Native disulfide bond formation in eukaryotes is dependent on protein-disulfide isomerase (PDI) and its homologs, which contain varying combinations of catalytically active and inactive thioredoxin domains. However, the specific contribution of PDI to the formation of new disulfides versus reduction/rearrangement of non-native disulfides is poorly understood. We analyzed the role of individual PDI domains in disulfide bond formation in a reaction driven by their natural oxidant, Ero1p. We found that Ero1p oxidizes the isolated PDI catalytic thioredoxin domains, A and A' at the same rate. In contrast, we found that in the context of full-length PDI, there is an asymmetry in the rate of oxidation of the two active sites. This asymmetry is the result of a dual effect: an enhanced rate of oxidation of the second catalytic (A') domain and the substrate-mediated inhibition of oxidation of the first catalytic (A) domain. The specific order of thioredoxin domains in PDI is important in establishing the asymmetry in the rate of oxidation of the two active sites thus allowing A and A', two thioredoxin domains that are similar in sequence and structure, to serve opposing functional roles as a disulfide isomerase and disulfide oxidase, respectively. These findings reveal how native disulfide folding is accomplished in the endoplasmic reticulum and provide a context for understanding the proliferation of PDI homologs with combinatorial arrangements of thioredoxin domains.

Formation de la liaison disulfure natif chez les eucaryotes est tributaire de l&#39;isom&eacute;rase (PDI) de la prot&eacute;ine disulfure et ses homologues, qui contiennent des combinaisons vari&eacute;es de domaines thior&eacute;doxine catalytiquement actives et inactives. Cependant, la contribution sp&eacute;cifique du PDI &agrave; la formation de nouveaux disulfures versus r&eacute;duction/r&eacute;arrangement des allochtones disulfures est mal comprise. Nous avons analys&eacute; le r&ocirc;le des domaines individuels de PDI dans la formation de la liaison disulfure dans une r&eacute;action conduite par leur oxydant naturel, Ero1p. Nous avons trouv&eacute; que la Ero1p s&#39;oxyde les domaines isol&eacute;s de thioredoxin catalytique en PDI, A et A&#39; au m&ecirc;me rythme. En revanche, nous avons constat&eacute; que dans le cadre du PDI pleine longueur, il y a une asym&eacute;trie dans la vitesse d&#39;oxydation de deux sites actifs. Cette asym&eacute;trie est le r&eacute;sultat d&#39;un double effet : un taux major&eacute; de l&#39;oxydation de la deuxi&egrave;me catalytique (A&#39;) domaine et l&#39;inhibition induite par le substrat de l&#39;oxydation du premier catalytique (A) du domaine. L&#39;ordre sp&eacute;cifique des domaines thior&eacute;doxine dans PDI est important dans l&#39;&eacute;tablissement de l&#39;asym&eacute;trie dans le taux d&#39;oxydation des deux sites actifs permettant ainsi A et A&#39;, deux domaines thior&eacute;doxine qui sont semblables dans la s&eacute;quence et de la structure, pour servir de s&#39;opposer &agrave; des r&ocirc;les fonctionnels comme une isom&eacute;rase du bisulfure et oxydase disulfure, respectivement. Ces r&eacute;sultats r&eacute;v&egrave;lent comment native disulfure pliage s&#39;effectue dans le r&eacute;ticulum endoplasmique et de fournir un contexte pour comprendre la prolif&eacute;ration des homologues chez les PDI avec des arrangements combinatoires de domaines de thioredoxin.

Native Disulfid-Bond-Formation in Eukaryoten ist abh&auml;ngig von Protein-Disulfid-Isomerase (PDI) und seine UCP1, die verschiedene Kombinationen von katalytisch aktiven und inaktiven Thioredoxin-Dom&auml;nen enthalten. Allerdings ist der spezifische Beitrag der PDI zur Bildung des neuen Disulfiden gegen Reduktion/Umlagerung von Drittprogrammen Disulfiden schlecht verstanden. Wir analysierten die Rolle der einzelnen PDI-Dom&auml;nen in Disulfid-Bond-Formation in einer Reaktion, die angetrieben durch ihre nat&uuml;rliche Oxidationsmittel, Ero1p. Wir fanden, dass Ero1p die isolierten PDI katalytische Thioredoxin Dom&auml;nen, A und A oxidiert &#39; mit der gleichen Geschwindigkeit. Im Gegensatz dazu fanden wir, dass im Rahmen der in PDI, gibt es eine Asymmetrie in der Oxidation von den beiden Standorten aktiv. Diese Asymmetrie ist das Ergebnis eine doppelte Wirkung: eine erh&ouml;hte Rate von der zweiten katalytischen Oxidation (A&#39;) Dom&auml;ne und die Substrat-vermittelte Hemmung der Oxidation von der ersten katalytischen (A) Dom&auml;ne. Die spezifische Reihenfolge der Thioredoxin Domains in PDI ist wichtig bei der Festlegung der Asymmetrie in der Rate der Oxidation von den zwei aktiven Websites, so dass A und A&#39;, zwei Thioredoxin-Dom&auml;nen, die in Reihenfolge und Struktur, dienen der gegnerischen Funktionsrollen als Disulfid-Isomerase und Disulfid-Oxydase, bzw. &auml;hneln. Diese Ergebnisse zeigen, wie systemeigene Disulfid-Faltung erfolgt in dem Endoplasmatischen Retikulum und bieten einen Kontext f&uuml;r das Verst&auml;ndnis der Verbreitung von PDI UCP1 mit kombinatorischer Arrangements von Thioredoxin-Domains.

Formazione di legami disolfuro nativo negli eucarioti &egrave; dipendente isomerasi (PDI) di proteina-disolfuro e suoi omologhi, che contengono diverse combinazioni di domini tioredossina cataliticamente attivi e inattivi. Tuttavia, il contributo specifico di PDI per la formazione di nuovi disulfides versus riduzione/riarrangiamento di non-nativi disulfides &egrave; capito male. Abbiamo analizzato il ruolo dei singoli domini PDI nella formazione di legami disolfuro in una reazione guidata da loro ossidante naturale, Ero1p. Abbiamo trovato che Ero1p si ossida isolati domini catalitici tioredossina PDI, A e A&#39; allo stesso tasso. Al contrario, abbiamo trovato che nel contesto di PDI full-length, c&#39;&egrave; un&#39;asimmetria del tasso di ossidazione dei due siti attivi. Questa asimmetria &egrave; il risultato di un duplice effetto: un tasso maggiore di ossidazione del secondo catalitica (A&#39;) dominio e l&#39;inibizione mediata dal substrato di ossidazione catalitica primo (A) dominio. L&#39;ordine specifico della tioredossina domini in PDI &egrave; importante stabilire l&#39;asimmetria del tasso di ossidazione dei due siti attivi permettendo cos&igrave; A e A&#39;, due domini tioredossina che sono simili in sequenza e struttura, per servire opposti ruoli funzionali come un&#39;isomerasi disolfuro e disolfuro ossidasi, rispettivamente. Questi risultati rivelano come nativo disolfuro pieghevole &egrave; compiuta nel reticolo endoplasmatico e fornire un contesto per la comprensione della proliferazione di omologhi PDI con arrangiamenti combinatoriale di tioredossina domini.

Formaci&oacute;n de enlace de disulfuro nativa en eucariotas depende de isomerasa prote&iacute;na-disulfuro (PDI) y sus hom&oacute;logos, que contienen diferentes combinaciones de dominios tiorredoxina catal&iacute;tico activo e inactivo. Sin embargo, la contribuci&oacute;n espec&iacute;fica de PDI a la formaci&oacute;n de nuevos disulfuros versus reducci&oacute;n/cambio de disulfuros no nativos es mal entendida. Hemos analizado el papel de dominios individuales del PDI en la formaci&oacute;n del enlace de disulfuro en una reacci&oacute;n impulsada por su oxidante natural, Ero1p. Encontramos que Ero1p oxida los dominios aislados de catal&iacute;tico tiorredoxina PDI, A y A&#39; en la misma proporci&oacute;n. Por el contrario, encontramos que en el contexto de la PDI de cuerpo entero, hay una asimetr&iacute;a en la velocidad de oxidaci&oacute;n de los dos sitios activos. Esta asimetr&iacute;a es el resultado de un doble efecto: una mayor velocidad de oxidaci&oacute;n de la segunda catal&iacute;tica (A&#39;) dominio y la inhibici&oacute;n mediada por sustrato de oxidaci&oacute;n de la primera (A) catal&iacute;tico dominio. La orden espec&iacute;fica de tiorredoxina dominios en PDI es importante en el establecimiento de la asimetr&iacute;a en la velocidad de oxidaci&oacute;n de los dos sitios activos permitiendo A y A&#39;, dos dominios tiorredoxina que son similares en secuencia y estructura, para oponerse a roles funcionales como isomerasa disulfuro y oxidasa de disulfuro, respectivamente. Estos resultados revelan c&oacute;mo nativa disulfuro plegable se logra en el ret&iacute;culo endopl&aacute;smico y proporcionan un contexto para entender la proliferaci&oacute;n de hom&oacute;logos PDI con arreglos combinatorias de tiorredoxina dominios.

Skeletal muscle is a plastic tissue with known gender dimorphism, especially at the metabolic level. A proteomic comparison of male and female murine biceps brachii was undertaken, resolving an average of 600 protein spots of MW 15-150 kDa and pI 5-8. Twenty-six unique full-length proteins spanning 11 KOG groups demonstrated statistically significant (p<0.05) abundance differences between genders; the majority of these proteins have metabolic functions. Identified glycolytic enzymes demonstrated decreased abundance in females, while abundance differences in identified oxidative phosphorylation enzymes were specific to the proteins rather than to the functional group as a whole. Certain cytoskeletal and stress proteins showed specific expression differences, and all three phosphorylation states of creatine kinase showed significant decreased abundance in females. Expression differences were significant but many were subtle (< or = 2-fold), and known hormonally-regulated proteins were not identified. We conclude that while gender dimorphism is present in non-exercised murine skeletal muscle, the proteome comparison of male and female biceps brachii in exercise-naive mice indicates subtle differences rather than a large or obviously hormonal dimorphism.

Muscolo scheletrico &egrave; un tessuto in plastica con dimorfismo di genere conosciuto, soprattutto a livello metabolico. &Egrave; stato intrapreso un confronto proteomico del maschio e femmina murino bicipite brachiale, risolvendo una media di 600 punti di proteina di MW 15-150 kDa e pI 5-8. Venti-sei uniche full-length proteine spanning 11 gruppi KOG dimostrato statisticamente significativa (p &lt; 0.05) differenze di abbondanza tra i generi; la maggior parte di queste proteine hanno funzioni metaboliche.
