Amyotrophic lateral sclerosis (ALS) is a progressive, neurodegenerative disease characterized by the selective loss of motor neurons in the brain and spinal cord. Death due to respiratory failure occurs typically 2-5 years after disease onset. The pathogenic mechanism that underlies ALS remains largely unknown, but is known to include both genetic and environmental factors. At the cellular level, pathological changes in motor neuron connections and loss of neuromuscular contacts precede motor neuron degeneration and clinical symptoms. Several lines of recent evidence support the challenging hypothesis that aberrant expression or function of axon guidance proteins such as Semaphorins, Ephrins, Netrins and Slits, normally involved in sculpting and maintaining motor neuron circuits, may induce such pathological changes in motor neuron circuitry and contribute to the pathogenic mechanism involved in ALS. In the present review, we discuss the emerging roles of axon guidance proteins in the pathogenesis of ALS. First, we summarize our current understanding of the role of axon guidance proteins during the formation of motor neuron circuits. Subsequently, we present several lines of evidence showing an association between aberrant axon guidance protein function or expression and ALS. Finally, we discuss the therapeutic potential of axon guidance proteins in understanding and treating the changes in motor neuron connectivity that underlie this debilitating disease.

Axon guidance proteins: novel therapeutic targets for ALS?

Le m&#xE9;canisme pathog&#xE8;ne qui sous-tend la SLA reste largement inconnu, mais il est connu pour inclure des facteurs g&#xE9;n&#xE9;tiques et environnementaux. Plusieurs lignes de donn&#xE9;es r&#xE9;centes appuient l&#x26;#39;hypoth&#xE8;se difficile que l&#x26;#39;expression aberrante ou la fonction des prot&#xE9;ines de guidage axonal comme s&#xE9;maphorines, &#xE9;phrines, les n&#xE9;trines et Slits, normalement impliqu&#xE9; dans la sculpture et le maintien de circuits de neurones moteurs, peut induire des changements pathologiques dans des neurones moteurs circuits et de contribuer &#xE0; le m&#xE9;canisme pathog&#xE9;nique impliqu&#xE9; dans la SLA. Dans la pr&#xE9;sente &#xE9;tude, nous discutons les nouveaux r&#xF4;les de prot&#xE9;ines de guidage axonal dans la pathog&#xE9;n&#xE8;se de la SLA. Tout d&#x26;#39;abord, nous r&#xE9;sumons notre compr&#xE9;hension actuelle du r&#xF4;le des prot&#xE9;ines de guidage axonal au cours de la formation des circuits du motoneurone. La esclerosis lateral amiotr&#xF3;fica (ELA) es una enfermedad progresiva y neurodegenerativa que se caracteriza por la p&#xE9;rdida selectiva de neuronas motoras en el cerebro y la m&#xE9;dula espinal. La muerte por insuficiencia respiratoria generalmente ocurre 2-5 a&#xF1;os despu&#xE9;s del inicio de la enfermedad. El mecanismo patog&#xE9;nico que subyace a la ELA sigue siendo desconocida, pero se sabe que incluyen tanto los factores gen&#xE9;ticos y ambientales. A nivel celular, los cambios patol&#xF3;gicos en conexiones de neuronas motoras y la p&#xE9;rdida de contactos neuromusculares preceden degeneraci&#xF3;n de neuronas motoras y los s&#xED;ntomas cl&#xED;nicos. Varias l&#xED;neas de evidencia reciente apoya la hip&#xF3;tesis de un reto que la expresi&#xF3;n aberrante o funci&#xF3;n de las prote&#xED;nas para gu&#xED;a de axones como semaforinas y efrinas, netrinas y aberturas, que intervienen normalmente en la escultura y el mantenimiento de los circuitos neuronales de motor, pueden inducir estos cambios patol&#xF3;gicos en los circuitos de las neuronas motoras y contribuir a la el mecanismo patog&#xE9;nico implicado en la ELA. En la presente revisi&#xF3;n, se discuten las nuevas funciones de las prote&#xED;nas para gu&#xED;a de axones en la patogenia de la esclerosis lateral amiotr&#xF3;fica. En primer lugar, un resumen de nuestra actual comprensi&#xF3;n de la funci&#xF3;n de las prote&#xED;nas de guiado de los axones durante la formaci&#xF3;n de circuitos neuronales motoras. Posteriormente, presentamos varias l&#xED;neas de evidencia que muestra una asociaci&#xF3;n entre la funci&#xF3;n de la prote&#xED;na aberrante ax&#xF3;n orientaci&#xF3;n o expresi&#xF3;n y la esclerosis lateral amiotr&#xF3;fica. Finalmente, se discute el potencial terap&#xE9;utico de las prote&#xED;nas de guiado de los axones en la comprensi&#xF3;n y el tratamiento de los cambios en la neurona del motor de conectividad que subyacen a esta enfermedad debilitante.
