Titanium is one of the most commonly used materials for implantable devices in humans. Scanning electron microscopy (SEM) serves as an important tool for imaging titanium surfaces and analyzing cells and other organic matter adhering to titanium implants. However, high-vacuum SEM imaging of a nonconductive sample requires a conductive coating on the surface. A gold/palladium coating is commonly used and to date no method has been described to "clean" such gold/palladium covered surfaces for repeated experiments without etching the titanium itself. This constitutes a major problem with titanium-based implantable devices which are very expensive and thus in short supply. Our objective was to devise a protocol to regenerate titaniumsurfaces after SEM analysis. In a series of experiments, titanium samples from implantable cardiac assist devices were coated with fibronectin, seeded with cells and then coated with gold/palladium for SEM analysis. X-ray photoelectron spectroscopy spectra were obtained before and after five different cleaning protocols. Treatment with aqua regia (a 1:3 solution of concentrated nitric and hydrochloric acid), with or without ozonolysis, followed by sonication in soap solution and sonication in deionized water, allowed regenerating titanium surfaces to their original state. Atomic force microscopy confirmed that the established protocol did not alter the titanium microstructure. The protocol described herein is applicable to almost all titanium surfaces used in biomedical sciences and because of its short exposure time to aqua regia, will likely work for many titanium alloys as well.

Le titane est l&#x26;#39;un des mat&#xE9;riaux les plus couramment utilis&#xE9;s pour les dispositifs implantables chez l&#x26;#39;homme. Microscopie &#xE9;lectronique &#xE0; balayage (MEB) est un outil important pour les surfaces de titane et d&#x26;#39;imagerie des cellules d&#x26;#39;analyse et d&#x26;#39;autres mati&#xE8;res organiques adh&#xE9;rant &#xE0; des implants en titane. Cependant, un vide pouss&#xE9; imagerie SEM d&#x26;#39;un &#xE9;chantillon non conducteur exige un rev&#xEA;tement conducteur sur la surface. Un rev&#xEA;tement d&#x26;#39;or / palladium est couramment utilis&#xE9; et &#xE0; ce jour aucune m&#xE9;thode a &#xE9;t&#xE9; d&#xE9;crite pour &#xAB;nettoyer&#xBB; cet or / palladium surfaces couvertes pour des exp&#xE9;riences r&#xE9;p&#xE9;t&#xE9;es sans attaquer le titane lui-m&#xEA;me. Ceci constitue un probl&#xE8;me majeur avec le titane &#xE0; base de dispositifs implantables qui sont tr&#xE8;s co&#xFB;teux et donc rares. Notre objectif &#xE9;tait de mettre au point un protocole pour r&#xE9;g&#xE9;n&#xE9;rer titaniumsurfaces apr&#xE8;s analyse SEM. Dans une s&#xE9;rie d&#x26;#39;exp&#xE9;riences, les &#xE9;chantillons de titane &#xE0; partir de dispositifs implantables assistance cardiaque ont &#xE9;t&#xE9; recouvertes avec de la fibronectine, ensemenc&#xE9;s avec des cellules, puis recouvert d&#x26;#39;or / palladium pour l&#x26;#39;analyse SEM. Spectres de rayons X spectroscopie photo&#xE9;lectronique ont &#xE9;t&#xE9; obtenues avant et apr&#xE8;s cinq diff&#xE9;rents protocoles de nettoyage. Le traitement par l&#x26;#39;eau r&#xE9;gale (une solution 1:3 de nitrique concentr&#xE9; et d&#x26;#39;acide chlorhydrique), avec ou sans ozonolyse, suivie par sonication dans une solution de savon et de sonication dans l&#x26;#39;eau d&#xE9;min&#xE9;ralis&#xE9;e, a permis la r&#xE9;g&#xE9;n&#xE9;ration des surfaces de titane pour leur &#xE9;tat d&#x26;#39;origine. Microscopie &#xE0; force atomique a confirm&#xE9; que le protocole &#xE9;tabli n&#x26;#39;a pas modifi&#xE9; la microstructure de titane. Le protocole d&#xE9;crit ici est applicable &#xE0; presque toutes les surfaces de titane utilis&#xE9;s dans les sciences biom&#xE9;dicales et en raison de son temps d&#x26;#39;exposition court &#xE0; l&#x26;#39;eau r&#xE9;gale, sera probablement travailler pour de nombreux alliages de titane ainsi.

Titan ist eines der am h&auml;ufigsten verwendeten Materialien f&uuml;r implantierbare Ger&auml;te beim Menschen. Rasterelektronenmikroskopie (SEM) dient als ein wichtiges Instrument f&uuml;r imaging Titan-Oberfl&auml;chen und Analyse von Zellen und andere organischer Substanz, die Einhaltung von Titan-Implantaten. Hochvakuum-SEM Bildgebung in einem Kunststoffschraubenzieher Beispiel erfordert jedoch eine leitf&auml;hige Beschichtung auf der Oberfl&auml;che. Eine Gold/Palladium-Beschichtung wird h&auml;ufig verwendet, und bislang keine Methode ist beschrieben worden, um &quot;solche Gold/Palladium abgedeckt-Fl&auml;chen f&uuml;r die wiederholte Experimente clean&quot; ohne &Auml;tzen der Titan selbst. Dies stellt ein gro&szlig;es Problem mit Titan-basierte implantierbare Ger&auml;te, die sind sehr teuer und somit kurz liefern. Unser Ziel war es, ein Protokoll zum Regenerieren von Titaniumsurfaces nach SEM-Analyse zu entwickeln. In einer Reihe von Experimenten wurden Titan-Proben von implantierbaren kardialen Assist Ger&auml;te beschichtet mit Fibronektin, entkernt mit Zellen und dann beschichtet, mit Gold/Palladium f&uuml;r SEM-Analyse. R&ouml;ntgen-Photoelektronenspektroskopie-Spektroskopie-Spektren wurden vor und nach f&uuml;nf unterschiedliche Protokolle Reinigung erzielt. Behandlung mit K&ouml;nigswasser (1:3 L&ouml;sung von konzentrierter Salpeters&auml;ure und Salzs&auml;ure), mit oder ohne Ozonolyse, gefolgt von Sonorisierung in Seifenl&ouml;sung und Sonorisierung deionisiertem Wasser erlaubt, regenerierende Titan-Oberfl&auml;chen in ihrem urspr&uuml;nglichen Zustand. Rasterkraftmikroskopie best&auml;tigt, dass das erstelltes Protokoll die Titan-Mikrostruktur nicht ver&auml;ndert. Das Protokoll, die hierin beschriebenen auf fast allen Titan-Oberfl&auml;chen verwendet in biomedizinischen Wissenschaften und wegen seiner kurzen Belichtungszeit, K&ouml;nigswasser, anwendbar ist, wird wahrscheinlich Arbeit f&uuml;r viele Titan-Legierungen sowie.

Il titanio &egrave; uno dei materiali pi&ugrave; comunemente usati per i dispositivi impiantabili nell&#39;uomo. Scansione microscopia elettronica (SEM) serve come un importante strumento per le superfici di titanio di imaging e l&#39;analisi di cellule e altra materia organica aderendo al titanio impianti. Tuttavia, imaging SEM ad alto vuoto di un campione non conduttive richiede un rivestimento conduttivo sulla superficie. Un rivestimento oro/Palladio &egrave; comunemente usato e fino ad non oggi nessun metodo &egrave; stato descritto per &quot;pulire&quot; tali superfici coperta d&#39;oro/Palladio per esperimenti ripetuti senza incisione del titanio stesso. Questo costituisce un grave problema con dispositivi impiantabili a base di titanio che sono molto costosi e quindi fornire in breve. Il nostro obiettivo era quello di elaborare un protocollo per rigenerare titaniumsurfaces dopo analisi SEM. In una serie di esperimenti, campioni di titanio da dispositivi di assistenza cardiaco impiantabile erano rivestiti di fibronectina, seminati con le cellule e poi rivestiti con oro/Palladio per analisi SEM. Gli spettri di spettroscopia fotoelettronica a raggi x sono stati ottenuti prima e dopo cinque differenti protocolli di pulizie. Trattamento con acqua regia (1:3 soluzione di concentrato acido nitrico e acido cloridrico), con o senza ozonolisi, seguita da sonicazione nella soluzione di sapone e sonicazione in acqua deionizzata, ha permesso di rigenerare le superfici di titanio al loro stato originale. Microscopia a forza atomica ha confermato che il protocollo stabilito non alterava la microstruttura di titanio. Il protocollo descritto nel presente documento &egrave; applicabile a quasi tutte le superfici di titanio utilizzate nelle scienze biomediche e a causa del relativo tempo di esposizione breve a aqua regia, sar&agrave; probabile lavoro per molte leghe di titanio pure.

El titanio es uno de los materiales m&#xE1;s com&#xFA;nmente utilizados para dispositivos implantables en humanos. Microscop&#xED;a electr&#xF3;nica de barrido (MEB) sirve como una herramienta importante para las superficies de titanio de imagen y an&#xE1;lisis de las c&#xE9;lulas y otra materia org&#xE1;nica adherida a los implantes de titanio. Sin embargo, de alto vac&#xED;o im&#xE1;genes SEM de una muestra no conductor requiere un revestimiento conductor sobre la superficie. Un recubrimiento de oro / paladio se utiliza com&#xFA;nmente y hasta el momento no se ha descrito para &#x26;quot;limpiar&#x26;quot; el oro como / paladio superficies cubiertas de repetidos experimentos sin grabado el propio titanio. Esto constituye un problema importante con titanio basados &#x200B;&#x200B;en dispositivos implantables que son muy caros y, por tanto escasos. Nuestro objetivo fue elaborar un protocolo para regenerar titaniumsurfaces tras el an&#xE1;lisis SEM. En una serie de experimentos, las muestras de titanio de dispositivos de asistencia card&#xED;acos implantables fueron recubiertas con fibronectina, sembr&#xF3; con c&#xE9;lulas y luego revestida con oro / paladio para an&#xE1;lisis SEM. Espectros de rayos X espectroscopia de fotoelectrones se obtuvieron antes y despu&#xE9;s de cinco diferentes protocolos de limpieza. El tratamiento con agua regia (una soluci&#xF3;n 1:3 de n&#xED;trico concentrado y &#xE1;cido clorh&#xED;drico), con o sin ozon&#xF3;lisis, seguido por ultrasonidos en una soluci&#xF3;n de jab&#xF3;n y sonicaci&#xF3;n en agua desionizada, permite la regeneraci&#xF3;n de superficies de titanio a su estado original. Microscop&#xED;a de fuerza at&#xF3;mica confirm&#xF3; que el protocolo establecido no altera la microestructura de titanio. El protocolo se describe en este documento es aplicable a casi todas las superficies de titanio utilizadas en las ciencias biom&#xE9;dicas y debido a su corto tiempo de exposici&#xF3;n a agua regia, probablemente va a trabajar para las aleaciones de titanio muchos tambi&#xE9;n.

Effective ionic radius is found to be a primary determinant of the relative binding interactions of various guests, with small but measurable deviations in the case of nonspherical anions. Solvent effects are significant, and the strength of the binding interaction is found to depend directly on the donor ability of the solvent. A picture emerges in which anion binding can be effectively interpreted in terms of a competition between two solvation spheres: one provided by the solvent and a second dominated by a folded cavity lined with electropositive 1,2,3-triazole CH protons. Implications for rigid macrocycles and other multivalent hosts are discussed.

We evaluated the viability and adherence of peripheral blood-derived porcine endothelial progenitor cells (EPCs), seeded onto thin Ti layers on glass slides under static conditions and after exposure to fluid shear stresses. EPCs attached and grew to confluence on Ti in serum-free medium, without preadsorption of proteins. After attachment to Ti for 15 min, less than 5% of the cells detached at a shear stress of 100 dyne / cm(2). Confluent monolayers of EPCs on smooth Ti surfaces (Rq of 10 nm), exposed to 15 or 100 dyne/cm(2) for 48 h, aligned and elongated in the direction of flow and produced nitric oxide dependent on the level of shear stress. EPC-coated Ti surfaces had dramatically reduced platelet adhesion when compared to uncoated Ti surfaces. These results indicate that peripheral blood-derived EPCs adhere and function normally on Ti surfaces. Therefore EPCs may be used to seed cardiovascular devices prior to implantation to ameliorate platelet activation and thrombus formation.

Titanium (Ti) is commonly utilized in many cardiovascular devices, e.g. as a component of Nitinol stents, intra- and extracorporeal mechanical circulatory assist devices, but is associated with the risk of thromboemboli formation. We propose to solve this problem by lining the Ti blood-contacting surfaces with autologous peripheral blood-derived late outgrowth endothelial progenitor cells (EPCs) after having previously demonstrated that these EPCs adhere to and grow on Ti under physiological shear stresses and functionally adapt to their environment under flow conditions ex vivo. Autologous fluorescently-labeled porcine EPCs were seeded at the point-of-care in the operating room onto Ti tubes for 30 min and implanted into the pro-thrombotic environment of the inferior vena cava of swine (n = 8). After 3 days, Ti tubes were explanted, disassembled, and the blood-contacting surface was imaged. A blinded analysis found all 4 cell-seeded implants to be free of clot, whereas 4 controls without EPCs were either entirely occluded or partially thrombosed. Pre-labeled EPCs had spread and were present on all 4 cell-seeded implants while no endothelial cells were observed on control implants. These results suggest that late outgrowth autologous EPCs represent a promising source of lining Ti implants to reduce thrombosis in vivo.

Pre-etiquetados EPC se hab&#xED;a extendido y estaba presente en todos los cuatro cabeza de serie, los implantes de c&#xE9;lulas, mientras que no se observaron las c&#xE9;lulas endoteliales de los implantes de control. Estos resultados sugieren que las CPE excrecencia aut&#xF3;logas finales representan una fuente prometedora de revestimiento de los implantes de Ti para reducir la trombosis in vivo.
