Cela est r&eacute;alis&eacute;, en partie, par le biais de l&#39;action de nombreuses prot&eacute;ines r&eacute;gulatrices comme soluble cluster de diff&eacute;renciation 14 (sCD14) trouv&eacute;e en grande quantit&eacute; dans le lait maternel (~ 25 &agrave; 50 &mu;g/mL). Chez les adultes, CD14 stimule la production de cytokines en r&eacute;ponse aux lipopolysaccharides (LPS), le composant lipidique majeur se trouve dans la membrane externe des bact&eacute;ries Gram-n&eacute;gatives. Toutefois, le sort et la fonction de sCD14 dans le syst&egrave;me gastro-intestinal (GI) n&eacute;onatal sont inconnus et peut-&ecirc;tre la fonction diff&eacute;remment des adultes. Par cons&eacute;quent, nous administr&eacute; sCD14 humain pour les animaux de laboratoire et a fait observer qu&#39;il a persist&eacute; dans le tube digestif sup&eacute;rieur apr&egrave;s la t&eacute;t&eacute;e. Dans notre recherche d&#39;&eacute;ventuels protecteurs prot&eacute;olytiques, immunopr&eacute;cipitation de sCD14 du lait humain ont r&eacute;v&eacute;l&eacute; une nouvelle prot&eacute;ine de 15-kD qui co-purifi&eacute;e avec sCD14. Analyse de spectrom&eacute;trie de masse de la prot&eacute;ine identifi&eacute;e alpha-lactalbumine. CD14 a &eacute;t&eacute; &eacute;galement identifi&eacute; par immunoblot apr&egrave;s immunopr&eacute;cipitation de l&#39;alpha-lactalbumine du lait. Des essais de digestion in vitro r&eacute;v&egrave;lent qu&#39;alpha-lactalbumine purifi&eacute;e diminue la d&eacute;gradation prot&eacute;olytique du lait humain d&eacute;riv&eacute; sCD14 in vitro, ce qui sugg&egrave;re un m&eacute;canisme par lequel ce r&eacute;cepteur cl&eacute; de LPS peut-&ecirc;tre demeurer fonctionnel dans le tube digestif du nouveau-n&eacute;.

Mother's milk represents a foundational step in the proper development of newborn immunity. This is achieved, in part, through the action of numerous regulatory proteins such as soluble cluster of differentiation 14 (sCD14) found in significant quantities in human milk (~25-50 Î¼g/mL). In adults, CD14 stimulates cytokine production in response to lipopolysaccharide (LPS), the major lipid component found in the outer membrane of Gram-negative bacteria. However, the fate and function of sCD14 in the neonatal gastrointestinal (GI) tract are unknown and may function differently from adults. Therefore, we administered human sCD14 to experimental animals and observed that it persisted in the upper GI tract after feeding. In our search for potential proteolytic protectants, immunoprecipitation of sCD14 from human milk revealed a 15-kD novel protein that copurified with sCD14. Mass spectrometry analysis of the protein identified alpha-lactalbumin. CD14 was also identified by immunoblot after immunoprecipitation of alpha-lactalbumin from milk. In vitro digestion assays revealed that purified alpha-lactalbumin decreases the proteolytic degradation of human milk derived sCD14 in vitro, suggesting a mechanism by which this key LPS receptor may remain functional in the neonate gut.

In-vitro-Verdauung-Proben ergab, dass gereinigtes Alpha-Lactalbumin verringert den proteolytischen Abbau der Muttermilch abgeleitet sCD14 in vitro einen Mechanismus, mit dem dieser Schl&uuml;ssel LPS-Rezeptor funktionieren im Darm Neugeborener weiterhin kann, vorschlagen.

Leche de la madre representa un paso fundamental en el correcto desarrollo de la inmunidad del reci&eacute;n nacido. Esto se logra, en parte, a trav&eacute;s de la acci&oacute;n de numerosas prote&iacute;nas reguladoras como grupo soluble de diferenciaci&oacute;n 14 (sCD14) encontrada en cantidades significativas en la leche humana (~ 25-50 &mu;g/mL). En adultos, CD14 estimula la producci&oacute;n de citoquinas en respuesta a lipopolisac&aacute;rido (LPS), el componente lip&iacute;dico principales se encuentra en la membrana externa de las bacterias gramnegativas. Sin embargo, el destino y la funci&oacute;n de sCD14 en el tracto gastrointestinal (GI) neonatal son desconocidos y pueden funcionar de forma diferente a los adultos. Por ello, hab&iacute;a administrado sCD14 humana en animales experimentales y observ&oacute; que persist&iacute;a en el tracto gastrointestinal superior despu&eacute;s de la alimentaci&oacute;n. En nuestra b&uacute;squeda de potenciales productos proteol&iacute;ticas, inmunoprecipitaci&oacute;n de sCD14 de leche humana revel&oacute; una nueva prote&iacute;na 15-kD que copurified con sCD14. An&aacute;lisis de espectrometr&iacute;a de masas de la prote&iacute;na identifican alfa-lactoalb&uacute;mina. CD14 tambi&eacute;n fue identificada por el immunoblot despu&eacute;s de inmunoprecipitaci&oacute;n de alfa-lactoalb&uacute;mina de la leche. An&aacute;lisis de digesti&oacute;n in vitro revelaron alfa-lactoalb&uacute;mina purificada disminuye la degradaci&oacute;n proteol&iacute;tica de la leche humana derivada sCD14 in vitro, sugiriendo un mecanismo por el cual este receptor clave de LPS puede permanecer funcional en el intestino del reci&eacute;n nacido.

Il metodo &egrave; stato testato da indagare la biodistribuzione di C (14) in ratti che sono stati alimentati (14) con etichetta C umana solubile cluster della proteina (CD14) 14 di differenziazione, un recettore per lipopolisaccaride batterico. Due altre proteine di controllo, albumina di siero bovino (BSA) e la caseina, erano anche etichettati con (14) C e utilizzate per l&#39;analisi comparativa per determinare quanto segue: (i) l&#39;efficienza efficacia e i costi del procedimento sotto vuoto su radiolabeling e (ii) la misura dell&#39;incorporazione del C (14) etichetta in organi di orogastrically alimentato ratti Sprague-Dawley 10 giorni di et&agrave;. The economical preparation of microgram quantities of (14)C-labeled proteins by in vacuo methylation with methyl iodide is described. The (14)C radiolabeling was achieved by the covalent attachment of [(14)C]methyl groups onto amino and imidazole groups by reaction in vacuo with [(14)C]methyl iodide. The method was tested by investigating the biodistribution of (14)C in rats that were fed (14)C-labeled human soluble cluster of differentiation 14 (CD14) protein, a receptor for bacterial lipopolysaccharide. Two other control proteins, bovine serum albumin (BSA) and casein, were also labeled with (14)C and used for comparative analysis to determine the following: (i) the efficacy and cost efficiency of the in vacuo radiolabeling procedure and (ii) the extent of incorporation of the (14)C label into the organs of orogastrically fed 10-day-old Sprague-Dawley rats. [(14)C]BSA, [(14)C]casein, and [(14)C]CD14 were individually prepared with specific radioactivities of 34,400, 18,800, and 163,000 disintegrations per minute (dpm)/microg, respectively. It was found that the accumulation of (14)C label in the organs of [(14)C]CD14-fed rats, most notably the persistence of (14)C in the stomach 480 min postgavage, was temporally and spatially distinct from [(14)C]BSA and [(14)C]casein-fed rats.

Gastrointestinal transit times (GItts) were compared in separate litters of 10- and 15-day-old Sprague Dawley rats using barium sulphate. By tracking the leading front of the bolus on radiographs, the gastrocaecal transit times in pups were estimated. To measure the total GItt, the duration from orogastric gavage until an observable defecation of barium sulphate was recorded. The gastrocaecal times for 10-day-old pups maintained with their dam (n = 5) ranged from 4-5 h and those removed from the dam ranged from 2.5-5 h. For 15-day-old pups with their dam (n = 6) and without dam (n = 5), gastrocaecal times ranged from 4-6 h and 3.5-5 h, respectively. Ten-day-old pups that remained with the dam had a GItt of 13.8 Â± 0.9 h and those kept in the absence of the dam had a time of 9.3 Â± 0.7 h. This decrease (P < 0.05) in GItt in the absence of the dam was age-dependent in 10-day-old pups, and was not observed (P > 0.05) in 15-day-old pups. The results provide a basis, for the design of future studies involving neonate rat metabolism, to include maternal presence.

Tiempos de tr&aacute;nsito gastrointestinal (GItts) compararon diferentes camadas de ratas Sprague Dawley de 10 y 15 d&iacute;as de edad con sulfato de bario. Por el frente principal del bolo de seguimiento en las radiograf&iacute;as, se estimaron los tiempos de tr&aacute;nsito gastrocaecal en cachorros. Para medir la total GItt, se registr&oacute; la duraci&oacute;n de la sonda orog&aacute;strica hasta un observable defecaci&oacute;n de sulfato de bario. Los tiempos de gastrocaecal para 10 d&iacute;as de edad cachorros mantenidos con su presa (n = 5) oscil&oacute; entre 4-5 h y los quit&oacute; de la presa entre 2.5-5 h. Para cachorros de 15 d&iacute;as de edad con su presa (n = 6) y sin presa (n = 5), gastrocaecal veces oscilan entre 4-6 h y 3.5-5 h, respectivamente. Cachorros de diez d&iacute;as de edad que permanec&iacute;an con la presa ten&iacute;an un GItt de 13.8 &plusmn; 0,9 h y los mantuvo en la ausencia de la presa ten&iacute;an un tiempo de 9,3 &plusmn; 0,7 h. Esta disminuci&oacute;n (P &lt; 0.05) en GItt en ausencia de la presa fue dependiente de la edad en cr&iacute;as de 10 d&iacute;as de edad y no se observ&oacute; (P &gt; 0.05) en cachorros de 15 d&iacute;as de edad. Los resultados proporcionan una base, para el dise&ntilde;o de los futuros estudios que implican el metabolismo de la rata de reci&eacute;n nacido incluir la presencia materna.
