Cajal-Retzius cells are essential pioneer neurons that guide neuronal migration in the developing neocortex. During development, Cajal-Retzius cells arise from distinct progenitor domains that line the margins of the dorsal telencephalon, or pallium. Here, we show that the proneural gene Ascl1 is expressed in Cajal-Retzius cell progenitors in the pallial septum, ventral pallium, and cortical hem. Using a short-term lineage trace, we demonstrate that it is primarily the Ascl1-expressing progenitors in the pallial septum and ventral pallium that differentiate into Cajal-Retzius cells. Accordingly, we found a small, albeit significant reduction in the number of Reelin(+) and Trp73(+) Cajal-Retzius cells in the Ascl1(-/-) neocortex. Conversely, using a gain-of-function approach, we found that Ascl1 induces the expression of both Reelin, a Cajal-Retzius marker, and Tbr1, a marker of pallial-derived neurons, in a subset of early-stage pallial progenitors, an activity that declines over developmental time. Taken together, our data indicate that the proneural gene Ascl1 is required and sufficient to promote the differentiation of a subset of Cajal-Retzius neurons during early neocortical development. Notably, this is the first study that reports a function for Ascl1 in the pallium, as this gene is best known for its role in specifying subpallial neuronal identities.

Ascl1 participates in Cajal-Retzius cell development in the neocortex.

Notamment, il s&#x26;#39;agit de la premi&#xE8;re &#xE9;tude qui fait &#xE9;tat d&#x26;#39;une fonction pour Ascl1 dans le pallium, comme ce g&#xE8;ne est surtout connu pour son r&#xF4;le dans la sp&#xE9;cification souspallium identit&#xE9;s neuronales.

Ascl1 participe au d&#xE9;veloppement de cellules de Cajal-Retzius dans le n&#xE9;ocortex

Cajal-Retzius-Zellen sind wesentliche Pionier Neuronen, die in Entwicklungsl&auml;ndern Neocortex der neuronalen Migration f&uuml;hren. Las c&#xE9;lulas de Cajal-Retzius son neuronas pioneras esenciales que gu&#xED;an la migraci&#xF3;n neuronal en la corteza cerebral en desarrollo. Durante el desarrollo, las c&#xE9;lulas de Cajal-Retzius surgen de dominios progenitoras distintas que se alinean en las m&#xE1;rgenes del telenc&#xE9;falo dorsal, o el palio. En este sentido, muestran que el gen Ascl1 proneural se expresa en progenitores de c&#xE9;lulas de Cajal-Retzius en el tabique paleal, el palio ventral, y el dobladillo cortical. El uso de un origen del linaje de corto plazo, nos demuestran que se trata principalmente de los progenitores Ascl1 que expresan en el tabique paleal y el palio ventral, que se diferencian en c&#xE9;lulas de Cajal-Retzius. En consecuencia, se encontr&#xF3; una peque&#xF1;a, aunque significativa reducci&#xF3;n en el n&#xFA;mero de Reelin (+) y Trp73 (+) Cajal-Retzius c&#xE9;lulas en el Ascl1 (- / -) neoc&#xF3;rtex. A la inversa, utilizando un enfoque de ganancia de funci&#xF3;n, se encontr&#xF3; que Ascl1 induce la expresi&#xF3;n de ambos Reelin, un marcador de Cajal-Retzius, y Tbr1, un marcador de paleal neuronas derivadas, en un subconjunto de las primeras etapas progenitores paliales, un actividad que disminuye con el tiempo de desarrollo. En conjunto, nuestros datos indican que el gen Ascl1 proneural es necesaria y suficiente para promover la diferenciaci&#xF3;n de un subconjunto de neuronas Cajal-Retzius durante el desarrollo neocortical temprano. En particular, este es el primer estudio que informa de una funci&#xF3;n para Ascl1 en el palio, ya que este gen es mejor conocido por su papel en la especificaci&#xF3;n de identidades neuronales subpalio.

El cerebelo se compone de una amplia gama de subtipos neuronales.
