Antibody-based proteomics provides a powerful approach for the functional study of the human proteome involving the systematic generation of protein-specific affinity reagents. We used this strategy to construct a comprehensive, antibody-based protein atlas for expression and localization profiles in 48 normal human tissues and 20 different cancers. Here we report a new publicly available database containing, in the first version, approximately 400,000 high resolution images corresponding to more than 700 antibodies toward human proteins. Each image has been annotated by a certified pathologist to provide a knowledge base for functional studies and to allow queries about protein profiles in normal and disease tissues. Our results suggest it should be possible to extend this analysis to the majority of all human proteins thus providing a valuable tool for medical and biological research.

Ici, nous rapportons une nouvelle base de donn&#xE9;es contenant la disposition du public, dans la premi&#xE8;re version, environ 400.000 images &#xE0; haute r&#xE9;solution correspondant &#xE0; plus de 700 anticorps &#xE0; l&#x26;#39;&#xE9;gard des prot&#xE9;ines humaines. Chaque image a &#xE9;t&#xE9; annot&#xE9; par un pathologiste certifi&#xE9; pour fournir une base de connaissances pour des &#xE9;tudes fonctionnelles et &#xE0; autoriser les requ&#xEA;tes sur les profils prot&#xE9;iques dans les tissus normaux et pathologiques. Proteomica basata su anticorpi fornisce un approccio potente per lo studio funzionale del proteoma umano che coinvolge la generazione sistematica dei reagenti di proteine specifiche affinit&agrave;. Abbiamo usato questa strategia per costruire un atlante completo, basato su anticorpo proteina per i profili di espressione e localizzazione in 48 tessuti umani normali e 20 diversi tipi di cancro. Qui riportiamo un nuovo pubblicamente disponibile database contenente, nella prima versione, circa 400.000 immagini ad alta risoluzione, corrispondente a pi&ugrave; di 700 anticorpi verso proteine umane. Ogni immagine &egrave; stata annotata da un patologo certificato per fornire una conoscenza di base per gli studi funzionali e di consentire le query sui profili di proteine nei tessuti normali e malattia. I nostri risultati suggeriscono che dovrebbe essere possibile estendere questa analisi per la maggior parte di tutte le proteine umane fornendo cos&igrave; un prezioso strumento per la ricerca medica e biologica.

Basadas en anticuerpos prote&#xF3;mica ofrece un enfoque poderoso para el estudio funcional del proteoma humano que implique la generaci&#xF3;n sistem&#xE1;tica de los reactivos de afinidad espec&#xED;fico de la prote&#xED;na. Hemos utilizado esta estrategia para construir una paz general, basado en anticuerpos atlas de prote&#xED;nas para la expresi&#xF3;n y localizaci&#xF3;n de perfiles en 48 tejidos humanos normales y 20 tipos de c&#xE1;ncer diferentes. Aqu&#xED; mostramos una nueva base de datos que contiene a disposici&#xF3;n del p&#xFA;blico, en la primera versi&#xF3;n, alrededor de 400.000 im&#xE1;genes de alta resoluci&#xF3;n que corresponden a m&#xE1;s de 700 anticuerpos hacia las prote&#xED;nas humanas. Cada imagen ha sido anotado por un pat&#xF3;logo certificado para proporcionar una base de conocimientos para los estudios funcionales y para permitir las consultas sobre los perfiles de prote&#xED;nas en los tejidos normales y de enfermedad. Nuestros resultados sugieren que deber&#xED;a ser posible extender este an&#xE1;lisis a la mayor&#xED;a de todas las prote&#xED;nas humanas, proporcionando as&#xED; una herramienta valiosa para la investigaci&#xF3;n m&#xE9;dica y biol&#xF3;gica.
