HEPES is a favorable buffer for Ga-complexations in radiochemical laboratories. The drawback of this buffer is its prescribed limit of 200 μg per recommended application volume in the final formulation. Currently, a TLC test according to the European Pharmacopoeia (Ph. Eur.) has to be performed for quantification, but this analysis suffers from low reliability and reproducibility and is based on a subjective, semi-quantitative visual evaluation. In this study, the TLC method according to the Ph. Eur. and two literature-known HPLC assays for HEPES quantification were evaluated. Additionally, the development of an improved TLC method was performed.

Development and evaluation of a rapid analysis for HEPES determination in Ga-radiotracers.

Radiochemists/radiopharmacists, involved in the preparation of radiopharmaceuticals are regularly confronted with the requirement of continuous high quality productions in their day-to-day business. One of these requirements is high specific or molar activity of the radiotracer in order to avoid e.g. receptor saturation and pharmacological or even toxic effects of the applied tracer for positron emission tomography. In the case of C-labeled radiotracers, the reasons for low molar activity are manifold and often the search for potential C-contaminations is time-consuming.

Molar activity - The keystone in C-radiochemistry: An explorative study using the gas phase method.

The first aim of this study was to evaluate Ga-PSMA conjugate 11 positron emission tomography (PSMA PET) parameters for assessment of response to Lu-PSMA-617 radioligand therapy (RLT) in patients with metastatic castration-resistant prostate cancer (mCRPC). The second aim was to investigate factors associated with overall survival (OS).
