Die Division von Lungenkrankheiten von der National Heart, Lung und Blut-Institut, mit der Office von seltenen Krankheiten Forschung, hielt einen Workshop Schwerpunktbereiche und strategische Ziele zur Verbesserung und Beschleunigung der Forschung, die f&uuml;hren verbesserte Verst&auml;ndnis der Lunge Gef&auml;&szlig;system, translational-Research-Bed&uuml;rfnisse und letztlich die Pflege von Patienten mit pulmonalen vaskul&auml;ren Erkrankungen. Multidisziplin&auml;re Experten mit vielf&auml;ltigen Erfahrung in translational, Labor und klinische Studien identifiziert sieben Schwerpunktbereiche und Einschr&auml;nkungen in unserem derzeitigen Kenntnisstand, Technologien und Ans&auml;tze diskutiert. Der Fokus f&uuml;r zuk&uuml;nftige Forschungsbem&uuml;hungen umfassen Folgendes: (1) besser charakterisieren vaskul&auml;re Genotyp-Ph&auml;notyp-Beziehungen und die Einbeziehung Systembiologie n&auml;hert, wenn angemessen; (2) F&ouml;rderung unser Verst&auml;ndnis der pulmonalen vaskul&auml;ren metabolische regulatorischen signalisieren in Gesundheit und Krankheit; (3) erweitern unser Wissen &uuml;ber die biologischen Beziehungen zwischen den Kreislauf der Lunge und im Umlauf, Elemente, systemische vaskul&auml;re Funktion und richtige Herzfunktion und Krankheit; (4) Verbesserung der translationalen Forschung zur Identifizierung von krankheitsmodifizierenden Therapien f&uuml;r die pulmonale Hypertonie-Erkrankungen; (5) die Einrichtung einer angemessenen und effektiven Plattform f&uuml;r die F&ouml;rderung der translatorischen Erkenntnisse in klinische Studien testen; und (6) Entwicklung von spezifischen Technologien und Tools, die f&uuml;r diese Ziele, wie z. B. Frage-gef&uuml;hrte bildgebender Verfahren und Lunge vaskul&auml;re Ermittler Trainingsprogramme erm&ouml;glicht werden werden. Empfehlungen von diesem Workshop werden innerhalb der Lunge vaskul&auml;re Biologie und Krankheit extramuralem Forschungsprogramm f&uuml;r Planung und strategische Umsetzung Zwecke verwendet werden.

The Division of Lung Diseases of the National Heart, Lung, and Blood Institute, with the Office of Rare Diseases Research, held a workshop to identify priority areas and strategic goals to enhance and accelerate research that will result in improved understanding of the lung vasculature, translational research needs, and ultimately the care of patients with pulmonary vascular diseases. Multidisciplinary experts with diverse experience in laboratory, translational, and clinical studies identified seven priority areas and discussed limitations in our current knowledge, technologies, and approaches. The focus for future research efforts include the following: (1) better characterizing vascular genotype-phenotype relationships and incorporating systems biology approaches when appropriate; (2) advancing our understanding of pulmonary vascular metabolic regulatory signaling in health and disease; (3) expanding our knowledge of the biologic relationships between the lung circulation and circulating elements, systemic vascular function, and right heart function and disease; (4) improving translational research for identifying disease-modifying therapies for the pulmonary hypertensive diseases; (5) establishing an appropriate and effective platform for advancing translational findings into clinical studies testing; and (6) developing the specific technologies and tools that will be enabling for these goals, such as question-guided imaging techniques and lung vascular investigator training programs. Recommendations from this workshop will be used within the Lung Vascular Biology and Disease Extramural Research Program for planning and strategic implementation purposes.

La Divisi&#xF3;n de Enfermedades Pulmonares del Instituto Nacional del Coraz&#xF3;n, los Pulmones y la Sangre, con la Oficina de Investigaci&#xF3;n de Enfermedades Raras, que se celebr&#xF3; un taller para identificar las &#xE1;reas prioritarias y los objetivos estrat&#xE9;gicos para mejorar y acelerar la investigaci&#xF3;n que se traducir&#xE1; en una mejor comprensi&#xF3;n de la vasculatura pulmonar, necesidades de investigaci&#xF3;n traslacional, y en &#xFA;ltima instancia, el cuidado de los pacientes con enfermedades vasculares pulmonares. Expertos multidisciplinar con experiencia diversa en el laboratorio, la traducci&#xF3;n, y los estudios cl&#xED;nicos identificaron siete &#xE1;reas prioritarias y las limitaciones discutidas en nuestro conocimiento actual, las tecnolog&#xED;as y enfoques. El enfoque para futuros esfuerzos de investigaci&#xF3;n son las siguientes: (1) caracterizar mejor vasculares relaciones genotipo-fenotipo y la incorporaci&#xF3;n de la biolog&#xED;a de sistemas se acerca cuando sea apropiado, (2) avanzar en nuestra comprensi&#xF3;n de la vasculares pulmonares de se&#xF1;alizaci&#xF3;n metab&#xF3;licas reguladoras en materia de salud y la enfermedad, (3) ampliar nuestro conocimiento de las relaciones biol&#xF3;gicas entre la circulaci&#xF3;n pulmonar y de los elementos circulantes, funci&#xF3;n vascular sist&#xE9;mica y la funci&#xF3;n del coraz&#xF3;n derecho y la enfermedad, (4) la mejora de la investigaci&#xF3;n traslacional para la identificaci&#xF3;n de modificaci&#xF3;n de la enfermedad de las terapias para las enfermedades hipertensivas pulmonares; (5) establecer un sistema adecuado y eficaz plataforma para el avance de los resultados de traslaci&#xF3;n en las pruebas de los ensayos cl&#xED;nicos, y (6) desarrollo de las tecnolog&#xED;as y herramientas espec&#xED;ficas que ser&#xE1;n propicios para que estas metas, tales como t&#xE9;cnicas de imagen con gu&#xED;a de preguntas y pulmonares programas de formaci&#xF3;n de investigadores vasculares. The contribution of the Wnt pathway has been extensively characterized in embryogenesis, differentiation, and stem cell biology but not in mammalian metabolism. Here, using in vivo gain- and loss-of-function models, we demonstrate an important role for Wnt signaling in hepatic metabolism. In particular, Î²-catenin, the downstream mediator of canonical Wnt signaling, altered serum glucose concentrations and regulated hepatic glucose production. Î²-Catenin also modulated hepatic insulin signaling. Furthermore, Î²-catenin interacted with the transcription factor FoxO1 in livers from mice under starved conditions. The interaction of FoxO1 with Î²-catenin regulated the transcriptional activation of the genes encoding glucose-6-phosphatase (G6Pase) and phosphoenolpyruvate carboxykinase (PEPCK), the two rate-limiting enzymes in hepatic gluconeogenesis. Moreover, starvation induced the hepatic expression of mRNAs encoding different Wnt isoforms. In addition, nutrient deprivation appeared to favor the association of Î²-catenin with FoxO family members, rather than with members of the T cell factor of transcriptional activators. Notably, in a model of diet-induced obesity, hepatic deletion of Î²-catenin improved overall metabolic homeostasis. These observations implicate Wnt signaling in the modulation of hepatic metabolism and raise the possibility that Wnt signaling may play a similar role in the metabolic regulation of other tissues.

Death from chronic lung disease is increasing and chronic obstructive pulmonary disease has become the third leading cause of death in the United States in the past decade. Both chronic and acute lung diseases disproportionately affect elderly individuals, making it likely that these diseases will become more frequent and severe as the worldwide population ages. Chronic lung diseases are associated with substantial morbidity, frequently resulting in exercise limiting dyspnea, immobilization, and isolation. Therefore, effective strategies to prevent or treat lung disease are likely to increase healthspan as well as life span. This review summarizes the findings of a joint workshop sponsored by the NIA and NHLBI that brought together investigators focused on aging and lung biology. These investigators encouraged the use of genetic systems and aged animals in the study of lung disease and the development of integrative systems-based platforms that can dynamically incorporate data sets that describe the genomics, transcriptomics, epigenomics, metabolomics, and proteomics of the aging lung in health and disease. Further research was recommended to integrate benchmark biological hallmarks of aging in the lung with the pathobiology of acute and chronic lung diseases with divergent pathologies for which advanced age is the most important risk factor.

The Intersection of Aging Biology and the Pathobiology of Lung Diseases: A Joint NHLBI/NIA Workshop.

Endothelial-to-mesenchymal transition (EndoMT) is a cellular process often initiated by the transforming growth factor β (TGF-β) family of ligands. Although required for normal heart valve development, deregulated EndoMT is linked to a wide range of pathological conditions. Here, we demonstrate that endothelial fatty acid oxidation (FAO) is a critical in vitro and in vivo regulator of EndoMT. We further show that this FAO-dependent metabolic regulation of EndoMT occurs through alterations in intracellular acetyl-CoA levels. Disruption of FAO via conditional deletion of endothelial carnitine palmitoyltransferase II (Cpt2) augments the magnitude of embryonic EndoMT, resulting in thickening of cardiac valves. Consistent with the known pathological effects of EndoMT, adult Cpt2 mice demonstrate increased permeability in multiple vascular beds. Taken together, these results demonstrate that endothelial FAO is required to maintain endothelial cell fate and that therapeutic manipulation of endothelial metabolism could provide the basis for treating a growing number of EndoMT-linked pathological conditions.
