DNA vaccination is an effective means of eliciting both humoral and cellular immunity. Most of influenza vaccines targeted at hemagglutinin (HA) show efficient immunogenicity for protecting subjects against influenza virus infection. However, major antigenic variations of HA may facilitate the virus in developing resistance against such vaccines. DNA vaccines encoding conserved antigens protect animals against diverse viral subtypes, but their potency requires further improvement. In the present study, a DNA vaccine encoding the conserved nucleoprotein (NP) with a tissue plasminogen activator (tPA) signal sequence (ptPAs/NP) was generated, and immune responses were examined in vaccinated mice. A higher level of NP expression and secretion was observed in lysates and supernatants of the cells transfected with ptPAs/NP when compared to a plasmid encoding the wild-type full-length NP (pflNP). Immunofluorescence studies showed the cytoplasmic localization of the NP protein expressed from ptPAs/NP, but not from pflNP. In mice, the ptPAs/NP vaccine elicited higher levels of the NP-specific IgG and CD8(+) T cell-stimulating responses than that of pflNP. Vaccination with ptPAs/NP efficiently cleared the homologous H5N1 influenza virus in the infected lungs and induced partial cross-protection against heterologous, highly pathogenic H5N1 strains in mice. Our results may contribute to the development of protective immunity against diverse, highly pathogenic H5N1 virus subtypes.

La vaccination par ADN est un moyen efficace de susciter &#xE0; la fois humorale et l&#x26;#39;immunit&#xE9; cellulaire. La plupart des vaccins contre la grippe destin&#xE9;s aux h&#xE9;magglutinine (HA) montrent immunog&#xE9;nicit&#xE9; efficace pour prot&#xE9;ger les sujets contre les virus de la grippe. Cependant, les variations antig&#xE9;niques majeures de HA peut faciliter le virus dans le d&#xE9;veloppement de la r&#xE9;sistance contre ces vaccins. Les vaccins &#xE0; ADN codant pour les antig&#xE8;nes conserv&#xE9;s prot&#xE9;ger les animaux contre diverses sous-types viraux, mais leur puissance doit encore &#xEA;tre am&#xE9;lior&#xE9;. Vacunaci&#xF3;n de ADN es un medio eficaz de provocar tanto humoral y la inmunidad celular. La mayor&#xED;a de vacunas contra la influenza dirigidos a la hemaglutinina (HA) muestran inmunogenicidad eficaz para proteger a los sujetos contra la infecci&#xF3;n por virus de la influenza. Sin embargo, grandes variaciones antig&#xE9;nicas de HA puede facilitar el virus en el desarrollo de resistencia contra dichas vacunas. Las vacunas de ADN que codifican ant&#xED;genos conservados proteger a los animales contra diversos subtipos virales, pero su potencia requiere m&#xE1;s mejoras. En el presente estudio, una vacuna de ADN que codifica para la nucleoprote&#xED;na conservado (NP), con una secuencia de activador tisular del plasmin&#xF3;geno de la se&#xF1;al (tPA) (ptPAs / NP) se ha generado, y la respuesta inmune se examinaron en ratones vacunados. Using the complete genome sequences of 35 classical swine fever viruses (CSFV) representing all three genotypes and all three kinds of virulence, we analyzed synonymous codon usage and the relative dinucleotide abundance in CSFV. The general correlation between base composition and codon usage bias suggests that mutational pressure rather than natural selection is the main factor that determines the codon usage bias in CSFV. Furthermore, we observed that the relative abundance of dinucleotides in CSFV is independent of the overall base composition but is still the result of differential mutational pressure, which also shapes codon usage. In addition, other factors, such as the subgenotypes and aromaticity, also influence the codon usage variation among the genomes of CSFV. This study represents the most comprehensive analysis to date of CSFV codon usage patterns and provides a basic understanding of the mechanisms for codon usage bias.

DNA vaccination is an effective means of eliciting both humoral and cellular immune responses. The hemagglutinin (HA) surface protein of influenza A virus is a major target of protective antibody responses induced by virus infection or by vaccination and is widely considered to be the antigen of choice for an influenza vaccine. Cytotoxic T lymphocyte (CTL) responses directed against the conserved nucleoprotein (NP) are thought to play an important role in clearing virus and promoting survival and recovery from influenza. In this study, we developed a novel DNA vaccine approach using a chimeric plasmid consisting of the HA of H5N1 influenza virus in which an MHC class I-restricted NP-specific CTL epitope (NP147-155) was inserted. Immunogenicity and antiviral efficacy of this vaccine was assessed in mouse models. A similar level of HA expression was achieved in 293T cells transfected with pHA/NP(147-155) compared to that with pHA. Besides eliciting the specific anti-HA antibody responses, vaccination using pHA/NP(147-155) in mice induced NP epitope-specific CD8(+) T cell responses, which are generally not inducible by vaccination with pHA alone. After H5N1 influenza virus challenge, BALB/c mice vaccinated with pHA/NP(147-155) exhibited reduced inflammation severity and lung viral titers compared to those vaccinated with pHA. Our work may contribute to improvement of HA-based influenza DNA vaccines.

Enhanced protective immunity against H5N1 influenza virus challenge by vaccination with DNA expressing a chimeric hemagglutinin in combination with an MHC class I-restricted epitope of nucleoprotein in mice.

La vaccination par ADN est un moyen efficace de susciter &#xE0; la fois humorales et cellulaires des r&#xE9;ponses immunitaires. La prot&#xE9;ine h&#xE9;magglutinine de surface (HA) du virus grippal A est un objectif majeur des r&#xE9;ponses d&#x26;#39;anticorps protecteurs induits par une infection virale ou par la vaccination et est largement consid&#xE9;r&#xE9; comme l&#x26;#39;antig&#xE8;ne de choix pour un vaccin contre la grippe. Lymphocytes T cytotoxiques (CTL) dirig&#xE9;s contre les r&#xE9;ponses de la nucl&#xE9;oprot&#xE9;ine conserv&#xE9; (NP) sont suppos&#xE9;s jouer un r&#xF4;le important dans l&#x26;#39;&#xE9;limination du virus et la promotion de la survie et le r&#xE9;tablissement de la grippe. Dans cette &#xE9;tude, nous avons d&#xE9;velopp&#xE9; une approche de l&#x26;#39;ADN roman vaccin en utilisant un plasmide chim&#xE9;rique constitu&#xE9; de la HA du virus de la grippe H5N1 dans lequel un CMH de classe I-restreint NP-CTL sp&#xE9;cifique &#xE9;pitope (NP147-155) a &#xE9;t&#xE9; ins&#xE9;r&#xE9;. Immunog&#xE9;nicit&#xE9; et l&#x26;#39;efficacit&#xE9; antivirale de ce vaccin a &#xE9;t&#xE9; &#xE9;valu&#xE9;e dans des mod&#xE8;les murins. Un niveau similaire d&#x26;#39;expression HA a &#xE9;t&#xE9; r&#xE9;alis&#xE9;e dans des cellules 293T transfect&#xE9;es avec de la PHA / NP (147-155) par rapport &#xE0; celle avec de la PHA. Outre susciter des r&#xE9;ponses sp&#xE9;cifiques d&#x26;#39;anticorps anti-HA, la vaccination &#xE0; l&#x26;#39;aide PHA / NP (147-155) chez la souris induit NP &#xE9;pitope CD8 sp&#xE9;cifiques (+) les r&#xE9;ponses des cellules T, qui ne sont g&#xE9;n&#xE9;ralement pas inductible par la vaccination avec de la PHA seule. Apr&#xE8;s la grippe d&#xE9;fi virus H5N1, souris BALB / c vaccin&#xE9;es avec de la PHA / NP (147-155) ont montr&#xE9; la gravit&#xE9; l&#x26;#39;inflammation r&#xE9;duite et les titres du poumon virales par rapport &#xE0; ceux vaccin&#xE9;s avec de la PHA. Notre travail peut contribuer &#xE0; l&#x26;#39;am&#xE9;lioration de la base d&#x26;#39;HA vaccins &#xE0; ADN de la grippe.

DNA-Impfung ist ein wirksames Mittel zur sowohl humorale und zellul&auml;re Immunantworten hervorrufen. Das H&auml;magglutinin (HA) Oberfl&auml;che Protein des Influenza-A-Virus ist ein zentrales Ziel der sch&uuml;tzende Antik&ouml;rperantworten induziert durch Infektion oder durch Impfung und gilt weithin als das Antigen der Wahl f&uuml;r eine Influenza-Impfstoff. Zytotoxische T-Lymphozyten (CTL) Antworten gegen die konservierten Nukleoprotein (NP) werden gedacht, um eine wichtige Rolle im clearing Virus und F&ouml;rderung des &Uuml;berlebens und Erholung von Grippe. In dieser Studie entwickelten wir einen Roman DNA-Impfstoff-Ansatz mit einem Chim&auml;ren Plasmid, bestehend aus HA des Influenza-Virus H5N1 in die eine MHC Klasse ich beschr&auml;nkt NP-spezifische CTL Epitope (NP147-155) eingef&uuml;gt wurde. Immunogenit&auml;t und antivirale Wirksamkeit von diesen Impfstoff wurde in Mausmodelle bewertet. Ein &auml;hnliches Ma&szlig; an HA Ausdruck wurde im 293T erreicht, die transfected Zellen mit pHA/NP(147-155) gegen&uuml;ber, die mit pHA. Le risposte dei linfociti T citotossici (CTL) dirette contro il conservato nucleoproteina (NP) sono pensate di giocare un ruolo importante nel recupero dall&#39;influenza e virus e promuovere la sopravvivenza di compensazione. In questo studio, abbiamo sviluppato un approccio di vaccinazione del DNA usando un plasmide chimerico costituito l&#39;HA del virus dell&#39;influenza H5N1 in cui &egrave; stato inserito un MHC classe I-limitato NP-specifici CTL epitopo (NP147-155) romanzo. La vacuna de ADN es un medio eficaz de obtener respuestas inmunes humoral y celular. La hemaglutinina (HA) prote&#xED;na de superficie del virus de influenza A es el principal objetivo de las respuestas de anticuerpos protectores inducidos por la infecci&#xF3;n por el virus o por vacunaci&#xF3;n y es ampliamente considerado como el ant&#xED;geno de elecci&#xF3;n para una vacuna contra la influenza. Linfocitos T citot&#xF3;xicos (CTL) dirigidos contra la nucleoprote&#xED;na conservado (NP) se cree que desempe&#xF1;an un papel importante en la limpieza de virus y promover la supervivencia y la recuperaci&#xF3;n de la influenza. En este estudio, hemos desarrollado un enfoque de ADN nueva vacuna usando un pl&#xE1;smido quim&#xE9;rico que consiste en la HA del virus de la influenza H5N1 en el que se inserta un MHC de clase I-NP-espec&#xED;fica restringido ep&#xED;topo CTL (NP147-155). La inmunogenicidad y la eficacia antiviral de esta vacuna se evalu&#xF3; en modelos de rat&#xF3;n. Un nivel similar de la expresi&#xF3;n de HA se logr&#xF3; en las c&#xE9;lulas 293T transfectadas con PHA / NP (147-155) en comparaci&#xF3;n con con PHA. Despu&#xE9;s de la exposici&#xF3;n el virus H5N1 de la gripe, ratones Balb / c vacunados con PHA / NP (147-155) mostraron la gravedad de la inflamaci&#xF3;n y reduce los t&#xED;tulos pulmonares virales en comparaci&#xF3;n con los vacunados con PHA. Highly pathogenic avian influenza H5N1 virus represents a growing threat for an influenza pandemic. Development of effective vaccines for H5N1 is a priority for pandemic preparedness. Focusing on influenza virus-like particles (VLPs) has been suggested as a promising vaccine approach. Recent VLP vaccination efforts have been concentrated on the H5N1 strains isolated from humans. Because all confirmed cases of human H5N1 infection were directly transmitted from infected poultry, it is of interest to develop VLP vaccines comprised of antigenic proteins of avian H5N1 strains in order to compare their efficacy in fighting diverse H5N1 strains with vaccines developed using human isolates. In this study, we generated a VLP vaccine composed of the HA, NA, and M1 proteins of the avian H5N1 influenza virus isolate A/chicken/Hubei/489/2004, which seems to occupy a unique phylogenetic position; it belongs to neither clade 1 nor clade 2. Upon infection of Sf9 insect cells using recombinant baculoviruses, the co-expressed HA, NA, and M1 proteins self-assembled and released into the culture medium as VLPs. In a mouse model, purified VLPs elicited an effective antibody response and conferred complete protection against heterologous human H5N1 influenza virus, as well as a homologous avian H5N1 influenza virus isolate. Our work provides further evidence that vaccination with influenza VLPs may be a productive approach to achieve protection against diverse H5N1 strains.

D&#xE9;veloppement de vaccins efficaces pour le virus H5N1 est une priorit&#xE9; pour la pr&#xE9;paration aux pand&#xE9;mies. En se concentrant sur la grippe virus-like particles (VLP) a &#xE9;t&#xE9; sugg&#xE9;r&#xE9; comme une approche vaccin prometteur. Les r&#xE9;cents efforts de vaccination VLP ont &#xE9;t&#xE9; concentr&#xE9;s sur les souches de virus H5N1 isol&#xE9;s chez l&#x26;#39;homme. Parce que tous les cas confirm&#xE9;s d&#x26;#39;infection humaine par le H5N1 ont &#xE9;t&#xE9; transmis directement de la volaille infect&#xE9;e, il est int&#xE9;ressant de d&#xE9;velopper des vaccins VLP comprenant des prot&#xE9;ines antig&#xE9;niques de souches de virus H5N1 aviaire, afin de comparer leur efficacit&#xE9; dans la lutte contre diverses souches de virus H5N1 avec des vaccins d&#xE9;velopp&#xE9;s &#xE0; partir d&#x26;#39;isolats humains. Konzentration auf Grippe Virus-&auml;hnliche Partikel (VLPs) wurde als ein vielversprechender Impfstoff Ansatz vorgeschlagen. Die j&uuml;ngsten Bem&uuml;hungen der VLP-Impfung haben auf die H5N1-St&auml;mme isoliert von Menschen konzentriert. Da alle best&auml;tigte F&auml;lle von H5N1 Infektion direkt von infizierten Gefl&uuml;gel &uuml;bertragen wurden, ist es von Interesse, VLP-Vakzine bestehend aus Antigene Proteine der Vogelgrippe H5N1-St&auml;mme um ihre Wirksamkeit im Kampf gegen diverse H5N1-St&auml;mme mit Impfstoffen, die mit menschliche Isolaten entwickelt zu vergleichen zu entwickeln. In dieser Studie wir generiert einen VLP-Impfstoff, bestehend aus HA, NA, und M1 Proteine des Vogelgrippevirus H5N1 Influenza-Virus A/Huhn/Hubei/489/2004, die anscheinend eine einzigartige phylogenetische Position einnehmen zu isolieren; Es geh&ouml;rt zur Gruppe 1 weder Klade 2. Sforzi di vaccinazione recenti VLP sono concentrati su ceppi H5N1 isolati dagli esseri umani. Los virus altamente pat&#xF3;genos de la gripe aviar H5N1 representa una amenaza creciente para una pandemia de influenza. Desarrollo de vacunas eficaces contra el virus H5N1 es una prioridad para la preparaci&#xF3;n para una pandemia. Centr&#xE1;ndose en la gripe part&#xED;culas similares a virus (VLP) ha sido sugerido como un enfoque de la vacuna prometedora. Los recientes esfuerzos de vacunaci&#xF3;n VLP se han concentrado en las cepas H5N1 aisladas en seres humanos. Debido a que todos los casos confirmados de infecci&#xF3;n humana por virus H5N1 se transmite directamente de aves de corral infectadas, es de inter&#xE9;s para desarrollar vacunas VLP compuestas de prote&#xED;nas antig&#xE9;nicas de las cepas de H5N1 aviar a fin de comparar su eficacia en la lucha contra diversas cepas del virus H5N1 con vacunas desarrolladas con las cepas humanas. En este estudio, se gener&#xF3; una vacuna VLP compuesto de la HA, NA, y las prote&#xED;nas M1 del virus de la gripe aviar H5N1 aislado A/chicken/Hubei/489/2004, que parece ocupar una posici&#xF3;n filogen&#xE9;tica &#xFA;nica, que no pertenece a ninguna clado 1 ni el clado 2. Tras la infecci&#xF3;n de c&#xE9;lulas de insecto Sf9 utilizando baculovirus recombinantes, el HA co-expresado, NA, y las prote&#xED;nas M1 auto-ensamblado y se libera en el medio de cultivo como VLP. As an adaptor molecule in the retinoic acid-inducible gene-I (RIG-I) signaling pathway, the virus-induced signaling adaptor (VISA) molecule activates NF-κB and IRF3 and thereby leads to the production of type I interferons (IFNs). To explore the potential of VISA as a genetic adjuvant for DNA vaccines, a eukaryotic expression plasmid, pVISA, was generated by cloning the VISA gene into the pVAX1vector. For comparison, the pTRIF plasmid was similarly constructed, encoding the known genetic adjuvant TRIF (TIR-domain-containing adapter-inducing interferon-β), an adapter in the Toll-like receptor (TLR) signaling pathway. Mice were immunized with the chimeric DNA vaccine pHA/NP(147-155), which encodes the HA (hemagglutinin) fused with NP (nucleoprotein) CTL epitope (NP(147-155)) of H5N1 influenza virus, either alone or in combination with pVISA or pTRIF. Antigen-specific immune responses were examined in immunized mice. Our results demonstrate that co-immunization of the pHA/NP(147-155) plasmid with the VISA adjuvant augmented DNA-raised cellular immune responses and provided protection against H5N1 influenza virus challenge in mice. In addition, our data suggest that VISA acts as a stronger adjuvant for DNA immunization than TRIF. We conclude that co-inoculation with a vector expressing the adaptor molecule VISA enhanced the protective immunity against H5N1 infection induced by pHA/NP(147-155) and that VISA could be developed as a novel genetic adjuvant for DNA vaccines.

The virus-induced signaling adaptor molecule enhances DNA-raised immune protection against H5N1 influenza virus infection in mice.

Prenatal dexamethasone exposure (PDE) induces multiorgan developmental toxicities in offspring. Here we verified the transgenerational inheritance effect of ovarian developmental toxicity by PDE and explored its intrauterine programming mechanism. Pregnant rats subcutaneously received 0.2 mg/kg/d dexamethasone from gestational day (GD) 9 to GD20. A subgroup was euthanized for fetuses on GD20, and the other group went on to spontaneous labor to produce F1 offspring. The adult F1 females were mated with normal males to produce the F2 and F3 generations. The PDE fetal rats exhibited ovarian mitochondrial structural abnormalities, decreased serum estradiol (E2) levels, and lower expression levels of ovarian steroidogenic factor 1 (SF1), steroidal synthetases, and insulinlike growth factor 1 (IGF1). On postnatal week (PW) 6 and PW12, the PDE F1 offspring showed altered reproductive behavior and ovarian morphology. The serum E2 level and ovarian expression of SF1, steroidal synthetases, and IGF1 were also decreased. The adult F3 offspring showed alterations in reproductive phenotype and ovarian IGF1, SF1, and steroidal synthetase expression similar to those of F1. PDE induces ovarian developmental toxicity and transgenerational inheritance effects. The mechanism by which this toxicity occurs may be related to PDE-induced low-functional programming of fetal ovarian IGF1/SF1 and steroidal synthetases.

Prenatal Dexamethasone Exposure Induced Ovarian Developmental Toxicity and Transgenerational Effect in Rat Offspring.

Meiotic recombination permits exchange of genetic material between homologous chromosomes. The replication protein A (RPA) complex, the predominant ssDNA-binding complex, is required for nearly all aspects of DNA metabolism, but its role in mammalian meiotic recombination remains unknown due to the embryonic lethality of RPA mutant mice. RPA is a heterotrimer of RPA1, RPA2, and RPA3. We find that loss of RPA1, the largest subunit, leads to disappearance of RPA2 and RPA3, resulting in the absence of the RPA complex. Using an inducible germline-specific inactivation strategy, we find that loss of RPA completely abrogates loading of RAD51/DMC1 recombinases to programmed meiotic DNA double strand breaks, thus blocking strand invasion required for chromosome pairing and synapsis. Surprisingly, loading of MEIOB, SPATA22, and ATR to DNA double strand breaks is RPA-independent and does not promote RAD51/DMC1 recruitment in the absence of RPA. Finally, inactivation of RPA reduces crossover formation. Our results demonstrate that RPA plays two distinct roles in meiotic recombination: an essential role in recombinase recruitment at early stages and an important role in promoting crossover formation at later stages.

Dual functions for the ssDNA-binding protein RPA in meiotic recombination.

RNA regulation by RNA-binding proteins (RBPs) involve extremely complicated mechanisms. MOV10 and MOV10L1 are two homologous RNA helicases implicated in distinct intracellular pathways. MOV10L1 participates specifically in Piwi-interacting RNA (piRNA) biogenesis and protects mouse male fertility. In contrast, the functional complexity of MOV10 remains incompletely understood, and its role in the mammalian germline is unknown. Here, we report a study of the biological and molecular functions of the RNA helicase MOV10 in mammalian male germ cells.
