Siderophore biosynthesis by the highly lethal mould Aspergillus fumigatus is essential for virulence, but non-existent in humans, presenting a rare opportunity to strategize therapeutically against this pathogen. We have previously demonstrated that A. fumigatus excretes fusarinine C and triacetylfusarinine C to capture extracellular iron, and uses ferricrocin for hyphal iron storage. Here, we delineate pathways of intra- and extracellular siderophore biosynthesis and show that A. fumigatus synthesizes a developmentally regulated fourth siderophore, termed hydroxyferricrocin, employed for conidial iron storage. By inactivation of the nonribosomal peptide synthetase SidC, we demonstrate that the intracellular siderophores are required for germ tube formation, asexual sporulation, resistance to oxidative stress, catalase A activity, and virulence. Restoration of the conidial hydroxyferricrocin content partially rescues the virulence of the apathogenic siderophore null mutant Delta sidA, demonstrating an important role for the conidial siderophore during initiation of infection. Abrogation of extracellular siderophore biosynthesis following inactivation of the acyl transferase SidF or the nonribosomal peptide synthetase SidD leads to complete dependence upon reductive iron assimilation for growth under iron-limiting conditions, partial sensitivity to oxidative stress, and significantly reduced virulence, despite normal germ tube formation. Our findings reveal distinct cellular and disease-related roles for intra- and extracellular siderophores during mammalian Aspergillus infection.

The protein phosphatase calcineurin is an important mediator connecting calcium-dependent signalling to various cellular responses in multiple organisms. In fungi calcineurin acts largely through regulating Crz1p-like transcription factors. Here we characterize an Aspergillus fumigatus CRZ1 homologue, CrzA and demonstrate its mediation of cellular tolerance to increased concentrations of calcium and manganese. In addition to acute sensitivity to these ions, and decreased conidiation, the crzA null mutant suffers altered expression of calcium transporter mRNAs under high concentrations of calcium, and loss of virulence when compared with the corresponding complemented and wild-type strains. We use multiple expression analyses to probe the transcriptional basis of A. fumigatus calcium tolerance identifying several genes having calA and/or crzA dependent mRNA accumulation patterns. We also demonstrate that contrary to previous findings, the gene encoding the Aspergillus nidulans calcineurin subunit homologue, cnaA, is not essential and that the cnaA deletion mutant shares the morphological phenotypes observed in the corresponding A. fumigatus mutant, DeltacalA. Exploiting the A. nidulans model system, we have linked calcineurin activity with asexual developmental induction, finding that CrzA supports appropriate developmental induction in a calcineurin and brlA-dependent manner in both species.

Functional characterization of the Aspergillus fumigatus CRZ1 homologue, CrzA.

La calcineurine phosphatase de prot&eacute;ine est un m&eacute;diateur important reliant calcium-d&eacute;pendante de signalisation &agrave; diverses r&eacute;actions cellulaires dans plusieurs organismes. Chez les champignons calcineurine agit en grande partie par la r&eacute;gulation des facteurs de transcription de type Crz1p. Ici, nous caract&eacute;risons un Aspergillus fumigatus homologue CRZ1, CrzA et d&eacute;montrer sa m&eacute;diation cellulaire tol&eacute;rance &agrave; une augmentation des concentrations de calcium et de mangan&egrave;se. En plus de la sensibilit&eacute; aigu&euml; de ces ions et une diminution de la conidiation, le mutant nul crzA souffre une expression alt&eacute;r&eacute;e des ARNm de transporteur de calcium sous des concentrations &eacute;lev&eacute;es de calcium et la perte de virulence en comparaison avec les souches compl&eacute;t&eacute;es et sauvage correspondants. Caract&eacute;risation fonctionnelle de l&#39;Aspergillus fumigatus homologue CRZ1, CrzA.

Das Protein Phosphatase Calcineurin ist ein wichtiger Vermittler Verbindung Kalzium-abh&auml;ngige Signalisierung zu verschiedenen zellul&auml;ren Antworten in mehrere Organismen. In Pilzen fungiert Calcineurin weitgehend durch Crz1p-wie Transkriptionsfaktoren regulieren. Hier wir ein Aspergillus Fumigatus CRZ1 Homolog, CrzA charakterisieren und ihrer Vermittlung der zellul&auml;re Toleranz zu erh&ouml;hten Konzentrationen von Kalzium und Mangan zu demonstrieren. Neben akuten Sensibilit&auml;t f&uuml;r diese Ionen und verminderte Conidiation leidet die CrzA null Mutant ver&auml;nderten Ausdruck Kalzium Transporter mRNAs unter hohen Konzentrationen von Calcium und Verlust der Virulenz im Vergleich mit den entsprechenden erg&auml;nzten und Wildtyp-St&auml;mmen. Wir verwenden mehrere Ausdruck-Analysen, um die transkriptionelle Basis A. Fumigatus Kalzium Toleranz identifizieren mehrere Gene mit CalA und/oder CrzA abh&auml;ngige mRNA-Ansammlung-Muster. Wir zeigen auch, dass im Gegensatz zu fr&uuml;heren Feststellungen, gen, das den Aspergillus Nidulans Calcineurin Untereinheit Homolog, CnaA, ist nicht unbedingt erforderlich und das CnaA L&ouml;schung Mutant die morphologischen Ph&auml;notypen, die in den entsprechenden A. Fumigatus Mutant, DeltacalA beobachtet teilt. A. Nidulans-Modell-System nutzen, haben wir verkn&uuml;pft Calcineurin-Aktivit&auml;t mit ungeschlechtliche Entwicklungs-Induktion, CrzA unterst&uuml;tzt entsprechende Entwicklungsst&ouml;rungen Induktion in eine Calcineurin und BrlA-abh&auml;ngigen Weise bei beiden Spezies zu finden.

Funktionelle Charakterisierung der Aspergillus Fumigatus CRZ1 Homolog, CrzA.

La proteina fosfatasi calcineurina &egrave; un mediatore importante collegamento calcio-dipendente di segnalazione a varie risposte cellulari in molteplici organismi. Nei funghi calcineurina agisce in gran parte attraverso la regolazione di fattori di trascrizione come Crz1p. Qui ci caratterizzano un Aspergillus fumigatus CRZ1 omologo, CrzA e dimostrare la sua mediazione di tolleranza cellulare all&#39;aumento delle concentrazioni di calcio e manganese. Oltre a acuta sensibilit&agrave; di questi ioni e conidiation &egrave; diminuito, il mutante null crzA soffre alterata espressione dei mRNAs transporter calcio sotto alte concentrazioni di calcio e la perdita di virulenza quando confrontati con i corrispondenti ceppi integrati e selvaggio-tipo. Usiamo pi&ugrave; analisi di espressione per sondare la base transcriptional di tolleranza di calcio di a. fumigatus identificazione di diversi geni avendo calA o crzA modelli di accumulo di mRNA dipendente. Dimostreremo anche che contrariamente alle precedenti scoperte, il gene che codifica per l&#39;omologo di subunit&agrave; Aspergillus nidulans calcineurina, cnaA, non &egrave; essenziale e che il mutante di delezione cnaA condivide i fenotipi morfologici osservati nel corrispondente a. fumigatus mutante, DeltacalA. Sfruttando il sistema di modello a. nidulans, noi abbiamo collegato attivit&agrave; della calcineurina con induzione dello sviluppo asessuale, trovando che CrzA supporta induzione dello sviluppo appropriato in modo calcineurina e brlA-dipendente in entrambe le specie.

La prote&iacute;na fosfatasa calcineurina es un mediador importante conexi&oacute;n se&ntilde;alizaci&oacute;n dependiente de calcio a varias respuestas celulares en varios organismos. En hongos calcineurina act&uacute;a en gran parte a trav&eacute;s de la regulaci&oacute;n de factores de transcripci&oacute;n Crz1p-como. Aqu&iacute; se caracterizan un Aspergillus fumigatus CRZ1 hom&oacute;logo, CrzA y demostrar su mediaci&oacute;n de tolerancia celular al aumento de las concentraciones de calcio y magnesio. Adem&aacute;s de aguda sensibilidad a estos iones y disminuci&oacute;n de la conidiation, el mutante nulo crzA sufre la expresi&oacute;n alterada de los mRNAs de transportador de calcio bajo altas concentraciones de calcio y p&eacute;rdida de virulencia en comparaci&oacute;n con las cepas complementadas y salvaje-tipo correspondientes. Utilizamos m&uacute;ltiples an&aacute;lisis de expresi&oacute;n para sondear la base transcripcional de a. fumigatus tolerancia de calcio identificaci&oacute;n de varios genes que calA y/o crzA patrones de acumulaci&oacute;n dependientes de mRNA. Tambi&eacute;n demostramos que contrariamente a los resultados anteriores, el gene que codifica el hom&oacute;logo de subunidad Aspergillus nidulans calcineurina, cnaA, no es esencial y que los mutantes de deleci&oacute;n de cnaA comparten los fenotipos morfol&oacute;gicos observados en los correspondientes mutantes de a. fumigatus, DeltacalA. Aprovechando el sistema de modelo de a. nidulans, hemos vinculado actividad de calcineurina con inducci&oacute;n del desarrollo asexual, encontrando que CrzA apoya la inducci&oacute;n del desarrollo apropiada de manera calcineurina y brlA-dependiente en ambas especies.

Phosphate is an ion that is essential for fungal growth. The systems for inorganic phosphate (P(i)) acquisition in eukaryotic cells (PHO) have been characterized as a low-affinity (that assures a supply of P(i) at normal or high external P(i) concentrations) and a high-affinity (activated in response to P(i) starvation). Here, as an initial step to understand the PHO pathway in Aspergillus fumigatus, we characterized the PHO80 homologue, PhoB(PHO80). We show that the DeltaphoB(PHO80) mutant has a polar growth defect (i.e., a delayed germ tube emergence) and, by phenotypic and phosphate uptake analyses, establish a link between PhoB(PHO80), calcineurin and calcium metabolism. Microarray hybridizations carried out with RNA obtained from wild-type and DeltaphoB(PHO80) mutant cells identify Afu4g03610 (phoD(PHO84)), Afu7g06350 (phoE(PHO89)), Afu4g06020 (phoC(PHO81)), and Afu2g09040 (vacuolar transporter Vtc4) as more expressed both in the DeltaphoB(PHO80) mutant background and under phosphate-limiting conditions of 0.1mM P(i). Epifluorescence microscopy revealed accumulation of poly-phosphate in DeltaphoB(PHO80) vacuoles, which was independent of extracellular phosphate concentration. Surprisingly, a phoD(PHO84) deletion mutant is indistinguishable phenotypically from the corresponding wild-type strain. mRNA analyses suggest that protein kinase A absence supports the expression of PHO genes in A. fumigatus. Furthermore, DeltaphoB(PHO80) and DeltaphoD(PHO84) mutant are fully virulent in a murine low dose model for invasive aspergillosis.

Aspergillus fumigatus is a common mould whose spores are a component of the normal airborne flora. Immune dysfunction permits developmental growth of inhaled spores in the human lung causing aspergillosis, a significant threat to human health in the form of allergic, and life-threatening invasive infections. The success of A. fumigatus as a pathogen is unique among close phylogenetic relatives and is poorly characterised at the molecular level. Recent genome sequencing of several Aspergillus species provides an exceptional opportunity to analyse fungal virulence attributes within a genomic and evolutionary context. To identify genes preferentially expressed during adaptation to the mammalian host niche, we generated multiple gene expression profiles from minute samplings of A. fumigatus germlings during initiation of murine infection. They reveal a highly co-ordinated A. fumigatus gene expression programme, governing metabolic and physiological adaptation, which allows the organism to prosper within the mammalian niche. As functions of phylogenetic conservation and genetic locus, 28% and 30%, respectively, of the A. fumigatus subtelomeric and lineage-specific gene repertoires are induced relative to laboratory culture, and physically clustered genes including loci directing pseurotin, gliotoxin and siderophore biosyntheses are a prominent feature. Locationally biased A. fumigatus gene expression is not prompted by in vitro iron limitation, acid, alkaline, anaerobic or oxidative stress. However, subtelomeric gene expression is favoured following ex vivo neutrophil exposure and in comparative analyses of richly and poorly nourished laboratory cultured germlings. We found remarkable concordance between the A. fumigatus host-adaptation transcriptome and those resulting from in vitro iron depletion, alkaline shift, nitrogen starvation and loss of the methyltransferase LaeA. This first transcriptional snapshot of a fungal genome during initiation of mammalian infection provides the global perspective required to direct much-needed diagnostic and therapeutic strategies and reveals genome organisation and subtelomeric diversity as potential driving forces in the evolution of pathogenicity in the genus Aspergillus.

Um die Identifizierung von Genen, die bevorzugt in Anpassung an die S&auml;ugetier-Host-Nische ge&auml;u&szlig;erten, haben wir mehrere Gen-Ausdruck-Profile von Minute Stichproben von A. Inorganic polyphosphate (poly-P) consists of just a chain of phosphate groups linked by high energy bonds. It is found in every organism and is implicated in a wide variety of cellular processes (e.g. phosphate storage, blood coagulation, and pathogenicity). Its metabolism has been studied mainly in bacteria while remaining largely uncharacterized in eukaryotes. It has recently been suggested that poly-P metabolism is connected to that of highly phosphorylated inositol species (inositol pyrophosphates). Inositol pyrophosphates are molecules in which phosphate groups outnumber carbon atoms. Like poly-P they contain high energy bonds and play important roles in cell signaling. Here, we show that budding yeast mutants unable to produce inositol pyrophosphates have undetectable levels of poly-P. Our results suggest a prominent metabolic parallel between these two highly phosphorylated molecules. More importantly, we demonstrate that DDP1, encoding diadenosine and diphosphoinositol phosphohydrolase, possesses a robust poly-P endopolyphosphohydrolase activity. In addition, we prove that this is an evolutionarily conserved feature because mammalian Nudix hydrolase family members, the three Ddp1 homologues in human cells (DIPP1, DIPP2, and DIPP3), are also capable of degrading poly-P.

Polifosfato inorg&#xE1;nico (poli-P) consta de s&#xF3;lo una cadena de grupos fosfato unidos por enlaces de alta energ&#xED;a. Se encuentra en todos los organismos y est&#xE1; implicado en una amplia variedad de procesos celulares (por ejemplo fosfato de almacenamiento, coagulaci&#xF3;n de la sangre, y la patogenicidad). Su metabolismo se ha estudiado principalmente en las bacterias sin dejar de ser en gran medida no caracterizados en las c&#xE9;lulas eucariotas. Recientemente se ha sugerido que la poli-P metabolismo est&#xE1; conectado a la de las especies altamente inositol fosforilado (pirofosfatos inositol). Pirofosfatos inositol son mol&#xE9;culas en las que los grupos fosfato son m&#xE1;s numerosos que los &#xE1;tomos de carbono. Aqu&#xED;, mostramos que los mutantes de levadura en ciernes que no pueden producir pirofosfatos inositol tener niveles indetectables de poli-P. Nuestros resultados sugieren un paralelismo metab&#xF3;lica m&#xE1;s importante entre estas dos mol&#xE9;culas altamente fosforilados. M&#xE1;s importante a&#xFA;n, nos demuestran que DDP1, que codifica fosfohidrolasa diadenosine y diphosphoinositol, posee una s&#xF3;lida actividad de poli-P endopolyphosphohydrolase. Adem&#xE1;s, se demuestra que esta es una caracter&#xED;stica conservada evolutivamente, ya miembros de la familia de mam&#xED;feros NUDIX hidrolasa, los tres Ddp1 hom&#xF3;logos en las c&#xE9;lulas humanas (DIPP1, DIPP2 y DIPP3), tambi&#xE9;n son capaces de degradar poli-P.
