Loss-of-function, dominant-negative, and change-of-function genetic approaches were used to investigate the role played by the Arabidopsis mitogen-activated protein (MAP) kinase MPK6 throughout development. Plants homozygous for T-DNA null alleles of MPK6 displayed reduced male fertility and abnormal anther development. In addition, a portion of the seed produced by mpk6 plants was found to contain embryos that had burst out of their seed coats. To address potential functional redundancy, a dominant-negative version of MPK6 was constructed by changing the TEY activation loop motif to the amino acid sequence AEF. Plants expressing MPK6AEF via the MPK6 native promoter were found to produce excessive stomata, consistent with the recently described role of MPK6 in stomatal patterning. A novel floral phenotype characterized by abnormal sepal development was also observed in MPK6AEF lines. The gene expression pattern of the MPK6 native promoter was determined using a YFP-MPK6 fusion construct, and expression was observed throughout most plant tissues, consistent with a role for MPK6 in multiple developmental processes. The YFP-MPK6 construct was found to rescue the fertility phenotype of mpk6 null alleles, indicating that the fusion protein retains its biological activity. It was also observed, however, that plants expressing YFP-MPK6 displayed reduced apical dominance and a shortening of inflorescence internodes. These results suggest that the YFP tag modifies the activity of MPK6 in a manner that affects inflorescence development but not anther development. Taken together, the present results indicate that MPK6 is involved in the regulation of multiple aspects of plant development.

La p&#xE9;rdida de la funci&#xF3;n, dominante-negativa, y el cambio de la funci&#xF3;n de los enfoques gen&#xE9;ticos han sido utilizados para investigar el papel desempe&#xF1;ado por la Arabidopsis activadas por mit&#xF3;genos (MAP) quinasa MPK6 lo largo del desarrollo. Las plantas homocigotos para alelos nulos de T-ADN de MPK6 muestran reducci&#xF3;n de la fertilidad masculina y desarrollo de la antera anormal. Adem&#xE1;s, una porci&#xF3;n de la semilla producida por mpk6 plantas se encontr&#xF3; que conten&#xED;a embriones que hab&#xED;an salido de sus capas de semillas. Para hacer frente a la redundancia funcional potencial, una versi&#xF3;n dominante negativo de MPK6 se construy&#xF3; cambiando el motivo de activaci&#xF3;n bucle TEY a la AEF secuencia de amino&#xE1;cidos. Las plantas que expresan a trav&#xE9;s de la promotora MPK6AEF MPK6 nativa se encontr&#xF3; que producen exceso de estomas, en consonancia con el papel de la recientemente descrita MPK6 en los patrones de los estomas. Un nuevo fenotipo floral caracterizado por el desarrollo anormal de los s&#xE9;palos tambi&#xE9;n se observ&#xF3; en l&#xED;neas MPK6AEF. TILLING (for Targeting Induced Local Lesions IN Genomes) is a well-established method for identifying plants carrying point mutations in genes of interest. A traditional TILLING project requires a significant investment of time and resources to establish the mutant population and screening infrastructure. Here, we describe a modified TILLING procedure that substantially reduces the investment needed to perform mutation screening. Our motivation for developing iTILLING was to make it practical for individual laboratories to rapidly perform mutation screens using specialized genetic backgrounds. With iTILLING, M2 seeds are collected in bulk from the mutagenized population of plants, greatly reducing the labor needed to manage the mutant lines. Growth of the M2 seedlings for mutation screening, tissue collection, and DNA extraction are all performed in 96-well format. Mutations are then identified using high-resolution melt-curve analysis of gene-specific polymerase chain reaction products. Individual plants carrying mutations of interest are transferred from the 96-well growth plates to soil. One scientist can complete an iTILLING screen in less than 4 months. As a proof-of-principle test, we applied iTILLING to Arabidopsis (Arabidopsis thaliana) plants that were homozygous for the mekk1-1 (for MAPK/ERK kinase kinase 1) mutation and also carried a MEKK1 rescue construct. The goal of our screen was to identify mutations in the closely linked MEKK2 and MEKK3 loci. We obtained five mutations in MEKK2 and seven mutations in MEKK3, all located within 20 kb of the mekk1-1 T-DNA insertion. Using repeated iterations of the iTILLING process, mutations in three or more tandemly duplicated genes could be generated.

La croissance des plants M2 pour le d&#xE9;pistage de mutation, la collecte de tissus, et l&#x26;#39;extraction d&#x26;#39;ADN sont tous r&#xE9;alis&#xE9;s en format 96 puits. Comme un test de validation de principe, nous avons appliqu&#xE9; &#xE0; iTILLING (Arabidopsis thaliana) Arabidopsis plantes qui &#xE9;taient homozygotes pour la MEKK1-1 (pour MAPK / ERK kinase kinase 1) mutation et &#xE9;galement r&#xE9;alis&#xE9; une op&#xE9;ration de sauvetage MEKK1 construire. ITILLING: une approche personnalis&#xE9;e pour l&#x26;#39;identification des mutations induites chez Arabidopsis

TILLING (f&uuml;r Targeting Induced Local L&auml;sionen IN Genomen) ist eine allgemein anerkannte Methode zur Identifizierung von Pflanzen, die mit Punkt-Mutationen in Genen von Interesse. Ein traditionelles TILLING-Projekt erfordert eine bedeutende Investition von Zeit und Ressourcen zur Etablierung mutant Bev&ouml;lkerung und Screening-Infrastruktur. Hier beschreiben wir eine modifizierte TILLING-Prozedur, die die Mutation Screening durchf&uuml;hren erforderlichen Investitionen erheblich reduziert. Unsere Motivation f&uuml;r die Entwicklung von iTILLING wurde um es praktisch f&uuml;r einzelne Laboratorien Mutation Bildschirme verwenden spezielle genetische Hintergr&uuml;nde schnell durchf&uuml;hren. Mit iTILLING sind mutagenized Bev&ouml;lkerung der Pflanzen stark reduziert die Arbeit ben&ouml;tigt, um die Mutanten Linien M2 Samen in gro&szlig;en Mengen gesammelt. Wachstum der S&auml;mlinge M2 f&uuml;r Mutation Screening, Gewebe-Auflistung und DNA-Extraktion werden in 96-Well-Format ausgef&uuml;hrt. Die Mutationen sind dann mit hochaufl&ouml;senden Schmelze-Kurvenanalyse Gen-spezifischen Polymerase-Kettenreaktion-Produkte. Einzelpflanzen mit Mutationen des Interesses werden von den Wachstum 96-Well-Platten zum Boden &uuml;bertragen. Ein Wissenschaftler kann einen iTILLING-Bildschirm in weniger als 4 Monaten abgeschlossen werden. Als Beweis-des-Prinzip Test wir iTILLING auf Arabidopsis (Arabidopsis Thaliana) Pflanzen, die homozygot f&uuml;r die Mutation mekk1-1 (f&uuml;r MAPK/ERK-Kinase-Kinase 1) wurden und auch ein MEKK1-Rettung-Konstrukt angewendet. Das Ziel von unserem Bildschirm war Mutationen in der eng verbundenen MEKK2 und MEKK3 Loci identifiziert. Wir erhielten f&uuml;nf Mutationen im MEKK2 und sieben Mutationen im MEKK3, alle gelegen innerhalb der mekk1-1 T-DNA Einf&uuml;gung 20 kb. Mit wiederholte Iterationen des iTILLING Prozesses, konnten Mutationen in drei oder mehr tandemly duplizierten Genen generiert werden.

iTILLING: eine personalisierte Ansatz zur Identifizierung der induzierten Mutationen in Arabidopsis.

TILLING (per Targeting Induced Local lesioni IN genomi) &egrave; un metodo consolidato per l&#39;identificazione di piante che trasportano le mutazioni puntiformi nei geni di interesse. Un progetto TILLING tradizionale richiede un notevole investimento di tempo e risorse per stabilire la popolazione mutante e infrastruttura di screening. Qui, descriviamo una procedura TILLING modificata che riduce notevolmente gli investimenti necessari per eseguire lo screening della mutazione. La nostra motivazione per lo sviluppo di iTILLING era di rendere pratico per singoli laboratori eseguire rapidamente schermi mutazione utilizzando specializzati background genetico. Con iTILLING, M2 semi sono raccolti in massa della popolazione mutagenized delle piante, riducendo notevolmente il lavoro necessario per gestire le linee mutanti. Crescita dei semenzali M2 per lo screening della mutazione, tessuto insieme ed estrazione del DNA sono tutte eseguite in formato a 96 pozzetti. iTILLING: Arabidopsis &#xC720;&#xBC1C;&#xB41C; &#xB3CC;&#xC5F0;&#xBCC0;&#xC774;&#xC758; &#xC2DD;&#xBCC4;&#xC5D0; &#xB9DE;&#xCDA4;&#xD615;&#xB41C; &#xC811;&#xADFC; &#xBC29;&#xC2DD;.

TILLING (de orientaci&#xF3;n inducida lesiones locales en los genomas) es un m&#xE9;todo bien establecido para la identificaci&#xF3;n de las plantas portadoras de mutaciones puntuales en los genes de inter&#xE9;s. Un proyecto TILLING tradicional requiere una importante inversi&#xF3;n de tiempo y recursos para establecer la poblaci&#xF3;n mutante y la infraestructura de detecci&#xF3;n. A continuaci&#xF3;n, describimos un procedimiento TILLING modificado que reduce sustancialmente la inversi&#xF3;n necesaria para llevar a cabo la detecci&#xF3;n de mutaciones.
