclin-dependent kinase inhibitor p21(Cip1), tumor suppressor p19(ARF), and proapoptotic genes bax and IGFBP-3. Despite the pX-mediated induction of growth arrest and apoptotic genes and the absence of pX-dependent Cdc2 activation, 4pX-1 cells do not undergo pX-dependent G(2)/M arrest or apoptosis. Nocodazole-treated, G(2)/M-arrested 4pX-1 cells exhibit pX-dependent formation of multinucleated cells, similar to human T-cell lymphotropic virus type I Tax-expressing cells. We propose that in 4pX-1 cells, pX deregulates the G(2)/M checkpoint, thus rescuing cells from pX-mediated apoptosis.

Hepatitis B virus X protein differentially regulates cell cycle progression in X-transforming versus nontransforming hepatocyte (AML12) cell lines.

L&#x26;#39;h&#xE9;patite B (VHB) la prot&#xE9;ine X (pX) est impliqu&#xE9; dans l&#x26;#39;h&#xE9;patocarcinogen&#xE8;se des patients chroniquement infect&#xE9;s par le VHB. Pour comprendre le m&#xE9;canisme (s) de pX-m&#xE9;diation transformation cellulaire, nous avons utilis&#xE9; deux t&#xE9;tracycline-r&#xE9;glement&#xE9;s, PX-exprimant les lign&#xE9;es cellulaires, construits dans les h&#xE9;patocytes immortalis&#xE9;s AML12: l&#x26;#39;un diff&#xE9;renci&#xE9;e (3px-1) et l&#x26;#39;autre un (d&#xE9;diff&#xE9;renci&#xE9;e 4PX- 1) lign&#xE9;e cellulaire d&#x26;#39;h&#xE9;patocytes. Seuls 3px-1 cellules subissent pX transformation m&#xE9;di&#xE9;e, par l&#x26;#39;interm&#xE9;diaire soutenue Ras-Raf-mitogen-activated protein kinase activation de la voie. pX-nontransforming 4PX-1 cellules affichent soutenue, pX-d&#xE9;pendante activation de la voie JNK. Pour comprendre comment la m&#xE9;diation pX diff&#xE9;rentes caract&#xE9;ristiques de croissance dans 3px-1-1 et 4PX cellules, que nous rapportons, ici, des analyses comparatives du cycle cellulaire. pX-transformation 3px-1 cellules affichent pX-d&#xE9;pendante G (1), S, et G (2) / M en t&#xE9;moigne la progression de la cycline D (1), A et B (1) l&#x26;#39;induction, et activation de la kinase Cdc2. pX-nontransforming 4PX-1 cellules afficher pX-d&#xE9;pendante U (1) et ouverture de la phase S, suivie par une pause de la phase S et en l&#x26;#39;absence d&#x26;#39;activation de la kinase Cdc2. Fait int&#xE9;ressant, 4PX-1 cellules pr&#xE9;sentent s&#xE9;lective pX-induite expression de la cycline-d&#xE9;pendantes inhibiteur de la kinase p21 (Cip1), suppresseur de tumeur p19 (ARF), et les g&#xE8;nes pro-apoptotiques bax et d&#x26;#39;IGFBP-3. Malgr&#xE9; l&#x26;#39;induction pX-m&#xE9;diation de l&#x26;#39;arrestation de croissance et les g&#xE8;nes apoptotiques et l&#x26;#39;absence de pX-activation d&#xE9;pendante Cdc2, 4PX-1 des cellules ne subissent pas de pX-d&#xE9;pendante G (2) / M arrestation ou de l&#x26;#39;apoptose. Nocodazole-trait&#xE9;e, G (2) / M-1-arr&#xEA;t&#xE9;s 4PX cellules pr&#xE9;sentent pX-d&#xE9;pendante de la formation de cellules multinucl&#xE9;&#xE9;es, semblable &#xE0; de lymphocytes T humains de type I du virus lymphotrope des cellules exprimant la taxe. Nous proposons que dans 4PX-1 des cellules, pX d&#xE9;r&#xE9;glemente le point de contr&#xF4;le G (2) / M, donc les cellules de sauvetage pX-apoptose.

Proteina di epatite B (HBV) virus X (pX) &egrave; implicata in hepatocarcinogenesis di pazienti HBV cronicamente infetti. Per comprendere i meccanismi comprendono di trasformazione cellulare mediata da pX, abbiamo impiegato due linee cellulari tetraciclina-regolato, esprimendo pX, costruiti nel AML12 immortalato epatociti: uno differenziato (3pX-1) e l&#39;altro un de-differentiated degli epatociti (1-4pX) cella linea. Solo 3pX-1 cellule subiscono trasformazione pX-mediata, tramite attivazione di pathway della chinasi proteina Ras-Raf-mitogene-attivata sostenuta. pX-nontransforming 4pX-1 cellule visualizzano attivazione di pathway JNK sostenuta, pX-dipendente. Per capire come pX da intermediario caratteristiche differenti di crescita nelle cellule 3pX-1 e 1-4pX, riportiamo qui, analisi comparativa ciclo cellulare. pX-trasformando 3pX-1 cellule visualizzano pX-dipendente g (1), S e G (2) m progressione evidenziato da cyclin d (1), A e B(1) ad induzione e attivazione della chinasi Cdc2. pX-nontransforming 4pX-1 cellule pX-dipendente g (1) e S fase voce viene visualizzata, seguita dalla pausa fase S e l&#39;assenza di attivazione della chinasi Cdc2. &Egrave; interessante notare, 4pX-1 cellule esibiscono selettiva espressione pX-indotta di chinasi ciclina-dipendente inibitore p21(Cip1), p19(ARF) di soppressore del tumore e Miselli geni bax e IGFBP-3. Nonostante l&#39;induzione di arresto di crescita e di apoptotic geni pX-mediata e l&#39;assenza di attivazione di Cdc2 pX-dipendente, 4pX-1 cellule non subiscono pX-dipendente/m G (2) arresto o apoptosi. Nocodazole-trattati, G (2) / M-arrestato 4pX-1 cellule esibiscono pX-dipendente dalla formazione di cellule multinucleate, simile al T-cellula umana virus linfotropico tipo I Tax-esprimendo le cellule. Noi proponiamo che in cellule 4pX-1, pX allenta il checkpoint/m G (2), salvando cos&igrave; le cellule dall&#39;apoptosi mediata da pX.

La hepatitis B (VHB), prote&#xED;na X (PX) est&#xE1; implicada en la carcinog&#xE9;nesis hep&#xE1;tica de pacientes con infecci&#xF3;n cr&#xF3;nica por VHB. Para entender el mecanismo (s) de PX-mediada por la transformaci&#xF3;n celular, se emplearon dos tetraciclina reguladas, PX-l&#xED;neas celulares que expresan, construido en AML12 hepatocitos inmortalizados: una forma diferenciada (3px-1), y el otro (diferenciado de 4px- 1) la l&#xED;nea celular de los hepatocitos. S&#xF3;lo 3px-1 c&#xE9;lulas se someten a pX transformaci&#xF3;n mediada, a trav&#xE9;s de sostenido Ras-Raf-activada por mit&#xF3;genos activaci&#xF3;n de la prote&#xED;na quinasa. pX-nontransforming 4PX-1 en las c&#xE9;lulas muestran sostenido, pX dependiente de activaci&#xF3;n de la v&#xED;a JNK. Para entender c&#xF3;mo interviene en p&#xED;xeles diferentes caracter&#xED;sticas de crecimiento en 3px-1 y c&#xE9;lulas 4PX-1, se presenta, en este documento, los an&#xE1;lisis comparativos del ciclo celular. pX-transformando 3px-1 c&#xE9;lulas mostrar pX-dependiente G (1), S y T (2) / M progresi&#xF3;n evidenciada por ciclina D (1), A y B de inducci&#xF3;n (1), y la activaci&#xF3;n Cdc2 quinasa. pX-nontransforming 4PX-1 c&#xE9;lulas mostrar pX-dependiente G (1) y la entrada de la fase S, seguido de una pausa fase S y ausencia de activaci&#xF3;n quinasa Cdc2. Curiosamente, 4PX-1 en las c&#xE9;lulas exhiben pX selectiva inducida por la expresi&#xF3;n de la ciclina dependiente de inhibidor de la quinasa p21 (Cip1), supresor de tumores p19 (ARF), y los genes pro-apopt&#xF3;ticos bax y la IGFBP-3. Pluripotent neural crest (NC) cells differentiate to diverse lineages, including the neuronal, sympathoadrenal lineage. In primary NC cultures, bone morphogenetic protein 2 (BMP2) requires moderate activation of cAMP signaling for induction of the sympathoadrenal lineage. However, the mechanism by which cAMP signaling synergizes with BMP2 to induce the sympathodrenal lineage is unknown. Herein, we demonstrate that moderate activation of cAMP signaling induces both transcription and activity of proneural transcription factor Phox2a. In NC cultures inhibition of cAMP-response element-binding protein (CREB)-mediated transcription by expression of dominant-negative CREB suppresses Phox2a transcription and sympathoadrenal lineage development. Interestingly, the constitutively active CREB(DIEDML), despite inducing Phox2a transcription, is insufficient for sympathoadrenal lineage development, requiring activation of the cAMP pathway. Because CREB(DIEDML)-mediates cAMP-dependent transcription without requiring activation by the cAMP-dependent protein kinase A (PKA), these results identify PKA activation as necessary in sympathoadrenal lineage development. Treatment of NC cultures with the PKA inhibitor H89 or 1-10 nm okadaic acid (OA), a serine/threonine PP2A-like phosphatase inhibitor, suppresses sympathoadrenal lineage development. Likewise, OA treatment of the CNS-derived catecholaminergic CAD cell line inhibits cAMP-mediated neuronal differentiation. Specifically, OA inhibits cAMP-mediated Phox2a dephosphorylation, cAMP-dependent Phox2a DNA binding in vitro, and cAMP- and Phox2a-dependent dopamine-beta-hydroxylase-luciferase reporter expression. Together, these results support cAMP-dependent Phox2a dephosphorylation is required for its activation. We conclude that moderate activation of cAMP signaling has dual inputs in catecholaminergic, sympathoadrenal lineage development; that is, regulation of both Phox2a transcription and activity. These results provide the first mechanistic understanding of how moderate activation of the cAMP pathway in synergy with BMP2 promotes sympathoadrenal lineage development.

Axonal transport of mitochondria is critical for proper neuronal function. However, little is known about the extracellular signals that regulate this process. In the present study, we show that the neuromodulator serotonin (5-HT) greatly enhances mitochondrial movement in the axons of rat hippocampal neurons in vitro. Administration of a 5-HT1A receptor antagonist inhibited mitochondrial movement, whereas addition of fluoxetine, a selective serotonin reuptake inhibitor, promoted mitochondrial movement. 5-HT receptors are known to activate the Akt/Protein kinase B pathway. Consistent with this, directional mitochondrial movement was almost completely blocked by a specific Akt inhibitor. Moreover, an inhibitor of glycogen synthase kinase-3beta (GSK3beta), a kinase whose activity is blocked by Akt-mediated phosphorylation, promoted mitochondrial movement. These findings show that 5-HT1A receptor activation stimulates mitochondrial movement in hippocampal neurons by inhibiting GSK3beta activity via Akt. Our findings suggest that 5-HT may mediate the redistribution of energy sources within responsive neurons, a possibility that has significant implications for understanding the global biological effects of this important neuromodulator.

Serotonin stimulates mitochondrial transport in hippocampal neurons.

En el presente estudio, mostramos que la serotonina neuromodulador (5-HT) mejora en gran medida el movimiento mitocondrial en los axones de las neuronas del hipocampo de rata in vitro. The trafficking of mitochondria within neurons is a highly regulated process. In an earlier study, we found that serotonin (5-HT), acting through the 5-HT1A receptor subtype, promotes axonal transport of mitochondria in cultured hippocampal neurons by increasing Akt activity, and consequently decreasing glycogen synthase kinase (GSK3beta) activity. This finding suggests a critical role for neuromodulators in the regulation of mitochondrial trafficking in neurons. In the present study, we investigate the effects of a second important neuromodulator, dopamine, on mitochondrial transport in hippocampal neurons.

Tubulin is a major substrate of the cytoplasmic class II histone deacetylase HDAC6. Inhibition of HDAC6 results in higher levels of acetylated tubulin and enhanced binding of the motor protein kinesin-1 to tubulin, which promotes transport of cargoes along microtubules. Microtubule-dependent intracellular trafficking may therefore be regulated by modulating the activity of HDAC6. We have shown previously that the neuromodulator serotonin increases mitochondrial movement in hippocampal neurons via the Akt-GSK3beta signaling pathway. Here, we demonstrate a role for HDAC6 in this signaling pathway.

L&#x26;#39;inhibition de HDAC6 r&#xE9;sultats dans des niveaux plus &#xE9;lev&#xE9;s de la tubuline ac&#xE9;tyl&#xE9;e et liaison am&#xE9;lior&#xE9;e de la prot&#xE9;ine motrice kin&#xE9;sine-1 &#xE0; la tubuline, qui favorise le transport des cargaisons le long des microtubules. Microtubules d&#xE9;pend trafic intracellulaire peut donc &#xEA;tre r&#xE9;gl&#xE9;e en modulant l&#x26;#39;activit&#xE9; de HDAC6. Nous avons montr&#xE9; pr&#xE9;c&#xE9;demment que la s&#xE9;rotonine augmente neuromodulateur mouvement mitochondrial dans les neurones hippocampiques via la voie Akt-GSK3b&#xEA;ta signalisation. Ici, nous d&#xE9;montrons un r&#xF4;le &#xE0; jouer dans HDAC6 cette voie de signalisation.

Tubulin ist ein wichtiger Substrat der zytoplasmatischen Klasse II Histon Deacetylase HDAC6. Hemmung der HDAC6 resultiert in h&ouml;heren Ebenen der acetylierten Tubulin und verst&auml;rkte Bindung der motor Protein Kinesin-1 Tubulin, die Bef&ouml;rderung von Frachten entlang der Mikrotubuli f&ouml;rdert. Microtubule-abh&auml;ngige intrazellul&auml;re Menschenhandel kann daher durch Modulation der Aktivit&auml;t des HDAC6 geregelt werden. Wir haben bereits gezeigt, dass das Serotonin oder Neuromodulatoren mitochondriale Bewegung in Neuronen &uuml;ber den Akt-GSK3beta-Signalweg erh&ouml;ht. Hier zeigen wir eine Rolle f&uuml;r die HDAC6 in dieser Signalweg.

Inibizione della HDAC6 si traduce in livelli pi&ugrave; elevati di tubulina acetilata e associazione avanzata del motore proteine chinesina-1 alla tubulina, che promuove il trasporto dei carichi lungo microtubuli. Traffico intracellulare di microtubule-dipendente quindi potrebbe essere regolamentata modulando l&#39;attivit&agrave; di HDAC6. Abbiamo dimostrato in precedenza che la serotonina Neuromodulatore aumenta movimento mitocondriale nei neuroni hippocampal tramite la via di segnalazione Akt-GSK3beta. Qui, noi dimostrare un ruolo per HDAC6 in questa via di segnalazione.

Tubulina es un sustrato importante de la clase frente al citoplasma de HDAC6 deacetilasa de histonas II. La inhibici&#xF3;n de HDAC6 se traduce en mayores niveles de tubulina acetilada y la mayor uni&#xF3;n de la prote&#xED;na motora quinesina-1 a la tubulina, que promueve el transporte de cargas a lo largo de los microt&#xFA;bulos. Microt&#xFA;bulos dependiente de tr&#xE1;fico intracelular por lo tanto, puede ser regulado por la modulaci&#xF3;n de la actividad de HDAC6. Hemos demostrado anteriormente que la serotonina aumenta el movimiento neuromodulador mitocondrial en las neuronas del hipocampo a trav&#xE9;s de la v&#xED;a Akt-GSK3beta se&#xF1;alizaci&#xF3;n.
