Implantable and extracorporeal cardiovascular devices are commonly made from titanium (Ti) (e.g. Ti-coated Nitinol stents and mechanical circulatory assist devices). Endothelializing the blood-contacting Ti surfaces of these devices would provide them with an antithrombogenic coating that mimics the native lining of blood vessels and the heart. We evaluated the viability and adherence of peripheral blood-derived porcine endothelial progenitor cells (EPCs), seeded onto thin Ti layers on glass slides under static conditions and after exposure to fluid shear stresses. EPCs attached and grew to confluence on Ti in serum-free medium, without preadsorption of proteins. After attachment to Ti for 15 min, less than 5% of the cells detached at a shear stress of 100 dyne / cm(2). Confluent monolayers of EPCs on smooth Ti surfaces (Rq of 10 nm), exposed to 15 or 100 dyne/cm(2) for 48 h, aligned and elongated in the direction of flow and produced nitric oxide dependent on the level of shear stress. EPC-coated Ti surfaces had dramatically reduced platelet adhesion when compared to uncoated Ti surfaces. These results indicate that peripheral blood-derived EPCs adhere and function normally on Ti surfaces. Therefore EPCs may be used to seed cardiovascular devices prior to implantation to ameliorate platelet activation and thrombus formation.

Implantables et dispositifs cardio-vasculaires sont extracorporelle g&#xE9;n&#xE9;ralement fabriqu&#xE9;s &#xE0; partir de titane (Ti) (par exemple Ti-stents en nitinol et une assistance circulatoire m&#xE9;canique des dispositifs). Endothelializing les surfaces contact avec le sang Ti de ces dispositifs serait de leur fournir avec un rev&#xEA;tement anti-thrombog&#xE8;ne qui imite la doublure natif de vaisseaux sanguins et le coeur. PEC et fix&#xE9; augment&#xE9; jusqu&#x26;#39;&#xE0; confluence de Ti dans un milieu sans s&#xE9;rum, sans pr&#xE9;adsorption de prot&#xE9;ines. Apr&#xE8;s la fixation de Ti pendant 15 min, inf&#xE9;rieur &#xE0; 5% des cellules d&#xE9;tach&#xE9;es sous une contrainte de cisaillement de 100 dynes / cm (2). Monocouches confluentes de PEC sur les surfaces lisses (Rq Ti de 10 nm), expos&#xE9;s &#xE0; 15 ou 100 dynes / cm (2) pendant 48 h, align&#xE9;s et allong&#xE9;e dans la direction d&#x26;#39;&#xE9;coulement et de l&#x26;#39;oxyde nitrique produit en fonction du niveau de contrainte de cisaillement. CBE surfaces rev&#xEA;tues de Ti avait consid&#xE9;rablement r&#xE9;duit l&#x26;#39;adh&#xE9;sion des plaquettes par rapport aux surfaces Ti non couch&#xE9;s. Ces r&#xE9;sultats indiquent que p&#xE9;riph&#xE9;riques d&#xE9;riv&#xE9;s du sang EPC respecter et de fonctionner normalement sur les surfaces Ti. Implantierbare und extrakorporalen Kreislauf-Ger&auml;te bestehen h&auml;ufig aus Titan (Ti) (z.B. Ti-beschichtete Nitinol-Stents und mechanische Kreislauf unterst&uuml;tzen Ger&auml;te). Endothelializing die Blut-Kontakt-Ti-Fl&auml;chen dieser Ger&auml;te w&uuml;rden sie mit Antithrombogenic Beschichtung darstellen, die die native Auskleidung der Blutgef&auml;&szlig;e und das Herz imitiert. Wir evaluieren die Lebensf&auml;higkeit und die Einhaltung der peripheren Blut abgeleitete porcine endothelialen Vorl&auml;uferzellen (EPCs), entkernt, in d&uuml;nnen Schichten von Ti auf Objekttr&auml;gern unter statischen Bedingungen und nach Exposition mit Fl&uuml;ssigkeit Scherspannungen. EPK befestigt und wuchs bis M&uuml;ndung auf Ti in serumfreien Medium, ohne Preadsorption von Proteinen. Nach der Anlage zu Ti 15 min, weniger als 5 % der Zellen getrennt bei einer Scherspannung des 100 dyn / cm(2). Konfluierende Monolayers von EPCs auf glatten Oberfl&auml;chen in Ti (Rq 10 nm), 15 oder 100 dyne/cm(2) f&uuml;r 48 h ausgesetzt, ausgerichtet und l&auml;nglich in Flie&szlig;richtung und produzierte Stickstoffmonoxid auf der Ebene der Schubspannung angewiesen. EPC-beschichtete Ti Fl&auml;chen hatte Pl&auml;ttchen Adh&auml;sion im Vergleich zu unbeschichteten Ti Fl&auml;chen drastisch reduziert. Diese Ergebnisse zeigen, dass die peripheren Blut abgeleitete EPCs halten und normal funktionieren auf Ti-Oberfl&auml;chen. Daher d&uuml;rfen EPCs Seeding von Herz-Kreislauf-Ger&auml;ten vor der Einnistung zur Besserung der Thrombozyten Aktivierung und Thrombus-Bildung eingesetzt werden.

Die Biokompatibilit&auml;t von Titan-Herz-Kreislauf-Ger&auml;ten mit autologer Blut abgeleitete endothelialen Vorl&auml;uferzellen Agarn&auml;hrboden: EPC-bes&auml;te antithrombotischen Ti Implantate.

Dispositivi impiantabili ed extracorporee cardiovascolare sono comunemente realizzati in titanio (Ti) (ad esempio gli stent in Nitinol rivestiti di Ti e meccanica circolatoria assistere dispositivi). Endothelializing le superfici di contatto con il sangue Ti di questi dispositivi li fornirebbe con rivestimento antithrombogenic che imita il nativo rivestimento dei vasi sanguigni e il cuore. Abbiamo valutato la vitalit&agrave; e l&#39;aderenza delle cellule periferiche emoderivati suina progenitrici endoteliali (EPC), seminati su strati sottili di Ti su vetrini in condizioni statiche e dopo l&#39;esposizione a liquidi shear stress. EPCs associato e crebbe alla confluenza su Ti in media serum-free, senza preadsorption di proteine. Dopo l&#39;attacco a Ti per 15 min, meno del 5% delle cellule staccate a una sollecitazione di taglio di 100 dyne / cm(2). Monostrati confluenti di EPCs su superfici lisce Ti (Rq di 10 nm), esposti a 15 o 100 dyne/cm(2) per 48 h, allineati e allungata in direzione del flusso e prodotto ossido nitrico dipendente dal livello di sollecitazione di taglio. Superfici verniciate a EPC Ti avevano drasticamente ridotto adesione piastrinica rispetto alle superfici Ti non patinate. Questi risultati indicano che EPCs derivati da sangue periferico aderiscono e funzionare normalmente su superfici di Ti. Pertanto EPCs pu&ograve; essere utilizzato per seme dispositivi cardiovascolari prima dell&#39;impianto per migliorare la attivazione piastrinica e la formazione di trombi.

Titanium (Ti) is commonly utilized in many cardiovascular devices, e.g. as a component of Nitinol stents, intra- and extracorporeal mechanical circulatory assist devices, but is associated with the risk of thromboemboli formation. We propose to solve this problem by lining the Ti blood-contacting surfaces with autologous peripheral blood-derived late outgrowth endothelial progenitor cells (EPCs) after having previously demonstrated that these EPCs adhere to and grow on Ti under physiological shear stresses and functionally adapt to their environment under flow conditions ex vivo. Autologous fluorescently-labeled porcine EPCs were seeded at the point-of-care in the operating room onto Ti tubes for 30 min and implanted into the pro-thrombotic environment of the inferior vena cava of swine (n = 8). After 3 days, Ti tubes were explanted, disassembled, and the blood-contacting surface was imaged. A blinded analysis found all 4 cell-seeded implants to be free of clot, whereas 4 controls without EPCs were either entirely occluded or partially thrombosed. Pre-labeled EPCs had spread and were present on all 4 cell-seeded implants while no endothelial cells were observed on control implants. These results suggest that late outgrowth autologous EPCs represent a promising source of lining Ti implants to reduce thrombosis in vivo.

Use of autologous blood-derived endothelial progenitor cells at point-of-care to protect against implant thrombosis in a large animal model.

De titanio (Ti) se utiliza com&#xFA;nmente en muchos dispositivos cardiovasculares, por ejemplo, como un componente de los stents de Nitinol, circulatorio mec&#xE1;nico intra y extracorp&#xF3;rea dispositivos de asistencia, pero se asocia con el riesgo de formaci&#xF3;n trombo&#xE9;mbolos. Proponemos para resolver este problema por el forro los Ti contacto de sangre con superficies perif&#xE9;ricas aut&#xF3;logas derivadas de la sangre c&#xE9;lulas excrecencia finales progenitoras endoteliales (CPE) despu&#xE9;s de haber demostrado previamente que estas se adhieren a las CFP y crecer en Ti bajo esfuerzos cortantes fisiol&#xF3;gicas y funcionalmente adaptarse a su medio ambiente bajo condiciones de flujo ex vivo. Aut&#xF3;logo marcadas con fluorescencia EPC porcina fueron sembradas en el punto de atenci&#xF3;n en la sala de operaciones en Ti tubos durante 30 minutos y se implanta en el medio ambiente pro-tromb&#xF3;tico de la vena cava inferior de la especie porcina (n = 8). Despu&#xE9;s de 3 d&#xED;as, los tubos de Ti se explantaron, desmontado, y la superficie de contacto con la sangre fue fotografiada. Un an&#xE1;lisis ciego encontr&#xF3; los 4 cabezas de serie de c&#xE9;lulas implantes para estar libre de co&#xE1;gulos, mientras que los 4 controles sin EPC eran totalmente ocluido o parcialmente trombosado. Pre-etiquetados EPC se hab&#xED;a extendido y estaba presente en todos los cuatro cabeza de serie, los implantes de c&#xE9;lulas, mientras que no se observaron las c&#xE9;lulas endoteliales de los implantes de control. Estos resultados sugieren que las CPE excrecencia aut&#xF3;logas finales representan una fuente prometedora de revestimiento de los implantes de Ti para reducir la trombosis in vivo.
