Fat is an atypical cadherin that controls both cell growth and planar polarity. Atrophin is a nuclear co-repressor that is also essential for planar polarity; however, it is not known what genes Atrophin controls in planar polarity, or how Atrophin activity is regulated during the establishment of planar polarity. We show that Atrophin binds to the cytoplasmic domain of Fat and that Atrophin mutants show strong genetic interactions with fat. We find that both Atrophin and fat clones in the eye have non-autonomous disruptions in planar polarity that are restricted to the polar border of clones and that there is rescue of planar polarity defects on the equatorial border of these clones. Both fat and Atrophin are required to control four-jointed expression. In addition our mosaic analysis demonstrates an enhanced requirement for Atrophin in the R3 photoreceptor. These data lead us to a model in which fat and Atrophin act twice in the determination of planar polarity in the eye: first in setting up positional information through the production of a planar polarity diffusible signal, and later in R3 fate determination.

En outre, notre analyse d&#xE9;montre une exigence de la mosa&#xEF;que am&#xE9;lior&#xE9; pour Atrophin dans le photor&#xE9;cepteur R3. Recently, the epidermal growth factor receptor (EGFR) pathway has been implicated in determining ommatidial rotation. We have isolated a new allele of echinus, a gene known to control the patterning and number of interommatidial cells. We show that echinus displays defects in the rotation of ommatidia that are not evident until mid-pupal stages, and we propose that echinus action is that of opposing EGFR by an unknown mechanism and that this can explain both its influence in ommatidial rotation and lattice programmed cell death (PCD).

La orientaci&#xF3;n precisa de los omatidios en el ojo de Drosophila se logra a trav&#xE9;s de un proceso especializado de la migraci&#xF3;n celular que tiene lugar en el disco de los ojos de tercer estadio imaginal cuando grupos ommatidial rotar 90 grados. Este proceso est&#xE1; estrictamente coordinado con el establecimiento de la polaridad celular plana (PCP), pero se basa en un conjunto espec&#xED;fico de genes que controlan su mecanismo de se&#xF1;alizaci&#xF3;n independientemente de PCP. Recientemente, el receptor de factor de crecimiento epid&#xE9;rmico (EGFR) ha sido implicado en la determinaci&#xF3;n de la rotaci&#xF3;n ommatidial. ISWI is an evolutionarily conserved ATP-dependent chromatin remodeling factor playing central roles in DNA replication, RNA transcription, and chromosome organization. The variety of biological functions dependent on ISWI suggests that its activity could be highly regulated. Our group has previously isolated and characterized new cellular activities that positively regulate ISWI in Drosophila melanogaster. To identify factors that antagonize ISWI activity we developed a novel in vivo eye-based assay to screen for genetic suppressors of ISWI. Our screen revealed that ISWI interacts with an evolutionarily conserved network of cellular and nuclear factors that escaped previous genetic and biochemical analyses.

These flies display severe lysosomal dysfunction caused by the Atrophin protein, as a distinctive feature that modulates polyglutamine toxicity. The autophagic flux is blocked after autophagosome-lysosome fusion, leading to clearance defects, and further induction of autophagy does not rescue neurodegeneration but is instead detrimental. This highlights the importance for the full cycle to be completed to achieve neuroprotection by autophagy, much like waste recycling, more than disposal, benefits society.

Large alterations in transcription accompany neurodegeneration in polyglutamine (polyQ) diseases. These pathologies manifest both general polyQ toxicity and mutant protein-specific effects. In this study, we report that the fat tumour suppressor gene mediates neurodegeneration induced by the polyQ protein Atrophin. We have monitored early transcriptional alterations in a Drosophila model of Dentatorubral-pallidoluysian Atrophy and found that polyQ Atrophins downregulate fat. Fat protects from neurodegeneration and Atrophin toxicity through the Hippo kinase cascade. Fat/Hippo signalling does not provoke neurodegeneration by stimulating overgrowth; rather, it alters the autophagic flux in photoreceptor neurons, thereby affecting cell homeostasis. Our data thus provide a crucial insight into the specific mechanism of a polyQ disease and reveal an unexpected neuroprotective role of the Fat/Hippo pathway.

Nous avons suivi d&#xE9;but des alt&#xE9;rations transcriptionnelles dans un mod&#xE8;le de drosophile de Dentatorubral-pallido atrophie et a constat&#xE9; que Atrophins polyQ r&#xE9;guler n&#xE9;gativement la graisse. Fat prot&#xE8;ge de la toxicit&#xE9; et la neurod&#xE9;g&#xE9;n&#xE9;rescence Atrophin &#xE0; travers la cascade Hippo kinase. Fat / Hippo signalisation ne provoque pas la neurod&#xE9;g&#xE9;n&#xE9;rescence par la prolif&#xE9;ration stimulant, mais plut&#xF4;t, il modifie le flux autophagique dans les neurones photor&#xE9;cepteurs, ce qui affecte l&#x26;#39;hom&#xE9;ostasie cellulaire. In dieser Studie berichten wir, dass die Fett-Tumor-Suppressor-Gens Neurodegeneration induziert durch das PolyQ-Protein Atrophin vermittelt. Wir haben fr&uuml;he transkriptionelle &Auml;nderungen in einem Drosophila-Modell der dentato-rubro-palidal &uuml;berwacht und gefunden, dass PolyQ Atrophins gulieren Fett. Fett sch&uuml;tzt vor Neurodegeneration und Atrophin Toxizit&auml;t durch die Hippo-Kinase-Kaskade. Fett/Hippo Signalisierung nicht provozieren zu Neurodegeneration durch Stimulierung &Uuml;berwuchterung; vielmehr &auml;ndert sie die autophagic Flux in Photorezeptoren Neuronen, und damit die Hom&ouml;ostase der Zelle. Grandi alterazioni nella trascrizione accompagnano Neurodegenerazione nelle malattie delle poliglutamine (polyQ). Queste patologie si manifestano sia generale polyQ tossicit&agrave; e gli effetti specifici della proteina mutanti. In questo studio, si segnala che il gene soppressore tumorale grasso da intermediario neurodegenerazione indotta dalla proteina polyQ Atrophin. Abbiamo monitorato le prime alterazioni trascrizionali in un modello di Drosophila di atrofia Dentatorubral-rubro e trovato quello polyQ Atrophins sottoregola grasso. Grasso protegge dalla tossicit&agrave; Atrophin attraverso la cascata di chinasi Hippo e neurodegenerazione. Grasso/Hippo segnalazione non provocano neurodegenerazione da stimolare la crescita eccessiva; piuttosto, altera il flusso autophagic in neuroni fotorecettore, influenzando di conseguenza l&#39;omeostasi cellulare. I nostri dati cos&igrave; forniscono una comprensione fondamentale il meccanismo specifico di una malattia polyQ e rivelano un ruolo inaspettato neuroprotective del percorso grasso/Hippo.

Polyglutamine diseases are accompanied by large-scale alterations of transcription that may be responsible for neurodegeneration. We have monitored early transcriptional alterations in a Drosophila model for dentatorubral-pallidoluysian atrophy (DRPLA) and reported the critical downregulation of the fat tumor suppressor gene. We show that, besides partially mediating neurodegeneration in the Drosophila model for DRPLA, fat and the downstream hippo pathway are essential for adult neuronal homeostasis. This function of the Fat/Hippo pathway is independent of the well-established actions on proliferation and cell polarity; rather it has an impact upon autophagy, which is blocked and ineffective in fat/hippo mutants. These findings reveal the unexpected role in postmitotic neuronal homeostasis of a tumor suppressor pathway and hint at a new player involved directly or indirectly in the regulation of autophagy.

Enfermedades poliglutaminas se acompa&#xF1;an de alteraciones a gran escala de transcripci&#xF3;n que pueden ser responsables de la neurodegeneraci&#xF3;n. Hemos monitoreado primeras alteraciones de la transcripci&#xF3;n en un modelo de Drosophila para Dentatorubral-pallidoluysian atrofia (DRPLA) e inform&#xF3; de la regulaci&#xF3;n a la baja cr&#xED;tica del gen supresor de tumores de grasa. Se demuestra que, adem&#xE1;s de mediar parcialmente la neurodegeneraci&#xF3;n en el modelo de Drosophila para DRPLA, la grasa y la posterior v&#xED;a hipop&#xF3;tamos son esenciales para la homeostasis neuronal adulta. Esta funci&#xF3;n de la v&#xED;a de Grasa / Hipona es independiente de las acciones bien establecidas en la proliferaci&#xF3;n y la polaridad celular, sino que tiene un impacto sobre la autofagia, que est&#xE1; bloqueado e ineficaz en grasa / hipop&#xF3;tamos mutantes. Estos resultados ponen de manifiesto el papel inesperado en la homeostasis postmitotic neuronal de una v&#xED;a supresora de tumores y apuntan a un nuevo jugador que participan directa o indirectamente en la regulaci&#xF3;n de la autofagia.
