The nerve-cell cytoskeleton is essential for the regulation of intrinsic neuronal activity. For example, neuronal migration defects are associated with microtubule regulators, such as LIS1 and dynein, as well as with actin regulators, including Rac GTPases and integrins, and have been thought to underlie epileptic seizures in patients with cortical malformations. However, it is plausible that post-developmental functions of specific cytoskeletal regulators contribute to the more transient nature of aberrant neuronal activity and could be masked by developmental anomalies. Accordingly, our previous results have illuminated functional roles, distinct from developmental contributions, for Caenorhabditis elegans orthologs of LIS1 and dynein in GABAergic synaptic vesicle transport. Here, we report that C. elegans with function-altering mutations in canonical Rac GTPase-signaling-pathway members demonstrated a robust behavioral response to a GABA(A) receptor antagonist, pentylenetetrazole. Rac mutants also exhibited hypersensitivity to an acetylcholinesterase inhibitor, aldicarb, uncovering deficiencies in inhibitory neurotransmission. RNA interference targeting Rac hypomorphs revealed synergistic interactions between the dynein motor complex and some, but not all, members of Rac-signaling pathways. These genetic interactions are consistent with putative Rac-dependent regulation of actin and microtubule networks and suggest that some cytoskeletal regulators cooperate to uniquely govern neuronal synchrony through dynein-mediated GABAergic vesicle transport in C. elegans.

Pharmacogenetic analysis reveals a post-developmental role for Rac GTPases in Caenorhabditis elegans GABAergic neurotransmission.

Wir berichten hier, dass C. Pharmacogenetic &#xBD84;&#xC11D; Caenorhabditis elegans GABAergic neurotransmission&#xC5D0;&#xC11C;&#xC5D0;&#xC11C; Rac GTPases &#xC704;&#xD55C; post-developmental &#xC5ED;&#xD560;&#xC744; &#xBCF4;&#xC5EC;&#xC900;&#xB2E4;.

El citoesqueleto de las c&#xE9;lulas nerviosas es esencial para la regulaci&#xF3;n de la actividad neuronal intr&#xED;nseca. Por ejemplo, defectos de migraci&#xF3;n neuronal se asocian con los reguladores de los microt&#xFA;bulos, como LIS1 y dine&#xED;na, as&#xED; como con los reguladores de actina, incluyendo GTPasas Rac y las integrinas, y se han pensado que la base de las crisis epil&#xE9;pticas en pacientes con malformaciones corticales. Sin embargo, es posible que despu&#xE9;s del desarrollo de las funciones espec&#xED;ficas reguladores del citoesqueleto contribuyen a la naturaleza m&#xE1;s transitoria de actividad neuronal aberrante y podr&#xED;a ser enmascarada por anomal&#xED;as del desarrollo. En consecuencia, nuestros resultados anteriores han arrojado luz sobre los roles funcionales, distintas de las contribuciones en el desarrollo, para Caenorhabditis elegans orthologs de LIS1 y dine&#xED;na en el transporte de ves&#xED;culas sin&#xE1;pticas GABA&#xE9;rgicas. En este caso, nos informan de que C. elegans, con alteraci&#xF3;n de la funci&#xF3;n de las mutaciones en can&#xF3;nicos Rac GTPasa de la v&#xED;a de se&#xF1;alizaci&#xF3;n miembros demostraron una respuesta contundente a un comportamiento GABA (A) antagonista de los receptores, pentilentetrazol. Rac mutantes tambi&#xE9;n mostraron hipersensibilidad a un inhibidor de la acetilcolinesterasa, el aldicarb, la detecci&#xF3;n de deficiencias en la neurotransmisi&#xF3;n inhibitoria. La interferencia del ARN dirigida hypomorphs Rac revel&#xF3; interacciones sin&#xE9;rgicas entre el complejo dine&#xED;na motor y algunos, pero no todos, los miembros de Rac-las v&#xED;as de se&#xF1;alizaci&#xF3;n. Estas interacciones gen&#xE9;ticas son consistentes con la supuesta Rac-dependen de la regulaci&#xF3;n de la actina y las redes de microt&#xFA;bulos, y sugieren que algunos reguladores del citoesqueleto cooperar para regular &#xFA;nicamente la sincron&#xED;a neuronal a trav&#xE9;s de dine&#xED;na mediada por el transporte de ves&#xED;culas GABA&#xE9;rgica en C. elegans.
