A heteropolymer (HP) is a unique dual antibody conjugate composed of specific, chemically cross-linked monoclonal antibodies (mAbs). In this study we have demonstrated that HPs can be purified using hydrophobic interaction chromatography (HIC). Two propyl HIC resins; [PolyPropyl A and EMD Fractogel Propyl (S)] were evaluated in this study. Phosphate buffers, pH 6.5 containing ammonium sulfate or sodium sulfate were used to bind the HP to the column. A descending sulfate gradient or step gradient was used to elute the bound HP species from the column. The HP reaction mixture typically contains multiple conjugated HP species, as well as unreacted monomer mAbs. Conjugated HP product was successfully separated from unreacted antibody monomers with both propyl resins using buffers with ammonium sulfate. There was no monomer separation from HP using buffers with sodium sulfate. The purification processes, presented in this study allows the non-cross-linked antibodies to pass through the column without being bound to the resin, while the cross-linked antibodies (the HP product) bound to the column were subsequently eluted by decreasing the ammonium sulfate concentration in the running buffer. HP product was efficiently separated from free mAbs using Propyl HIC resins at both analytical and preparative scales.

Purification of antibody heteropolymers using hydrophobic interaction chromatography.

Dans cette &eacute;tude, nous avons d&eacute;montr&eacute; que les HPs peuvent &ecirc;tre purifi&eacute;s &agrave; l&#39;aide de la chromatographie hydrophobe (HIC). Ein Heteropolymer (HP) ist eine einzigartige dual Antik&ouml;rper konjugiert bestehend aus bestimmten, chemisch vernetzte monoklonale Antik&ouml;rper (mAbs). In dieser Studie haben wir gezeigt, dass HPs mit hydrophobe Interaktion Chromatographie (HIC) gereinigt werden k&ouml;nnen. Zwei Propyl HIC Harze; [PolyPropyl A und EMD Fractogel Propyl (S)] wurden in dieser Studie evaluiert. Phosphat-Puffer, pH 6.5 mit Ammoniumsulfat oder Natriumsulfat dienten die HP an die Spalte zu binden. Eine absteigende Sulfat Farbverlauf oder einen Schritt Verlauf wurde verwendet, um die gebundenen HP-Arten aus der Spalte zu eluieren. Das Reaktionsgemisch HP enth&auml;lt normalerweise mehrere konjugierte HP-Arten sowie Spurenverunreinigungen Monomer mAbs. Konjugierte HP-Produkt wurde erfolgreich von Spurenverunreinigungen Antik&ouml;rper Monomere mit beiden Propyl-Harzen mit Puffer mit Ammonsulfat getrennt. Es gab keine Monomer-Trennung von HP mit Puffer mit Natriumsulfat. Die Reinigungsverfahren, die in dieser Studie vorgestellt erm&ouml;glicht die Kreuz-chromosomale Antik&ouml;rper, ohne an das Harz gebunden wird, w&auml;hrend die vernetzte Antik&ouml;rper (das HP-Produkt) an die Spalte gebunden anschlie&szlig;end durch eine Verringerung der Ammonium-Sulfat-Konzentration im laufenden Puffer eluiert wurden durch die Spalte &uuml;bergeben. HP Produkt wurde vom freien mAbs pr&auml;parative und Analytische Waage Propyl HIC Harze mit effizient abgetrennt.

Reinigung von Antik&ouml;rper-Heteropolymers mit hydrophobe Interaktion Chromatographie.

Un heteropolymer (HP) &egrave; un unico doppio anticorpo coniugato composto specifici, reticolati chimicamente anticorpi monoclonali (mAbs). In questo studio abbiamo dimostrato che HPs pu&ograve; essere purificato mediante cromatografia di interazione idrofobica (HIC). Due resine di propile HIC; [PolyPropyl A ed EMD Fractogel propilico (S)] sono stati valutati in questo studio. Buffer fosfato, pH 6.5 contenente solfato di ammonio o solfato di sodio sono stati utilizzati per associare l&#39;HP per la colonna. Un gradiente decrescente di solfato o gradiente di passaggio &egrave; stato utilizzato per eluire la specie HP vincolato dalla colonna. The productivity of stably transfected cell lines is of critical importance for the manufacturing of therapeutic proteins. Various methods have been successfully implemented to increase the production output of mammalian cell cultures. Increasing evidence suggests that optimization of the gene coding sequences of an expression vector can improve specific cell line yield of the recombinant protein. Here we demonstrate that gene optimization substantially enhances antibody production in Chinese hamster ovary cells. When gene optimization was applied to the heavy and light chain genes of a therapeutic antibody, we observed increased antibody production in transient transfection. Elevated heavy chain mRNA level was associated with the increase of antibody production. Further analysis suggested that the increased antibody expression is attributable to enhanced mRNA stability resulting from gene optimization. Gene optimization also led to increased antibody production in stable clones.

La productividad de las l&#xED;neas celulares transfectadas establemente es de importancia cr&#xED;tica para la fabricaci&#xF3;n de prote&#xED;nas terap&#xE9;uticas. Varios m&#xE9;todos han sido aplicado con &#xE9;xito para aumentar el rendimiento de producci&#xF3;n de cultivos celulares de mam&#xED;feros. Aumento de la evidencia sugiere que la optimizaci&#xF3;n del gen que codifica secuencias de un vector de expresi&#xF3;n puede mejorar el rendimiento espec&#xED;fico l&#xED;nea celular de la prote&#xED;na recombinante. Aqu&#xED; demostramos que la optimizaci&#xF3;n del gen mejora sustancialmente la producci&#xF3;n de anticuerpos en las c&#xE9;lulas de ovario de h&#xE1;mster chino. Cuando gen optimizaci&#xF3;n se aplic&#xF3; a los genes de las cadenas pesada y ligera de un anticuerpo terap&#xE9;utico, se observ&#xF3; aumento de la producci&#xF3;n de anticuerpos en transfecci&#xF3;n transitoria.
