An abnormal growth form called mound has been hypothesized to be a neoplasm in the filamentous fungus Schizophyllum commune. An alternative hypothesis is that mounds represent some unusual developmental form in the fruiting body morphogenetic pathway. Hydrophobin proteins have been found in fruiting bodies where they line the surface of gas exchange pores and function to keep the pores hydrophobic. To further determine possible relationships between mounds and fruiting bodies, mound tissue was examined for gas exchange pores and the presence of hydrophobins. Cryoscanning electron microscopic images revealed the presence of channels in mound tissue and presumptive hydrophobin rodlets similar to the air channels in fruiting bodies. Hydrophobin gene expression was also measured in mound tissue using quantitative real-time PCR and showed both monokaryotic and dikaryotic mound tissue exhibited high expression of the dikaryotic specific Sc4 hydrophobin gene. In contrast, Sc4 hydrophobin expression was barely detectable in monokaryotic fruiting bodies. The expression of Sc4 hydrophobin genes in mounds suggests mound development uses this aspect of the dikaryotic fruiting developmental pathway.

The high cost of enzymes is a major bottleneck preventing the development of an economically viable lignocellulosic ethanol industry. Commercial enzyme cocktails for the conversion of plant biomass to fermentable sugars are complex mixtures containing more than 80 proteins of suboptimal activities and relative proportions. As a step toward the development of a more efficient enzyme cocktail for biomass conversion, we have developed a platform, called GENPLAT, that uses robotic liquid handling and statistically valid experimental design to analyze synthetic enzyme mixtures. Commercial enzymes (Accellerase 1000 +/- Multifect Xylanase, and Spezyme CP +/- Novozyme 188) were used to test the system and serve as comparative benchmarks. Using ammonia-fiber expansion (AFEX) pretreated corn stover ground to 0.5 mm and a glucan loading of 0.2%, an enzyme loading of 15 mg protein/g glucan, and 48 h digestion at 50 degrees C, commercial enzymes released 53% and 41% of the available glucose and xylose, respectively. Mixtures of three, five, and six pure enzymes of Trichoderma species, expressed in Pichia pastoris, were systematically optimized. Statistical models were developed for the optimization of glucose alone, xylose alone, and the average of glucose + xylose for two digestion durations, 24 and 48 h. The resulting models were statistically significant (P < 0.0001) and indicated an optimum composition for glucose release (values for optimized xylose release are in parentheses) of 29% (5%) cellobiohydrolase 1, 5% (14%) cellobiohydrolase 2, 25% (25%) endo-beta1,4-glucanase 1, 14% (5%) beta-glucosidase, 22% (34%) endo-beta1,4-xylanase 3, and 5% (17%) beta-xylosidase in 48 h at a protein loading of 15 mg/g glucan. Comparison of two AFEX-treated corn stover preparations ground to different particle sizes indicated that particle size (100 vs. 500 microm) makes a large difference in total digestibility. The assay platform and the optimized "core" set together provide a starting point for the rapid testing and optimization of alternate core enzymes from other microbial and recombinant sources as well as for the testing of "accessory" proteins for development of superior enzyme mixtures for biomass conversion.

Als Schritt zur Entwicklung einer effizienteren Enzym cocktail f&uuml;r Biomasse-Konversion entwickelten wir eine Plattform, genannt GENPLAT, das Roboter-liquid-Handling und statistisch valide experimentelles Design verwendet, um synthetische Enzym Mischungen zu analysieren. Kommerzielle Enzyme (Accellerase 1000 +/-Multifect Xylanase) und Spezyme CP +/-Novozyme 188 wurden verwendet, um das System zu testen und dienen als vergleichenden Benchmarks. El alto costo de las enzimas es un cuello de botella importante que impide el desarrollo de una industria de etanol lignocelul&#xF3;sico econ&#xF3;micamente viable. C&#xF3;cteles comerciales de enzimas para la conversi&#xF3;n de la biomasa vegetal en az&#xFA;cares fermentables son mezclas complejas que contienen m&#xE1;s de 80 prote&#xED;nas de actividades sub&#xF3;ptimas y proporciones relativas. Como un paso hacia el desarrollo de un c&#xF3;ctel de enzimas m&#xE1;s eficientes para la conversi&#xF3;n de la biomasa, hemos desarrollado una plataforma, llamada GENPLAT, que utiliza la manipulaci&#xF3;n rob&#xF3;tica l&#xED;quido y el dise&#xF1;o experimental estad&#xED;sticamente v&#xE1;lida para analizar mezclas sint&#xE9;ticas de enzimas. Enzimas comerciales (Accellerase 1000 + / - Multifect xilanasa, y CP Spezyme + / - Novozyme 188) se utilizaron para probar el sistema y sirven como puntos de referencia comparativos. Usando amon&#xED;aco-fibra de expansi&#xF3;n (AFEX) ma&#xED;z pretratado suelo rastrojo a 0,5 mm y una carga de glucano de 0,2%, una carga enzima de 15 mg de prote&#xED;na / g glucano, y 48 h de digesti&#xF3;n a 50 grados C, enzimas comerciales liberado el 53% y 41 % de la glucosa disponible y xilosa, respectivamente. Las mezclas de tres, cinco, seis y enzimas puras de especies de Trichoderma, expresada en Pichia pastoris, se optimizaron sistem&#xE1;ticamente. Los modelos estad&#xED;sticos fueron desarrollados para la optimizaci&#xF3;n de la glucosa sola, xilosa solo, y el promedio de glucosa + xilosa por dos duraciones de digesti&#xF3;n, 24 y 48 h. Los modelos resultantes fueron estad&#xED;sticamente significativas (P &#x26;lt;0,0001) y se indica una composici&#xF3;n &#xF3;ptima para la liberaci&#xF3;n de glucosa (cifras para la versi&#xF3;n optimizada de la xilosa se encuentran en par&#xE9;ntesis) de 29% (5%) celobiohidrolasa 1, 5% (14%) celobiohidrolasa 2, el 25% (25%) endo-beta 1 ,4-glucanasa 1, 14% (5%) de beta-glucosidasa, 22% (34%) endo-beta 1 ,4-xilanasa 3, y 5% (17%) de beta-xilosidasa en 48 h con una carga de prote&#xED;na de 15 mg / g de glucano. Comparaci&#xF3;n de dos AFEX tratado con suelo el rastrojo de ma&#xED;z preparativos para diferentes tama&#xF1;os de part&#xED;culas indican que el tama&#xF1;o de part&#xED;cula (100 frente a 500 micras) hace una gran diferencia en la digestibilidad total. A high throughput enzyme assay platform, called GENPLAT, was used to guide the development of an optimized mixture of individual purified enzymes from ten "accessory" and six "core" enzymes. Enzyme mixtures were optimized for release of Glu, Xyl, or a combination of the two from corn stover pretreated by ammonia-fiber expansion (AFEX). Assay conditions were a fixed enzyme loading of 15 mg/g glucan, 48 h digestion, and 50 degrees C. Five of the ten tested accessory proteins enhanced Glu or Xyl yield compared to the core set alone, and five did not. An 11-component mixture containing the core set and five accessory enzymes optimized for Glu released 52.1% of the available Glu, compared to 38.5% with the core set alone. A mixture optimized for Xyl released 39.9% of the Xyl, compared to 26.4% with the core set alone. We predict that there is still considerable opportunity for further improvement of synthetic mixtures. Furthermore, the strategy described here is applicable to the development of more efficient enzyme cocktails for any pretreatment/biomass combination and for detecting enzymes that make a heretofore unrecognized contribution to lignocellulose deconstruction.

Enzymes for plant cell wall deconstruction are a major cost in the production of ethanol from lignocellulosic biomass. The goal of this research was to develop optimized synthetic mixtures of enzymes for multiple pretreatment/substrate combinations using our high-throughput biomass digestion platform, GENPLAT, which combines robotic liquid handling, statistical experimental design and automated Glc and Xyl assays. Proportions of six core fungal enzymes (CBH1, CBH2, EG1, Î²-glucosidase, a GH10 endo-Î²1,4-xylanase, and Î²-xylosidase) were optimized at a fixed enzyme loading of 15 mg/g glucan for release of Glc and Xyl from all combinations of five biomass feedstocks (corn stover, switchgrass, Miscanthus, dried distillers' grains plus solubles [DDGS] and poplar) subjected to three alkaline pretreatments (AFEX, dilute base [0.25% NaOH] and alkaline peroxide [AP]). A 16-component mixture comprising the core set plus 10 accessory enzymes was optimized for three pretreatment/substrate combinations. Results were compared to the performance of two commercial enzymes (Accellerase 1000 and Spezyme CP) at the same protein loadings.

Enzyme f&uuml;r Pflanze Zellwand Dekonstruktion sind einen hohen Kostenfaktor bei der Herstellung von Ethanol aus Lignocellulose. Das Ziel dieser Forschung war, optimierte synthetische Mischungen von Enzymen f&uuml;r mehrere Vorbehandlung/Substrat-Kombinationen, die mit unseren Hochdurchsatz-Biomasse-Verdauung-Plattform, GENPLAT, robot, handling, Statistische Versuchsplanung und automatisierte Glc und Xyl Assays Fl&uuml;ssigkeit kombiniert zu entwickeln. Proportionen der sechs Kern Pilz Enzyme (CBH1, CBH2, EG1, &beta;-Glucosidase, ein GH10 Endo-&beta;1 4-Xylanase und &beta;-Xylosidase) wurden optimiert, bei einer festen Enzym Laden von 15 mg/g-Glucan Freilassung der Glc und Xyl von allen Kombinationen von f&uuml;nf Biomasse Rohstoffen (Mais Stover, Switchgrass, Miscanthus, getrocknete Brenner K&ouml;rner plus Mehlen [DDGS] und Pappel) drei Alkali Vorbehandlungen unterworfen (AFEX, verd&uuml;nnten Base [0,25 % NaOH] und Alkali Peroxid [AP]). Eine 16-Komponenten-Mischung, bestehend aus der Core set plus 10 Zubeh&ouml;r Enzyme wurde f&uuml;r drei Vorbehandlung/Substrat-Kombinationen optimiert. Ergebnisse wurden mit der Leistung von zwei kommerzielle Enzyme (Accellerase 1000 und Spezyme CP) an die gleiche Protein-Belastungen verglichen.

Gli enzimi per la decostruzione di parete delle cellule vegetali sono un costo maggiore nella produzione di etanolo da biomassa lignocellulosica. L&#39;obiettivo di questa ricerca era quello di sviluppare miscele sintetiche ottimizzate degli enzimi per molteplici combinazioni di pretrattamento/substrato utilizzando la nostra piattaforma di high-throughput biomassa digestione, GENPLAT, che combina robot liquido gestione statistico del disegno sperimentale e analisi Glc e Xyl automatizzate. Proporzioni di sei core fungine enzimi (CBH1, CBH2, EG1, &beta;-glucosidasi, un GH10 endo-&beta;1, 4-xilanasi e &beta;-xylosidase) sono stati ottimizzati in un carico fisso enzima del glucano di 15 mg/g per il rilascio del Glc e Xyl da tutte le combinazioni di materie cinque biomassa prime (stover mais, switchgrass, Miscanthus, secchi distillatori grani pi&ugrave; solubili [DDGS] e pioppo) sottoposti a pretrattamenti alcalini tre (AFEX, base diluita [0.25% NaOH] e perossido alcalino [AP]). Una miscela di 16-componente comprendente il core set plus 10 enzimi accessori &egrave; stata ottimizzata per le tre combinazioni di pretrattamento/substrato. Enzimas para la deconstrucci&#xF3;n la pared celular vegetal es un costo importante en la producci&#xF3;n de etanol a partir de biomasa lignocelul&#xF3;sica. El objetivo de esta investigaci&#xF3;n fue desarrollar mezclas sint&#xE9;ticas optimizadas para m&#xFA;ltiples enzimas de pretratamiento / sustrato combinaciones con nuestra plataforma de alto rendimiento de la digesti&#xF3;n de biomasa, GENPLAT, que combina el manejo de l&#xED;quidos rob&#xF3;tica, el dise&#xF1;o estad&#xED;stico experimental y Glc automatizado y ensayos XYL. Pretreatment is a critical step in the conversion of lignocellulose to fermentable sugars. Although many pretreatment processes are currently under investigation, none of them are entirely satisfactory in regard to effectiveness, cost, or environmental impact. The use of hydrogen peroxide at pH 11.5 (alkaline hydrogen peroxide (AHP)) was shown by Gould and coworkers to be an effective pretreatment of grass stovers and other plant materials in the context of animal nutrition and ethanol production. Our earlier experiments indicated that AHP performed well when compared against two other alkaline pretreatments. Here, we explored several key parameters to test the potential of AHP for further improvement relevant to lignocellulosic ethanol production.

Î±-Linked xylose is a major component of xyloglucans in the cell walls of higher plants. An Î±-xylosidase (AxlA) was purified from a commercial enzyme preparation from Aspergillus niger, and the encoding gene was identified. The protein is a member of glycosyl hydrolase family 31. It was active on p-nitrophenyl-Î±-d-xyloside, isoprimeverose, xyloglucan heptasaccharide (XXXG), and tamarind xyloglucan. When expressed in Pichia pastoris, AxlA had activity comparable to the native enzyme on pNPÎ±X and IP despite apparent hyperglycosylation. The pH optimum of AxlA was between 3.0 and 4.0. AxlA together with Î²-glucosidase depolymerized xyloglucan heptasaccharide. A combination of AxlA, Î²-glucosidase, xyloglucanase, and Î²-galactosidase in the optimal proportions of 51:5:19:25 or 59:5:11:25 could completely depolymerize tamarind XG to free Glc or Xyl, respectively. To the best of our knowledge, this is the first characterization of a secreted microbial Î±-xylosidase. Secreted Î±-xylosidases appear to be rare in nature, being absent from other tested commercial enzyme mixtures and from the genomes of most filamentous fungi.

Alkaline hydrogen peroxide (AHP) has several attractive features as a pretreatment in the lignocellulosic biomass-to-ethanol pipeline. Here, the feasibility of scaling-up the AHP process and integrating it with enzymatic hydrolysis and fermentation was studied. Corn stover (1â€‰kg) was subjected to AHP pretreatment, hydrolyzed enzymatically, and the resulting sugars fermented to ethanol. The AHP pretreatment was performed at 0.125â€‰g H(2) O(2) /g biomass, 22Â°C, and atmospheric pressure for 48â€‰h with periodic pH readjustment. The enzymatic hydrolysis was performed in the same reactor following pH neutralization of the biomass slurry and without washing. After 48â€‰h, glucose and xylose yields were 75% and 71% of the theoretical maximum. Sterility was maintained during pretreatment and enzymatic hydrolysis without the use of antibiotics. During fermentation using a glucose- and xylose-utilizing strain of Saccharomyces cerevisiae, all of the Glc and 67% of the Xyl were consumed in 120â€‰h. The final ethanol titer was 13.7â€‰g/L. Treatment of the enzymatic hydrolysate with activated carbon prior to fermentation had little effect on Glc fermentation but markedly improved utilization of Xyl, presumably due to the removal of soluble aromatic inhibitors. The results indicate that AHP is readily scalable and can be integrated with enzyme hydrolysis and fermentation. Compared to other leading pretreatments for lignocellulosic biomass, AHP has potential advantages with regard to capital costs, process simplicity, feedstock handling, and compatibility with enzymatic deconstruction and fermentation. Per&#xF3;xido de hidr&#xF3;geno alcalino (AHP) tiene varias caracter&#xED;sticas interesantes como un pretratamiento de biomasa lignocelul&#xF3;sica en el a etanol tuber&#xED;a. En este caso, la viabilidad de la ampliaci&#xF3;n del proceso de AHP y su integraci&#xF3;n con la hidr&#xF3;lisis enzim&#xE1;tica y fermentaci&#xF3;n fue estudiada. El rastrojo de ma&#xED;z (1 kg) se someti&#xF3; a tratamiento previo AHP, hidrolizado enzim&#xE1;ticamente, y los az&#xFA;cares resultantes fermentada en etanol. El pretratamiento AHP se realiz&#xF3; a 0,125 g de H (2) de la biomasa de O (2) / g, 22 &#xB0; C, ya presi&#xF3;n atmosf&#xE9;rica durante 48 h con reajuste peri&#xF3;dico del pH. La hidr&#xF3;lisis enzim&#xE1;tica se realiz&#xF3; en la neutralizaci&#xF3;n del pH mismo reactor siguiente de la suspensi&#xF3;n de biomasa y sin lavar. Despu&#xE9;s de 48 h, el rendimiento de glucosa y xilosa fueron 75% y 71% del m&#xE1;ximo te&#xF3;rico. Esterilidad se mantuvo durante el pretratamiento y la hidr&#xF3;lisis enzim&#xE1;tica sin el uso de antibi&#xF3;ticos. Durante la fermentaci&#xF3;n usando una glucosa-xilosa y-utilizando la cepa de Saccharomyces cerevisiae, todos de la Glc y 67% de la XYL se consume en 120 h. El t&#xED;tulo final de etanol fue de 13,7 g / L. El tratamiento del hidrolizado enzim&#xE1;tico con carb&#xF3;n activado antes de la fermentaci&#xF3;n tuvo poco efecto sobre la fermentaci&#xF3;n Glc pero la utilizaci&#xF3;n marcadamente mejorado de XYL, presumiblemente debido a la eliminaci&#xF3;n de los inhibidores solubles arom&#xE1;ticos.
