Whether the recently identified innate lymphocyte population coexpressing natural killer cell receptors (NKRs) and the nuclear receptor RORÎ³t is part of the NK or lymphoid tissue inducer (LTi) cell lineage remains unclear. By using adoptive transfer of genetically tagged LTi-like cells, we demonstrate that NKRâ»RORÎ³t(+) innate lymphocytes but not NK cells were direct progenitors to NKR(+)RORÎ³t(+) cells in vivo. Genetic lineage tracing revealed that the differentiation of LTi-like cells was characterized by the stable upregulation of NKRs and a progressive loss of RORÎ³t expression. Whereas interleukin-7 (IL-7) and intestinal microbiota stabilized RORÎ³t expression within such NKR-LTi cells, IL-12 and IL-15 accelerated RORÎ³t loss. RORÎ³t(+) NKR-LTi cells produced IL-22, whereas RORÎ³tâ» NKR-LTi cells released IFN-Î³ and were potent inducers of colitis. Thus, the RORÎ³t gradient in NKR-LTi cells serves as a tunable rheostat for their functional program. Our data also define a previously unappreciated role of RORÎ³tâ» NKR-LTi cells for the onset or maintenance of inflammatory bowel diseases.

Lignage cellulaire reste peu clair si les r&eacute;cepteurs de cellule tueuse naturelle coexpressing de lymphocytes inn&eacute;s identifi&eacute; r&eacute;cemment population (NKRs) et le r&eacute;cepteur nucl&eacute;aire ROR&gamma;t appartient &agrave; la NK ou inducteur de tissu lympho&iuml;de (LTi). En utilisant le transfert adoptif de cellules g&eacute;n&eacute;tiquement marqu&eacute;s LTi, nous d&eacute;montrer que les lymphocytes inn&eacute;s NKR&#x207B;ROR&gamma;t(+) mais pas les cellules NK &eacute;taient prog&eacute;niteurs directs aux cellules NKR(+)ROR&gamma;t(+) in vivo. Tra&ccedil;age de la lign&eacute;e g&eacute;n&eacute;tique a r&eacute;v&eacute;l&eacute; que la diff&eacute;renciation des cellules LTi se caract&eacute;rise par l&#39;augmentation stable de NKRs et une perte progressive de l&#39;expression de ROR&gamma;t. Alors que l&#39;interleukine-7 (IL-7) et le microbiote intestinal stabilis&eacute; ROR&gamma;t expression dans les cellules de ces couronnes-LTi, IL-12 et IL-15 acc&eacute;l&eacute;r&eacute; la perte de ROR&gamma;t. ROR&gamma;t(+) couronnes-LTi cellules produisent IL-22, alors que les cellules ROR&gamma;t&#x207B; couronnes-LTi lib&eacute;r&eacute;s d&#39;IFN-&gamma; et puissants inducteurs de colite. Ainsi, le gradient de ROR&gamma;t dans les cellules de couronnes-LTi sert un rh&eacute;ostat accordable pour leur programme fonctionnel. Nos donn&eacute;es &eacute;galement d&eacute;finissent un r&ocirc;le pr&eacute;c&eacute;demment incompris des cellules ROR&gamma;t&#x207B; couronnes-LTi d&#39;apparition ou entretien des maladies inflammatoires de l&#39;intestin.

Expression r&eacute;gul&eacute;e de r&eacute;cepteurs nucl&eacute;aires ROR&gamma;t conf&egrave;re des destins fonctionnels distincts &agrave; NK cellules exprimant le r&eacute;cepteur ROR&gamma;t(+) inn&eacute;es lymphocytes.

Genetische Abstammung Ablaufverfolgung ergab, dass die Differenzierung der LTi-&auml;hnlichen Zellen von der stabilen Hochregulation des NKRs und einen progressiven Verlust des ROR&gamma;t Ausdruck gepr&auml;gt war. W&auml;hrend Interleukin-7 (IL-7) und Darm Microbiota ROR&gamma;t Ausdruck innerhalb solcher NKR-LTi-Zellen stabilisiert, beschleunigt die IL-12 und IL-15 ROR&gamma;t Verlust. Somit dient die Steigung ROR&gamma;t NKR-LTi-Zellen als eine einstellbare Rheostat f&uuml;r ihre funktionale Programm. &#xD575; &#xC218;&#xC6A9; &#xCCB4; Ror&gamma;t&#xC758; &#xB808; &#xADE4; &#xB808;&#xC774;&#xD2B8; &#xB41C; &#xC2DD; NK &#xC138;&#xD3EC; &#xC218;&#xC6A9; &#xCCB4;&#xB97C; &#xD45C;&#xD604; ROR&gamma;t(+) &#xD0C0;&#xACE0; &#xB09C; &#xC138;&#xD3EC;&#xC5D0; &#xB69C;&#xB837;&#xD55C; &#xAE30;&#xB2A5;&#xC801; &#xC6B4;&#xBA85;&#xC744; &#xC218; &#xC5EC;.

Si las linfocitos innata recientemente identificado poblaci&oacute;n coexpressing c&eacute;lula de asesino natural receptores (NKRs) y el receptor nuclear ROR&gamma;t es parte del NK o inductor de tejido linfoide (LTi) linaje celular sigue siendo confuso. Mediante la transferencia adoptiva de c&eacute;lulas gen&eacute;ticamente etiquetas LTi, demostramos que los linfocitos innatos de NKR&#x207B;ROR&gamma;t(+) pero no las c&eacute;lulas NK fueron progenitores directos a NKR(+)ROR&gamma;t(+) c&eacute;lulas in vivo. Seguimiento de linaje gen&eacute;tico revel&oacute; que la diferenciaci&oacute;n de LTi-como las c&eacute;lulas se caracteriz&oacute; por la regulaci&oacute;n al alza estable de NKRs y una p&eacute;rdida progresiva de la expresi&oacute;n de ROR&gamma;t. Considerando que la interleucina-7 (IL-7) y la microbiota intestinal estabilizaron ROR&gamma;t expresi&oacute;n dentro de tales c&eacute;lulas NKR-LTi, IL-12 y la IL-15 aceleran p&eacute;rdida de ROR&gamma;t. ROR&gamma;t(+) NKR-LTi c&eacute;lulas producen IL-22, mientras que las c&eacute;lulas ROR&gamma;t&#x207B; NKR-LTi lanzado IFN-&gamma; y eran potentes inductores de la colitis. As&iacute;, el gradiente de ROR&gamma;t en c&eacute;lulas de Nagorno-Karabaj-LTi sirve como un re&oacute;stato armonioso para su programa funcional. La expresi&oacute;n regulada de receptor nuclear ROR&gamma;t confiere distintos destinos funcionales a NK que expresan el receptor ROR&gamma;t(+) innatos linfocitos de c&eacute;lulas.
