L&#39;obiettivo di questo studio era quello di sviluppare un sistema per generare rapidamente costrutti tessuto ingegnerizzato da aggregati di cellule e cellule derivate di matrice extracellulare (ECM) per consentire la valutazione delle cellule derivate tessuto struttura e funzione. Cellule del muscolo liscio aortiche ratto teste di serie dell&#39;agarosi anulare pozzi (2, 4 o 6 mm diametro interno) aggregati e formano anelli di tessuto spesso entro 2 settimane di coltura statica (0,76 mm a 8 giorni; 0,94 mm a 14 giorni). The goal of this study was to develop a system to rapidly generate engineered tissue constructs from aggregated cells and cell-derived extracellular matrix (ECM) to enable evaluation of cell-derived tissue structure and function. Rat aortic smooth muscle cells seeded into annular agarose wells (2, 4 or 6 mm inside diameter) aggregated and formed thick tissue rings within 2 weeks of static culture (0.76 mm at 8 days; 0.94 mm at 14 days). Overall, cells appeared healthy and surrounded by ECM comprised of glycosoaminoglycans and collagen, although signs of necrosis were observed near the centers of the thickest rings. Tissue ring strength and stiffness values were superior to those reported for engineered tissue constructs cultured for comparable times. The strength (100-500 kPa) and modulus (0.5-2 MPa) of tissue rings increased with ring size and decreased with culture duration. Finally, tissue rings cultured for 7 days on silicone mandrels fused to form tubular constructs. Ring margins were visible after 7 days, but tubes were cohesive and mechanically stable, and histological examination confirmed fusion between ring subunits. This unique system provides a versatile new tool for optimization and functional assessment of cell-derived tissue, and a new approach to creating tissue-engineered vascular grafts.

Das Ziel dieser Studie war es, die Entwicklung eines Systems zum schnell generieren ver&auml;nderte Gewebe Konstrukte aus aggregierten Zellen und Zellen abgeleiteten extrazellul&auml;ren Matrix (ECM) Bewertung der Cell-derived Gewebestruktur und Funktion zu aktivieren. Die St&auml;rke (100-500 kPa) und Modulo (0.5-2 MPa) des Gewebes Ringe mit Ringgr&ouml;&szlig;e erh&ouml;ht und sank mit Kultur-Dauer. Schlie&szlig;lich verschmolzen Gewebe Ringe kultiviert f&uuml;r 7 Tage auf Silikon Dorne auf tubul&auml;re Form-Konstrukte. Ring R&auml;nder sichtbar waren, nach 7 Tagen, aber Schl&auml;uche wurden Zusammenhalt und mechanisch stabil und histologische Untersuchung best&auml;tigt Fusion zwischen Ring-Untereinheiten. Dieses einzigartige System bietet ein vielseitiges neues Werkzeug f&uuml;r die Optimierung und funktionale Bewertung der Cell-derived Gewebe und einen neuen Ansatz zum Erstellen von Tissue-engineered vaskul&auml;rer Prothesen.

El objetivo de este estudio fue desarrollar un sistema para generar r&#xE1;pidamente las construcciones de ingenier&#xED;a de tejidos de c&#xE9;lulas agregados y derivado de las c&#xE9;lulas de la matriz extracelular (MEC) para permitir la evaluaci&#xF3;n de la estructura del tejido derivado de c&#xE9;lulas y la funci&#xF3;n. Las c&#xE9;lulas del m&#xFA;sculo liso de aorta de rata sembradas en anular los pozos de agarosa (2, 4 o 6 mm de di&#xE1;metro interior) agregan y forman anillos gruesos de tejido dentro de 2 semanas de la cultura est&#xE1;tica (0,76 mm en 8 d&#xED;as, 0,94 mm en 14 d&#xED;as). En general, las c&#xE9;lulas apareci&#xF3; sano y rodeado por ECM compuesto de glycosoaminoglycans y el col&#xE1;geno, aunque los signos de necrosis se observaron cerca de los centros de los anillos m&#xE1;s gruesos. La fuerza del tejido del anillo y valores de rigidez fueron superiores a los reportados para construcciones de ingenier&#xED;a tisular se cultivaron durante tiempos similares. La fuerza (100-500 kPa) y el m&#xF3;dulo (0.5-2 MPa) de los anillos de tejido aumenta con el tama&#xF1;o del anillo y disminuy&#xF3; con la duraci&#xF3;n cultivo. Finalmente, los anillos de tejidos cultivados durante 7 d&#xED;as sobre mandriles de silicona fundida para formar construcciones tubulares. M&#xE1;rgenes del anillo eran visibles despu&#xE9;s de 7 d&#xED;as, pero los tubos eran cohesionada y estable mec&#xE1;nicamente, y el examen histol&#xF3;gico confirm&#xF3; la fusi&#xF3;n entre las subunidades del anillo.
