&#x53D1;&#x5C55;&#x548C;&#x73AF;&#x5883;&#x4FE1;&#x53F7;&#x8BF1;&#x5BFC; C4 Pepc &#x57FA;&#x56E0;&#x5728;&#x7389;&#x7C73;&#x4E2D;&#x7684;&#x8FDC;&#x7AEF;&#x548C;&#x8FD1;&#x7AEF;&#x542F;&#x52A8;&#x5B50;&#x5143;&#x7D20;&#x4E0A;&#x7684;&#x4E0D;&#x540C;&#x7684;&#x7EC4;&#x86CB;&#x767D;&#x4E59;&#x9170;&#x5316;&#x914D;&#x7F6E;&#x6587;&#x4EF6;&#x3002;

The maize C(4)-Pepc gene is expressed in an organ- and cell-type-specific manner, inducible by light and modulated by nutrient availability and the metabolic state of the cell. We studied the contribution of histone acetylation at five lysine residues to the integration of these signals into a graduated promoter response. In roots and coleoptiles, where the gene is constitutively inactive, three of the five lysines were acetylated and the modifications showed unique patterns with respect to their distribution on the gene. A similar pattern was observed in etiolated leaves, where the gene is poised for activation by light. Here, illumination selectively induced the acetylation of histone H4 lysine 5 and histone H3 lysine 9 in both the promoter and the transcribed region, again with unique distribution patterns. Induction was independent of transcription and fully reversible in the dark. Nitrate and hexose availability modulated acetylation of all five lysines restricted to a distal promoter region, whereas proximal promoter acetylation was highly resistant to these stimuli. Our data suggest that light induction of acetylation is controlled by regulating HDAC activity, whereas metabolic signals regulate HAT activity. Acetylation turnover rates were high in the distal promoter and the transcribed regions, but low on the proximal promoter. On the basis of these results, we propose a model with three levels of stimulus-induced histone modifications that collectively adjust promoter activity. The results support a charge neutralization model for the distal promoter and a stimulus-mediated, but transcription-independent, histone acetylation pattern on the core promoter, which might be part of a more complex histone code.

Developmental and environmental signals induce distinct histone acetylation profiles on distal and proximal promoter elements of the C4-Pepc gene in maize.

Ma&iuml;s C 4-Pepc g&egrave;ne s&#39;exprime d&#39;une mani&egrave;re d&#39;orgue et cellule-type-sp&eacute;cifiques, inductible par la lumi&egrave;re et modul&eacute;e par la disponibilit&eacute; des nutriments et de l&#39;&eacute;tat m&eacute;tabolique de la cellule. Nous avons &eacute;tudi&eacute; la contribution de l&#39;ac&eacute;tylation des histones &agrave; cinq r&eacute;sidus de lysine &agrave; l&#39;int&eacute;gration de ces signaux dans une r&eacute;ponse gradu&eacute;e promoteur. Dans les racines et les col&eacute;optiles, o&ugrave; le g&egrave;ne est constitutivement inactif, trois des cinq lysines sont ac&eacute;tyl&eacute;s et les modifications ont montr&eacute; des profils uniques en ce qui concerne leur r&eacute;partition sur le g&egrave;ne. Sur la base de ces r&eacute;sultats, nous proposons un mod&egrave;le avec trois niveaux des modifications d&#39;histone induite par la stimulation qui s&#39;ajustent collectivement activit&eacute; du promoteur. Les r&eacute;sultats confirment un mod&egrave;le de neutralisation de charge pour le promoteur distal et un motif d&#39;ac&eacute;tylation des histones induite par la stimulation, mais ind&eacute;pendant de la transcription, sur le promoteur du core, qui peut-&ecirc;tre faire partie d&#39;un code histone plus complexe.

Les signaux environnementaux et de d&eacute;veloppement induisent des profils ac&eacute;tylation histone distinctes sur les &eacute;l&eacute;ments du promoteur distale et proximale du g&egrave;ne C4-Pepc dans le ma&iuml;s.

Das Maize C 4-Pepc gen dr&uuml;ckt sich in einer Orgel und Zelle-Typ-spezifische Weise durch Induktion durch Licht und modulierte von N&auml;hrstoff-Verf&uuml;gbarkeit und die metabolischen Zustand der Zelle. Wir studierten den Beitrag der Histon-Acetylierung auf f&uuml;nf Lysin R&uuml;ckst&auml;nde zur Integration dieser Signale in einer abgestuften Projekttr&auml;ger-Antwort. Unsere Daten zeigen, dass Licht Induktion der Acetylierung durch Regulierung HDAC-Aktivit&auml;t, kontrolliert wird, w&auml;hrend die metabolische Signale Hut-Aktivit&auml;t zu regulieren. Entwicklungs- und umweltpolitischen Signale induzieren unterschiedliche Histon Acetylierung Profile auf distale und proximale Promotor Elemente im C4-Pepc-gen in Mais.

Il mais C (4)-Pepc gene &egrave; espresso in maniera organo e cellula-tipo-specifici, inducibile da luce e modulato dalla disponibilit&agrave; dei nutrienti e lo stato metabolico della cellula. Abbiamo studiato il contributo di acetilazione dell&#39;istone presso cinque residui di lisina per l&#39;integrazione di questi segnali in una risposta graduata promotore. Nelle radici e Coleottili, dove il gene &egrave; costitutivamente attivo, tre le cinque Lisine erano acetilato e le modifiche hanno mostrato modelli unici per quanto riguarda la loro distribuzione sul gene. Un modello simile &egrave; stato osservato nelle foglie etiolated, dove il gene &egrave; pronta per l&#39;attivazione di luce. Qui, illuminazione indotta selettivamente l&#39;acetilazione della lisina di istone H4 5 e istone H3 lisina 9 nella regione trascritta, nuovo con modelli di distribuzione esclusiva e promotore. Acetilazione fatturato tassi erano alti in regioni trascritte e promotore distale, ma bassa sul promotore prossimale. El ma&iacute;z C 4-Pepc gen se expresa de una manera de &oacute;rgano - y c&eacute;lula-tipo-espec&iacute;ficas, inducible por la luz y modulada por la disponibilidad de nutrientes y el estado metab&oacute;lico de la c&eacute;lula. Estudiamos la contribuci&oacute;n de la acetilaci&oacute;n de histonas en cinco residuos de lisina para la integraci&oacute;n de estas se&ntilde;ales en una respuesta graduada del promotor. En ra&iacute;ces y cole&oacute;ptilos, donde el gen es constitutivamente inactivo, tres de los cinco lisinas fueron acetilado y las modificaciones mostraron patrones &uacute;nicos con respecto a su distribuci&oacute;n en el gen. Un patr&oacute;n similar se observ&oacute; en hojas descoloridas, donde el gen est&aacute; preparado para la activaci&oacute;n por la luz. Aqu&iacute;, la iluminaci&oacute;n hab&iacute;a inducida selectivamente la acetilaci&oacute;n de lisina de histona H4 5 y la histona H3 lisina 9 en el promotor y la regi&oacute;n transcrita, otra vez con los patrones de distribuci&oacute;n &uacute;nica. Inducci&oacute;n era independiente de la transcripci&oacute;n y completamente reversible en la oscuridad. Se&ntilde;ales de desarrollo y medio ambientales inducen perfiles de acetilaci&oacute;n de histonas distintas en elementos distal y proximal del promotor del gen C4-Pepc en ma&iacute;z.

In plants and animals, induced resistance (IR) to biotic and abiotic stress is associated with priming of cells for faster and stronger activation of defense responses. It has been hypothesized that cell priming involves accumulation of latent signaling components that are not used until challenge exposure to stress. However, the identity of such signaling components has remained elusive. Here, we show that during development of chemically induced resistance in Arabidopsis thaliana, priming is associated with accumulation of mRNA and inactive proteins of mitogen-activated protein kinases (MPKs), MPK3 and MPK6. Upon challenge exposure to biotic or abiotic stress, these two enzymes were more strongly activated in primed plants than in nonprimed plants. This elevated activation was linked to enhanced defense gene expression and development of IR. Strong elicitation of stress-induced MPK3 and MPK6 activity is also seen in the constitutive priming mutant edr1, while activity was attenuated in the priming-deficient npr1 mutant. Moreover, priming of defense gene expression and IR were lost or reduced in mpk3 or mpk6 mutants. Our findings argue that prestress deposition of the signaling components MPK3 and MPK6 is a critical step in priming plants for full induction of defense responses during IR.

Tras la exposici&#xF3;n desaf&#xED;o a estr&#xE9;s bi&#xF3;tico o abi&#xF3;tico, estas dos enzimas fueron m&#xE1;s fuertemente activado en las plantas de imprimaci&#xF3;n que en las plantas nonprimed. Esta activaci&#xF3;n elevada se relacion&#xF3; con una mayor expresi&#xF3;n de genes de defensa y el desarrollo de la IR. Priming of defence genes for amplified response to secondary stress can be induced by application of the plant hormone salicylic acid or its synthetic analogue acibenzolar S-methyl. In this study, we show that treatment with acibenzolar S-methyl or pathogen infection of distal leaves induce chromatin modifications on defence gene promoters that are normally found on active genes, although the genes remain inactive. This is associated with an amplified gene response on challenge exposure to stress. Mutant analyses reveal a tight correlation between histone modification patterns and gene priming. The data suggest a histone memory for information storage in the plant stress response.
