We previously reported that nanoceria can slow retinal degeneration in the tubby mouse for two weeks by multiple systemic injections. However, the long-term protection of retinal structure and function by directly deliver of nanoceria to the eye had not been explored. In this study, 172 ng of nanoceria in 1 μl saline (1 mm) were intravitreally injected into tubby P7 pups and assays were performed at P28, P49, P80 and P120. The expression of antioxidant associated genes and photoreceptor-specific genes was significantly up regulated, the mislocalization of rod and cone opsins was decreased, and retinal structure and function were protected. These findings demonstrate that nanoceria can function as catalytic antioxidants in vivo and may be broad spectrum therapeutic agents for multiple types of ocular diseases.

Sustained protection against photoreceptor degeneration in tubby mice by intravitreal injection of nanoceria.

Tubby is the founding member of the tubby-like family of proteins. The naturally occurring tubby mutation in mice causes retinitis pigmentosa, hearing loss and obesity. Tubby has been proposed to function as an accessory factor in ciliary trafficking. We directly examined a role for tubby in ciliary trafficking in vivo.

Tubby is required for trafficking G protein-coupled receptors to neuronal cilia.

Cerium oxide nanoparticles (nanoceria) are direct antioxidants; they inhibit pathological neovascularization following a single intravitreal injection into new born very low density lipoprotein receptor knockout (vldlr(-/-)) mice. However, the long-term therapeutic effects and mechanisms of nanoceria action on regression of the existing pathologic neovascularization in the eyes are unknown. We intravitreally injected P28 vldlr(-/-) mice and extended the endpoint for analysis until P70. The data demonstrate that nanoceria sustained their therapeutic function up to 6 weeks. Multiple parameters for nanoceria effects were examined including: regression of existing abnormal blood vessels, reduction of vascular leakage, down-regulation of the expression of vascular endothelial growth factor (VEGF), acrolein, glial fibrillary acidic protein (GFAP) and caspase 3 as well as up-regulation of the expression of rod- and cone-opsin genes. Regulation of ASK1-P38/JNK-NF-κB signaling pathway by nanoceria was investigated. Our data demonstrated that a single intravitreal injection of nanoceria in P28 vldlr(-/-) mice produced sustained regression of existing oxidative stress-induced neovascularizations, prevented blood vessel leakage and inhibited apoptosis via down-regulation of the ASK1-P38/JNK-NF-κB signaling pathway.

Sustained inhibition of neovascularization in vldlr-/- mice following intravitreal injection of cerium oxide nanoparticles and the role of the ASK1-P38/JNK-NF-κB pathway.

Oxidative stress is a node common to the causes and effects of various ocular diseases. We have shown that thioredoxin has neuroprotective effects on tubby photoreceptors. We also demonstrated that nanoceria (cerium oxide nanoparticles), which are direct antioxidants, have long-term effects on prevention of retinal degeneration in tubby mice. Here, using commercially available PCR array plates, we surveyed the regulation in expression of 89 oxidative stress-associated genes in the eyes of P12 tubby mice which are either intravitreally injected with nanoceria or in which the Trx gene is overexpressed. Our data demonstrate that nanoceria and Trx regulate the same group of genes associated with antioxidative stress and antioxidant defense. 

Nanoceria and thioredoxin regulate a common antioxidative gene network in tubby mice.

Mutation of the Tub gene results in the mislocalization of photoreceptor-specific proteins and eventually retinal degeneration. However, the exact mechanism underlying the retinal degeneration remains largely unknown. In this study, we discovered that the expression of endoplasmic reticulum (ER) stress markers, IRE1, ATF6, eIF2α, GRP78/BiP, and XBP-1, is up regulated during tubby retinal development. The dynamics of the expression of these genes are time-dependent and coincided with the time-course of photoreceptor death. Our data also demonstrated that ER stress triggers apoptosis via down-regulation of Bcl2, up-regulation of CHOP and the activation of NF-кB signaling. 

Correlation of ER stress and retinal degeneration in tubby mice.

Age-related macular degeneration (AMD) is the leading cause of blinding diseases. The "dry" form of AMD is the most common form of AMD. In contrast to the treatable neovascular (wet) AMD, no effective treatment is available for dry AMD. In this review, we summarize the animal models and therapeutic strategies for dry AMD. The novel candidates as potential treatment targets and the potential effectiveness of nanoceria as a treatment of dry AMD are also discussed. 

Nanoceria: a Potential Therapeutic for Dry AMD.

Diabetic retinopathy (DR) is one of the major complications of diabetes. Although great efforts have been made to uncover the mechanisms underlying the pathology of DR, the exact causes of DR remain largely unknown. Because of multifactor involvement in DR etiology, currently no effective therapeutic treatments for DR are available. In this paper, we review the pathology of DR, commonly used animal models, and novel therapeutic approaches. Perspectives and future directions for DR treatment are discussed.
