We have recently reported that two Rho family GTPases, Rac1 and Cdc42, are intimately involved in the control of cell survival of murine fibroblasts linked to adherence to the extracellular matrix. Inhibition of either Rac1 or Cdc42 signaling in adherent cells mimics the loss of anchorage and efficiently induces apoptosis in both immortalized and primary cells. In both cases cell death is dependent on the wild-type p53 tumor suppressor and is accompanied by activation of endogenous p53. Here, we describe that the inhibition of Rac1 or Cdc42 signaling leads to MAPK ERK activation via a pathway involving PI(3)K, Akt, Raf, and MEK, but not Ras. The moderate level of ERK activation that accompanies anoikis is an essential component of proapoptotic signaling; whereas sustained, high-intensity ERK signaling promotes survival in the same experimental system.

Activation of ERK, controlled by Rac1 and Cdc42 via Akt, is required for anoikis.

The Hedgehog (Hh) signaling pathway promotes pattern formation and cell proliferation in Drosophila and vertebrates. Hh is a ligand that binds and represses the Patched (Ptc) receptor and thereby releases the latent activity of the multipass membrane protein Smoothened (Smo), which is essential for transducing the Hh signal. In Caenorhabditis elegans, the Hh signaling pathway has undergone considerable divergence. Surprisingly, obvious Smo and Hh homologs are absent whereas PTC, PTC-related (PTR), and a large family of nematode Hh-related (Hh-r) proteins are present. We find that the number of PTC-related and Hh-r proteins has expanded in C. elegans, and that this expansion occurred early in Nematoda. Moreover, the function of these proteins appears to be conserved in Caenorhabditis briggsae. Given our present understanding of the Hh signaling pathway, the absence of Hh and Smo raises many questions about the evolution and the function of the PTC, PTR, and Hh-r proteins in C. elegans. To gain insights into their roles, we performed a global survey of the phenotypes produced by RNA-mediated interference (RNAi). Our study reveals that these genes do not require Smo for activity and that they function in multiple aspects of C. elegans development, including molting, cytokinesis, growth, and pattern formation. Moreover, a subset of the PTC, PTR, and Hh-r proteins have the same RNAi phenotypes, indicating that they have the potential to participate in the same processes.

The Hedgehog (Hh) v&#xED;a de se&#xF1;alizaci&#xF3;n promueve la formaci&#xF3;n de patrones y la proliferaci&#xF3;n celular en Drosophila y vertebrados. Hh es un ligando que se une y reprime el parcheado (PTC) del receptor y por lo tanto libera la actividad latente de la prote&#xED;na de la membrana multipaso Smoothened (SMO), que es esencial para transducir la se&#xF1;al Hh. En Caenorhabditis elegans, la v&#xED;a de se&#xF1;alizaci&#xF3;n Hh ha experimentado una considerable divergencia. Ces derni&#xE8;res ann&#xE9;es, des &#xE9;tudes avec des organismes mod&#xE8;les invert&#xE9;br&#xE9;s ont donn&#xE9; un aper&#xE7;u de l&#x26;#39;origine, la fonction et l&#x26;#39;&#xE9;volution de l&#x26;#39;immunit&#xE9; inn&#xE9;e. Ici, nous utilisons l&#x26;#39;&#xE9;chelle du g&#xE9;nome analyse du transcriptome pour caract&#xE9;riser la cons&#xE9;quence de l&#x26;#39;infection fongique naturel dans Caenorhabditis elegans. Encounters with pathogens provoke changes in gene transcription that are an integral part of host innate immune responses. In recent years, studies with invertebrate model organisms have given insights into the origin, function, and evolution of innate immunity. Here, we use genome-wide transcriptome analysis to characterize the consequence of natural fungal infection in Caenorhabditis elegans. We identify several families of genes encoding putative antimicrobial peptides (AMPs) and proteins that are transcriptionally up-regulated upon infection. Many are located in small genomic clusters. We focus on the nlp-29 cluster of six AMP genes and show that it enhances pathogen resistance in vivo. The same cluster has a different structure in two other Caenorhabditis species. A phylogenetic analysis indicates that the evolutionary diversification of this cluster, especially in cases of intra-genomic gene duplications, is driven by natural selection. We further show that upon osmotic stress, two genes of the nlp-29 cluster are strongly induced. In contrast to fungus-induced nlp expression, this response is independent of the p38 MAP kinase cascade. At the same time, both involve the epidermal GATA factor ELT-3. Our results suggest that selective pressure from pathogens influences intra-genomic diversification of AMPs and reveal an unexpected complexity in AMP regulation as part of the invertebrate innate immune response.

Anti-fungal innate immunity in C. elegans is enhanced by evolutionary diversification of antimicrobial peptides.

In den letzten Jahren haben Studien mit Wirbellosen Modellorganismen Einblicke in die Herkunft, Funktion und Entwicklung der angeborenen Immunit&auml;t gegeben. Demselben Cluster hat eine andere Struktur in zwei weitere Caenorhabditis-Arten. Eine phylogenetische Analyse zeigt, dass die evolution&auml;re Diversifizierung dieses Clusters, insbesondere in F&auml;llen von Intra-genomische gen Duplikationen, durch nat&uuml;rliche Selektion getrieben wird. Wir zeigen weiter, dass bei der osmotische Druck, zwei Gene des Clusters Nlp-29 stark umweltbedingt sind. Im Gegensatz zu Pilz-induzierte Nlp Ausdruck ist diese Antwort unabh&auml;ngig von der p38-MAP-Kinase-Kaskade. Zur gleichen Zeit betreffen sowohl den epidermalen GATA-Faktor ELT-3. Aqu&#xED;, nosotros usamos todo el genoma an&#xE1;lisis de transcriptoma para caracterizar la consecuencia de la infecci&#xF3;n f&#xFA;ngica natural en Caenorhabditis elegans. Identificamos varias familias de genes que codifican p&#xE9;ptidos antimicrobianos putativos (AMP) y las prote&#xED;nas que est&#xE1;n transcripcionalmente regulados hasta despu&#xE9;s de la infecci&#xF3;n. Muchos est&#xE1;n situados en peque&#xF1;os racimos gen&#xF3;micas. Nos centramos en el PNL-29 grupo de seis genes AMP y demostrar que mejora la resistencia a los pat&#xF3;genos in vivo. El mismo grupo tiene una estructura diferente en dos especies Caenorhabditis otros. After being infected by the fungus Drechmeria coniospora, Caenorhabditis elegans produces antimicrobial peptides in its epidermis, some regulated by a signaling cascade involving a p38 mitogen-activated protein kinase. Here we show that infection-induced expression of peptides of the Caenacin family occurred independently of the p38 pathway. The caenacin (cnc) genes enhanced survival after fungal infection, and neuronal expression of the transforming growth factor-beta homolog DBL-1 promoted cnc-2 expression in the epidermis in a dose-dependent paracrine way. Our results lead to a model in which antifungal defenses are coordinately regulated by a cell-autonomous p38 cascade and a distinct cytokine-like transforming growth factor-beta signal from the nervous system, each of which controls distinct sets of antimicrobial peptide-encoding genes in the epidermis.

For almost four decades, the nematode Caenorhabditis elegans has been of great value in many fields of biological research. It is now used extensively in studies of microbial pathogenesis and innate immunity. The worm lacks an adaptive immune system and relies solely on its innate immune defences to cope with pathogen attack. Infectious microbes, many of which are of clinical interest, trigger specific mechanisms of innate immunity, and provoke the expression of antifungal or antibacterial polypeptides. In this review, we highlight some of these families of antimicrobial peptides (AMPs) and proteins that are candidates for the development of novel antibiotics. In addition, we describe how systems of C. elegans infection provide an increasing number of possibilities for large-scale in vivo screens for the discovery of new antimicrobial drugs. These systems open promising perspectives for innovative human therapies.

C. elegans: model host and tool for antimicrobial drug discovery.

Durante casi cuatro d&#xE9;cadas, el nematodo Caenorhabditis elegans ha sido de gran valor en muchos campos de la investigaci&#xF3;n biol&#xF3;gica. En la actualidad se utiliza ampliamente en estudios de patog&#xE9;nesis microbiana y la inmunidad innata. El gusano no tiene un sistema inmune adaptativo, y se basa &#xFA;nicamente en sus defensas inmunitarias innatas para hacer frente al ataque de pat&#xF3;genos. Los microbios infecciosos, muchos de los cuales son de inter&#xE9;s cl&#xED;nico, poner en marcha mecanismos espec&#xED;ficos de la inmunidad innata, y provocar la expresi&#xF3;n de polip&#xE9;ptidos antif&#xFA;ngicos o antibacterianos. En esta revisi&#xF3;n, se destacan algunas de estas familias de p&#xE9;ptidos antimicrobianos (AMP) y las prote&#xED;nas que son candidatos para el desarrollo de nuevos antibi&#xF3;ticos. Adem&#xE1;s, se describe c&#xF3;mo los sistemas de la infecci&#xF3;n por C. elegans proporcionar un n&#xFA;mero cada vez mayor de posibilidades de gran escala en las pantallas in vivo para el descubrimiento de nuevos f&#xE1;rmacos antimicrobianos. Estos sistemas abren perspectivas prometedoras para las terapias humanas.
