We report the synthesis, spectroscopic properties and intracellular imaging of recombinant antibody single chain fragment (scFv) conjugates with photosensitizers used for photodynamic therapy of cancer (PDT). Two widely-studied photosensitizers have been selected: preclinical pyropheophorbide-a (PPa) and verteporfin (VP), which has been clinically approved for the treatment of acute macular degeneration (Visudyne). Pyropheophorbide-a and verteporfin have been conjugated to an anti-HER2 scFv containing on average ten photosensitizer molecules per scFv with a small contribution (<or=20%) from non-covalently bound molecules. Confocal fluorescence microscopy demonstrates good cellular uptake of PPa conjugate with the HER2-positive cell line, SKOV-3, while negligible cell uptake is demonstrated for the HER2-negative cell line, KB. For the VP conjugate, increased rate of cellular uptake and prolonged retention in SKOV-3 cells is observed compared to free photosensitizer. In clinical applications this could provide increased potency and desired selectivity towards malignant tissue, leaving surrounding healthy tissue unharmed and reducing skin photosensitivity. The present study highlights the usefulness of photosensitizer immunoconjugates with scFvs for targeted PDT.

Sono stati selezionati due ampiamente studiato fotosensibilizzanti: studi preclinici pyropheophorbide-a (PPa) e verteporfin (VP), che &egrave; stato clinicamente approvato per il trattamento della degenerazione maculare acuta (Visudyne). Pyropheophorbide-a e verteporfin hanno stato coniugato a un scFv anti-HER2 contenente mediamente dieci molecole fotosensibilizzatore per scFv con una piccola donazione (&lt; o = 20%) da molecole non covalentemente. Microscopia di fluorescenza confocale dimostra buon assorbimento cellulare del coniugato PPa con la linea cellulare HER2-positivo, SKOV-3, mentre &egrave; dimostrato uptake cellulare trascurabile per la linea cellulare HER2-negativo, KB. Se presenta la s&#xED;ntesis, propiedades espectrosc&#xF3;picas y de imagen intracelular de fragmento de anticuerpo recombinante de cadena &#xFA;nica (scFv) se conjuga con fotosensibilizadores utilizados para la terapia fotodin&#xE1;mica del c&#xE1;ncer (PDT). Dos fotosensibilizadores ampliamente estudiados han sido seleccionados: precl&#xED;nica pyropheophorbide-a (PPA) y la verteporfina (VP), que ha sido cl&#xED;nicamente aprobado para el tratamiento de la degeneraci&#xF3;n macular aguda (Visudyne). Pyropheophorbide-a y con verteporfina se han conjugado con un scFv anti-HER2 que contiene un promedio de diez mol&#xE9;culas de fotosensibilizador por scFv con una peque&#xF1;a contribuci&#xF3;n ( &#x3C;or=20%) from non-covalently bound molecules. Confocal fluorescence microscopy demonstrates good cellular uptake of PPa conjugate with the HER2-positive cell line, SKOV-3, while negligible cell uptake is demonstrated for the HER2-negative cell line, KB. For the VP conjugate, increased rate of cellular uptake and prolonged retention in SKOV-3 cells is observed compared to free photosensitizer. In clinical applications this could provide increased potency and desired selectivity towards malignant tissue, leaving surrounding healthy tissue unharmed and reducing skin photosensitivity. The present study highlights the usefulness of photosensitizer immunoconjugates with scFvs for targeted PDT.

Current photodynamic therapy (PDT) of cancer is limited by inefficiencies involved in specifically targeting photosensitizers to tumors. Although antibodies are being explored as targeting vehicles, they present significant challenges, particularly in terms of pharmacokinetics and drug-coupling. We describe here a novel and effective system to covalently attach multiple photosensitizer molecules (both preclinical, pyropheophorbide-a and clinically approved, verteporfin photosensitizers) to single-chain Fvs. Further, we demonstrate that not only do the resulting photoimmunoconjugates retain photophysical functionality, they are more potent than either free photosensitizer, effectively killing tumor cells in vitro and in vivo. For example, treatment of human breast cancer xenografts with a photoimmunoconjugate comprising an anti-HER-2 scFv linked to 8-10 molecules of pyropheophorbide-a leads to significant tumor regression. These results give an insight into the important features that make scFvs good carriers for PDT drugs and provide proof of concept of our unique approach to targeted photodynamic therapy (tPDT). This promises to significantly improve on current photodynamic therapies for the treatment of cancer.

We propose a sensitive and versatile method for measuring the local microviscosity in biological systems, based on the determination of the fluorescence lifetime of 1. Fluorescence lifetime imaging (FLIM) performed on live cells incubated with 1 demonstrates the distinct intracellular lifetime of the molecular rotor of 1.6 +/- 0.2 ns corresponding to the intracellular viscosity of ca. 140 cP. Time-resolved fluorescence anisotropy of 1 in cells confirms insignificant binding of the fluorophore. The viscosity value obtained in the present study is considerably higher than that of water and of cellular cytoplasm. The high viscosity of intracellular compartments is likely to play an important role in vital intracellular processes, including the rate of diffusion of reactive oxygen species, causing programmed cell destruction.

Singlet molecular oxygen, O(2)(a(1)Delta(g)), can be created in a single cell from ground-state oxygen, O(2)(X(3)Sigma(g)(-)), upon focused laser irradiation of an intracellular sensitizer. This cytotoxic species can subsequently be detected by its 1270 nm phosphorescence (a(1)Delta(g) --> X(3)Sigma(g)(-)) with subcellular spatial resolution. The singlet oxygen lifetime determines its diffusion distance and hence the intracellular volume element in which singlet-oxygen-initiated perturbation of the cell occurs. In this study, the time-resolved phosphorescence of singlet oxygen produced by the sensitizers chlorin (Chl) and 5,10,15,20-tetrakis(N-methyl-4-pyridyl)-21H,23H-porphine (TMPyP) was monitored. These molecules localize in different domains of a living cell. The data indicate that (i) the singlet oxygen lifetime and (ii) the rate constant for singlet oxygen quenching by added NaN(3) depend on whether Chl or TMPyP was the photosensitizer. These observations likely reflect differences in the chemical and physical constituency of a given subcellular domain (e.g., spatially dependent oxygen and NaN(3) diffusion coefficients), thereby providing evidence that singlet oxygen responds to the inherent heterogeneity of a cell. Thus, despite a relatively long intracellular lifetime, singlet oxygen does not diffuse a great distance from its site of production. This is a consequence of an apparent intracellular viscosity that is comparatively large.

In dieser Studie die Zeit aufgel&ouml;st Phosphoreszenz von Singulett-Sauerstoff durch die Sensibilisatoren Chlorin (Chl) und 5,10,15,20-tetrakis(N-methyl-4-pyridyl) - 21 H, 23 H-Porphinliganden (TMPyP) wurde &uuml;berwacht. Diese Molek&uuml;le, die in verschiedenen Dom&auml;nen einer lebenden Zelle lokalisiert werden. The sol-to-gel transition was monitored via the use of time-resolved recording of the fluorescence emission of viscosity-sensitive probes. Two dyes were chosen for the study, water-soluble DASPMI and a hydrophobic BODIPY, and steady-state, time-resolved and time-tagged fluorescence measurements were performed. These techniques, coupled with the probes different solubility, allowed complementary fluorescence lifetime and intensity data to be obtained from the dyes introduced into the matrix-forming mixture to produce sol-gel derived monoliths. Two different precursors were used as examples. A hydrogel was formed from a commercially available gellan gum (Gelrite), and a glass-like monolith was formed using tetraethyl orthosilicate. Changes in fluorescence lifetime could be related to those in the local viscosity sensed by the probe. The combination of this type of probe with time-resolved measurements is extremely useful in monitoring the microscopic changes that occur during the sol-to-gel transition within this important class of materials.

Photodynamic Therapy (PDT) is a minimally invasive procedure used for treating a range of neoplastic diseases, which utilises combined action of light and a PDT drug called a photosensitiser. The efficiency of this treatment depends crucially on the properties of the photosensitiser used, namely on its efficient uptake by cells or by the surrounding vasculature, intracellular localisation, minimal dark toxicity and substantial phototoxicity. In this report we compare the spectroscopic properties, cell uptake and in vitro phototoxicity of two novel hydrophilic photosensitisers derived from pyropheophorbide-a (PPa). Both new photosensitisers have the potential to form bioconjugates with antibody fragments for targeted PDT. We find that the photophysical properties of both new photosensitisers are favourable compared to the parent PPa, including enhanced absorption in the red spectral region and substantial singlet oxygen quantum yields. Both molecules show efficient cellular uptake, but display a different intracellular localisation. Both new photosensitisers exhibit no significant dark-toxicity at concentrations of up to 100 microM. The phototoxicity of the two photosensitisers is strikingly different, with one derivative being 13 times more efficient than the parent PPa and another derivative being 18 times less efficient in SKOV3 ovarian cancer cells. We investigate the reasons behind such drastic differences in phototoxicity using confocal fluorescence microscopy and conclude that intracellular localisation is a crucial factor in the photodynamic efficiency of pheophorbide derivatives. These studies highlight the underlying factors behind creating more potent photosensitisers through synthetic manipulation.

Novel photosensitisers derived from pyropheophorbide-a: uptake by cells and photodynamic efficiency in vitro.

La th&#xE9;rapie photodynamique (PDT) est une proc&#xE9;dure mini-invasive utilis&#xE9;e pour traiter un &#xE9;ventail de maladies n&#xE9;oplasiques, qui utilise l&#x26;#39;action combin&#xE9;e de la lumi&#xE8;re et un m&#xE9;dicament appel&#xE9; HAP un photosensibilisateur. L&#x26;#39;efficacit&#xE9; de ce traitement d&#xE9;pend essentiellement des propri&#xE9;t&#xE9;s de l&#x26;#39;agent photosensibilisant utilis&#xE9;, &#xE0; savoir sur son absorption par les cellules efficace ou par le syst&#xE8;me vasculaire entourant, la localisation intracellulaire, la toxicit&#xE9; minimale sombre et phototoxicit&#xE9; substantielle. Dans ce rapport, nous comparons les propri&#xE9;t&#xE9;s spectroscopiques, l&#x26;#39;absorption cellulaire et de phototoxicit&#xE9; in vitro de deux nouveaux photosensibilisateurs hydrophiles d&#xE9;riv&#xE9;s de pyropheophorbide-un (PPA). Les deux nouveaux photosensibilisateurs ont le potentiel pour former des fragments d&#x26;#39;anticorps avec bioconjugu&#xE9;s pour cible HAP. Nous constatons que les propri&#xE9;t&#xE9;s photophysiques des deux photosensibilisateurs nouvelles sont favorables par rapport &#xE0; la soci&#xE9;t&#xE9; m&#xE8;re, y compris l&#x26;#39;absorption PPa renforc&#xE9;e dans la r&#xE9;gion rouge du spectre et substantielles oxyg&#xE8;ne singulet rendements quantiques. Die Wirksamkeit dieser Behandlung h&auml;ngt entscheidend von den Eigenschaften der Photosensitiser verwendet, n&auml;mlich auf die effiziente Aufnahme von Zellen oder die umliegenden Gef&auml;&szlig;system, intrazellul&auml;re Lokalisation, minimal dunkel Toxizit&auml;t und erhebliche Phototoxizit&auml;t. Wir untersuchen die Gr&uuml;nde f&uuml;r solche drastische Unterschiede bei Phototoxizit&auml;t mit konfokale Fluoreszenzmikroskopie und feststellen, dass die intrazellul&auml;re Lokalisation ist entscheidend f&uuml;r die photodynamische Effizienz von Pheophorbide Derivaten. Diese Studien unterstreichen die zugrunde liegenden Faktoren hinter erstellen potenter Photosensitisers durch synthetische Manipulation.

L&#39;efficacia di questo trattamento dipende fondamentalmente dalle propriet&agrave; di photosensitiser utilizzati, vale a dire sul suo assorbimento efficiente di cellule o dal sistema vascolare circostante, localizzazione intracellulare, tossicit&agrave; minima scura e fototossicit&agrave; sostanziale. La terapia fotodin&#xE1;mica (TFD) es un procedimiento m&#xED;nimamente invasivo que se utiliza para el tratamiento de una amplia gama de enfermedades neopl&#xE1;sicas, que utiliza la acci&#xF3;n combinada de luz y un medicamento llamado PDT fotosensibilizador. La eficacia de este tratamiento depende fundamentalmente de las propiedades del fotosensibilizador utilizado, es decir, sobre su absorci&#xF3;n eficiente por las c&#xE9;lulas o por la vasculatura circundante, localizaci&#xF3;n intracelular, la toxicidad oscuro m&#xED;nima y fototoxicidad sustancial. We present polarization-resolved fluorescence measurements of fluorescent molecular rotors 9-(2-carboxy-2-cyanovinyl)julolidine (CCVJ), 9-(2,2-dicyanovinyl)julolidine (DCVJ), and a meso-substituted boron dipyrromethene (BODIPY-C(12)). The photophysical properties of these molecules are highly dependent on the viscosity of the surrounding solvent. The relationship between their quantum yields and the viscosity of the surrounding medium is given by an equation first described and presented by FÃ¶rster and Hoffmann and can be used to determine the microviscosity of the environment around a fluorophore. Herein we evaluate the applicability of molecular rotors as probes of apparent viscosity on a microscopic scale based on their viscosity dependent fluorescence depolarization. We develop a theoretical framework, combining the FÃ¶rster-Hoffmann equation with the Perrin equation and compare the dynamic ranges and usable working regimes for these dyes in terms of utilising fluorescence anisotropy as a measure of viscosity. We present polarization-resolved fluorescence spectra and steady-state fluorescence anisotropy imaging data for measurements of intracellular viscosity. We find that the dynamic range for fluorescence anisotropy for CCVJ and DCVJ is significantly lower than that of BODIPY-C(12) in the viscosity range 0.6<Î·<600 cP. Moreover, using steady-state anisotropy measurements to probe microviscosity in the low (<3 cP) viscosity regime, the molecular rotors can offer a better dynamic range in anisotropy compared with a rigid dye as a probe of microviscosity, and a higher total working dynamic range in terms of viscosity.

&#x5206;&#x5B50;&#x30ED;&#x30FC;&#x30BF;&#x30FC;&#x306E;&#x86CD;&#x5149;&#x7570;&#x65B9;&#x6027;

Se presenta la polarizaci&#xF3;n resuelto medidas de fluorescencia de rotores moleculares fluorescentes 9 - (2-carboxi-2-cianovinil) julolidina (CCVJ), 9 - (2,2-dicyanovinyl) julolidina (DCVJ), y un meso-boro sustituido dipyrromethene (BODIPY -C (12)). Las propiedades fotof&#xED;sicas de estas mol&#xE9;culas son altamente dependientes de la viscosidad del disolvente circundante. La relaci&#xF3;n entre sus rendimientos cu&#xE1;nticos y la viscosidad del medio circundante est&#xE1; dada por una ecuaci&#xF3;n descrita por primera vez y presentada por F&#xF6;rster y Hoffmann y se puede utilizar para determinar el microviscosidad del medio ambiente en torno a un fluor&#xF3;foro. Aqu&#xED; se eval&#xFA;a la aplicabilidad de los rotores moleculares como sondas de viscosidad aparente a una escala microsc&#xF3;pica en base a su viscosidad fluorescencia despolarizaci&#xF3;n dependiente. Se desarrolla un marco te&#xF3;rico, combinando la ecuaci&#xF3;n F&#xF6;rster-Hoffmann con la ecuaci&#xF3;n Perrin y comparar los rangos din&#xE1;micos y utilizables reg&#xED;menes de trabajo para estos colorantes en t&#xE9;rminos de utilizaci&#xF3;n de anisotrop&#xED;a de fluorescencia como una medida de la viscosidad. Se presenta la polarizaci&#xF3;n resolver espectros de fluorescencia y en estado constante de datos de im&#xE1;genes de fluorescencia de anisotrop&#xED;a para la medici&#xF3;n de la viscosidad intracelular. Se encuentra que el rango din&#xE1;mico de la anisotrop&#xED;a de fluorescencia para CCVJ y DCVJ es significativamente menor que la de BODIPY-C (12) en el rango de viscosidad 0,6 &#x26;lt;&#x3B7; &#x26;lt;600 cP. Por otra parte, utilizando las mediciones de estado estable de anisotrop&#xED;a para investigar microviscosidad en el r&#xE9;gimen de baja viscosidad (&#x26;lt;3 CP), los rotores moleculares pueden ofrecer un mejor rango din&#xE1;mico de la anisotrop&#xED;a en comparaci&#xF3;n con un tinte r&#xED;gida como una sonda de microviscosidad, y un total m&#xE1;s alto de trabajo din&#xE1;mico rango en t&#xE9;rminos de viscosidad.
