Many developing tissues require programmed cell death (PCD) for proper formation. In mice and C. elegans, developmental PCD is regulated by the Bcl-2 family of proteins. Two bcl-2 genes are encoded in the Drosophila genome (debcl/dBorg1/Drob-1/dBok and buffy/dBorg2) and previous RNAi-based studies suggested a requirement for these in embryonic development. However, we report here that, despite the fact that many tissues in fruit flies are shaped by PCD, deletion of the bcl-2 genes does not perturb normal development. We investigated whether the fly bcl-2 genes regulate non-apoptotic processes that require caspases, but found these to be bcl-2 gene-independent. However, irradiation of the mutants demonstrates that DNA damage-induced apoptosis, mediated by Reaper, is blocked by buffy and that debcl is required to inhibit buffy. Our results demonstrate that developmental PCD regulation in the fly does not rely upon the Bcl-2 proteins, but that they provide an added layer of protection in the apoptotic response to stress.

Drosophila Bcl-2 proteins participate in stress-induced apoptosis, but are not required for normal development.

Beaucoup de tissus en d&#xE9;veloppement ont besoin de mort cellulaire programm&#xE9;e (PCD) pour la formation appropri&#xE9;e. Chez la souris et C. elegans, le d&#xE9;veloppement PCD est r&#xE9;gie par la famille Bcl-2 des prot&#xE9;ines. Cependant, l&#x26;#39;irradiation des mutants montre que l&#x26;#39;ADN des dommages-apoptose induite, m&#xE9;di&#xE9;e par Reaper, est bloqu&#xE9; par buffy et que debcl est requise pour inhiber la leuco-plaquettaire. Vielen Entwicklungsl&auml;ndern Geweben erfordern f&uuml;r ordnungsgem&auml;&szlig;e Bildung programmierten Zelltod (PCD). Entwicklungsbiologie PCD wird in M&auml;usen und C. Elegans von der Familie Bcl-2 Proteine geregelt. Zwei Gene der Bcl-2 sind in der Taufliege Genom (Debcl/dBorg1/Drob-1/dBok und buffy/dBorg2) codiert und fr&uuml;here RNAi-basierten Studien vorgeschlagen eine Voraussetzung f&uuml;r diese in der embryonalen Entwicklung. Hier berichten wir jedoch, dass trotz der Tatsache, die vielen Geweben in Fruchtfliegen von PCD gepr&auml;gt sind, L&ouml;schung der Bcl-2-Gene das von der normalen Entwicklung nicht verl&auml;ngern. Wir wurde untersucht, ob die fliegen Bcl-2-Gene nicht apoptotische Prozesse regulieren, die Caspases, aber fand diese als Bcl-2 gen-unabh&auml;ngig. Muchos tejidos en desarrollo necesitan muerte celular programada (PCD) para la formaci&#xF3;n adecuada. En los ratones y los C. elegans, el desarrollo de PCD es regulada por la familia Bcl-2 de prote&#xED;nas. Dos Bcl-2 genes est&#xE1;n codificados en el genoma de Drosophila (debcl/dBorg1/Drob-1/dBok y buffy/dBorg2) y los estudios previos basados &#x200B;&#x200B;en RNAi sugiri&#xF3; un requisito para &#xE9;stas en el desarrollo embrionario. Sin embargo, aqu&#xED; nos informe que, a pesar del hecho de que en muchos tejidos en moscas de la fruta est&#xE1;n conformadas por PCD, la supresi&#xF3;n de los genes bcl-2 no perturbar el desarrollo normal. Hemos investigado si la mosca de Bcl-2 genes regulan la no-apopt&#xF3;ticos procesos que requieren las caspasas, pero se encontr&#xF3; que &#xE9;stos sean gen bcl-2-independiente. Sin embargo, la irradiaci&#xF3;n de los mutantes demuestra que da&#xF1;an el ADN inducido por la apoptosis, mediada por segador, est&#xE1; bloqueada por leucocitaria y que debcl se requiere para inhibir leucocitaria. Nuestros resultados demuestran que la regulaci&#xF3;n del desarrollo de PCD en la mosca no se basa en las prote&#xED;nas Bcl-2, pero que proporcionan una capa adicional de protecci&#xF3;n en la apoptosis respuesta a estr&#xE9;s.
