Advances in predicting responses to therapies in ovarian cancer have not matched progress seen in other solid-organ tumours: ovarian cancer remains a poor-prognosis disease. There has been a paradigm shift in molecular therapeutics away from targeting individual molecules to whole biological pathways. The aim of this study was to quantitatively measure the activation state of druggable oncogenic pathways by generating a phosphoprotein profile in cancer tissues, in order to establish associations with clinicopathological parameters and to identify treatment groups for targeted therapy. In total we analysed the expression of ten phosphoproteins within eight signalling pathways (PI3K, MAPK, Î²-catenin, STAT, NFÎºB, ER, cell cycle and DNA damage response), proliferation (phospho-histone H3 and Ki67) and apoptosis (activated caspase 3), in two independent cohorts of ovarian cancers using quantitative immunofluorescence image analysis. Data were analysed by unsupervised and K-means clustering to determine new biologically relevant groups. Expression of markers of the five main pathways deregulated by mutation or copy number changes was different between histological subtypes. Four main clusters with distinct phosphoprotein profiles were identified, which were significantly associated with survival in univariate analysis, and which had distinct patterns of pathway expression reproducible between clinical cohorts. These pathway profiles suggest novel therapeutic regimens for the treatment of ovarian cancer, such as MAPK-inhibition in serous or clear cell carcinomas, or combined inhibition of STAT, NFÎºB and WNT signalling.

Phosphoprotein pathway profiling of ovarian carcinoma for the identification of potential new targets for therapy.

Les progr&#xE8;s dans la pr&#xE9;diction des r&#xE9;ponses &#xE0; des th&#xE9;rapies de cancer de l&#x26;#39;ovaire n&#x26;#39;ont pas &#xE9;gal&#xE9; les progr&#xE8;s observ&#xE9;s dans d&#x26;#39;autres organes solides tumeurs: cancer de l&#x26;#39;ovaire reste une maladie de mauvais pronostic. Il ya eu un changement de paradigme dans des th&#xE9;rapies mol&#xE9;culaires loin de cibler des mol&#xE9;cules individuelles &#xE0; l&#x26;#39;ensemble des voies biologiques. L&#x26;#39;objectif de cette &#xE9;tude &#xE9;tait de mesurer quantitativement l&#x26;#39;&#xE9;tat d&#x26;#39;activation des voies oncog&#xE9;niques druggable en g&#xE9;n&#xE9;rant un profil phosphoprot&#xE9;ine dans les tissus canc&#xE9;reux, afin de cr&#xE9;er des associations avec les param&#xE8;tres clinicopathologiques et d&#x26;#39;identifier les groupes de traitement pour la th&#xE9;rapie cibl&#xE9;e. Au total, nous avons analys&#xE9; l&#x26;#39;expression de dix phosphoprot&#xE9;ines dans les huit voies de signalisation (PI3K, MAPK, &#x3B2;-cat&#xE9;nine, STAT, NFkB, ER, le cycle cellulaire et la r&#xE9;ponse dommages &#xE0; l&#x26;#39;ADN), la prolif&#xE9;ration (phospho-histone H3 et Ki67) et de l&#x26;#39;apoptose (caspase activ&#xE9;e 3), dans deux cohortes ind&#xE9;pendantes de cancers de l&#x26;#39;ovaire &#xE0; l&#x26;#39;aide d&#x26;#39;analyse d&#x26;#39;image quantitative immunofluorescence. Les donn&#xE9;es ont &#xE9;t&#xE9; analys&#xE9;es par classification non supervis&#xE9;e et K-means pour d&#xE9;terminer de nouveaux groupes biologiquement pertinentes. Expression de marqueurs des cinq principales voies d&#xE9;r&#xE9;gul&#xE9;s par les changements de num&#xE9;ro de mutation ou d&#x26;#39;une copie &#xE9;tait diff&#xE9;rente entre les sous-types histologiques. Quatre groupes principaux avec des profils distincts ont &#xE9;t&#xE9; identifi&#xE9;s phosphatases, qui ont &#xE9;t&#xE9; significativement associ&#xE9;s &#xE0; la survie en analyse univari&#xE9;e, et qui avaient des mod&#xE8;les distincts d&#x26;#39;expression voie reproductible entre les cohortes cliniques. Fortschritte in der Vorhersage von Antworten zu Therapien bei Eierstockkrebs haben Fortschritte in anderen Solid-Orgel-Tumoren gesehen nicht &uuml;bereinstimmend: Eierstockkrebs bleibt eine Armen-Prognose-Krankheit. Es gab ein Paradigmenwechsel in molecular Therapeutics entfernt gezielt einzelne Molek&uuml;le zu ganzen biologischen Bahnen. Ziel dieser Studie war der Aktivierungsstatus druggable Onkogene Pathways quantitativ zu messen, indem ein Phosphoprotein-Profil im Krebs-Gewebe, generieren, um Zuordnungen mit Clinicopathological Parametern herstellen und Behandlungsgruppen f&uuml;r gezielte Therapie zu identifizieren. Insgesamt analysiert wir den Ausdruck der zehn Phosphoproteine innerhalb acht Signalgebung Wege (PI3K, MAPK, &beta;-Catenin, STAT, NF&kappa;B, ER, Zellzyklus und DNA-Sch&auml;den-Reaktion), Verbreitung (Phospho-Histon H3 und Ki67) und Apoptose (aktiviert Caspase 3), in zwei unabh&auml;ngige Kohorten von Eierstockkrebs mit quantitativen Immunofluoreszenz Bildanalyse. Daten wurden analysiert, durch unkontrollierte und K-Means-Algorithmus um neue biologisch relevante Gruppen zu bestimmen. Ausdruck von Markern der f&uuml;nf wichtigsten Bahnen dereguliert durch Mutation oder Kopie &auml;ndert Unterschied zwischen histologischen Subtypen. Vier wichtigste Clustern mit unterschiedlichen Phosphoprotein-Profile wurden identifiziert, die waren deutlich in Zusammenhang mit dem &Uuml;berleben in der univariaten Analyse, und hatte verschiedene Muster der Stoffwechselweg Ausdruck zwischen klinischer Kohorten reproduzierbar. Diese Profile Weg schlagen neuartige therapeutische Therapien f&uuml;r die Behandlung von Eierstockkrebs, wie MAPK-Hemmung in ser&ouml;se oder klare Zelle Karzinome, oder kombinierten Hemmung der STAT, NF&kappa;B und WNT signalisieren.

Progressi nel predire le risposte alle terapie nel cancro ovarico non hanno corrispondenza progresso visto in altri tumori solidi-organo: carcinoma ovarico rimane una malattia poveri prognosi. C&#39;&egrave; stato un cambiamento di paradigma in molecular therapeutics lontano dal targeting singole molecole a percorsi tutta biologiche. Lo scopo di questo studio era di misurare quantitativamente lo stato di attivazione di percorsi oncogenica druggable generando un profilo fosfoproteina in tessuti tumorali, al fine di stabilire associazioni con parametri Filipek e di identificare i gruppi di trattamento per la terapia mirata. In totale abbiamo analizzato l&#39;espressione di dieci fosfoproteine entro otto segnalazione percorsi (PI3K, MAPK, &beta;-catenina, STAT, NF&kappa;B, ER, ciclo cellulare e risposta di danni del DNA), proliferazione (fosfo-istone H3 e Ki67) e apoptosi (activated caspase 3), in due coorti indipendente dei tumori ovarici mediante analisi immunofluorescenza quantitativa delle immagini. Dati sono stati analizzati da unsupervised e K-means clustering per determinare nuovi gruppi biologicamente rilevanti. Espressione di marcatori dei cinque percorsi principali deregolamentati di mutazione o copia numero modifiche era diverso tra sottotipi istologici. Sono stati identificati quattro cluster principale con profili fosfoproteina distinti, che erano significativamente associati con la sopravvivenza in analisi univariata, e che aveva distinto pattern di espressione di percorso riproducibile tra coorti cliniche. Questi profili pathway suggeriscono nuovi regimi terapeutici per il trattamento del cancro ovarico, come MAPK-inibizione in carcinomi a cellule chiare o sierosa, o inibizione combinata di STAT, NF&kappa;B e WNT segnalazione.

Los avances en la predicci&#xF3;n de las respuestas a las terapias en c&#xE1;ncer de ovario no han alcanzado el progreso observado en otros tumores de &#xF3;rganos s&#xF3;lidos: el c&#xE1;ncer de ovario sigue siendo una enfermedad de mal pron&#xF3;stico. Se ha producido un cambio de paradigma en terapias moleculares lejos de la orientaci&#xF3;n de las mol&#xE9;culas individuales enteros v&#xED;as biol&#xF3;gicas. El objetivo de este estudio fue medir cuantitativamente el estado de activaci&#xF3;n de las v&#xED;as oncog&#xE9;nicas druggable mediante la generaci&#xF3;n de un perfil fosfoprote&#xED;na en el c&#xE1;ncer de los tejidos, a fin de establecer asociaciones con los par&#xE1;metros cl&#xED;nico-patol&#xF3;gicos e identificar los grupos de tratamiento para la terapia dirigida. En total se analiz&#xF3; la expresi&#xF3;n de los diez fosfoprote&#xED;nas dentro de las ocho v&#xED;as de se&#xF1;alizaci&#xF3;n (PI3K, MAPK, &#x3B2;-catenina, STAT, NFkB, ER, ciclo celular y la respuesta al da&#xF1;o del ADN), la proliferaci&#xF3;n (fosfo-histona H3 y Ki67) y la apoptosis (caspasa 3), en dos cohortes independientes de los c&#xE1;nceres de ovario mediante el an&#xE1;lisis cuantitativo de la imagen de inmunofluorescencia. Los datos fueron analizados por el agrupamiento no supervisado y la K significa-para determinar los nuevos grupos biol&#xF3;gicamente relevantes. Expresi&#xF3;n de marcadores de las cinco v&#xED;as principales del mercado liberalizado mediante cambios en el n&#xFA;mero de mutaciones o copia fue diferente entre los subtipos histol&#xF3;gicos. Cuatro grupos principales con diferentes perfiles de fosfoprote&#xED;na fueron identificados, que se asociaron significativamente con la supervivencia en el an&#xE1;lisis univariado, y que ten&#xED;an distintos patrones de expresi&#xF3;n v&#xED;a reproducible entre cohortes cl&#xED;nicas.
