Transient receptor potential A1 (TRPA1) forms nonselective cation channels implicated in acute inflammatory pain and nociception. The mechanism of ligand activation of TRPA1 may involve either covalent modification of cysteine residues or conventional reversible ligand-receptor interactions. For certain electrophilic prostaglandins, covalent modification has been considered as the main mechanism involved in their stimulatory effect on TRPA1. Because some nonsteroidal anti-inflammatory drugs (NSAIDs) are structural analogs of prostaglandins, we examined several nonelectrophilic NSAIDs on TRPA1 activation using electrophysiological techniques and intracellular Ca(2+) measurements and found that a selected group of NSAIDs can act as TRPA1 agonists. Extracellularly applied flufenamic, niflumic, and mefenamic acid, as well as flurbiprofen, ketoprofen, diclofenac, and indomethacin, rapidly activated rat TRPA1 expressed in Xenopus oocytes and human TRPA1 endogenously expressed in WI-38 fibroblasts. Similarly, the NSAID ligands activated human TRPA1 inducibly expressed in HEK293 cells, but the responses were absent in uninduced and parental HEK293 cells. The response to fenamate agonists was blocked by TRPA1 antagonists, AP-18, HC-030031, and ruthenium red. At subsaturating concentrations, the fenamate NSAIDs also potentiate the activation of TRPA1 by allyl isothiocyanate, cinnamaldehyde, and cold, demonstrating positive synergistic interactions with other well-characterized TRPA1 activators. Importantly, among several thermosensitive TRP channels, the stimulatory effect is specific to TRPA1 because flufenamic acid inhibited TRPV1, TRPV3, and TRPM8. We conclude that fenamate NSAIDs are a novel class of potent and reversible direct agonists of TRPA1. This selective group of TRPA1-stimulating NSAIDs should provide a structural basis for developing novel ligands that noncovalently interact with TRPA1 channels.

Activation of TRPA1 channels by fenamate nonsteroidal anti-inflammatory drugs.

Transitoria receptor potencial A1 (TRPA1) forma los canales de cationes no selectivos implicados en el dolor inflamatorio agudo y la nocicepci&#xF3;n. El mecanismo de activaci&#xF3;n ligando de TRPA1 puede implicar modificaci&#xF3;n covalente de residuos de ciste&#xED;na o convencionales interacciones reversibles ligando-receptor. Para ciertas prostaglandinas electrof&#xED;licos, modificaci&#xF3;n covalente ha sido considerada como el principal mecanismo implicado en su efecto estimulante sobre la TRPA1. Debido a que algunos medicamentos anti-inflamatorios no esteroideos (AINE) son an&#xE1;logos estructurales de prostaglandinas, se examinaron varios AINE nonelectrophilic de TRPA1 activaci&#xF3;n mediante t&#xE9;cnicas electrofisiol&#xF3;gicas y de Ca intracelular (2 +) las mediciones y se encontr&#xF3; que un selecto grupo de los AINE pueden actuar como agonistas TRPA1. We previously reported that phosphatidylinositol 3,4,5-trisphosphate (PIP(3)), a lipid product of phosphoinositide 3-kinase (PI3K), induced Ca(2+) influx via a noncapacitative pathway in platelets, Jurkat T cells, and RBL-2H3 mast cells. The identity of this Ca(2+) influx system, however, remains unclear. Here, we investigate a potential link between PIP(3)-sensitive Ca(2+) entry and the canonical transient receptor potential (TRPC) channels by developing stable human embryonic kidney (HEK) 293 cell lines expressing TRPC1, TRPC3, TRPC5, and TRPC6. Two lines of evidence support TRPC6 as a putative target by which PIP(3) induces Ca(2+) influx. First, Fura-2 fluorometric Ca(2+) analysis shows the ability of PIP(3) to selectively stimulate [Ca(2+)](i) increase in TRPC6-expressing cells. Second, pull-down analysis indicates specific interactions between biotin-PIP(3) and TRPC6 protein. Our data indicate that PIP(3) activates store-independent Ca(2+) entry in TRPC6 cells via a nonselective cation channel. Although the activating effect of PIP(3) on TRPC6 is reminiscent to that of 1-oleoyl-2-acetyl-sn-glycerol, this activation is not attributable to the diacylglycerol substructure of PIP(3) since other phosphoinositides failed to trigger Ca(2+) responses. The PIP(3)-activated Ca(2+) entry is inhibited by known TRPC6 inhibitors such as Gd(3+) and SKF96365 and is independent of IP(3) production. Furthermore, we demonstrated that TRPC6 overexpression or antisense downregulation significantly alters the amplitude of PIP(3)- and anti-CD3-activated Ca(2+) responses in Jurkat T cells. Consequently, the link between TRPC6 and PIP(3)-mediated Ca(2+) entry provides a framework to account for an intimate relationship between PI3K and PLCgamma in initiating Ca(2+) response to agonist stimulation in T lymphocytes.

Members of the transient receptor potential (TRP) ion channel family mediate diverse sensory transduction processes in both vertebrates and invertebrates. In particular, members of the TRPA subfamily have distinct thermosensory roles in Drosophila, and mammalian TRPA1 is postulated to have a function in noxious cold sensation and mechanosensation. Here we show that mutations in trpa-1, the C. elegans ortholog of mouse Trpa1, confer specific defects in mechanosensory behaviors related to nose-touch responses and foraging. trpa-1 is expressed and functions in sensory neurons required for these mechanosensory behaviors, and contributes to neural responses of these cells to touch, particularly after repeated mechanical stimulation. Furthermore, mechanical pressure can activate C. elegans TRPA-1 heterologously expressed in mammalian cells. Collectively, these data demonstrate that trpa-1 encodes an ion channel that can be activated in response to mechanical pressure and is required for mechanosensory neuron function, suggesting a possible role in mechanosensory transduction or modulation.

Ion channels can be activated (gated) by a variety of stimuli, including chemicals, voltage, mechanical force or temperature. Although molecular mechanisms of ion channel gating by chemical and voltage stimuli are understood in principal, the mechanisms of temperature activation remain unknown. The transient receptor potential channel TRPV3 is a nonselective cation channel that is activated by warm temperatures and sensory chemicals such as camphor. Here we screened approcimately 14,000 random mutant clones of mouse TRPV3 and identified five single point mutations that specifically abolish heat activation but do not perturb chemical activation or voltage modulation. Notably, all five mutations are located in the putative sixth transmembrane helix and the adjacent extracellular loop in the pore region of mouse TRPV3. Although distinct in sequence, we found that the corresponding loop of frog TRPV3 is also specifically required for heat activation. These findings demonstrate that the temperature sensitivity of TRPV3 is separable from all other known activation mechanisms and implicate a specific region in temperature sensing.

Temperature-activated transient receptor potential ion channels (thermoTRPs) are polymodal detectors of various stimuli including temperature, voltage, and chemicals. To date, it is not known how TRP channels integrate the action of such disparate stimuli. Identifying specific residues required for channel-activation by distinct stimuli is necessary for understanding overall TRP channel function. TRPV3 is activated by warm temperatures and various chemicals, and is modulated by voltage. One potent activator of TRPV3 is 2-aminoethyl diphenylborinate (2-APB), a synthetic chemical that modulates many TRP channels. In a high-throughput mutagenesis screen of approximately 14,000 mutated mouse TRPV3 clones, we found 2 residues (H426 and R696) specifically required for sensitivity of TRPV3 to 2-APB, but not to camphor or voltage. The cytoplasmic N-terminal mutation H426N in human, dog, and frog TRPV3 also effectively abolished 2-APB activation without affecting camphor responses. Interestingly, chicken TRPV3 is weakly sensitive to 2-APB, and the equivalent residue at 426 is an asparagine (N). Mutating this residue to histidine induced 2-APB sensitivity of chicken TRPV3 to levels comparable for other TRPV3 orthologs. The cytoplasmic C-terminal mutation R696K in the TRP box displayed 2-APB specific deficits only in the presence of extracellular calcium, suggesting involvement in gating. TRPV4, a related thermoTRP, is 2-APB insensitive and has variant sequences at both residues identified here. Remarkably, mutating these 2 residues in TRPV4 to TRPV3 sequences (N426H and W737R) was sufficient to induce TRPV3-like 2-APB sensitivity. Here we show that zinc excites nociceptive somatosensory neurons and causes nociception in mice through TRPA1, a cation channel previously shown to mediate the pungency of wasabi and cinnamon through cysteine modification. Zinc activates TRPA1 through a unique mechanism that requires zinc influx through TRPA1 channels and subsequent activation via specific intracellular cysteine and histidine residues. TRPA1 is highly sensitive to intracellular zinc, as low nanomolar concentrations activate TRPA1 and modulate its sensitivity. These findings identify TRPA1 as an important target for the sensory effects of zinc and support an emerging role for zinc as a signaling molecule that can modulate sensory transmission.

Objective:â€ƒ To compare the diagnostic yield of transrectal ultrasonography and transurethral seminal vesiculoscopy in patients with persistent hematospermia, and to determine the advantages and disadvantages of both modalities. Methods:â€ƒ We prospectively enrolled 106 patients with persistent hematospermia of mean duration 20.5â€ƒmonths. All patients were evaluated by both transrectal ultrasonography and transurethral seminal vesiculoscopy after excluding definite etiological lesions beyond the reproductive duct system. The diagnostic yield and other technical parameters of both modalities were compared. Results:â€ƒ Final diagnoses were made in 93 patients (87.7%), with transrectal ultrasonography and transurethral seminal vesiculoscopy showing overall diagnostic yields of 45.3% and 74.5%, respectively (Pâ€ƒ<â€ƒ0.001). The diagnostic yield of combining transrectal ultrasonography and transurethral seminal vesiculoscopy was significantly higher than that of each modality alone (both Pâ€ƒ<â€ƒ0.001). Of the 114 findings of diagnostic value, the most frequent was calculus (47.4%, nâ€ƒ=â€ƒ54), followed by obstruction/stricture (37.7%, nâ€ƒ=â€ƒ43), cyst (8.8%, nâ€ƒ=â€ƒ10), dysplasia (3.5%, nâ€ƒ=â€ƒ4), polyp (1.8%, nâ€ƒ=â€ƒ2) and benign mass (0.9%, nâ€ƒ=â€ƒ1). Transurethral seminal vesiculoscopy showed significant superiority in detecting calculi and obstruction/stricture. Hematospermia disappeared in 95.3% (101/106) of all patients and in 97.6% (83/85) of patients receiving transurethral seminal vesiculoscopy therapy during follow up. No major adverse effects occurred during and after examination. Conclusions:â€ƒ The diagnostic yield of transurethral seminal vesiculoscopy for persistent hematospermia was significantly superior to that of transrectal ultrasonography, especially in lesions diagnosed with calculi and obstruction/stricture. Combining both modalities might provide extra benefits for patients with persistent hematospermia.

R&#xE9;sultats: le diagnostic final a &#xE9;t&#xE9; faite chez 93 patients (87,7%), avec l&#x26;#39;&#xE9;chographie transrectale et transur&#xE9;trale vesiculoscopy s&#xE9;minale montrant les rendements globaux de diagnostic de 45,3% et 74,5%, respectivement (P &#x26;lt;0,001). Le rendement diagnostique de la combinaison &#xE9;chographie transrectale et transur&#xE9;trale vesiculoscopy s&#xE9;minal &#xE9;tait significativement plus &#xE9;lev&#xE9; que celui de chaque modalit&#xE9; seule (P &#x26;lt;0,001). Sur les 114 conclusions de valeur diagnostique, la plus fr&#xE9;quente &#xE9;tait le calcul (47,4%, n = 54), suivie par une obstruction / st&#xE9;nose (37,7%, n = 43), kyste (8,8%, n = 10), la dysplasie (3,5% , n = 4), un polype (1,8%, n = 2) et la masse b&#xE9;nigne (0,9%, n = 1). Transur&#xE9;trale vesiculoscopy s&#xE9;minale a montr&#xE9; une sup&#xE9;riorit&#xE9; significative dans la d&#xE9;tection des calculs et l&#x26;#39;obstruction / st&#xE9;nose. H&#xE9;matospermie disparu chez 95,3% (101/106) de tous les patients et chez 97,6% (83/85) des patients recevant la th&#xE9;rapie transur&#xE9;trale vesiculoscopy s&#xE9;minal au cours du suivi. Pas d&#x26;#39;effets ind&#xE9;sirables majeurs a eu lieu pendant et apr&#xE8;s l&#x26;#39;examen. Conclusions: Le rendement diagnostique de vesiculoscopy s&#xE9;minale transur&#xE9;trale pour h&#xE9;matospermie persistante &#xE9;tait significativement sup&#xE9;rieure &#xE0; celle de l&#x26;#39;&#xE9;chographie transrectale, en particulier dans les l&#xE9;sions diagnostiqu&#xE9;es avec des calculs et l&#x26;#39;obstruction / st&#xE9;nose. &#xC9;tude prospective comparant &#xE9;chographie transrectale et transur&#xE9;trale Vesiculoscopy s&#xE9;minale pour h&#xE9;matospermie persistante

Ziel: Um die diagnostische Ausbeute der Transrektale Ultraschall und Transurethrale bahnbrechenden Vesiculoscopy bei Patienten mit anhaltenden Hematospermia zu vergleichen und die vor- und Nachteile der beiden Modalit&auml;ten zu bestimmen. Methoden: Wir nahmen prospektiv 106 Patienten mit anhaltenden Hematospermia der mittlere Dauer 20,5 Monate. Alle Patienten wurden durch Transrektale Ultraschall und Transurethrale bahnbrechenden Vesiculoscopy ausgewertet, bereinigt um definitive &auml;tiologische L&auml;sionen jenseits der reproduktiven Kanalsystem. Die diagnostische Ausbeute und andere technische Parameter der beiden Modalit&auml;ten wurden verglichen. Ergebnisse: Endg&uuml;ltige Diagnosen waren in 93 Patienten (87,7 %), mit Transrektale Ultraschall und Transurethrale bahnbrechenden Vesiculoscopy zeigen insgesamt Diagnose ergibt 45,3 % und 74,5 %, bzw. (P &lt; 0,001). Die diagnostische Ausbeute des Kombinierens der Transrektale Ultraschall und Transurethrale bahnbrechenden Vesiculoscopy war deutlich h&ouml;her als die der jede Modalit&auml;t allein (beide P &lt; 0,001). 114 Ergebnisse der diagnostischen Wert, war die h&auml;ufigste Kalk&uuml;l (47,4 %, n = 54), gefolgt von Obstruktion/Striktur (37,7 %, n = 43), Zyste (8,8 %, n = 10), Dysplasie (3,5 %, n = 4), Polyp (1,8 %, n = 2) und gutartige Masse (0,9 %, n = 1). Transurethrale bahnbrechenden Vesiculoscopy zeigten signifikante &Uuml;berlegenheit Kalk&uuml;le und Behinderung/Striktur zu erkennen. Hematospermia verschwanden 95,3 % (101/106) aller Patienten und bei 97,6 % (83/85) der Patienten Transurethrale bahnbrechenden Vesiculoscopy Therapie w&auml;hrend der follow-up. Keine gro&szlig;en Nebenwirkungen aufgetreten w&auml;hrend und nach der Pr&uuml;fung. Schlussfolgerungen: Die diagnostische Ausbeute der Transurethrale bahnbrechenden Vesiculoscopy f&uuml;r persistente Hematospermia war deutlich h&ouml;her als die der Transrektale Ultraschall, vor allem in L&auml;sionen Kalk&uuml;le und Behinderung/Striktur diagnostiziert. Kombinieren beide Modalit&auml;ten m&ouml;glicherweise zus&auml;tzliche Vorteile f&uuml;r Patienten mit anhaltenden Hematospermia bereit.

Prospektive Studie Vergleich Transrektale Ultraschall und Transurethrale bahnbrechenden Vesiculoscopy f&uuml;r permanente Hematospermia.

Obiettivo: Confrontare la resa diagnostica dell&#39;ultrasonografia transrettale e transuretrale vesiculoscopy seminali nei pazienti con emospermia persistente e per determinare i vantaggi e gli svantaggi di entrambe le modalit&agrave;. Metodi: Abbiamo prospetticamente iscritti 106 pazienti con emospermia persistente di media durata mesi 20,5. Tutti i pazienti sono stati valutati sia l&#39;ecografia transrettale e transuretrale seminale vesiculoscopy dopo aver escluso lesioni eziologica precise oltre il sistema riproduttivo del condotto. La resa diagnostica e altri parametri tecnici di entrambe le modalit&agrave; sono stati confrontati. Risultati: La diagnosi finale sono state fatte in 93 pazienti (87.7%), con l&#39;ecografia transrettale e transuretrale seminale vesiculoscopy mostrando diagnostico complessivo rendimenti del 45,3% e 74,5%, rispettivamente (P &lt; 0.001). La resa diagnostica combinando l&#39;ecografia transrettale e transuretrale vesiculoscopy seminale era significativamente superiore a quello di ogni modalit&agrave; da solo (entrambi P &lt; 0.001). Dei 114 risultati valore diagnostico, la pi&ugrave; frequente era calcolo (47,4%, n = 54), seguita da ostruzione/stenosi (37,7%, n = 43), cisti (8,8%, n = 10), displasia (3.5%, n = 4), Polipo (1,8%, n = 2) e massa benigna (0,9%, n = 1). Vesiculoscopy transuretrale seminale ha mostrato notevole superiorit&agrave; nella rilevazione di calculi e ostruzione/stenosi. Emospermia scomparve nel 95,3% (101/106) di tutti i pazienti e 97,6% (83/85) dei pazienti terapia transuretrale seminale vesiculoscopy durante il follow-up. Senza grandi effetti negativi si &egrave; verificato durante e dopo l&#39;esame. Conclusioni: Il rendimento diagnostico di transurethral vesiculoscopy seminali per emospermia persistente era significativamente superiore a quello dell&#39;ecografia transrettale, soprattutto nelle lesioni con diagnosticata di calcoli e ostruzione/stenosi. Combinando entrambe le modalit&agrave; potrebbe fornire ulteriori benefici per i pazienti con emospermia persistente.

&#xC608;&#xBE44; &#xC2E4;&#xD5D8; transrectal &#xCD08;&#xC74C;&#xD30C; &#xBC0F; transurethral &#xC815;&#xC561; vesiculoscopy &#xC601;&#xAD6C; hematospermia&#xC5D0; &#xB300; &#xD55C; &#xBE44;&#xAD50;.

Objetivo: Comparar el rendimiento diagn&#xF3;stico de la ecograf&#xED;a transrectal y vesiculoscopy seminal transuretral de pr&#xF3;stata en pacientes con hemospermia persistente, y para determinar las ventajas y desventajas de ambas modalidades. M&#xE9;todos: Estudio prospectivo en 106 pacientes con hemospermia persistente de la duraci&#xF3;n media de 20,5 meses. Todos los pacientes fueron evaluados tanto por ecograf&#xED;a transrectal y transuretral vesiculoscopy seminal despu&#xE9;s de excluir determinadas lesiones etiol&#xF3;gicas m&#xE1;s all&#xE1; del sistema de conductos reproductiva. El rendimiento diagn&#xF3;stico y otros par&#xE1;metros t&#xE9;cnicos de ambas modalidades se compararon. Resultados: Los diagn&#xF3;sticos se realizaron en 93 pacientes (87,7%), con la ecograf&#xED;a transrectal y transuretral vesiculoscopy fundamental que muestra en general el rendimiento diagn&#xF3;stico de 45,3% y 74,5%, respectivamente (P &#x26;lt;0,001). La rentabilidad diagn&#xF3;stica de la combinaci&#xF3;n de la ecograf&#xED;a transrectal y vesiculoscopy seminal transuretral fue significativamente mayor que la de cada modalidad por s&#xED; solo (ambos p &#x26;lt;0,001). De los 114 resultados de valor diagn&#xF3;stico, la m&#xE1;s frecuente fue el c&#xE1;lculo (47,4%, n = 54), seguida de la obstrucci&#xF3;n / estenosis (37,7%, n = 43), quiste (8,8%, n = 10), displasia (3,5% , n = 4), p&#xF3;lipo (1,8%, n = 2) y la masa benigna (0,9%, n = 1). Vesiculoscopy seminal transuretral demostr&#xF3; una superioridad significativa en la detecci&#xF3;n de c&#xE1;lculos y obstrucci&#xF3;n / estenosis. Hematospermia desapareci&#xF3; en el 95,3% (101/106) de todos los pacientes y en el 97,6% (83/85) de los pacientes que reciben terapia transuretral vesiculoscopy seminal durante el seguimiento. No se observaron efectos adversos importantes durante y despu&#xE9;s del examen. Conclusiones: El rendimiento diagn&#xF3;stico de vesiculoscopy seminal transuretral por hemospermia persistente fue significativamente superior a la de la ecograf&#xED;a transrectal, especialmente en las lesiones con diagn&#xF3;stico de litiasis y la obstrucci&#xF3;n / estenosis. La combinaci&#xF3;n de ambas modalidades puede proporcionar beneficios adicionales para los pacientes con hemospermia persistente.
