The mycotoxin deoxynivalenol (DON), a contaminant of certain foods and feeds, is cytotoxic and genotoxic to mammalians cells. Exposure of human embryonic kidney (Hek-293) cells to DON led to a dose- and time-dependent decrease in cell viability, with an IC(50) about 7.6 Î¼M. The DON effects on Hek-293 morphology, reactive oxygen species, lipid peroxidation and antioxidative system and caspase 3 and bcl-2 expression were studied. Cells became round and in some are progressive loss of cell attachment appeared. These biochemical parameters were assessed after 6, 12 and 24 h of treatment with 2.5 and 5 Î¼M DON. An increase in superoxide dismutase activity within the interval 6-12 h and almost complete recovery by the end of experiment for both concentrations was observed, whereas the profile of catalase activity was the same with the superoxide dismutase one for 2.5 Î¼M and decreased in a time-dependent manner for 5 Î¼M. A temporary activation of glutathione peroxidase and glutathione reductase was recorded at 12 h post-exposure, while the glutathione-S-transferase activity was unchanged for both concentrations. The NADP(+)-dependent isocitrate dehydrogenase activity showed a transient increase at the 12 h post-exposure. The caspase 3 expression remained unchanged and the bcl-2 one decreased after 24 h of exposure for the two concentrations. Our results showed the dose- and time specific changes in the antioxidants system of Hek-293 cells, which could not counteract efficiently the effects DON exposure. The different types of cell death which could be activated by this DON induced changes are mentioned.

Die Mykotoxin-Deoxynivalenol (DON), Verunreinigung bestimmter Nahrungs- und f&uuml;ttert, ist zytotoxisch und genotoxische S&auml;ugetiere Zellen. Menschlichen embryonalen Nierenzellen (Hek-293) Don f&uuml;hrte dazu eine Dosis und zeitabh&auml;ngige Abnahme der Zellviabilit&auml;t, mit einem IC (50) &uuml;ber 7,6 &mu;M. Die DON-Auswirkungen auf die Morphologie, reaktive Sauerstoffspezies der Hek-293, Lipidperoxidation und antioxidativen System und Caspase 3 und Bcl-2 Ausdruck wurden untersucht. Der Ausdruck Caspase 3 blieb unver&auml;ndert und der Bcl-2 eine verringerte sich nach 24 h der Exposition f&uuml;r die beiden Konzentrationen. Unsere Ergebnisse zeigten die Dosis und Zeit bestimmten &Auml;nderungen in der Antioxidantien-System von Hek-293-Zellen, die die Auswirkungen nicht effizient entgegenwirken k&ouml;nnte DON Exposition. Die verschiedenen Arten von Zelltod, die von dieser DON aktiviert werden k&ouml;nnte induzierten Ver&auml;nderungen sind erw&auml;hnt.

La micotossina deossinivalenolo (DON), un contaminante di determinati cibi e mangimi, &egrave; citotossico e genotossico in cellule di mammiferi. L&#39;esposizione delle cellule renali embrionali umane (Hek-293) di DON ha condotto ad una diminuzione dose - e tempo-dipendente della vitalit&agrave; cellulare, con un IC(50) circa 7,6 &mu;M. Sono stati studiati gli effetti di DON sulla morfologia Hek-293, specie reattive dell&#39;ossigeno, perossidazione lipidica e antiossidanti, sistema e caspase 3 e bcl-2 expression. Cellule divennero rotonde e in alcuni sono comparso progressiva perdita di attacco della cellula. Questi parametri biochimici sono stati valutati dopo 6, 12 e 24 ore di trattamento con 2,5 e 5 &mu;M DON. La micotoxinas deoxinivalenol (DON), un contaminante de ciertos alimentos y piensos, es citot&oacute;xico y genot&oacute;xico en c&eacute;lulas de mam&iacute;feros. Exposici&oacute;n de las c&eacute;lulas de ri&ntilde;&oacute;n embrionario humano (Hek-293) a DON condujo a una disminuci&oacute;n de dependientes de la dosis y tiempo en la viabilidad celular, con una IC(50) sobre 7.6 &mu;M. Se estudiaron los efectos de DON Hek-293 morfolog&iacute;a, especies reactivas de ox&iacute;geno, peroxidaci&oacute;n de l&iacute;pidos y antioxidante expresi&oacute;n sistema y caspasa 3 y bcl-2. Las c&eacute;lulas se convirti&oacute; en redondas y en algunos son p&eacute;rdida progresiva del accesorio celular apareci&oacute;. Estos par&aacute;metros bioqu&iacute;micos fueron evaluados despu&eacute;s de 6, 12 y 24 h de tratamiento con 2.5 y 5 &mu;M DON. Se observ&oacute; un aumento en la actividad de super&oacute;xido dismutasa dentro del intervalo de 6-12 h y recuperaci&oacute;n casi completa al final del experimento para ambas concentraciones, mientras que el perfil de actividad de la catalasa era lo mismo con la super&oacute;xido dismutasa uno de 2,5 &mu;M y disminuy&oacute; de manera dependiente del tiempo para 5 &mu;M. Una activaci&oacute;n temporal de la glutati&oacute;n peroxidasa y glutati&oacute;n reductasa se registr&oacute; en postexposici&oacute;n de 12 h, mientras que la actividad de la glutati&oacute;n-S-transferasa era igual para ambas concentraciones. El NADP (+)-actividad de la deshidrogenasa dependiente de isocitrato demostr&oacute; un aumento transitorio en la postexposici&oacute;n de 12 h. La expresi&oacute;n de caspasa 3 se mantuvo sin cambios y el bcl-2 uno disminuido despu&eacute;s de 24 h de exposici&oacute;n para las dos concentraciones. Nuestros resultados mostraron los cambios espec&iacute;ficos de dosis y tiempo en el sistema de antioxidantes de las c&eacute;lulas Hek-293, que no pudo contrarrestar eficazmente los efectos DON exposici&oacute;n. Los diferentes tipos de muerte celular que podr&iacute;a activarse por este DON cambios inducidos se mencionan.
