The mammalian vomeronasal organ detects complex chemical signals that convey information about gender, strain, and the social and reproductive status of an individual. How these signals are encoded is poorly understood. We developed transgenic mice expressing the calcium indicator G-CaMP2 and analyzed population responses of vomeronasal neurons to urine from individual animals. A substantial portion of cells was activated by either male or female urine, but only a small population of cells responded exclusively to gender-specific cues shared across strains and individuals. Female cues activated more cells and were subject to more complex hormonal regulations than male cues. In contrast to gender, strain and individual information was encoded by the combinatorial activation of neurons such that urine from different individuals activated distinctive cell populations.

L&#x26;#39;organe vom&#xE9;ronasal des mammif&#xE8;res d&#xE9;tecte les signaux chimiques complexes qui transmettent des informations sur le sexe, la souche, et le statut social et de la reproduction d&#x26;#39;un individu. Comment ces signaux sont cod&#xE9;s est mal comprise. Nous avons d&#xE9;velopp&#xE9; des souris transg&#xE9;niques exprimant l&#x26;#39;indicateur de calcium G-camp2 et analys&#xE9; les r&#xE9;ponses de la population de neurones Vomeronasal &#xE0; l&#x26;#39;urine d&#x26;#39;animaux individuels. Une partie importante de cellules a &#xE9;t&#xE9; activ&#xE9;e par l&#x26;#39;urine soit m&#xE2;le ou femelle, mais seulement une petite population de cellules r&#xE9;pondu exclusivement &#xE0; sexosp&#xE9;cifiques indices partag&#xE9;s entre les souches et les individus. Indices Femme activ&#xE9; plus de cellules et ont &#xE9;t&#xE9; soumis &#xE0; plus de complexes r&#xE9;gulations hormonales que les indices de sexe masculin. En revanche le sexe, la souche et l&#x26;#39;information individuelle a &#xE9;t&#xE9; cod&#xE9;e par l&#x26;#39;activation des neurones combinatoire tels que l&#x26;#39;urine provenant de diff&#xE9;rents individus des populations de cellules activ&#xE9;es distinctifs.

Das S&auml;ugetier-Vomeronasal Organ erkennt komplexe chemische Signale, die Informationen &uuml;ber Geschlecht, Stamm und den sozialen und reproduktiven Status eines Individuums zu vermitteln. Oft nicht ganz ist verstanden, wie diese Signale codiert werden. Wir entwickelten transgene M&auml;usen zum Ausdruck bringen des Kalzium-Kennzeichen G-CaMP2 und analysiert Bev&ouml;lkerung Antworten der Vomeronasal Neuronen zu Urin aus einzelnen Tieren. Ein erheblicher Anteil der Zellen durch entweder m&auml;nnlichen oder weiblichen Urin aktiviert wurde, aber nur eine kleine Population von Zellen reagiert ausschlie&szlig;lich auf Gender-spezifische Hinweise f&uuml;r Belastungen und Einzelpersonen freigegeben. Weibliche Signale aktiviert mehr Zellen und komplexere hormonelle Vorschriften als m&auml;nnlichen Signale unterstanden. Im Gegensatz zu Geschlecht Stamm und individuelle Informationen war codiert durch die kombinatorische Aktivierung von Neuronen, dass Urin aus verschiedenen Individuen charakteristischen Zelle Bev&ouml;lkerungen aktiviert.

L&#39;organo vomeronasale mammiferi rileva segnali complessi chimici che trasmettono informazioni sul genere, ceppo e lo status sociale e riproduttivo di un individuo. Come questi segnali sono codificati &egrave; scarsamente compreso. Abbiamo sviluppato i topi transgenici che esprimono l&#39;indicatore di calcio G-CaMP2 e analizzato le risposte dei neuroni vomeronasal popolazione di urina da singoli animali. Una parte sostanziale delle cellule &egrave; stata attivata da urina o maschio o femmina, ma solo una piccola popolazione di cellule ha risposto esclusivamente al genere specifici spunti condivisi in ceppi e gli individui. Stecche femminile attivato pi&ugrave; celle ed erano conforme alle regolazioni ormonali pi&ugrave; complessi rispetto a stecche maschi. El &#xF3;rgano vomeronasal detecta se&#xF1;ales de mam&#xED;feros complejos qu&#xED;micos que transmiten informaci&#xF3;n sobre el g&#xE9;nero, raza y el estatus social y reproductiva de un individuo. &#xBF;C&#xF3;mo estas se&#xF1;ales son codificadas es poco conocida. Hemos desarrollado ratones transg&#xE9;nicos que expresan el indicador de calcio G-camp2 y se analizaron las respuestas de la poblaci&#xF3;n de neuronas vomeronasal a la orina de los animales individuales. Una parte sustancial de las c&#xE9;lulas se activa por la orina ya sea hombre o mujer, pero s&#xF3;lo una peque&#xF1;a poblaci&#xF3;n de c&#xE9;lulas respond&#xED;an exclusivamente a las se&#xF1;ales espec&#xED;ficas de g&#xE9;nero en todas las cepas comunes y particulares. Se&#xF1;ales femeninas activa m&#xE1;s c&#xE9;lulas y estaban sujetos a regulaciones m&#xE1;s complejas que las se&#xF1;ales hormonales masculinos. En contraste con el g&#xE9;nero, la tensi&#xF3;n y la informaci&#xF3;n individual fue codificado por la activaci&#xF3;n de las neuronas combinatoria de tal manera que la orina de individuos diferentes poblaciones distintivas de c&#xE9;lulas activadas.

In mammalian species, detection of pheromone cues by the vomeronasal organ (VNO) at different concentrations can elicit distinct behavioral responses and endocrine changes. It is not well understood how concentration-dependent activation of the VNO impacts innate behaviors. In this study, we find that, when mice investigate the urogenital areas of a conspecific animal, the urinary pheromones can reach the VNO at a concentration of approximately 1% of that in urine. At this level, urinary pheromones elicit responses from a subset of cells that are tuned to sex-specific cues and provide unambiguous identification of the sex and strain of animals. In contrast, low concentrations of urine do not activate these cells. Strikingly, we find a population of neurons that is only activated by low concentrations of urine. The properties of these neurons are not found in neurons responding to putative single-compound pheromones. Additional analyses show that these neurons are masked by high-concentration pheromones. Thus, an antagonistic interaction in natural pheromones results in the activation of distinct populations of cells at different concentrations. The differential activation is likely to trigger different downstream circuitry and underlies the concentration-dependent pheromone perception.

Zus&auml;tzliche Analysen zeigen, dass diese Neuronen durch hoch-Konzentration-Pheromone maskiert werden. Daher f&uuml;hrt eine antagonistische Aktivit&auml;t in nat&uuml;rlichen Pheromone die Aktivierung der unterschiedlichen Populationen von Zellen in verschiedenen Konzentrationen. Die differentielle Aktivierung wahrscheinlich andere nachgeschaltete Schaltung ausl&ouml;sen und zugrunde liegt die Konzentration abh&auml;ngige Pheromon-Wahrnehmung.

In terrestrial vertebrates, the vomeronasal organ (VNO) detects and transduces pheromone signals. VNO activation is thought to be mediated by the transient receptor potential C2 (TRPC2) channel. The aberrant behavioural phenotypes observed in TRPC2-/- mice are generally attributed to the lost VNO function. Recently, calcium-activated chloride channels have been shown to contribute to VNO activation. Here we show that CACCs can be activated in VNO slice preparations from the TRPC2-/- mice and this activation is blocked by pharmacological agents that inhibit intracellular Ca(2+) release. Urine-evoked Cl(-) current is sufficient to drive spiking changes in VNO neurons from both wild-type (WT) and TRPC2-/- mice. Moreover, blocking Cl(-) conductance essentially abolishes VNO activation in WT neurons. These results suggest a TRPC2-independent signalling pathway in the VNO and the requirement of calcium-activated chloride channels currents to mediate pheromone activation. Our data further suggest that TRPC2-/- mice retain partial VNO function.

En los vertebrados terrestres, el &#xF3;rgano vomeronasal (VNO) detecta y transduce se&#xF1;ales de las feromonas. La activaci&#xF3;n de VNO se cree que est&#xE1; mediada por el receptor de potencial transitorio C2 (trpC2) del canal. Los fenotipos aberrantes de comportamiento observados en trpC2-/ - ratones se atribuyen generalmente a la funci&#xF3;n perdida VNO. Recientemente, activados por calcio-canales de cloro se ha demostrado que contribuyen a la activaci&#xF3;n del VNO. Aqu&#xED; nos muestran que CACCs se puede activar en los preparativos trozo del OVN de los trpC2-/ - ratones y esta activaci&#xF3;n est&#xE1; bloqueada por los agentes farmacol&#xF3;gicos que inhiben la acumulaci&#xF3;n intracelular de Ca (2 +) de liberaci&#xF3;n. Orina-evoc&#xF3; Cl (-) actual es suficiente para impulsar los cambios enriquecidas en neuronas del OVN de ambos de tipo salvaje (WT) y trpC2-/ - ratones. Por otra parte, el bloqueo de Cl (-) de la conductancia en esencia suprime la activaci&#xF3;n de VNO en las neuronas WT. Estos resultados sugieren una v&#xED;a de se&#xF1;alizaci&#xF3;n trpC2-independiente en el OVN y el requisito de calcio activados-canales de cloruro de corrientes para mediar la activaci&#xF3;n feromona. Nuestros datos sugieren que trpC2-/ - ratones conservan parcial de la funci&#xF3;n de VNO.
