Malaria parasite transmission depends on the successful transition of Plasmodium through discrete developmental stages in the lumen of the mosquito midgut. Like the human intestinal tract, the mosquito midgut contains a diverse microbial flora, which may compromise the ability of Plasmodium to establish infection. We have identified an Enterobacter bacterium isolated from wild mosquito populations in Zambia that renders the mosquito resistant to infection with the human malaria parasite Plasmodium falciparum by interfering with parasite development before invasion of the midgut epithelium. Phenotypic analyses showed that the anti-Plasmodium mechanism requires small populations of replicating bacteria and is mediated through a mosquito-independent interaction with the malaria parasite. We show that this anti-Plasmodium effect is largely caused by bacterial generation of reactive oxygen species.

Nous avons identifi&#xE9; une bact&#xE9;rie Enterobacter isol&#xE9;e des populations de moustiques sauvages en Zambie qui rend le moustique r&#xE9;sistant &#xE0; l&#x26;#39;infection par le parasite du paludisme humain Plasmodium falciparum en interf&#xE9;rant avec le d&#xE9;veloppement du parasite avant l&#x26;#39;invasion de l&#x26;#39;&#xE9;pith&#xE9;lium de l&#x26;#39;intestin. Analyses ph&#xE9;notypiques ont montr&#xE9; que le m&#xE9;canisme anti-Plasmodium exige de petites populations de bact&#xE9;ries et de la r&#xE9;plication est m&#xE9;di&#xE9;e par une interaction moustiques ind&#xE9;pendant avec le parasite du paludisme. Nous montrons que cet effet anti-Plasmodium est en grande partie due &#xE0; la production bact&#xE9;rienne d&#x26;#39;esp&#xE8;ces r&#xE9;actives de l&#x26;#39;oxyg&#xE8;ne.

Malaria-Parasiten-&Uuml;bertragung h&auml;ngt von den erfolgreichen &Uuml;bergang von Plasmodium durch diskrete Entwicklungsstadien in das Lumen des der Moskito Mitteldarm. Wie den menschlichen Darm-Trakt enth&auml;lt die Moskito Mitteldarm eine vielf&auml;ltige mikrobielle Flora, die die M&ouml;glichkeit von Plasmodium, Infektion herzustellen gef&auml;hrden kann. Wir haben festgestellt, dass ein Enterobacter-Bakterium aus wilden Moskito-Populationen in Sambia, die die M&uuml;cke widerstandsf&auml;hig gegen Infektion mit der menschlichen Malaria Parasit Plasmodium Falciparum rendert durch Eingriffe in Parasit Entwicklung vor den Mitteldarm Epithelinvasion isoliert. Ph&auml;notypische Analysen zeigten, dass der anti-Plasmodium Mechanismus kleine Populationen erfordert der Replikation von Bakterien und durch einen Moskito-unabh&auml;ngige Interaktion mit der Malaria-Parasiten vermittelt wird. Wir zeigen, dass diese anti-Plasmodium Wirkung weitgehend durch bakterielle Generation reaktive Sauerstoffspezies zur&uuml;ckzuf&uuml;hren ist.

Come il tratto intestinale umano, l&#39;intestino della zanzara contiene una flora microbica varia, che pu&ograve; compromettere la capacit&agrave; di stabilire infezione da Plasmodium. Abbiamo individuato un batterio Enterobacter isolato da popolazioni selvatiche zanzara in Zambia che esegue il rendering della zanzara resistente all&#39;infezione con il parassita Plasmodium falciparum malaria umana interferendo con lo sviluppo del parassita prima invasione dell&#39;epitelio intestino. Analisi fenotipiche ha dimostrato che il meccanismo di anti-Plasmodium richiede piccole popolazioni di batteri la replica ed &egrave; mediato attraverso un&#39;interazione di zanzara-indipendente con il parassita della malaria. Surveillance for drug-resistant parasites in human blood is a major effort in malaria control. Here we report contrasting antifolate resistance polymorphisms in Plasmodium falciparum when parasites in human blood were compared with parasites in Anopheles vector mosquitoes from sleeping huts in rural Zambia. DNA encoding P. falciparum dihydrofolate reductase (EC 1.5.1.3) was amplified by PCR with allele-specific restriction enzyme digestions. Markedly prevalent pyrimethamine-resistant mutants were evident in human P. falciparum infections--S108N (>90%), with N51I, C59R, and 108N+51I+59R triple mutants (30-80%). This resistance level may be from selection pressure due to decades of sulfadoxine/pyrimethamine use in the region. In contrast, cycloguanil-resistant mutants were detected in very low frequency in parasites from human blood samples-S108T (13%), with A16V and 108T+16V double mutants (âˆ¼4%). Surprisingly, pyrimethamine-resistant mutants were of very low prevalence (2-12%) in the midguts of Anopheles arabiensis vector mosquitoes, but cycloguanil-resistant mutants were highly prevalent-S108T (90%), with A16V and the 108T+16V double mutant (49-57%). Structural analysis of the dihydrofolate reductase by in silico modeling revealed a key difference in the enzyme within the NADPH binding pocket, predicting the S108N enzyme to have reduced stability but the S108T enzyme to have increased stability. We conclude that P. falciparum can bear highly host-specific drug-resistant polymorphisms, most likely reflecting different selective pressures found in humans and mosquitoes. Thus, it may be useful to sample both human and mosquito vector infections to accurately ascertain the epidemiological status of drug-resistant alleles.

Plasmodium falciparum dihydrofolate reductase (DHFR) polymorphisms &#xC778;&#xAC04;&#xACFC; Anopheles &#xBAA8;&#xAE30;&#xC5D0; &#xB300;&#xC870;&#xC640; &#xB9D0;&#xB77C;&#xB9AC;&#xC544; antifolate &#xC800;&#xD56D;.

La vigilancia de la droga-resistentes a los par&#xE1;sitos en la sangre humana es un esfuerzo importante en el control de la malaria. Aqu&#xED; mostramos que contrasta polimorfismos antifolatos de resistencia en los par&#xE1;sitos Plasmodium falciparum, cuando en la sangre humana se compararon con los par&#xE1;sitos en los mosquitos Anopheles vectores de dormir en caba&#xF1;as de zonas rurales de Zambia. ADN que codifica el P. falciparum dihidrofolato reductasa (CE 1.5.1.3) se amplific&#xF3; por PCR con alelo-espec&#xED;ficas digestiones de enzimas de restricci&#xF3;n. Marcadamente predominantes pirimetamina mutantes resistentes fueron evidentes en humanos infecciones por P. falciparum - S108N (&#x26;gt; 90%), con N51I, C59R, y 108N +51 +59 I mutantes R triples (30-80%). Este nivel de resistencia puede ser de la presi&#xF3;n de selecci&#xF3;n debido a d&#xE9;cadas de sulfadoxina / pirimetamina uso en la regi&#xF3;n. En contraste, cicloguanil mutantes resistentes se detectaron en frecuencia muy baja en los par&#xE1;sitos de la sangre humana muestras-S108T (13%), con A16V y 108T +16 V mutantes dobles (~ 4%). Sorprendentemente, los mutantes resistentes a la pirimetamina eran de muy baja prevalencia (2-12%) en el intestino medio de los mosquitos Anopheles arabiensis vector, pero cicloguanil mutantes resistentes tuvieron una alta prevalencia-S108T (90%), con A16V y el 108T +16 V doble mutante (49-57%). El an&#xE1;lisis estructural de la dihidrofolato reductasa por el modelado in silico revel&#xF3; una diferencia fundamental en la enzima en el bolsillo de uni&#xF3;n de NADPH, la predicci&#xF3;n de la enzima S108N haber reducido la estabilidad, pero la enzima S108T que ha aumentado la estabilidad. Llegamos a la conclusi&#xF3;n que el P. falciparum puede soportar gran acogida espec&#xED;ficos resistentes a los medicamentos polimorfismos, m&#xE1;s probable es que reflejen las distintas presiones selectivas que se encuentran en los seres humanos y los mosquitos. Por lo tanto, puede ser &#xFA;til para muestrear las infecciones vector tanto humanos como mosquitos para determinar con precisi&#xF3;n la situaci&#xF3;n epidemiol&#xF3;gica de los alelos resistentes a los f&#xE1;rmacos.
