Wir &uuml;berpr&uuml;fen die Einzelkomponenten zu menschlichen embryonalen Stammzellen (ESC) und induzierte pluripotente Stammzellen (iPSC) Kultur und formulieren ein Zellkultursystem, in welche alle Protein Reagenzien f&uuml;r fl&uuml;ssige Medien, Anlage Oberfl&auml;chen und Aufteilung chemisch definiert sind. Eine wesentliche Verbesserung ist das Fehlen einer Serum-Albumin-Komponente, wie Variationen entweder Tier oder Mensch-Quellen Albumin stapelweise zuvor menschlichen ESC und iPSC-Kultur mit Inkonsistenzen geplagt haben. Mit diesem neuen Medium (E8) und spezifischer-beschichteten Oberfl&auml;chen, demonstrieren wir verbesserte Ableitung Wirkungsgrade von Vektor-freien menschlichen iPSCs mit einem episomale Ansatz. Dieses vereinfachte E8-Medium sollte Forschung Verwendung sowohl die klinischen Anwendungen der menschlichen WSR und iPSCs und ihre Derivate erleichtern und auf andere Neuprogrammierung Methoden anwendbar sein sollte.

We re-examine the individual components for human embryonic stem cell (ESC) and induced pluripotent stem cell (iPSC) culture and formulate a cell culture system in which all protein reagents for liquid media, attachment surfaces and splitting are chemically defined. A major improvement is the lack of a serum albumin component, as variations in either animal- or human-sourced albumin batches have previously plagued human ESC and iPSC culture with inconsistencies. Using this new medium (E8) and vitronectin-coated surfaces, we demonstrate improved derivation efficiencies of vector-free human iPSCs with an episomal approach. This simplified E8 medium should facilitate both the research use and clinical applications of human ESCs and iPSCs and their derivatives, and should be applicable to other reprogramming methods.

Combining high-mass-accuracy mass spectrometry, isobaric tagging and software for multiplexed, large-scale protein quantification, we report deep proteomic coverage of four human embryonic stem cell and four induced pluripotent stem cell lines in biological triplicate. This 24-sample comparison resulted in a very large set of identified proteins and phosphorylation sites in pluripotent cells. The statistical analysis afforded by our approach revealed subtle but reproducible differences in protein expression and protein phosphorylation between embryonic stem cells and induced pluripotent cells. Merging these results with RNA-seq analysis data, we found functionally related differences across each tier of regulation. We also introduce the Stem Cell-Omics Repository (SCOR), a resource to collate and display quantitative information across multiple planes of measurement, including mRNA, protein and post-translational modifications.

Cette comparaison de 24 &#xE9;chantillons abouti &#xE0; un ensemble tr&#xE8;s grand nombre de prot&#xE9;ines identifi&#xE9;es et des sites de phosphorylation dans les cellules pluripotentes. L&#x26;#39;analyse statistique offerte par notre approche a r&#xE9;v&#xE9;l&#xE9; des diff&#xE9;rences subtiles mais reproductible &#xE0; l&#x26;#39;expression des prot&#xE9;ines et la phosphorylation des prot&#xE9;ines entre les cellules souches embryonnaires et les cellules pluripotentes induites. Kombination von Masse-hochpr&auml;zise Massenspektrometrie, Isobare Etikettieren und Software f&uuml;r Multiplex, gro&szlig; angelegte Protein Quantifizierung, berichten wir Tiefe Proteomic Abdeckung der vier menschlichen embryonalen Stammzellen und vier induzierten pluripotenten Stammzelllinien in biologischen dreifacher. Dieser 24-Stichproben-Vergleich ergab eine sehr gro&szlig;e Gruppe von identifizierten Proteine und Phosphorylierung Standorte in pluripotente Zellen. Die statistische Analyse von unserem Ansatz geboten offenbart subtil, aber reproduzierbare Unterschiede Proteinexpression und Proteinphosphorylierung zwischen embryonalen Stammzellen und induzierte pluripotente Zellen. Combinando la spettrometria di massa ad alta precisione-massa, tagging isobarico e il software per multiplex, quantificazione di proteine su larga scala, riportiamo proteomic profonda copertura delle quattro cellule staminali embrionali e quattro linee di cellule staminali pluripotenti indotte in triplice copia biologica. Questo confronto 24-campione ha provocato un insieme molto ampio di proteine identificate e siti di fosforilazione nelle cellule pluripotenti. L&#39;analisi statistica fornita dal nostro approccio ha rivelato differenze sottili ma riproducibile nell&#39;espressione della proteina e fosforilazione della proteina tra le cellule staminali embrionali e cellule pluripotenti indotte. La combinaci&#xF3;n de alta precisi&#xF3;n la masa de espectrometr&#xED;a de masas, el etiquetado isob&#xE1;rica y software para la multiplexaci&#xF3;n, la cuantificaci&#xF3;n de prote&#xED;nas a gran escala, se presenta una cobertura profunda prote&#xF3;mico de cuatro c&#xE9;lulas madre embrionarias humanas pluripotentes inducidas y cuatro l&#xED;neas de c&#xE9;lulas madre biol&#xF3;gica, por triplicado. Esta comparaci&#xF3;n de 24 muestras dio como resultado un conjunto muy grande de las prote&#xED;nas identificadas y los sitios de fosforilaci&#xF3;n en las c&#xE9;lulas pluripotentes.
