&#x77ED;&#x80BD;&#x914D;&#x4F53;&#x7EB3;&#x5165;&#x8981;&#x91CD;&#x5B9A;&#x5411;&#x7684; AAV8 &#x548C; AAV9 &#x5728;&#x4F53;&#x5185;&#x57FA;&#x56E0;&#x8F6C;&#x5BFC;&#x7684;&#x4E09;&#x91CD;&#x7A57;&#x8863;&#x58F3;&#x57DF;&#x3002;

Efficiency and specificity of viral vectors are vital issues in gene therapy. Insertion of peptide ligands into the adeno-associated viral (AAV) capsid at receptor binding sites can re-target AAV2-derived vectors to alternative cell types. Also, the use of serotypes AAV8 and -9 is more efficient than AAV2 for gene transfer to certain tissues in vivo. Consequently, re-targeting of these serotypes by ligand insertion could be a promising approach but has not been explored so far. Here, we generated AAV8 and -9 vectors displaying peptides in the threefold spike capsid domain. These peptides had been selected from peptide libraries displayed on capsids of AAV serotype 2 to optimize systemic gene delivery to murine lung tissue and to breast cancer tissue in PymT transgenic mice (PymT). Such peptide insertions at position 590 of the AAV8 capsid and position 589 of the AAV9 capsid changed the transduction properties of both serotypes. However, both peptides inserted in AAV8 did not result in the same changes of tissue tropism as they did in AAV2. While the AAV2 peptides selected on murine lung tissue did not alter tropism of serotypes 8 and -9, insertion of the AAV2-derived peptide selected on breast cancer tissue augmented tumor gene delivery in both serotypes. Further, this peptide mediated a strong but unspecific in vivo gene transfer for AAV8 and abrogated transduction of various control tissues for AAV9. Our findings indicate that peptide insertion into defined sites of AAV8 and -9 capsids can change and improve their efficiency and specificity compared to their wild type variants and to AAV2, making these insertion sites attractive for the generation of novel targeted vectors in these serotypes.

Peptide ligands incorporated into the threefold spike capsid domain to re-direct gene transduction of AAV8 and AAV9 in vivo.

L&#x26;#39;efficacit&#xE9; et la sp&#xE9;cificit&#xE9; de vecteurs viraux sont des questions vitales de la th&#xE9;rapie g&#xE9;nique. Insertion de ligands peptidiques dans la viral ad&#xE9;no-associ&#xE9; (AAV) capside &#xE0; des sites de liaison aux r&#xE9;cepteurs cibles peut r&#xE9;-AAV2 vecteurs d&#xE9;riv&#xE9;s &#xE0; des types de cellules de remplacement. En outre, l&#x26;#39;utilisation des s&#xE9;rotypes AAV8 et -9 est plus efficace que AAV2 de transfert de g&#xE8;nes &#xE0; certains tissus in vivo. Ici, nous avons g&#xE9;n&#xE9;r&#xE9; AAV8 et -9 vecteurs pr&#xE9;sentant des peptides dans le domaine triple pic capside. Ces peptides ont &#xE9;t&#xE9; s&#xE9;lectionn&#xE9;s &#xE0; partir des banques de peptides affich&#xE9;s sur capsides d&#x26;#39;AAV de s&#xE9;rotype 2 pour optimiser la livraison de g&#xE8;nes syst&#xE9;mique du tissu pulmonaire murin et de tissus du cancer du sein dans PyMT souris transg&#xE9;niques (PyMT). Ces insertions peptidiques en position 590 de la capside et la position AAV8 589 de la capside AAV9 modifi&#xE9; les propri&#xE9;t&#xE9;s de transduction des deux s&#xE9;rotypes. Toutefois, les deux peptides ins&#xE9;r&#xE9;s dans AAV8 n&#x26;#39;a pas entra&#xEE;n&#xE9; dans les m&#xEA;mes changements de tropisme tissulaire comme ils le faisaient dans AAV2. Tandis que les peptides s&#xE9;lectionn&#xE9;s AAV2 sur le tissu pulmonaire murin ne pas modifier le tropisme de s&#xE9;rotypes 8 et -9, insertion du peptide d&#xE9;riv&#xE9; AAV2 s&#xE9;lectionn&#xE9; sur le tissu du cancer du sein augment&#xE9;e d&#x26;#39;apport de g&#xE8;ne dans les deux s&#xE9;rotypes tumeur. En outre, ce peptide m&#xE9;diation non sp&#xE9;cifique, mais une forte en mati&#xE8;re de transfert de g&#xE8;ne in vivo pour AAV8 et de la transduction des tissus abrog&#xE9; contr&#xF4;le diff&#xE9;rents pour AAV9. Nos r&#xE9;sultats indiquent que l&#x26;#39;insertion peptide dans des sites d&#xE9;finis de AAV8 et -9 capsides peuvent modifier et am&#xE9;liorer leur efficacit&#xE9; et leur sp&#xE9;cificit&#xE9; par rapport &#xE0; leurs variantes de type sauvage et de AAV2, ce qui rend ces sites d&#x26;#39;insertion int&#xE9;ressante pour la g&#xE9;n&#xE9;ration de nouveaux vecteurs cibl&#xE9;s dans ces s&#xE9;rotypes.

Effizienz und Besonderheit des viralen Vektoren sind wichtige Themen in der Gentherapie. Einf&uuml;gen von Peptid-Liganden in das Adeno-assoziierte Virus (AAV) Kapsid am Rezeptor Bindungsstellen kann AAV2 abgeleitete Vektoren auf alternative Zelltypen re-target. Auch ist die Verwendung von Serotypen AAV8 und-9 effizienter als AAV2 Gentransfer in bestimmten Geweben in-vivo. Folglich k&ouml;nnte Vermarktungsm&ouml;glichkeiten diese Serotypen von Ligand einf&uuml;gen ein vielversprechender Ansatz aber hat bisher nicht erforscht. Hier erzielten wir im AAV8 und-9 Vektoren Peptide in der dreifachen Spike-Kapsid-Dom&auml;ne angezeigt. Diese Peptide hatte von Peptid-Bibliotheken auf Capsids des AAV Serotyp 2 ausgew&auml;hlt wurde, systemische gen Lieferung murinen Lungengewebe und Krebs Brustgewebe in PymT transgene M&auml;use (PymT) zu optimieren. Solche Peptid-Einf&uuml;gevorg&auml;ngen in 590 von der AAV8-Kapsid und Position, dass 589 von der AAV9-Kapsid die Transduktion Eigenschaften beider Serotypen ge&auml;ndert. Jedoch beide Peptide in AAV8 eingef&uuml;gt werden, nicht die gleichen Ver&auml;nderungen des Gewebes Tropismus f&uuml;hren wie in AAV2. W&auml;hrend die AAV2 Peptide auf murinen Lungengewebe ausgew&auml;hlt Tropismus Serotypen 8 und 9, Einf&uuml;gung der AAV2 abgeleitete Peptide Nachnahme Brust Krebs Gewebe erg&auml;nzt Tumor-gen in beiden Serotypen ausgew&auml;hlt nicht ge&auml;ndert. Dar&uuml;ber hinaus vermittelt dieses Peptid f&uuml;r AAV8 und aufgehobene Transduktion der verschiedenen Steuerelement Gewebe f&uuml;r AAV9 eine starke aber unspezifisch in-vivo Gentransfer. Unsere Ergebnisse zeigen, dass Peptid einf&uuml;gen in definierte Standorte von AAV8 und-9 Capsids &auml;ndern und verbessern Sie ihre Effizienz und Spezifit&auml;t im Vergleich zu ihren Wildtyp-Varianten und AAV2, Attraktivit&auml;t dieser Einf&uuml;gung-Sites f&uuml;r die Generierung von Roman gezielte Vektoren in diese Serotypen.

L&#39;efficienza e la specificit&agrave; dei vettori virali sono questioni vitali in terapia genica. Inserimento dei ligandi peptidici in l&#39;adeno-associati (AAV) capside virale presso i siti di legame del recettore pu&ograve; modificare la destinazione degli vettori AAV2-derivato di tipi cellulari alternativi. Inoltre, l&#39;uso di sierotipi AAV8 e -9 &egrave; pi&ugrave; efficiente di AAV2 per trasferimento del gene ad alcuni tessuti in vivo. Ulteriormente, questo peptide mediata da un forte ma trasferimento genico in vivo non specifico per AAV8 e abrogata trasduzione dei vari tessuti di controllo per AAV9. I nostri risultati indicano che inserimento del peptide in siti definiti di AAV8 e -9 capsidi pu&ograve; cambiare e migliorare la loro efficienza e la specificit&agrave; rispetto alle loro varianti di tipo selvatico e di AAV2, rendendo questi siti inserimento attraente per la generazione di nuovi vettori mirati in questi sierotipi.

La eficiencia y la especificidad de los vectores virales son cuestiones vitales en la terapia g&#xE9;nica. La inserci&#xF3;n de ligandos pept&#xED;dicos en el virus adeno-asociado (AAV) c&#xE1;pside en los sitios de uni&#xF3;n al receptor puede volver a destino AAV2 vectores derivados de tipos de c&#xE9;lulas a otras. Adem&#xE1;s, el uso de los serotipos AAV8 y -9 es m&#xE1;s eficiente que AAV2 de transferencia de genes a ciertos tejidos in vivo. En consecuencia, re-focalizaci&#xF3;n de estos serotipos mediante la inserci&#xF3;n ligando puede ser un enfoque prometedor, pero no ha sido explorada hasta ahora. Aqu&#xED;, hemos generado AAV8 y -9 vectores que muestran p&#xE9;ptidos en la c&#xE1;pside tres picos. Estos p&#xE9;ptidos han sido seleccionados de bibliotecas de p&#xE9;ptidos que se muestran en c&#xE1;psides de AAV serotipo 2 para optimizar la entrega de genes en el tejido pulmonar sist&#xE9;mica murino y en el tejido de c&#xE1;ncer de mama en ratones transg&#xE9;nicos PYMT (PYMT). Estas inserciones de p&#xE9;ptidos en la posici&#xF3;n 590 de la c&#xE1;pside AAV8 y la posici&#xF3;n 589 de la c&#xE1;pside AAV9 cambiado las propiedades de transducci&#xF3;n de ambos serotipos. Sin embargo, ambos p&#xE9;ptidos insertados en AAV8 no dio lugar a los mismos cambios de tropismo tisular como lo hicieron en AAV2. Mientras que los AAV2 p&#xE9;ptidos seleccionados en el tejido pulmonar murino no alter&#xF3; tropismo de los serotipos 8 y -9, inserci&#xF3;n del p&#xE9;ptido AAV2 derivado seleccionado en el c&#xE1;ncer de mama entrega tejido aumentada gen tumor en ambos serotipos. Adem&#xE1;s, este p&#xE9;ptido mediado una fuerte, pero no espec&#xED;fica en la transferencia de genes in vivo para AAV8 y transducci&#xF3;n abrogada de tejidos de control diferentes para AAV9. Nuestros hallazgos indican que la inserci&#xF3;n del p&#xE9;ptido en sitios definidos de AAV8 y -9 c&#xE1;psidas pueden cambiar y mejorar su eficiencia y especificidad en comparaci&#xF3;n con sus variantes de tipo salvaje y AAV2, por lo que estos sitios de inserci&#xF3;n atractivo para la generaci&#xF3;n de nuevos vectores dirigidos en estos serotipos.
