The glycosaminoglycan, heparan sulfate (HS), binds proteins to modulate signaling events in embryogenesis. All identified protein-binding HS epitopes contain l-iduronic acid (IdoA). We report that targeted disruption of the murine d-glucuronyl C5-epimerase gene results in a structurally altered HS lacking IdoA. The corresponding phenotype is lethal, with renal agenesis, lung defects, and skeletal malformations. Unexpectedly, major organ systems, including the brain, liver, gastrointestinal tract, skin, and heart, appeared normal. We find that IdoA units are essential for normal kidney, lung, and skeletal development, albeit with different requirement for 2-O-sulfation. By contrast, major early developmental events known to critically depend on heparan sulfate apparently proceed normally even in the absence of IdoA.

Divulgamos que interrupci&oacute;n espec&iacute;fica del gen murino d-glucuronil C5-Epimerasa da lugar a un cap&iacute;tulo estructuralmente alterado falta IdoA. El fenotipo correspondiente es letal, con agenesia renal, defectos del pulm&oacute;n y malformaciones esquel&eacute;ticas. Inesperadamente, los sistemas de &oacute;rganos principales, incluyendo el cerebro, tracto gastrointestinal, piel, h&iacute;gado y coraz&oacute;n, aparec&iacute;an normales. Glucuronyl C5-epimerase catalyzes the conversion of d-glucuronic acid to l-iduronic acid units in heparan sulfate biosynthesis. Substrate recognition depends on the N-substituent pattern of the heparan sulfate precursor polysaccharide and requires the adjacent glucosamine residue toward the non-reducing end to be N-sulfated. Epimerization of an appropriately N-sulfated substrate is freely reversible in a soluble system, with equilibrium favoring retention of d-gluco configuration (Hagner-McWhirter, A., Lindahl, U., and Li, J.-P. (2000) Biochem. J. 347, 69-75). We studied the reversibility of the epimerase reaction in a cellular system, by incubating human embryonic kidney 293 cells with d-[5-(3)H]galactose. The label was incorporated with glucuronic acid units into the heparan sulfate precursor polysaccharide and was lost upon subsequent C5-epimerization to iduronic acid. However, analysis of oligosaccharides obtained by deaminative cleavage of the mature heparan sulfate chains indicated that all glucuronic acid units retained their C5-(3)H label, irrespective of whether they had occurred in sequences susceptible or resistant to the epimerase. All (3)H-labels of the final products resisted incubation with epimerase in a soluble system, apparently due to blocking O-sulfate groups. These results indicate that glucuronic acid C5-epimerization is effectively irreversible in vivo and argue for a stringent organization of the biosynthetic machinery.

Glucoronil C5-epimarasi catalizza la conversione dell&#39;acido d-glucuronico alle unit&agrave; di acido l-iduronico nella biosintesi di eparan solfato. Riconoscimento del substrato dipende il modello N-sostituente il polisaccaride precursore di eparan solfato e richiede il residuo di glucosamina adiacenti verso l&#39;estremit&agrave; non riducenti ad essere N-solfato. Epimerization di un substrato opportunamente N-solfato &egrave; liberamente reversibile in un sistema solubile, con equilibrio, favorendo la ritenzione di configurazione d-gluco (Hagner-McWhirter, A., Lindahl, U. e Li, J.-P. Biochem (2000). J. 347, 69-75). Abbiamo studiato la reversibilit&agrave; della reazione epimarasi in un sistema cellulare, incubando le cellule renali embrionali umane 293 con [5-(3) H] d-galattosio. L&#39;etichetta fu incorporata con unit&agrave; di acido glucuronico il polisaccaride precursore di eparan solfato e perse su successive C5-epimerization acido iduronico. Glucuronil C5-Epimerasa cataliza la conversi&oacute;n de &aacute;cido d-glucur&oacute;nico en unidades de &aacute;cido l-idur&oacute;nico en bios&iacute;ntesis de hepar&aacute;n sulfato. Reconocimiento del sustrato depende el patr&oacute;n N-sustituto del polisac&aacute;rido de precursor de hepar&aacute;n sulfato y requiere el residuo de glucosamina adyacentes hacia el extremo no reductor que N-sulfatada. Epimerizaci&oacute;n de un sustrato N-sulfatada apropiadamente es libremente reversible en un sistema soluble, con equilibrio, favoreciendo la retenci&oacute;n de la configuraci&oacute;n d-gluco (Hagner-McWhirter, A., Lindahl, U. y Li, j.-P. Biochem (2000). J. 347, 69-75). Estudiamos la reversibilidad de la reacci&oacute;n Epimerasa en un sistema celular, por incubaci&oacute;n de las c&eacute;lulas de ri&ntilde;&oacute;n embrionario humano 293 con d - galactosa [5-(3) H]. La etiqueta se incorpor&oacute; con unidades de &aacute;cido glucur&oacute;nico el polisac&aacute;rido de precursor de hepar&aacute;n sulfato y se perdi&oacute; en C5-epimerizaci&oacute;n posterior al &aacute;cido idur&oacute;nico. Sin embargo, el an&aacute;lisis de oligosac&aacute;ridos obtenidos por escote deaminative de las cadenas de maduro hepar&aacute;n sulfato indic&oacute; que todas las unidades de &aacute;cido glucur&oacute;nico conservan su etiqueta de C5 - 3 H, independientemente de si hab&iacute;an producido en secuencias susceptibles o resistentes a la Epimerasa. Western blotting is a powerful and widely used method, but limitations in detection sensitivity and specificity, and dependence upon high quality antibodies to detect targeted proteins, are hurdles to overcome. The in situ proximity ligation assay, based on dual antibody recognition and powerful localized signal amplification, offers increased detection sensitivity and specificity, along with an ability to identify complex targets such as phosphorylated or interacting proteins. Here we have applied the in situ proximity ligation assay mechanism in Western blotting. This combination allowed the use of isothermal rolling circle amplification of DNA molecules formed in target-specific ligation reaction, for 16-fold or greater increase in detection sensitivity. The increased specificity because of dual antibody recognition ensured highly selective assays, detecting the specific band when combinations of two cross-reactive antitubulin antibodies were used (i.e. both producing distinct nonspecific bands in traditional Western blotting). We also demonstrated detection of phosphorylated platelet-derived growth factor receptor Î² by proximity ligation with one antibody directed against the receptor and another directed against the phosphorylated tyrosine residue. This avoided the need for stripping and re-probing the membrane or aligning two separate traditional blots. We demonstrate that the high-performance in situ proximity ligation-based Western blotting described herein is compatible with detection via enhanced chemiluminescence and fluorescence detection systems, and can thus be readily employed in any laboratory.

Western Blot es un de gran alcance y m&eacute;todo ampliamente utilizado, pero con limitaciones en la sensibilidad de detecci&oacute;n y especificidad y dependencia de los anticuerpos de alta calidad para detectar prote&iacute;nas espec&iacute;ficas, son obst&aacute;culos que superar. El ensayo de ligadura de proximidad in situ, basado en el reconocimiento de doble anticuerpo y amplificaci&oacute;n de la se&ntilde;al localizada potente, ofrece detecci&oacute;n mayor sensibilidad y especificidad, junto con una capacidad para identificar objetivos complejos como las prote&iacute;nas fosforiladas o interacci&oacute;n. Aqu&iacute; hemos aplicado el mecanismo de an&aacute;lisis in situ de proximidad ligadura en Western blotting. Esta combinaci&oacute;n permite el uso de isot&eacute;rmico balanceo amplificaci&oacute;n de c&iacute;rculo de mol&eacute;culas de ADN formadas en reacci&oacute;n de ligadura espec&iacute;fico del destino, para 16-fold o mayor incremento en la sensibilidad de detecci&oacute;n. La mayor especificidad por reconocimiento de doble anticuerpo asegur&oacute; ensayos altamente selectivos, detectar la banda espec&iacute;fica cuando se utilizan combinaciones de dos anticuerpos antitubulin cruz-reactivos (es decir, ambos productores inespec&iacute;ficos vendas distintas en Western blotting tradicional). Tambi&eacute;n demostramos la detecci&oacute;n de factor de crecimiento derivado de plaquetas fosforilado del receptor &beta; por ligadura de proximidad con un anticuerpo dirigido contra el receptor y otra dirigida contra los residuos de tirosina fosforilados. Esto evita la necesidad de pelar y re-probing de la membrana o alinear dos lacras tradicionales separados. Demostramos que la proximidad de in situ de alto rendimiento basadas en la ligadura Western Blot descrito en este documento es compatible con la detecci&oacute;n mediante sistemas de detecci&oacute;n de fluorescencia y quimioluminiscencia mejorada y as&iacute; puede utilizarse f&aacute;cilmente en cualquier laboratorio.
