The development of modern neuroscience tools is critical for deciphering brain circuit organization and function. An important aspect for technical development is to enhance each technique's advantages and compensate for limitations. We developed a high-precision and fast functional mapping technique in brain slices that incorporates the spatial precision of activation that can be achieved by laser-scanning photostimulation with rapid and high-temporal resolution assessment of evoked network activity that can be achieved by voltage-sensitive dye imaging. Unlike combination of whole cell recordings with photostimulation for mapping local circuit inputs to individually recorded neurons, this innovation is a new photostimulation-based technique to map cortical circuit output and functional connections at the level of neuronal populations. Here we report on this novel technique in detail and show its effective applications in mapping functional connections and circuit dynamics in mouse primary visual cortex and hippocampus. Given that this innovation enables rapid mapping and precise evaluation of cortical organization and function, it can have broad impacts in the field of cortical circuitry.

Le d&#xE9;veloppement des neurosciences modernes outils est essentielle pour d&#xE9;chiffrer l&#x26;#39;organisation des circuits c&#xE9;r&#xE9;braux et de la fonction. Un aspect important pour le d&#xE9;veloppement technique est de renforcer les avantages de chaque technique et de compenser les limitations. Nous avons d&#xE9;velopp&#xE9; une technique de cartographie de haute pr&#xE9;cision et rapide fonctionnelle des tranches de cerveau qui int&#xE8;gre la pr&#xE9;cision spatiale de l&#x26;#39;activation qui peut &#xEA;tre atteint par balayage laser photostimulation &#xE0; l&#x26;#39;&#xE9;valuation la r&#xE9;solution rapide et de haute-temporelle de l&#x26;#39;activit&#xE9; du r&#xE9;seau &#xE9;voqu&#xE9;e qui peut &#xEA;tre atteint par une tension sensible colorant d&#x26;#39;image. Contrairement &#xE0; la combinaison des enregistrements de cellules enti&#xE8;res avec photostimulation pour la cartographie des entr&#xE9;es des circuits locaux de neurones enregistr&#xE9;s individuellement, cette innovation est une nouvelle technique bas&#xE9;e sur la photostimulation pour cartographier de sortie du circuit cortical et des connexions fonctionnelles au niveau des populations neuronales. Nous rapportons ici sur cette nouvelle technique en d&#xE9;tail et de montrer ses applications efficaces dans la cartographie des liens fonctionnels et de la dynamique de circuits dans le cortex visuel primaire de la souris et l&#x26;#39;hippocampe. &#xC9;tant donn&#xE9; que cette innovation permet la cartographie rapide et l&#x26;#39;&#xE9;valuation pr&#xE9;cise de l&#x26;#39;organisation corticale et la fonction, il peut avoir de vastes r&#xE9;percussions dans le domaine des circuits corticaux.

Die Entwicklung der modernen Neurowissenschaften Tools ist wichtig f&uuml;r dechiffrierende Gehirn Schaltung Organisation und Funktion. Wir entwickelten eine hochpr&auml;zise und schnelle funktionelle Zuordnung-Technik in Gehirn-Scheiben, die die r&auml;umliche Pr&auml;zision der Aktivierung beinhaltet, die von Laserscanning erreicht werden kann Photostimulation mit schnellen und hoch-zeitlicher Aufl&ouml;sung Bewertung der evozierten Netzwerkaktivit&auml;t, die durch Spannung-empfindlichen Farbstoff-Bildgebung erreicht werden kann. Im Gegensatz zur Kombination der ganzen Zelle aufgenommen Aufnahmen mit Photostimulation f&uuml;r die Zuordnung von Rundkurs Eingaben individuell Neuronen, diese Neuerung ist eine neue Photostimulation basierende Technik kortikale Schaltung Ausgang und funktionale Verbindungen auf der Ebene der neuronalen Bev&ouml;lkerungen zugeordnet. Hier wir Bericht &uuml;ber diese neuartige Technik im Detail und zeigen ihre effektiven Anwendungen in der mapping-funktioneller Verbindungen und Schaltung Dynamik in Maus prim&auml;ren visuellen Kortex und Hippocampus. Angesichts der Tatsache, dass diese Innovation schnelle Zuordnung und pr&auml;zise Auswertung von kortikale Organisation und Funktion erm&ouml;glicht, kann es im Bereich der kortikalen Schaltung gro&szlig;e Auswirkungen haben.

Lo sviluppo di strumenti di neuroscienza moderna &egrave; critico per funzione e organizzazione di circuiti cerebrali decifrazione. Un aspetto importante per lo sviluppo tecnico &egrave; quello di valorizzare i vantaggi di ciascuna tecnica e compensare per le limitazioni. Abbiamo sviluppato una tecnica di mappatura ad alta precisione e veloce funzionale in fettine di cervello che incorpora la precisione spaziale di attivazione che pu&ograve; essere ottenuto mediante scansione laser fotostimolazione con valutazione rapida e temporale ad alta risoluzione dell&#39;attivit&agrave; di rete evocati che pu&ograve; essere raggiunto da imaging sensibile alla tensione di tintura. A differenza della combinazione dell&#39;intera cellula registrazioni con fotostimolazione per mappatura singolarmente gli ingressi del circuito locale a registrarono i neuroni, questa innovazione &egrave; una nuova tecnica di fotostimolazione a mappa uscita circuito corticale e connessioni funzionali a livello delle popolazioni neuronali. Qui abbiamo relazione su questa tecnica di romanzo in dettaglio e mostrare le applicazioni efficace in mappatura connessioni funzionali e dinamiche del circuito in mouse corteccia visiva primaria e ippocampo. Dato che questa innovazione consente la mappatura rapida e precisa valutazione dell&#39;organizzazione corticale e funzione, pu&ograve; avere impatti vasti nel campo dei circuiti corticali.

El desarrollo de herramientas de la neurociencia moderna es fundamental para descifrar la organizaci&#xF3;n del cerebro y la funci&#xF3;n del circuito. Un aspecto importante para el desarrollo t&#xE9;cnico es mejorar las ventajas de cada t&#xE9;cnica y compensar las limitaciones. Hemos desarrollado una t&#xE9;cnica de alta precisi&#xF3;n y r&#xE1;pido mapeo funcional en rodajas de cerebro que incorpora la precisi&#xF3;n espacial de activaci&#xF3;n que se puede lograr por fotoestimulaci&#xF3;n l&#xE1;ser de barrido con la evaluaci&#xF3;n r&#xE1;pida resoluci&#xF3;n y alta-temporal de la actividad de red evocado que se puede lograr por sensibles al voltaje colorante de imagen. A diferencia de combinaci&#xF3;n de las grabaciones de c&#xE9;lulas enteras con fotoestimulaci&#xF3;n para el mapeo de las entradas a las neuronas de circuitos locales registrados individualmente, esta innovaci&#xF3;n es una nueva t&#xE9;cnica basada en fotoestimulaci&#xF3;n para asignar salida del circuito cortical y conexiones funcionales en el nivel de poblaciones neuronales. Aqu&#xED; nos informe sobre esta novedosa t&#xE9;cnica en detalle y mostrar sus aplicaciones efectivas en el mapeo de las conexiones funcionales y la din&#xE1;mica de circuitos en la corteza visual primaria del rat&#xF3;n y el hipocampo. Teniendo en cuenta que esta innovaci&#xF3;n permite un r&#xE1;pido mapeo y evaluaci&#xF3;n precisa de la organizaci&#xF3;n cortical y la funci&#xF3;n, puede tener efectos amplios en el campo de los circuitos corticales.
