El crecimiento del sacculus acoplamiento-como peptidoglicano (PG) situado entre las membranas internas y externas bacterianas (OM) es regulado firmemente para asegurar la integridad celular, mantener la forma celular y orquestar la divisi&oacute;n. Elementos citoesquel&eacute;ticos dirigen la colocaci&oacute;n y la actividad de la PG Sintasas de dentro de la c&eacute;lula, pero preciso spatiotemporal control sobre este proceso es mal entendido. Demostramos que PG Sintasas tambi&eacute;n se controlan desde fuera el s&aacute;culo. Dos de las lipoprote&iacute;nas de OM, LpoA y LpoB, son esenciales para la funci&oacute;n, respectivamente, de PBP1A y PBP1B, los principales e. coli bifuncionales PG Sintasas. Growth of the mesh-like peptidoglycan (PG) sacculus located between the bacterial inner and outer membranes (OM) is tightly regulated to ensure cellular integrity, maintain cell shape, and orchestrate division. Cytoskeletal elements direct placement and activity of PG synthases from inside the cell, but precise spatiotemporal control over this process is poorly understood. We demonstrate that PG synthases are also controlled from outside of the sacculus. Two OM lipoproteins, LpoA and LpoB, are essential for the function, respectively, of PBP1A and PBP1B, the major E.Â coli bifunctional PG synthases. Each Lpo protein binds specifically to its cognate PBP and stimulates its transpeptidase activity, thereby facilitating attachment of new PG toÂ the sacculus. LpoB shows partial septal localization, and our data suggest that the LpoB-PBP1B complex contributes to OM constriction during cellÂ division. LpoA/LpoB and their PBP-docking regions are restricted to Î³-proteobacteria, providing models for niche-specific regulation of sacculus growth.

Les &eacute;l&eacute;ments du cytosquelette directement mise en place et l&#39;activit&eacute; des PG synthases d&#39;&agrave; l&#39;int&eacute;rieur de la cellule, mais contr&ocirc;le spatio-temporel pr&eacute;cis au cours de ce processus est mal compris. Nous d&eacute;montrons que PG synthases sont &eacute;galement contr&ocirc;l&eacute;s de l&#39;ext&eacute;rieur sacculus. Wachstum von der Masche-wie Peptidoglycan (PG)-Sacculus befindet sich zwischen die bakteriellen inneren und &auml;u&szlig;eren Membran (OM) ist streng reguliert, um zellul&auml;ren Integrit&auml;t sicherzustellen, Zelle Form beizubehalten und orchestrieren Abteilung. Zytoskelett Elemente sind direkte Platzierung und Aktivit&auml;t von PG-Synthasen aus innerhalb der Zelle, aber pr&auml;zise raumzeitliche Kontrolle &uuml;ber diesen Prozess wird oft missverstanden. LpoB zeigt teilweise Septumdefekt Lokalisierung und unsere Daten deuten darauf hin, dass der LpoB-PBP1B-Komplex zu OM Verengung w&auml;hrend der Zellteilung beitr&auml;gt. LpoA/LpoB und PBP-Andocken Regionen beschr&auml;nken sich auf &gamma;-Proteobacteria, die Modelle f&uuml;r Nischen-spezifische Regelung der Sacculus Wachstum.
