We previously developed a high-throughput system called 'Ice-Cap' for growing Arabidopsis seedlings in a 96-well format and rapidly collecting tissue for subsequent DNA extraction and genotyping. While the originally described Ice-Cap method is an effective tool for high-throughput genotyping, one shortcoming of the first version of Ice-Cap is that optimal seedling growth is highly dependent on specific environmental conditions. Here we describe several technical improvements to the Ice-Cap method that make it much more robust and provide a detailed protocol for implementing the method.

Hier beschreiben wir einige technische Verbesserungen der Eiskappe-Methode, die machen es viel robuster und bieten ein ausf&uuml;hrliches Protokoll f&uuml;r die Implementierung der Methode.

Abbiamo sviluppato in precedenza un sistema high-throughput chiamato &#39;Calotta&#39; per la coltivazione di piantine di Arabidopsis in un formato a 96 pozzetti e rapidamente raccogliendo il tessuto per la successiva estrazione del DNA e tipizzazione. Anteriormente hemos desarrollado un sistema de alto rendimiento llamada &#x26;quot;capa de hielo&#x26;quot; para el crecimiento de pl&#xE1;ntulas de Arabidopsis en un formato de 96 pocillos y r&#xE1;pido de recopilaci&#xF3;n de tejido para su posterior extracci&#xF3;n de ADN y el genotipado. Sammlungen von Arabidopsis Thaliana bestellt Zeilen mit zugeordnete T-DNA-Einf&uuml;gungen eine wichtige Ressource f&uuml;r Pflanze Wissenschaftler Durchf&uuml;hrung genetische Untersuchungen geworden. Fr&uuml;here Berichte gaben an, dass die T-DNA Einf&uuml;gung Linien k&ouml;nnen chromosomale Translokationen der T-DNA Einf&uuml;gung Website zugeordnet haben, aber die Pr&auml;valenz von diese Umlagerungen nicht gut dokumentiert. Um die Frequenz zu bestimmen mit der Umsiedlung in eine weit verbreitete Sammlung von T-DNA Einf&uuml;gung Zeilen vorhanden sind, analysierten wir 64 unabh&auml;ngige Leitungen aus der Salk T-DNA-mutant-Auflistung. Chromosomale Translokationen wurden bei 12 der Befragten 64 Linien (19 %) festgestellt. Zwei Proben wurden verwendet, um die T-DNA-Linien f&uuml;r Umsiedlung des Bildschirms: Bl&uuml;tenstaub Lebensf&auml;higkeit und genomweiten genetische Zuordnung. Obwohl die Messung der Bl&uuml;tenstaub Lebensf&auml;higkeit eine indirekte Bildschirm f&uuml;r das Vorhandensein einer Translokation ist, wurden alle 11 der T-DNA Linien zeigen eine abnorme Bl&uuml;tenstaub-Ph&auml;notyp festgestellt, dass eine Translokation bei Analyse der genetischen Zuordnung enthalten. Ein normalen Bl&uuml;tenstaub-Ph&auml;notyp garantiert jedoch nicht, ohne eine Translokation. Wir beobachteten eine T-DNA-Linie mit normalen Pollen, die dennoch eine Translokation hatte basierend auf genetische Zuordnung Ergebnisse. Eine zus&auml;tzliche Ph&auml;nomen, das wir durch unsere genetische Zuordnung-Experimenten beobachtet wurde, dass die T-DNA-Kreuzungen auf die 5&#39;-und 3&#39;-Seiten eines gezielten Gens k&ouml;nnen in einigen F&auml;llen genetisch voneinander trennen. Zwei Zeilen in unserer Umfrage angezeigt diese &#39; t-DNA Grenzen Separate&#39; Ph&auml;nomen. Experimentelle Verfahren f&uuml;r das screening von T-DNA-Linien f&uuml;r das Vorhandensein von Chromosomenanomalien effizient werden vorgestellt und diskutiert.

Ordered collections of Arabidopsis thaliana lines containing mapped T-DNA insertions have become an important resource for plant scientists performing genetic studies. Previous reports have indicated that T-DNA insertion lines can have chromosomal translocations associated with the T-DNA insertion site, but the prevalence of these rearrangements has not been well documented. To determine the frequency with which translocations are present in a widely-used collection of T-DNA insertion lines, we analyzed 64 independent lines from the Salk T-DNA mutant collection. Chromosomal translocations were detected in 12 of the 64 lines surveyed (19%). Two assays were used to screen the T-DNA lines for translocations: pollen viability and genome-wide genetic mapping. Although the measurement of pollen viability is an indirect screen for the presence of a translocation, all 11 of the T-DNA lines showing an abnormal pollen phenotype were found to contain a translocation when analyzed using genetic mapping. A normal pollen phenotype does not, however, guarantee the absence of a translocation. We observed one T-DNA line with normal pollen that nevertheless had a translocation based on genetic mapping results. One additional phenomenon that we observed through our genetic mapping experiments was that the T-DNA junctions on the 5'- and 3'-sides of a targeted gene can genetically separate from each other in some cases. Two of the lines in our survey displayed this 'T-DNA borders separate' phenomenon. Experimental procedures for efficiently screening T-DNA lines for the presence of chromosomal abnormalities are presented and discussed.

Chromosomal translocations are a common phenomenon in Arabidopsis thaliana T-DNA insertion lines.

&#xC5FC;&#xC0C9;&#xCCB4; translocations&#xB294; Arabidopsis thaliana T DNA &#xC0BD;&#xC785; &#xB77C;&#xC778;&#xC5D0;&#xC11C; &#xC77C;&#xBC18;&#xC801;&#xC778; &#xD604;&#xC0C1;.

Colecciones ordenadas de las l&#xED;neas de Arabidopsis thaliana que contienen asignadas inserciones de ADN-T se han convertido en un recurso importante para los cient&#xED;ficos que realizan estudios gen&#xE9;ticos de las plantas. Informes anteriores han indicado que el T-DNA de l&#xED;neas de inserci&#xF3;n puede tener translocaciones cromos&#xF3;micas asociadas con el sitio de inserci&#xF3;n de T-ADN, pero la prevalencia de estos reordenamientos no ha sido bien documentado. Para determinar la frecuencia con que las translocaciones est&#xE1;n presentes en una colecci&#xF3;n ampliamente utilizado de las l&#xED;neas de inserci&#xF3;n de T-ADN, se analizaron 64 l&#xED;neas independientes de la Salk T-DNA de la colecci&#xF3;n mutante. Translocaciones cromos&#xF3;micas fueron detectados en 12 de las 64 l&#xED;neas de encuestados (19%). Dos ensayos se utiliza para la inspecci&#xF3;n de las l&#xED;neas T-DNA de translocaciones: la viabilidad del polen y de todo el genoma de mapeo gen&#xE9;tico.
