After an initial increase, repeated exposure to a particular stimulus or familiarity with an event results in lower immediate early gene expression levels in relevant brain structures. We predicted that similar effects would occur in Japanese quail after repeated sexual experience within brain areas involved in sexual behavior, namely, the medial preoptic nucleus (POM), the bed nucleus of stria terminalis (BST), and the nucleus taeniae of the amygdala (TnA), an avian homolog of medial amygdala. High experience subjects copulated with a female once on each of 16 consecutive days, whereas low experience subjects were allowed to copulate either once or twice. Control subjects were never exposed to a female. High experience subjects were faster to initiate sexual interaction, performed more cloacal contacts, and completed each cloacal contact faster than low experience subjects. Low experience subjects showed an increase in egr-1 (ZENK) expression, an immediate early gene product used as marker of neural activation in birds, in the areas of interest. In contrast, in high experience animals, egr-1 expression in the POM, BST, and the periaqueductal gray (PAG) was not different than the level of expression in unmated controls. These results show that experience modulates the level of immediate early gene expression in the case of sexual behavior. Our results also indicate that immediate early gene expression in specific brain areas is not necessarily related to behavioral output but depends on the behavioral history of the subjects.

Sexual experience modulates neuronal activity in male Japanese quail.

Apr&egrave;s une augmentation initiale, l&#39;exposition r&eacute;p&eacute;t&eacute;e &agrave; un stimulus particulier et de la familiarit&eacute; avec un &eacute;v&eacute;nement entra&icirc;ne inf&eacute;rieurs imm&eacute;diat niveaux d&#39;expression g&eacute;nique pr&eacute;coce dans les structures c&eacute;r&eacute;brales pertinentes. Nous avons pr&eacute;dit que se produirait des effets similaires in Japanese quail apr&egrave;s plusieurs exp&eacute;rience sexuelle dans les zones du cerveau impliqu&eacute;s dans le comportement sexuel, &agrave; savoir, le noyau pr&eacute;optique m&eacute;dian (POM), le noyau de lit de la strie terminale (BST) et le taeniae du noyau de l&#39;amygdale (TnA), un homologue aviaire d&#39;amygdale m&eacute;dial. Des sujets de haute exp&eacute;rience copulaient avec une femelle fois sur chacun des 16 jours cons&eacute;cutifs, alors que la faible exp&eacute;rience des sujets &eacute;taient autoris&eacute;s &agrave; s&#39;accoupler soit une ou deux fois. Sujets t&eacute;moins n&#39;ont &eacute;t&eacute; jamais expos&eacute;s &agrave; une femelle. Faible exp&eacute;rience sujets ont montr&eacute; une augmentation dans l&#39;expression (ZENK) egr-1, un produit de g&egrave;ne pr&eacute;coce imm&eacute;diat utilis&eacute; comme marqueur de l&#39;activation neurale chez les oiseaux, dans les domaines d&#39;int&eacute;r&ecirc;t. En revanche, chez les animaux de grande exp&eacute;rience, egr-1 expression dans le POM, BST et la substance grise (PAG) n&#39;&eacute;tait pas diff&eacute;rente de celle du niveau d&#39;expression dans les contr&ocirc;les non accoupl&eacute;s. Ces r&eacute;sultats montrent qu&#39;exp&eacute;rience l&#39;module le niveau d&#39;expression de g&egrave;ne pr&eacute;coce imm&eacute;diat en cas de comportement sexuel. Wir voraus, dass &auml;hnliche Effekte, in Japanische Wachtel auftreten w&uuml;rde nach wiederholten, dass sexueller Erfahrung innerhalb Hirnareale Sexualverhalten, n&auml;mlich der medialen preoptic Kern (POM), den Bett-Kern der Stria Terminalis (BST) und der Zellkern-Taeniae von der Amygdala (TnA), eine Vogelgrippe Homolog des medialen Amygdala beteiligt. Hohe Erfahrung Themen unbegattet mit einer weiblichen einmal auf jedem der 16 aufeinanderfolgenden Tagen, w&auml;hrend wenig erfahrenen Untertanen durften entweder einmal oder zweimal zu kopulieren. Kontrollpersonen waren nie ein Weibchen ausgesetzt. Hohe Erfahrung Themen waren schneller sexuelle Interaktion, initiieren durchgef&uuml;hrt mehr Kloaken Kontakte und jeden Kloaken Kontakt schneller als wenig erfahrenen Themen abgeschlossen. Wenig erfahrenen Themen zeigten eine Zunahme Egr-1 (ZENK) Ausdruck, eine sofortige fr&uuml;he Genprodukt verwendet als Marker der neuronalen Aktivierung bei V&ouml;geln, in den Bereichen von Interesse. Im Gegensatz dazu war in hohe Erfahrung Tiere, Egr-1 Ausdruck in der POM, BST und die Periaqu&auml;duktales Grau (PAG) nicht anders als die Ebene des Ausdrucks in unverbundene Steuerelementen. Diese Ergebnisse zeigen, dass die Erfahrung die Ebene der unmittelbaren fr&uuml;hen Genexpression bei Sexualverhalten moduliert. Unsere Ergebnisse zeigen auch, dass sofortige fr&uuml;he Genexpression in bestimmten Hirnregionen sich nicht unbedingt an Verhaltens-Ausgang bezieht aber die verhaltensbasierte Geschichte der Themen h&auml;ngt.

Sexueller Erfahrung moduliert neuronalen Aktivit&auml;t in m&auml;nnliche Japanische Wachtel.

Dopo un aumento iniziale, un&#39;esposizione ripetuta a un particolare stimolo o familiarit&agrave; con un evento risultati nell&#39;immediato inizio del gene espressione livelli inferiori in strutture cerebrali pertinenti. Avevamo previsto che si sarebbero verificato effetti simili nella Quaglia giapponese dopo aver ripetuto l&#39;esperienza sessuale all&#39;interno di aree del cervello coinvolte nel comportamento sessuale, vale a dire, il nucleo mediale preoptic (POM), il nucleo del letto della stria terminalis (BST) e i taeniae nucleo dell&#39;amigdala (TnA), un omologo aviaria di amigdala mediale. Soggetti di elevata esperienza si accoppiassero con un femmina, una volta su ciascuno dei 16 giorni consecutivi, mentre scarsa esperienza soggetti potevano accoppiarsi sia una volta o due volte. Soggetti di controllo sono stati mai esposti a una femmina. Elevata esperienza soggetti erano pi&ugrave; veloce per avviare l&#39;interazione sessuale, eseguiti cloacali pi&ugrave; contatti e completato ogni contatto cloacali pi&ugrave; velocemente rispetto a soggetti di scarsa esperienza. Esperienza basso soggetti hanno mostrato un aumento nell&#39;espressione di egr-1 (ZENK), un prodotto del gene immediato inizio utilizzato come marcatore di attivazione neurale negli uccelli, nelle aree di interesse. Al contrario, negli animali di elevata esperienza, espressione di egr-1 nel POM BST, gray e il periacquiduttale (PAG) non era diverso rispetto al livello di espressione nei controlli spaiati. Questi risultati mostrano che esperienza modula il livello di espressione del gene inizio immediato in caso di comportamento sessuale. Anche i nostri risultati indicano che immediato inizio del gene expression in aree specifiche del cervello non &egrave; necessariamente correlata all&#39;output comportamentali ma dipende dalla storia del comportamento dei soggetti.

Sujetos de control nunca fueron expuestos a una mujer. Alta experiencia temas eran m&aacute;s r&aacute;pido iniciar la interacci&oacute;n sexual, realiza contactos m&aacute;s cloacales y completado cada contacto cloacal m&aacute;s r&aacute;pido que los sujetos bajo experiencia. Experiencia bajo sujetos mostraron un aumento en la expresi&oacute;n de egr-1 (ZENK), un producto de gen temprano inmediato utilizado como marcador de activaci&oacute;n neural en aves, en las &aacute;reas de inter&eacute;s. En cambio, en animales de alta experiencia, expresi&oacute;n de egr-1 en el POM, BST y la sustancia gris periacueductal (PAG) no fue diferente que el nivel de expresi&oacute;n en controles fecundados. Estos resultados muestran que la experiencia modula el nivel de expresi&oacute;n del gen temprano inmediato en el caso de comportamiento sexual. There is considerable evidence for the existence of comorbidity between alcohol-use disorders and depression in humans. One strategy to elucidate hereditary factors affecting the comorbidity of these disorders is to use genetic animal models, such as mouse lines selectively bred for voluntary ethanol consumption. We hypothesized that mice from lines that were bred for high-alcohol preference would manifest increased depression-like phenotypes compared to low-alcohol preferring mice. Mice that were bi-directionally selected and bred on the basis of their High- (HAP) or Low-Alcohol Preference (LAP) were tested in the open-field (OFT), dark-light box (DLB), forced swim (FST), and learned helplessness tests (LH). The study was conducted in two independently derived replicates. In the OFT, both HAP2 and HAP3 mice showed higher levels of general locomotion compared to LAP mice. However, only HAP2 mice spent more time in the center compared to LAP2 mice. In the DLB, there was a slightly higher anxiety-like phenotype in HAP mice. In both FST and LH, we observed higher depression-like behaviors in HAP mice compared to LAP mice, but this was limited to the Replicate 2 mice. Overall, we identified affect-related behavioral changes in mouse lines bred for high-alcohol preference. Notably, the Replicate 3 lines that showed fewer depression-like behaviors also manifest smaller differences in alcohol intake. These data suggest that there may be overlap between genes that predispose to excessive alcohol intake and those underlying affect-related behaviors in the mouse.

In beiden FST und LH beobachteten wir h&ouml;here Depression-&auml;hnliche Verhaltensweisen in HAP-M&auml;use im Vergleich zur LAP-M&auml;use, aber dies beschr&auml;nkte sich die replizieren 2 M&auml;use. Insgesamt haben wir festgestellt, Affekt-bezogene Verhaltens&auml;nderungen in Maus Linien gez&uuml;chtet f&uuml;r hoch-Alkohol-Einstellung. Insbesondere manifestieren die replizieren 3-Linien, die weniger Depression-&auml;hnliche Verhaltensweisen zeigte auch kleinere Unterschiede im Alkoholkonsum. Diese Daten legen nahe, dass es m&ouml;glicherweise &Uuml;berschneidungen zwischen Genen, die f&uuml;r &uuml;berm&auml;&szlig;igen Alkoholkonsum pr&auml;disponieren und die zugrunde liegenden Affekt-bezogene Verhaltensweisen in der Maus.

Abbiamo ipotizzato che topi da linee che sono stati allevati per alto-alcool preferenza sarebbero manifesto fenotipi di depressione-come maggiori rispetto ai topi preferendo bassa gradazione alcolica. Topi che bi-direzionale sono stati selezionati e allevati in base alla loro alta-(HAP) o preferenza di bassa gradazione alcolica (LAP) sono stati testati nel campo aperto (OFT), swim dark-light box (DLB), costretto (FST) e imparato test impotenza (LH). Lo studio &egrave; stato condotto in due repliche derivate in modo indipendente. Nell&#39;OFT, topi sia HAP2 e HAP3 hanno mostrato livelli pi&ugrave; elevati di locomozione generale rispetto ai topi in giro. Tuttavia, solo HAP2 topi speso pi&ugrave; tempo nel centro rispetto a topi di LAP2. In DLB, c&#39;era un fenotipo simile ansia leggermente superiore nei topi HAP. FST sia LH, abbiamo osservato maggiore depressione-come comportamenti nei topi HAP rispetto ai topi in giro, ma questo era limitato a replicare 2 topi. Nel complesso, abbiamo individuato cambiamenti del comportamento correlati a incidere nelle linee del mouse allevati per preferenza ad alta gradazione alcolica. In particolare, le linee di replicare 3 che ha mostrato meno depressione-come comportamenti manifestano anche piccole differenze nell&#39;assunzione di alcol. Questi dati suggeriscono che ci pu&ograve; essere sovrapposizione tra geni che predispongono all&#39;assunzione eccessiva di alcool e quelli sottostanti i comportamenti correlati a incidere nel topo.

Hay considerable evidencia de la existencia de comorbilidad entre los trastornos por consumo de alcohol y la depresi&#xF3;n en seres humanos. Una estrategia para dilucidar los factores hereditarios que afectan a la comorbilidad de estos trastornos es el uso de modelos gen&#xE9;ticos de los animales, tales como l&#xED;neas de ratones criados selectivamente para el consumo de etanol voluntaria. La hip&#xF3;tesis de que los ratones de las l&#xED;neas que fueron criados para la alta de alcohol en la preferencia manifiesta la depresi&#xF3;n mayor, como son los fenotipos en comparaci&#xF3;n con bajo contenido de alcohol que prefieren los ratones. Los ratones que fueron seleccionados de forma bidireccional y se han criado en la base de su preferencia alta (HAP) o bajo contenido de alcohol (LAP) se pusieron a prueba en el campo abierto (OFT), la caja de luz-oscuridad (DCL), nataci&#xF3;n forzada (FST ), y se enter&#xF3; de las pruebas de impotencia (LH). El estudio se realiz&#xF3; en dos de forma independiente derivada repeticiones. En la OFT, ambos HAP2 y HAP3 ratones mostraron niveles m&#xE1;s altos de la locomoci&#xF3;n en general en comparaci&#xF3;n con los ratones LAP. Sin embargo, s&#xF3;lo HAP2 los ratones pasan m&#xE1;s tiempo en el centro en comparaci&#xF3;n con los ratones vuelta 2. En la DCL, hubo un poco m&#xE1;s alto de ansiedad-como fenotipo en ratones HAP. En tanto FST y LH, se observ&#xF3; mayor, como la depresi&#xF3;n, conductas en los ratones de HAP en comparaci&#xF3;n con ratones vuelta, pero esto se limita a la r&#xE9;plica de 2 ratones.
