The characterization of virulence determinants of pathogenic agents is of utmost relevance for the design of disease control strategies. So far, two classes of virulence determinants have been characterized for viral populations: those imprinted in the nucleotide sequence of some specific genomic regions and those that depend on the complexity of the viral population as such. Here we provide evidence of a virulence determinant that depends neither on a genomic sequence nor on detectable differences in population complexity. Foot-and-mouth disease virus is lethal for C57BL/6 mice showing the highest viral load in pancreas. Virus isolated from pancreas after one passage in mice showed an attenuated phenotype, with no lethality even at the highest dose tested. By contrast, virus from sera of the same mice displayed a virulence similar to that of the parental wild-type clone and virus isolated from spleen displayed an intermediate phenotype. However, viral populations from pancreas, spleen, and serum showed indistinguishable consensus genomic nucleotide sequences and mutant spectrum complexities, as quantified according to the mutation frequencies of both entire genomic nucleotide sequences of biological clones. The results show that the populations with differing virulences cannot be distinguished either by the consensus sequence or by the average complexity of the mutant spectrum. Differential harvesting of virus generated by cell transfection of RNA from serum and pancreas failed to reveal genetic differences between subpopulations endowed with differing virulences. In addition to providing evidence of hidden virulence determinants, this study underlines the capacity of a clone of an RNA virus to rapidly diversify phenotypically in vivo.

La caract&eacute;risation des d&eacute;terminants de la virulence des agents pathog&egrave;nes est de la plus haute importance pour la conception de strat&eacute;gies de contr&ocirc;le des maladies. Jusqu&#39;ici, les deux classes de d&eacute;terminants de virulence ont &eacute;t&eacute; caract&eacute;ris&eacute;s pour populations virales : celles imprim&eacute;es dans la s&eacute;quence nucl&eacute;otidique de certaines r&eacute;gions g&eacute;nomiques sp&eacute;cifiques et ceux qui d&eacute;pendent de la complexit&eacute; de la population virale comme tel. Nous fournissons ici preuve d&#39;un d&eacute;terminant de la virulence qui repose sur une s&eacute;quence g&eacute;nomique, ni sur la diff&eacute;rence d&eacute;celable dans la complexit&eacute; de la population. Virus de la fi&egrave;vre aphteuse et est l&eacute;tal pour la souris C57BL/6 affichage de la charge virale la plus &eacute;lev&eacute;e dans le pancr&eacute;as. Virus isol&eacute; &agrave; partir du pancr&eacute;as apr&egrave;s qu&#39;un passage chez la souris ont montr&eacute; un ph&eacute;notype att&eacute;nu&eacute;, avec aucune l&eacute;talit&eacute; m&ecirc;me &agrave; la plus forte dose test&eacute;e. Die Charakterisierung der Determinanten der Virulenz von Krankheitserregern ist von gr&ouml;&szlig;ter Bedeutung f&uuml;r die Gestaltung von Strategien zur Krankheit. Bisher haben zwei Klassen von Virulenz Determinanten f&uuml;r virale Bev&ouml;lkerung gepr&auml;gt: diejenigen in der Nukleotidsequenz einige spezifischen genomischen Regionen und diejenigen, die abh&auml;ngig von der Komplexit&auml;t der viralen Bev&ouml;lkerung als solche eingepr&auml;gt. Hier bieten wir Beweise einer Virulenz-Determinante, die abh&auml;ngig ist, weder auf einer genomischen Sequenz noch auf erkennbare Unterschiede in der Bev&ouml;lkerung Komplexit&auml;t. Fu&szlig; - und - Klauenseuche-Virus ist t&ouml;dlich f&uuml;r C57BL/6 M&auml;usen zeigen die h&ouml;chste Viruslast in der Bauchspeicheldr&uuml;se. Virale Populationen von Pankreas, Milz und Serum zeigte allerdings nicht unterscheiden Konsens genomischen Nukleotid-Sequenzen und mutant Spektrum Komplexit&auml;ten, wie laut die Mutation-Frequenzen der beiden gesamten genomischen Nukleotidsequenzen biologische Klone quantifiziert. Die Ergebnisse zeigen, dass die Populationen mit unterschiedlichen Virulences die Konsensus-Sequenz oder durch die durchschnittliche Komplexit&auml;t des mutierten Spektrums unterschieden werden k&ouml;nnen. Differenzielle Ernte der Virus erzeugte von Zelltransfektion RNA aus Serum und der Bauchspeicheldr&uuml;se, die genetischen Unterschiede zwischen Teilpopulationen mit unterschiedlichen Virulences ausgestattet zeigen konnte. Zus&auml;tzlich zum Nachweis der verborgenen Virulenz Determinanten, unterstreicht diese Studie die Kapazit&auml;t eines Klons von einem RNA-Virus, rasch ph&auml;notypisch in-vivo zu diversifizieren.

La caratterizzazione dei determinanti di virulenza degli agenti patogeni &egrave; di massima importanza per la progettazione di strategie di controllo di malattia. Finora, due classi di determinanti di virulenza sono state caratterizzate per popolazioni virali: quelli impressi nella sequenza nucleotidica di alcune specifiche regioni genomiche e quelli che dipendono dalla complessit&agrave; della popolazione virale come tale. Qui forniamo prove di un determinante di virulenza che dipende da una sequenza genomica n&eacute; sulle differenze rilevabili nella complessit&agrave; della popolazione. Foot and mouth disease virus &egrave; letale per topi C57BL/6 mostrando la pi&ugrave; alta carica virale nel pancreas. Testato virus isolato da pancreas dopo un passaggio nei topi hanno mostrato un fenotipo attenuato, con no letalit&agrave; anche alla dose pi&ugrave; alta. La caracterizaci&oacute;n de los determinantes de virulencia de agentes pat&oacute;genos es de suma importancia para el dise&ntilde;o de estrategias de control de la enfermedad. Hasta ahora, se han caracterizado dos clases de determinantes de virulencia para poblaciones virales: los imprimieron en la secuencia de nucle&oacute;tido de algunas regiones gen&oacute;micas espec&iacute;ficas y aquellos que dependen de la complejidad de la poblaci&oacute;n viral como tal. ABSTRACT: Foot-and-mouth disease virus (FMVD), one of the most contagious viruses of cloven-hoofed animals, may cause a prolonged, asymptomatic but persistent infection in ruminants, named the "carrier state". However, it remains an open question whether this carrier state occurs in pigs. Here we present quantitative analyses of the duration of FMDV RNA and infectivity in lymphoid and epithelial tissues in experimentally infected pigs with FMDV C-S8c1. The data indicated that although FMDV RNA remained in blood until day 14 post-infection (pi), viremia was cleared by day 7 pi. However, all tissues tested were positive for FMDV until day 14-17 pi. Interestingly, the specific infectivity of FMDV in these tissues was in some cases even higher than the FMDV C-S8c1. We therefore propose that a "pseudopersistent state" may occur in pigs in which virus replicates in lymphoid tissues for a prolonged period of time, thereby representing a potential source of virus.

A replication analysis of foot-and-mouth disease virus in swine lymphoid tissue might indicate a putative carrier stage in pigs.

Nous proposons donc qu&#39;un &laquo; Etat pseudopersistent &raquo; peut se produire chez les porcs dans lequel r&eacute;plique de virus dans les tissus lympho&iuml;des pendant une p&eacute;riode prolong&eacute;e de temps, ce qui repr&eacute;sente une source potentielle de virus.

Une analyse de la r&eacute;plication du virus de la fi&egrave;vre aphteuse et dans le tissu lympho&iuml;de porcine pourrait indiquer une &eacute;tape de transporteur putatif chez les porcs.

Zusammenfassung: Fu&szlig; - und - Klauenseuche-Virus (FMVD), einer der am meisten ansteckenden Viren von Paarhufern, m&ouml;glicherweise eine verl&auml;ngerte, asymptomatische, aber anhaltende Infektion bei Wiederk&auml;uern, genannt den &quot;Tr&auml;ger-Staat&quot;. Eine offene Frage bleibt allerdings, ob dieser Tr&auml;ger-Status bei Schweinen auftritt. Hier pr&auml;sentieren wir Quantitative Analysen der Dauer der FMDV RNA und Infektiosit&auml;t im lymphatischen und epitheliale Gewebe experimentell infizierten Schweinen mit FMDV C-S8c1. Die Daten zeigten, dass trotz FMDV RNA in Blut bis Tag 14 Stunden (Pi) Vir&auml;mie 7.Tag Pi ger&auml;umt wurde. Allerdings waren alle Gewebe getestet f&uuml;r FMDV positiv bis Tag 14-17 Pi. Interessanterweise war die spezifische Infektiosit&auml;t des FMDV in diesen Geweben in einigen F&auml;llen sogar noch h&ouml;her als die FMDV C-S8c1. Deshalb schlagen wir ein &quot;pseudopersistent Zustand&quot; bei Schweinen in welcher Virus repliziert in lymphatischen Geweben f&uuml;r einen l&auml;ngeren Zeitraum, bef&uuml;rchten, darstellt und damit eine potenzielle Quelle des Virus.

Eine Replikation-Analyse des Fu&szlig; - und - Klauenseuche-Virus in Schweinen Lymphgewebe kann eine vermeintliche Tr&auml;ger-B&uuml;hne bei Schweinen hinweisen.

ABSTRACT: Foot and mouth disease virus (FMVD), uno dei virus pi&ugrave; contagioso di artiodattili, pu&ograve; causare un&#39;infezione prolungata, asintomatica, ma persistente nei ruminanti, denominati lo &quot;stato di vettore&quot;. Tuttavia, rimane aperta la questione se questo stato di vettore si verifica nei maiali. Qui vi presentiamo analisi quantitative della durata dell&#39;infettivit&agrave; e FMDV RNA nei tessuti epiteliali e linfoidi nei suini infettati sperimentalmente con FMDV C-S8c1. I dati hanno indicato che sebbene FMDV RNA &egrave; rimasto nel sangue fino al post-infection giorno 14 (pi), viremia fu cancellata da pi di giorno 7. Proponiamo pertanto che uno &quot;stato pseudopersistent&quot; pu&ograve; verificarsi nei suini in cui replicazione del virus nei tessuti linfoidi per un periodo prolungato di tempo, che rappresenta quindi una potenziale fonte di virus.

Un&#39;analisi della replica del foot and mouth disease virus nel tessuto linfoide suina potrebbe indicare una fase portante putativo nei suini.

Resumen: pie - y - boca disease virus (FMVD), uno de los virus m&aacute;s contagiosos de animales de pezu&ntilde;a hendida, puede causar una infecci&oacute;n prolongada, asintom&aacute;tica pero persistente en rumiantes, llamados el &quot;estado de portador&quot;. Sin embargo, sigue siendo una inc&oacute;gnita si este estado de portador ocurre en cerdos. Aqu&iacute; presentamos los an&aacute;lisis cuantitativos de la duraci&oacute;n del VFA RNA e infectividad en tejidos linfoides y epiteliales en cerdos infectados experimentalmente con VFA C-S8c1. Los datos indicaron que aunque VFA RNA permaneci&oacute; en sangre hasta post-infecci&oacute;n (pi) del d&iacute;a 14, la viremia despejaron por pi d&iacute;a 7. Sin embargo, todos los tejidos probados eran positivos para VFA hasta pi d&iacute;a 14-17. Curiosamente, la infectividad espec&iacute;fica del VFA en estos tejidos era en algunos casos incluso m&aacute;s altos que el VFA C-S8c1. Por lo tanto, proponemos que un &quot;estado pseudopersistent&quot; puede ocurrir en cerdos en que muestras del virus en tejidos linfoides durante un per&iacute;odo prolongado de tiempo, lo que representa una fuente potencial de virus.
