Survival signaling by the erythropoietin (Epo) receptor (EpoR) is essential for erythropoiesis and for its acceleration in hypoxic stress. Several apparently redundant EpoR survival pathways were identified in vitro, raising the possibility of their functional specialization in vivo. Here we used mouse models of acute and chronic stress, including a hypoxic environment and Î²-thalassemia, to identify two markedly different response dynamics for two erythroblast survival pathways in vivo. Induction of the antiapoptotic protein Bcl-x(L) is rapid but transient, while suppression of the proapoptotic protein Bim is slower but persistent. Similar to sensory adaptation, however, the Bcl-x(L) pathway "resets," allowing it to respond afresh to acute stress superimposed on a chronic stress stimulus. Using "knock-in" mouse models expressing mutant EpoRs, we found that adaptation in the Bcl-x(L) response occurs because of adaptation of its upstream regulator Stat5, both requiring the EpoR distal cytoplasmic domain. We conclude that survival pathways show previously unsuspected functional specialization for the acute and chronic phases of the stress response. Bcl-x(L) induction provides a "stop-gap" in acute stress, until slower but permanent pathways are activated. Furthermore, pathologic elevation of Bcl-x(L) may be the result of impaired adaptation, with implications for myeloproliferative disease mechanisms.

Supervivencia de la se&#xF1;alizaci&#xF3;n de la eritropoyetina (EPO) del receptor (EpoR) es esencial para la eritropoyesis y su aceleraci&#xF3;n en el estr&#xE9;s hip&#xF3;xico. Varias v&#xED;as de EpoR aparentemente redundantes de supervivencia fueron identificados in vitro, aumentando la posibilidad de su especializaci&#xF3;n funcional in vivo. Aqu&#xED; hemos utilizado modelos de rat&#xF3;n de estr&#xE9;s agudo y cr&#xF3;nico, incluyendo un ambiente hip&#xF3;xico y la &#x3B2;-talasemia, para identificar dos din&#xE1;micas muy diferentes de respuesta de dos v&#xED;as eritroblasto supervivencia in vivo. La inducci&#xF3;n de la prote&#xED;na antiapopt&#xF3;tica Bcl-x (L) es r&#xE1;pida, pero transitoria, mientras que la supresi&#xF3;n de la prote&#xED;na Bim proapopt&#xF3;tico es m&#xE1;s lento, pero persistente. Al igual que la adaptaci&#xF3;n sensorial, sin embargo, la Bcl-x (L) y la v&#xED;a &#x26;quot;restablece&#x26;quot;, a fin de responder de nuevo a estr&#xE9;s agudo superpuesto a un est&#xED;mulo de estr&#xE9;s cr&#xF3;nico. El uso de &#x26;quot;knock-in&#x26;quot; modelos de rat&#xF3;n que expresan EpoRs mutantes, se encontr&#xF3; que la adaptaci&#xF3;n de la Bcl-x (L) la respuesta se debe a la adaptaci&#xF3;n de su regulador de aguas arriba STAT5, tanto que requiere el dominio de EpoR citoplasm&#xE1;tica distal. Llegamos a la conclusi&#xF3;n de que las v&#xED;as de supervivencia muestran especializaci&#xF3;n funcional insospechado de las fases aguda y cr&#xF3;nica de la respuesta al estr&#xE9;s. Bcl-x (L) de inducci&#xF3;n ofrece una &#x26;quot;soluci&#xF3;n temporal&#x26;quot; en el estr&#xE9;s agudo, hasta que las v&#xED;as m&#xE1;s lentas pero permanente se activan. Por otra parte, la elevaci&#xF3;n patol&#xF3;gica de Bcl-x (L) puede ser el resultado de la adaptaci&#xF3;n deficiente, con implicaciones para los mecanismos de las enfermedades mieloproliferativas.
