Profiles of antibodies to the nucleocapsid protein of the severe acute respiratory syndrome (SARS)-associated coronavirus in 445 probable SARS patients and 3,749 healthy people or non-SARS patients were analyzed by antigen-capturing enzyme-linked immunosorbent assay. Antinucleocapsid antibodies were elucidated in 17.5% of the probable SARS patients 1 to 7 days after the onset of symptoms and in 80% of the patients 8 to 14 days after the onset. About 90% of the probable SARS patients were positive 15 or more days after illness. Antibody titers increased up to 70 days, and high antibody titers were maintained at least for another 3 months. Of the healthy people and non-SARS patients, only seven (0.187%) were weakly positive.

Efficient and dependable methods for detection and measurement of synaptic events are important for studies of synaptic physiology and neuronal circuit connectivity. As the published methods with detection algorithms based upon amplitude thresholding and fixed or scaled template comparisons are of limited utility for detection of signals with variable amplitudes and superimposed events that have complex waveforms, previous techniques are not applicable for detection of evoked synaptic events in photostimulation and other similar experimental situations. Here we report on a novel technique that combines the design of a bank of approximate matched filters with the detection and estimation theory to automatically detect and extract photostimluation-evoked excitatory postsynaptic currents (EPSCs) from individually recorded neurons in cortical circuit mapping experiments. The sensitivity and specificity of the method were evaluated on both simulated and experimental data, with its performance comparable to that of visual event detection performed by human operators. This new technique was applied to quantify and compare the EPSCs obtained from excitatory pyramidal cells and fast-spiking interneurons. In addition, our technique has been further applied to the detection and analysis of inhibitory postsynaptic current (IPSC) responses. Given the general purpose of our matched filtering and signal recognition algorithms, we expect that our technique can be appropriately modified and applied to detect and extract other types of electrophysiological and optical imaging signals.

Des m&#xE9;thodes efficaces et fiables pour la d&#xE9;tection et la mesure des &#xE9;v&#xE9;nements synaptiques sont importants pour les &#xE9;tudes de la physiologie synaptique et de la connectivit&#xE9; circuit neuronal. Comme les m&#xE9;thodes publi&#xE9;es avec des algorithmes de d&#xE9;tection bas&#xE9;es sur le seuillage d&#x26;#39;amplitude et de comparaisons de mod&#xE8;les fixes ou &#xE9;chelle sont d&#x26;#39;une utilit&#xE9; limit&#xE9;e pour la d&#xE9;tection des signaux avec des amplitudes variables et des &#xE9;v&#xE9;nements superpos&#xE9;s qui ont des formes d&#x26;#39;onde complexes, techniques pr&#xE9;c&#xE9;dentes ne sont pas applicables pour la d&#xE9;tection des &#xE9;v&#xE9;nements &#xE9;voqu&#xE9;s synaptiques dans photostimulation et d&#x26;#39;autres semblables situations exp&#xE9;rimentales. Nous rapportons ici une nouvelle technique qui combine la conception d&#x26;#39;une banque de filtres adapt&#xE9;s approximatives avec la d&#xE9;tection et la th&#xE9;orie de l&#x26;#39;estimation de d&#xE9;tecter automatiquement et extraire photostimluation-courants postsynaptiques excitateurs &#xE9;voqu&#xE9;s (EPSCs) &#xE0; partir de neurones enregistr&#xE9;s individuellement dans des exp&#xE9;riences de cartographie corticale circuit. La sensibilit&#xE9; et la sp&#xE9;cificit&#xE9; de la m&#xE9;thode ont &#xE9;t&#xE9; &#xE9;valu&#xE9;es sur des donn&#xE9;es simul&#xE9;es et exp&#xE9;rimentales, avec sa performance comparable &#xE0; celle de la d&#xE9;tection d&#x26;#39;&#xE9;v&#xE9;nements visuelle effectu&#xE9;e par des op&#xE9;rateurs humains. Cette nouvelle technique a &#xE9;t&#xE9; appliqu&#xE9;e pour quantifier et comparer les EPSCs obtenus &#xE0; partir de cellules pyramidales excitatrices et rapide avec enrichissement des interneurones. En outre, notre technique a ensuite &#xE9;t&#xE9; appliqu&#xE9;e &#xE0; la d&#xE9;tection et l&#x26;#39;analyse des inhibiteurs postsynaptiques actuels (IPSC) r&#xE9;ponses. Compte tenu de l&#x26;#39;objectif g&#xE9;n&#xE9;ral de nos filtrage adapt&#xE9; et des algorithmes de reconnaissance de signaux, nous nous attendons &#xE0; ce que notre technique peut &#xEA;tre modifi&#xE9; en cons&#xE9;quence et appliqu&#xE9;es afin de d&#xE9;tecter et d&#x26;#39;extraire d&#x26;#39;autres types de signaux d&#x26;#39;imagerie &#xE9;lectrophysiologique et optique.

Da die ver&ouml;ffentlichten Methoden mit Algorithmen auf Amplitude Thresholding basierend und festen oder skalierte Vorlage Vergleiche sind begrenzte Dienstprogramm zur Erkennung von Signalen mit variabler Amplituden und &uuml;berlagerten Ereignisse mit komplexer Signalverl&auml;ufe, gelten nicht fr&uuml;here Techniken zur Erkennung von evozierte synaptische Ereignisse in Photostimulation und &auml;hnlichen experimentellen Situationen. Hier berichten wir &uuml;ber eine neuartige Technik, die verbindet das Design einer Bank der ungef&auml;hre &uuml;bereinstimmende Filter mit der Erkennung und Einsch&auml;tzung Theorie automatisch erkennen und individuell aufgenommenen Neuronen im kortikalen Schaltung Zuordnung Experimente Photostimluation evoziert exzitatorischen postsynaptisches Str&ouml;mungen (EPSCs) extrahieren. Metodi efficienti e affidabili per il rilevamento e misurazione di eventi sinaptici sono importanti per gli studi di fisiologia sinaptica e connettivit&agrave; circuito neuronale. Come i metodi pubblicati con algoritmi di rilevamento basano su soglia di ampiezza e confronti di modello in scala o fisso sono di utilit&agrave; limitata per la rilevazione di segnali con ampiezze variabili ed eventi sovrapposti che hanno forme d&#39;onda complesse, tecniche precedenti non sono applicabili per il rilevamento di eventi evocati sinaptici fotostimolazione e altre simili situazioni sperimentali. Riportiamo su una romanzo tecnica che combina il design di una banca di approssimativi filtri adattati con la teoria di rilevazione e stima automaticamente rilevare ed estrarre evocato photostimluation eccitatorie postsinaptiche correnti (EPSCs) da neuroni individualmente registrati negli esperimenti di mappatura del circuito corticale. La sensibilit&agrave; e la specificit&agrave; del metodo sono state valutate su dati simulati e sperimentali, con le sue prestazioni paragonabile a quello di rilevamento evento visual eseguita da operatori umani. Questa nuova tecnica &egrave; stata applicata per quantificare e confrontare il EPSCs ottenuto da cellule piramidali eccitatorie e chiodare veloce interneurons. M&#xE9;todos eficaces y fiables para la detecci&#xF3;n y medici&#xF3;n de los eventos sin&#xE1;pticos son importantes para los estudios de la fisiolog&#xED;a sin&#xE1;ptica y la conectividad del circuito neuronal. Como los m&#xE9;todos publicados con algoritmos de detecci&#xF3;n basados &#x200B;&#x200B;en umbralizaci&#xF3;n amplitud y comparaciones de plantillas fijas o escamas son de utilidad limitada para la detecci&#xF3;n de se&#xF1;ales con amplitudes variables y eventos superpuestas que tienen formas de onda complejas, las t&#xE9;cnicas anteriores no son aplicables para la detecci&#xF3;n de evoc&#xF3; eventos sin&#xE1;pticas en fotoestimulaci&#xF3;n y otras situaciones experimentales similares. Aqu&#xED; nos informe sobre una nueva t&#xE9;cnica que combina el dise&#xF1;o de un banco de filtros adaptados aproximados con la detecci&#xF3;n y la teor&#xED;a de la estimaci&#xF3;n para detectar autom&#xE1;ticamente y extraer photostimluation-evoc&#xF3; corrientes postsin&#xE1;pticas excitatorias (EPSCs) de las neuronas corticales individuales registradas en los experimentos de circuitos de mapeo. La sensibilidad y especificidad del m&#xE9;todo se evaluaron en los datos tanto simulados y experimentales, con su rendimiento comparable a la de detecci&#xF3;n de eventos visual realizado por los operadores humanos. Esta nueva t&#xE9;cnica se ha aplicado para cuantificar y comparar los EPSCs obtenidos a partir de las c&#xE9;lulas piramidales excitatorias y las interneuronas r&#xE1;pido clavar. Adem&#xE1;s, nuestra t&#xE9;cnica ha sido m&#xE1;s aplicada a la detecci&#xF3;n y an&#xE1;lisis de inhibitoria postsin&#xE1;ptica corrientes (IPSC) las respuestas. Teniendo en cuenta el objetivo general de nuestro filtro adaptado y algoritmos de reconocimiento de se&#xF1;ales, esperamos que nuestra t&#xE9;cnica puede ser modificado apropiadamente y se aplica para detectar y extraer otros tipos de se&#xF1;ales electrofisiol&#xF3;gicas y de imagen &#xF3;ptico.

Recent technical advances enable the regulation of neuronal circuit activity with high spatial and temporal resolution through genetic delivery of molecular activation or inactivation systems.Among them, the allatostatin receptor (AlstR)/ligand system has been developed for selective and quickly reversible silencing of mammalian neurons. However, targeted AlstR-mediated inactivation of specific neuronal types, particularly diverse types of inhibitory interneurons, remains to be established. In the present study, we achieved Cre-directed expression of AlstRs to excitatory and inhibitory cell-types in the cortex, and found that the AlstR-mediated inactivation was specific and robust at single-cell and neuronal population levels. Bath application of the allatostatin peptide markedly reduced spiking activity of AlstR-expressing excitatory and inhibitory neurons in response to intrasomatic current injections and laser photostimulation via glutamate uncaging, but control neurons without AlstR expression were not affected. As for the cortical network activity, the peptide application constrained photostimulation-evoked excitatory activity propagation detected by fast voltage-sensitive dye (VSD) imaging of the slices expressing AlstRs selectively in excitatory neurons, while it augmented excitatory activity in those slices with inhibitory neurons expressing AlstRs. In addition, AlstR-mediated inactivation effectively suppressed pharmacologically induced seizure activity in the slices targeting AlstRs to excitatory neurons. Taken together, our work demonstrated that the genetic delivery of AlstRs can be used for regulation of cortical excitability in a cell-type specific manner, and suggested that the AlstR system can be potentially used for fast seizure control.

Bath aplicaci&#xF3;n del p&#xE9;ptido allatostatin reduce notablemente la actividad de la adici&#xF3;n AlstR que expresan las neuronas excitadoras e inhibidoras en respuesta a las inyecciones de intrasomatic actuales y fotoestimulaci&#xF3;n con l&#xE1;ser a trav&#xE9;s de uncaging glutamato, pero las neuronas de control sin expresi&#xF3;n AlstR no se vieron afectados. En cuanto a la actividad de la red cortical, la aplicaci&#xF3;n de p&#xE9;ptidos restringidos fotoestimulaci&#xF3;n evocada por la propagaci&#xF3;n de la actividad excitatoria detectado por la v&#xED;a r&#xE1;pida sensibles al voltaje medio de contraste (CIV) de im&#xE1;genes de los cortes que expresan AlstRs selectivamente en neuronas excitadoras, al tiempo que aument&#xF3; la actividad excitatoria en los cortes con las neuronas inhibitorias que expresa AlstRs.
