Circulating epithelial progenitor cells are important for repair of the airway epithelium in a mouse model of tracheal transplantation. We therefore hypothesized that circulating epithelial progenitor cells would also be present in normal human subjects and could be important for repair of the airway after lung injury. As lung transplantation is associated with lung injury, which is severe early on and exacerbated during episodes of infection and rejection, we hypothesized that circulating epithelial progenitor cell levels could predict clinical outcome following lung transplantation.

La sindrome da iper IgE (HIES) &egrave; caratterizzata da ascessi, eczema, infezioni ricorrenti, scheletriche e anomalie del tessuto connettivo, elevato siero IgE e diminuito le risposte infiammatorie. Esiste come forma autosomica dominante e autosomica recessiva che manifesta segni clinici comuni e distintivi. La maggioranza di quelli con HIES autosomica dominante hanno mutazioni eterozigoti nel trasduttore di segnale e attivatore di trascrizione (STAT) -3 e alterata differenziazione T (H) 17.

Difetti lungo il percorso di differenziazione di T (H) 17 sottostanno geneticamente distinte forme di iper sindrome di IgE.

The hyper IgE syndrome (HIES) is characterized by abscesses, eczema, recurrent infections, skeletal and connective tissue abnormalities, elevated serum IgE, and diminished inflammatory responses. It exists as autosomal-dominant and autosomal-recessive forms that manifest common and distinguishing clinical features. A majority of those with autosomal-dominant HIES have heterozygous mutations in signal transducer and activator of transcription (STAT)-3 and impaired T(H)17 differentiation.

Defects along the T(H)17 differentiation pathway underlie genetically distinct forms of the hyper IgE syndrome.

Hyper-IgE-Syndroms (HIES) zeichnet sich durch Abszesse, Ekzeme, rezidivierende Infekte, Skelett- und Bindegewebes Abnormit&auml;ten, erh&ouml;hte Serum-IgE und vermindert entz&uuml;ndliche Reaktionen. Es existiert als autosomal-Dominant und autosomal-rezessive Formen, die gemeinsame und charakteristischen klinischen Symptome manifestieren. Eine Mehrheit derjenigen mit autosomal-dominanten HIES haben heterozygote Mutationen im Signal-Wandler und Aktivator der Transkription (STAT)-3 und gest&ouml;rter T (H) 17-Differenzierung.

The aryl hydrocarbon receptor (AhR) mediates induction of CYP1A1 and CYP1B1 by 2,3,7,8-tetrachlorodibenzo-Ï-dioxin (dioxin) via binding to xenobiotic-responsive elements (XREs) in their enhancer regions. CYP1A1 and CYPIB1 were both inducible by dioxin in human MCF-7 cells. However, only CYP1A1 was inducible in human HepG2 cells. Further experiments focused on providing an explanation for this last observation. Dioxin induced the recruitment of AHR and the transcriptional coactivators p300 and p300/cAMP response element-binding protein binding protein-associated factor (PCAF) to the CYP1B1 enhancer in HepG2 cells but failed to induce recruitment of RNA polymerase II (polII) or the TATA binding protein (TBP) and acetylations of histones 3 and 4 or methylation of histone 3 at the promoter. Because p300 was required for dioxin induction of the aforementioned histone modifications at the CYP1B1 promoter and for induction of CYP1B1 transcription (in MCF-7 cells), the recruitments of p300 and AhR, although necessary, are not sufficient for eliciting the above responses to dioxin. Cytosine residues within CpG dinucleotides at the enhancer, including those within the XREs, were partially methylated, whereas those at the promoter were fully methylated. Treatment of HepG2 cells with 5-aza-2'-deoxycytidine led to partial demethylation of the promoter, restored polII and TBP binding, and CYP1B1 inducibility. Thus, the deficiency of CYP1B1 induction in HepG2 cells is ascribable to cytosine methylation at the promoter, which prevents recruitment of TBP and polII. It is noteworthy that our data indicate that stable recruitment of p300 and PCAF to the CYP1B1 gene does not require their tethering to the promoter and to the enhancer.

Role of epigenetic mechanisms in differential regulation of the dioxin-inducible human CYP1A1 and CYP1B1 genes.

El receptor de hidrocarburo de aril (AhR) media inducci&oacute;n de CYP1A1 y CYP1B1 por 2.378-tetrachlorodibenzo-&rho;-dioxinas (dioxinas) mediante la Uni&oacute;n a elementos xenobi&oacute;ticos-sensibles (XREs) en sus regiones de potenciador. CYP1A1 y CYPIB1 eran ambos inducible por dioxina en c&eacute;lulas humanas de MCF-7. Sin embargo, s&oacute;lo CYP1A1 fue inducible en las c&eacute;lulas HepG2 humanas. Otros experimentos enfocaron en proveer una explicaci&oacute;n para esta &uacute;ltima observaci&oacute;n. Dioxina inducida por el reclutamiento de AHR y la activaci&oacute;n transcripcional p300 y p300/campamento respuesta elemento vinculante prote&iacute;na asociada a la prote&iacute;na factor de uni&oacute;n (PCAF) al optimizador de CYP1B1 en c&eacute;lulas HepG2 pero fallido para inducir el reclutamiento de la ARN polimerasa II (polII) o la prote&iacute;na de uni&oacute;n a TATA (TBP) y acetylations de histonas 3 y 4 o metilaci&oacute;n de histonas 3 en el promotor. Porque p300 fue requerida para la inducci&oacute;n de dioxinas de las modificaciones de las histonas mencionados en el promotor CYP1B1 y para inducir la transcripci&oacute;n CYP1B1 (en c&eacute;lulas MCF-7), los reclutamientos de p300 y AhR, aunque necesario, no son suficientes para que las respuestas anteriores a la dioxina. Residuos de citosina dentro de dinucle&oacute;tidos CpG en el reforzador, las de las XREs, incluidas fueron parcialmente desnaturalizados, mientras que &eacute;sos en el promotor fueron totalmente metilados. Tratamiento de las c&eacute;lulas HepG2 con 5-aza-2&#39;-desoxicitidina llev&oacute; a desmetilaci&oacute;n parcial del promotor, polII restaurada y Uni&oacute;n de TBP y CYP1B1 inducibilidad. As&iacute;, la deficiencia de inducci&oacute;n CYP1B1 en c&eacute;lulas HepG2 es atribuible a la metilaci&oacute;n de la citosina en el promotor, que impide la contrataci&oacute;n de TBP y polII. Smoking is the most important known risk factor for the development of lung cancer. Tobacco exposure results in chronic inflammation, tissue injury, and repair. A recent hypothesis argues for a stem/progenitor cell involved in airway epithelial repair that may be a tumor-initiating cell in lung cancer and which may be associated with recurrence and metastasis. We used immunostaining, quantitative real-time PCR, Western blots, and lung cancer tissue microarrays to identify subpopulations of airway epithelial stem/progenitor cells under steady-state conditions, normal repair, aberrant repair with premalignant lesions and lung cancer, and their correlation with injury and prognosis. We identified a population of keratin 14 (K14)-expressing progenitor epithelial cells that was involved in repair after injury. Dysregulated repair resulted in the persistence of K14+ cells in the airway epithelium in potentially premalignant lesions. The presence of K14+ progenitor airway epithelial cells in NSCLC predicted a poor prognosis, and this predictive value was strongest in smokers, in which it also correlated with metastasis. This suggests that reparative K14+ progenitor cells may be tumor-initiating cells in this subgroup of smokers with NSCLC.

The airway epithelium is in direct contact with the environment and therefore constantly at risk for injury. Basal cells (BCs) have been found to repair the surface epithelium (SE), but the contribution of other stem cell populations to airway epithelial repair has not been identified. We demonstrated that airway submucosal gland (SMG) duct cells, in addition to BCs, survived severe hypoxic-ischemic injury. We developed a method to isolate duct cells from the airway. In vitro and in vivo models were used to compare the self-renewal and differentiation potential of duct cells and BCs. We found that only duct cells were capable of regenerating SMG tubules and ducts, as well as the SE overlying the SMGs. SMG duct cells are therefore a multipotent stem cell for airway epithelial repair This is of importance to the field of lung regeneration as determining the repairing cell populations could lead to the identification of novel therapeutic targets and cell-based therapies for patients with airway diseases.

Epigenetic changes have been implicated in the pathogenesis of asthma. We sought to determine if IL13, a key cytokine in airway inflammation and remodeling, induced epigenetic DNA methylation and miRNAs expression changes in the airways in conjunction with its transcriptional gene regulation. Inducible expression of an IL13 transgene in the airways resulted in significant changes in DNA methylation in 177 genes, most of which were associated with the IL13 transcriptional signature in the airways. A large number of genes whose expression was induced by IL13 were found to have decreased methylation, including those involved in tissue remodeling (Olr1), leukocyte influx (Cxcl3, Cxcl5, CSFr2b), and the Th2 response (C3ar1, Chi3l4). Reciprocally, some genes whose expression was suppressed were found to have increased methylation (e.g. Itga8). In addition, miRNAs were identified with targets for lung development and Wnt signaling, amongst others. These results indicate that IL13 confers an epigenetic methylation and miRNA signature that accompanies its transcriptional program in the airways, which may play a critical role in airway inflammation and remodeling.

Epigenetische Ver&auml;nderungen haben bei der Entstehung von Asthma verwickelt. Cambios epigen&eacute;ticos han sido implicados en la patogenia del asma. SUMMARY AT A GLANCE: The repair of the tracheal airway epithelium after hypoxic-ischemic injury was followed. In doing so we found a consistent tempero-spatial pattern of proliferation and differentiation of specific cell types of the airway epithelium thus increasing our understanding of thes process of repair and regeneration. ABSTRACT: Background and objective:â€ƒ The heterotopic syngeneic tracheal transplantmouse model is an acute hypoxic-ischemic injury model that undergoes complete repair and regeneration. We hypothesized that the repair and regeneration process of the surface epithelium and submucosal glands would occur in a reproducible pattern that could be followed by the expression of specific markers of epithelial cell types. Methods:â€ƒ We used a syngeneic heterotopic tracheal transplant model to develop a temporal and spatial map of cellular repair and regeneration by examining the tracheal grafts at post-transplant days 1, 3, 5, 7, 10 and 14. We used pulsed BrdU and immunofluorescent staining to identify and follow proliferating and repairing cell populations. Results:â€ƒ We confirmed the reproducibility of the injury and repair in the model and we found a distinct sequence of reappearance of the various stem/progenitor and differentiated cell populations of the tracheal surface epithelium and submucosal glands. In the initial phase, the basal and duct cells that survived the injury proliferated to re-epithelialize the basement membrane with K5 and K14 expressing cells. Then these cells proliferated further and differentiated to restore the function of the epithelium. During this repair process, TROP-2 marked all repairing submucosal gland tubules and ducts. Serous cells were found to differentiate 4-5 days before Clara, mucus, and ciliated cells. Conclusions:â€ƒ Improving our understanding of the reparative process of the airway will allow us to identify cell specific mechanisms of repair that could be used as novel therapeutic approaches for abnormal repair leading to airway diseases. Resumen de un vistazo: La reparaci&oacute;n del epitelio de la v&iacute;a a&eacute;rea traqueal despu&eacute;s sigui&oacute; la lesi&oacute;n hip&oacute;xico-isqu&eacute;mica. Al hacerlo as&iacute; encontramos un patr&oacute;n constante de tempero-espacial de proliferaci&oacute;n y diferenciaci&oacute;n de tipos espec&iacute;ficos de c&eacute;lulas del epitelio de las v&iacute;as respiratorias aumentando nuestra comprensi&oacute;n del proceso de reparaci&oacute;n y regeneraci&oacute;n de thes. Resumen: Antecedentes y objetivo: el modelo transplantmouse traqueal syngeneic heterot&oacute;pica es una lesi&oacute;n hip&oacute;xico-isqu&eacute;mica aguda que experimenta la reparaci&oacute;n completa y regeneraci&oacute;n. La hip&oacute;tesis de que el proceso de reparaci&oacute;n y regeneraci&oacute;n del epitelio superficial y las gl&aacute;ndulas de la submucosas se producir&iacute;a en un patr&oacute;n reproducible que podr&iacute;a ser seguido de la expresi&oacute;n de marcadores espec&iacute;ficos de tipos de c&eacute;lulas epiteliales. M&eacute;todos: Hemos utilizado un modelo de trasplante heterot&oacute;pico syngeneic de la tr&aacute;quea para desarrollar un mapa temporal y espacial de reparaci&oacute;n celular y regeneraci&oacute;n mediante el examen de los injertos traqueales en d&iacute;as post-trasplante 1, 3, 5, 7, 10 y 14. Utilizamos BrdU pulsada y tinci&oacute;n inmunofluorescente para identificar y seguir proliferando y reparaci&oacute;n de poblaciones celulares. Resultados: Confirmamos la reproducibilidad de la lesi&oacute;n y reparaci&oacute;n en el modelo y encontramos una secuencia distinta de reaparici&oacute;n de la madre/progenitoras diversos y diferenciados de poblaciones celulares del epitelio superficial traqueal y las gl&aacute;ndulas de la submucosas. En la fase inicial, las c&eacute;lulas basales y conducto que sobrevivieron la lesi&oacute;n proliferaron para re-epithelialize la membrana del s&oacute;tano con c&eacute;lulas expresa K5 y K14. Luego estas c&eacute;lulas proliferaron adicional y diferenciada para restaurar la funci&oacute;n del epitelio. Durante este proceso de reparaci&oacute;n, TROP-2 hab&iacute;a marcado toda reparaci&oacute;n de conductos y t&uacute;bulos de la submucosa de la gl&aacute;ndula. C&eacute;lulas serosas fueron encontradas para diferenciar 4-5 d&iacute;as antes de la Clara, moco y c&eacute;lulas ciliadas. CONCLUSIONES: Mejorar nuestra comprensi&oacute;n del proceso reparativo de la v&iacute;a a&eacute;rea nos permitir&aacute; identificar mecanismos espec&iacute;ficos de c&eacute;lulas de reparaci&oacute;n que podr&iacute;an ser utilizados como nuevos enfoques terap&eacute;uticos para reparaci&oacute;n anormal, lo que provoca enfermedades en las v&iacute;as respiratorias. &copy; 2012 Los autores.
