Prolonged use of the dopamine precursor l-DOPA for the treatment of Parkinson's disease commonly results in abnormal involuntary movements, which are termed l-DOPA-induced dyskinesia (LID). Over-activity at corticostriatal synapses onto neurons of the direct and indirect striatal output pathways has been implicated in the development of dyskinesia, but it has proved difficult to investigate the pathways separately due to their morphological similarities. The recent development of bacterial artificial chromosome mice that express green fluorescent protein in either the direct or indirect pathway allows visual identification of the output neurons in each pathway. Here we describe the use of two different strains of these transgenic mice (pure FVB and FVB crossed with C57B1/6) in the development of mouse models of l-DOPA-induced dyskinesia. This model will allow the direct and indirect pathways to be studied individually to delineate the cellular and molecular mechanisms underlying dyskinesias. These studies demonstrate that mouse strain impacts on behavioural responses and l-DOPA sensitivity. Therefore, when generating mouse models of LID, strain must be taken into consideration when choosing the l-DOPA dosing regimen.

L&#x26;#39;utilisation prolong&#xE9;e du pr&#xE9;curseur de la dopamine L-DOPA pour le traitement de la maladie de Parkinson entra&#xEE;ne g&#xE9;n&#xE9;ralement des mouvements anormaux involontaires, qui sont appel&#xE9;s L-DOPA-la dyskin&#xE9;sie induite par (LID). Au cours de l&#x26;#39;activit&#xE9; au niveau des synapses-corticostriatal SUR DES neurones des voies directes et indirectes de sortie du striatum a &#xE9;t&#xE9; impliqu&#xE9; dans le d&#xE9;veloppement de la dyskin&#xE9;sie, mais il s&#x26;#39;est av&#xE9;r&#xE9; difficile d&#x26;#39;enqu&#xEA;ter sur les voies s&#xE9;par&#xE9;ment en raison de leurs similitudes morphologiques. Le d&#xE9;veloppement r&#xE9;cent de souris chromosomes artificiels de bact&#xE9;ries qui expriment la prot&#xE9;ine fluorescente verte soit dans la voie directe ou indirecte permet une identification visuelle des neurones de sortie dans chaque voie. Nous d&#xE9;crivons ici l&#x26;#39;utilisation de deux souches diff&#xE9;rentes de ces souris transg&#xE9;niques (FVB pur et FVB crois&#xE9; avec C57B1 / 6) dans le d&#xE9;veloppement de mod&#xE8;les murins de la L-DOPA-la dyskin&#xE9;sie induite par. Ce mod&#xE8;le permettra aux voies directes et indirectes qui seront &#xE9;tudi&#xE9;s individuellement afin de d&#xE9;limiter les m&#xE9;canismes cellulaires et mol&#xE9;culaires qui sous-tendent les dyskin&#xE9;sies. Ces &#xE9;tudes d&#xE9;montrent que les impacts souche de souris sur les r&#xE9;ponses comportementales et la L-DOPA sensibilit&#xE9;. Par cons&#xE9;quent, lors de la g&#xE9;n&#xE9;ration des mod&#xE8;les murins de LID, souche doit &#xEA;tre pris en consid&#xE9;ration lors du choix de la posologie de L-DOPA.

L&auml;ngere Anwendung von l-DOPA die Dopamin-Vorstufe zur Behandlung der Parkinsonschen Krankheit f&uuml;hrt h&auml;ufig zu abnorme unwillk&uuml;rliche Bewegungen, die l-DOPA-induzierte Dyskinesien (LID) bezeichnet werden. Over-Activity am Corticostriatal Synapsen auf Neuronen von den direkten und indirekten Striatale Ausgang Wege hat verwickelt wurden, bei der Entwicklung von Dyskinesien, aber es erwies sich schwer, die Bahnen getrennt aufgrund ihrer morphologischen &Auml;hnlichkeiten zu untersuchen. Die aktuelle Entwicklung der bacterial artificial Chromosome-M&auml;use, die gr&uuml;nes fluoreszierendes Protein in beiden Ausdr&uuml;cken erm&ouml;glicht die direkte oder indirekte Pathway visuellen Identifikation von der Ausgabeneuronen in jeder Weg. Hier beschreiben wir die Verwendung von zwei verschiedenen St&auml;mmen von diese transgenen M&auml;usen (reine FVB und FVB gekreuzt mit C57B1/6) bei der Entwicklung von Mausmodelle von l-DOPA-induzierte Dyskinesien. Dieses Modell erlaubt die direkte und indirekte Wege in einzeln untersucht werden, um die zellul&auml;ren und molekularen Mechanismen Dyskinesien abzugrenzen. Diese Studien zeigen, dass die Auswirkungen von Maus-Stamm auf Verhaltensst&ouml;rungen Antworten und l-DOPA-Empfindlichkeit. Daher muss beim Generieren von Mausmodelle des LIDES Belastung ber&uuml;cksichtigt werden bei der Wahl der Dosierung l-DOPA-Therapie.

Erstellung eines Modells von L-DOPA-induzierte Dyskinesien bei zwei verschiedene Maus-St&auml;mme.

L&#39;uso prolungato del precursore l-DOPA dopamina per il trattamento della malattia di Parkinson comunemente si traduce in movimenti involontari anormali, che sono denominati discinesia indotta da l-DOPA (coperchio). Eccessiva attivit&agrave; presso corticostriatal sinapsi sui neuroni di striatal della diretta e indiretta di output percorsi &egrave; stato implicato nello sviluppo di discinesia, ma si &egrave; dimostrato difficile da indagare le vie separatamente a causa della loro somiglianze morfologiche. Il recente sviluppo di topi cromosoma artificiale batterico che esprimono la proteina fluorescente verde in entrambi il pathway diretto o indiretto consente identificazione visiva dei neuroni di output in ogni percorso. Qui descriviamo l&#39;uso di due diversi ceppi di questi topi transgenici (puro FVB e FVB attraversato con C57B1/6) nello sviluppo di modelli murini della discinesia indotta da l-DOPA. Questo modello permetter&agrave; i percorsi diretti e indiretti essere studiato individualmente per delineare i meccanismi cellulari e molecolari alla base di discinesie. Questi studi dimostrano che il ceppo del mouse gli impatti sulle risposte comportamentali e sensibilit&agrave; di l-DOPA. Pertanto, durante la generazione di modelli murini di coperchio, ceppo deve tener conto quando si sceglie il regime di somministrazione di l-DOPA.

El uso prolongado del precursor de la dopamina L-DOPA en el tratamiento de la enfermedad de Parkinson suele dar lugar a movimientos involuntarios anormales, que se denominan L-DOPA discinesia inducida (LID). El exceso de actividad en las sinapsis en las neuronas del corticostriatal de las v&#xED;as de salida directos e indirectos del estriado ha sido implicado en el desarrollo de la discinesia, pero ha sido dif&#xED;cil de investigar las v&#xED;as por separado debido a sus similitudes morfol&#xF3;gicas. El reciente desarrollo de los ratones de cromosomas artificiales bacterianos que expresan la prote&#xED;na verde fluorescente, ya sea en la v&#xED;a directa o indirecta permite la identificaci&#xF3;n visual de las neuronas de salida de cada v&#xED;a. A continuaci&#xF3;n se describe el uso de dos cepas diferentes de estos ratones transg&#xE9;nicos (FVB pura y FVB cruzado con C57B1 / 6) en el desarrollo de modelos de rat&#xF3;n de la L-dopa disquinesia inducida. Este modelo permitir&#xE1; que las v&#xED;as directas e indirectas que se estudiar&#xE1;n de forma individual para delinear los mecanismos celulares y moleculares que subyacen a las discinesias. Estos estudios demuestran que los impactos cepa de rat&#xF3;n en las respuestas de comportamiento y la sensibilidad de L-DOPA. Por lo tanto, cuando la generaci&#xF3;n de modelos de rat&#xF3;n de la tapa, la cepa se debe tener en consideraci&#xF3;n al elegir el r&#xE9;gimen de dosificaci&#xF3;n de L-DOPA.
