The goal of this study was to determine if selected mycotoxins (trichothecenes, aflatoxins, and ochratoxins) could be extracted and identified in human tissue and body fluids from patients exposed to toxin producing molds in their environment. Human urine and methanol extracted tissues and sputum were examined. Trichothecenes were tested using competitive ELISA techniques. Aflatoxins B1, B2, G1, and G2, and ochratoxin A were tested by using immunoaffinity columns and fluorometry. Test sensitivity and specificity were determined. Levels of detection for the various mycotoxins varied from 0.2 ppb for trichothecenes, 1.0 ppb for aflatoxins, and 2.0 ppb for ochratoxins. Trichothecene levels varied in urine, sputum, and tissue biopsies (lung, liver, brain) from undetectable (<0.2 ppb) to levels up to 18 ppb. Aflatoxin levels from the same types of tissues varied from 1.0 to 5.0 ppb. Ochratoxins isolated in the same type of tissues varied from 2.0 ppb to > 10.0 ppb. Negative control patients had no detectable mycotoxins in their tissues or fluids. These data show that mycotoxins can be detected in body fluids and human tissue from patients exposed to mycotoxin producing molds in the environment, and demonstrate which human tissues or fluids are the most likely to yield positive results.

El objetivo de este estudio fue determinar si las micotoxinas (tricotecenos, aflatoxinas y ocratoxinas) podr&iacute;an ser extra&iacute;das e identificadas en tejido humano y l&iacute;quidos corporales de pacientes expuestos a la toxina que produce moldes en su entorno. Orina humana y los tejidos extra&iacute;do de metanol y esputo fueron examinados. Tricotecenos fueron analizadas usando t&eacute;cnicas de ELISA competitivas. Aflatoxinas B1, B2, G1 y G2 y ocratoxina A se analizaron mediante fluorometr&iacute;a y columnas de inmunoafinidad. Prueba de sensibilidad y especificidad se determinaron. Niveles de detecci&oacute;n para las diferentes micotoxinas variaron de 0.2 ppb de tricotecenos, 1,0 ppb de aflatoxinas y 2,0 ppb para ocratoxinas. Niveles de tricotecenos variaban en orina, esputo y tejidos biopsias (cerebro, h&iacute;gado, del pulm&oacute;n,) de indetectable (&lt; 0.2 ppb) a niveles de hasta 18 niveles de ppb. de Aflatoxinas de los mismos tipos de tejidos que vari&oacute; de 1.0 a 5.0 ppb. Ocratoxinas aislado en el mismo tipo de tejidos variada de 2,0 ppb a &gt; 10.0 ppb. Control negativo pacientes no ten&iacute;an detectables micotoxinas en sus tejidos o fluidos. Estos datos demuestran que las micotoxinas pueden detectarse en flu&iacute;dos corporales y tejidos humanos de pacientes expuestos a micotoxinas producir moldes en el medio ambiente y demuestran que los tejidos humanos o fluidos son los m&aacute;s propensos a producir resultados positivos.

Drywall from China has been reported to release sulfur producing products which are corrosive to metals, result in noxious odors, and represent a significant health risk. It has been reported that these emissions produce medical symptoms such as respiratory or asthma type problems, sinusitis, gastrointestinal disorders, and vision problems in home owners and their household pets. We report here a method of identifying a causative agent for these emissions by sampling affected gypsum wallboard and subjecting those samples to Real Time Polymerase Chain Reaction [RT-PCR] studies. Specific DNA probes and primers have been designed and patented that detect a specific iron and sulfur reducing bacterium (i.e., Thiobacillus ferrooxidans). One hundred percent of affected drywall samples obtained from homes located in the southeastern United States tested positive for the presence of T. ferrooxidans. All negative controls consisting of unaffected wallboard and internal controls, Geotrichum sp., tested negative within our limits of detection.

In this study we pursued a diagnostic target in Aspergillus fumigatus (AF) by using qualitative Realtime PCR combined with proprietary DNA primers and a hydrolysis probe specific for this fungal target. Qualitative Realtime PCR is a diagnostic tool that utilizes Realtime PCR technology and detects the presence or absence target specific DNA within a predetermined detection range. Respiratory tissue and fluids from experimentally infected guinea pigs were tested by extracting DNA from the samples which were amplified and detected using AF specific DNA primers and probe. This study included qualitative evaluations of all specimens for the presence of the DNA of AF. The findings in the tissues after AF infection were compared to the numbers of spores in aerosolized samples used to inoculate the animals. Results demonstrated that the specific probe and primer set could detect the presence or absence of AF DNA in the sample. The qualitative detection limit of the assay ranged from 6 Ã— 10(4) copies to 6 copies. Since blood cultures are rarely positive for Aspergillosis, our data indicate that qualitative Realtime PCR, in combination with the appropriate DNA primers and probe can serve as an effective diagnostic tool in the early detection of fungal infections.

Assessment of Aspergillus fumigatus in Guinea Pig Bronchoalveolar Lavages and Pulmonary Tissue by Culture and Realtime Polymerase Chain Reaction Studies.

Dans cette &eacute;tude, nous avons poursuivi une cible diagnostique dans Aspergillus fumigatus (AF) &agrave; l&#39;aide qualitative que Realtime PCR combin&eacute; avec des amorces ADN exclusifs et une sonde d&#39;hydrolyse sp&eacute;cifique pour cette cible fongique. Qualitative Realtime PCR est un outil de diagnostic qui utilise la technologie de Realtime PCR et d&eacute;tecte la pr&eacute;sence ou l&#39;absence cible ADN sp&eacute;cifique dans une zone de d&eacute;tection pr&eacute;d&eacute;termin&eacute;. Tissu respiratoire et fluides de cobayes exp&eacute;rimentalement infect&eacute;s ont &eacute;t&eacute; test&eacute;s en extrayant l&#39;ADN de l&#39;&eacute;chantillon qui ont &eacute;t&eacute; amplifi&eacute;es et d&eacute;tect&eacute; &agrave; l&#39;aide de la sonde et amorces ADN sp&eacute;cifiques de AF. Cette &eacute;tude comprend des &eacute;valuations qualitatives de tous les sp&eacute;cimens de la pr&eacute;sence de l&#39;ADN de l&#39;AF. In questo studio abbiamo perseguito un obiettivo diagnostico in Aspergillus fumigatus (AF) utilizzando qualitativa che realtime PCR combinato con il proprietario del DNA primer e una sonda di idrolisi specifica per questa destinazione fungina. Qualitativa Realtime PCR &egrave; uno strumento diagnostico che utilizza la tecnologia Realtime PCR e rileva la presenza o l&#39;assenza DNA bersaglio specifico all&#39;interno di un intervallo di rilevamento predeterminato. Tessuto delle vie respiratorie e fluidi da cavie infettati sperimentalmente sono stati testati con l&#39;estrazione del DNA dai campioni che sono stati amplificati e rilevati tramite sonda primer specifici di DNA AF. Questo studio ha incluso valutazioni qualitative di tutti i campioni per la presenza del DNA di AF. I risultati nei tessuti dopo infezione AF sono stati confrontati con i numeri delle spore in campioni aerosolized utilizzate per inoculare gli animali. En este estudio que hemos perseguido un objetivo diagn&oacute;stico en Aspergillus fumigatus (AF) utilizando cualitativa que Realtime PCR combinado con propiedad cartillas de ADN y una hidr&oacute;lisis sondeo espec&iacute;fico para este objetivo de hongo. Cualitativa Realtime PCR es una herramienta de diagn&oacute;stico que utiliza la tecnolog&iacute;a de PCR Realtime y detecta la presencia o ausencia ADN espec&iacute;fico de destino dentro de un rango de detecci&oacute;n predeterminado. Tejido respiratorio y fluidos de infectados experimentalmente de conejillos de Indias fueron probados mediante la extracci&oacute;n de ADN de las muestras que fueron amplificadas y detectan usando la sonda e iniciadores espec&iacute;ficos de ADN de AF. Este estudio incluy&oacute; evaluaciones cualitativas de todas las muestras para detectar la presencia de la DNA de AF. Los resultados en los tejidos despu&eacute;s de la infecci&oacute;n de la AF se compararon con los n&uacute;meros de esporas en aerosol muestras utilizadas para inocular a los animales. Los resultados demostraron que el conjunto espec&iacute;fico de la sonda y cartilla pudo detectar la presencia o ausencia de ADN de AF en la muestra. El l&iacute;mite de detecci&oacute;n cualitativa del ensayo oscilaron entre 6 &times; 4 10 copias y 6 copias. Ya que los hemocultivos son raramente positivos para aspergilosis, nuestros datos indican que cualitativa Realtime PCR, en combinaci&oacute;n con las cartillas de ADN y sonda apropiado puede servir como una herramienta de diagn&oacute;stico eficaz en la detecci&oacute;n precoz de las infecciones f&uacute;ngicas.

Evaluaci&oacute;n de Aspergillus fumigatus en lavados broncoalveolares conejillo de Indias y el tejido pulmonar por cultura y estudios de la reacci&oacute;n en cadena de polimerasa en tiempo real.
