Chondroitin/dermatan sulfate (CS/DS) is a glycosaminoglycan (GAG) found in abundance in extracellular matrices. In connective tissue, CS/DS proteoglycans play structural roles in maintaining viscoelasticity through the large number of immobilized sulfate groups on CS/DS chains. CS/DS chains also bind protein families including growth factors and growth factor receptors. Through such interactions, CS/DS chains play important roles in neurobiochemical processes, connective tissue homeostasis, coagulation, and cell growth regulation. Expression of DS has been observed to increase in cancerous tissue relative to controls. In earlier studies, MS(2) was used to compare the types of CS/DS isomers present in biological samples. The results demonstrated that product ion abundances reflect the types of CS/DS repeats present and can be used quantitatively. It was not clear, however, to which of the CS/DS repeats the product ions abundances were sensitive. The present work explores the utility of MS(3) for structural characterization of CS/DS oligosaccharides. The data show that MS(3) product ion abundances correlate with the presence of DS-like repeats in specific positions on the oligosaccharide chains.

CS/DS Proteoglykane spielen in Bindegewebe strukturelle Rollen bei der Aufrechterhaltung der Viskoelastizit&auml;t durch die gro&szlig;e Anzahl der immobilisierte Sulfat Gruppen auf CS/DS-Ketten. CS/DS Ketten binden ebenfalls Proteinfamilien einschlie&szlig;lich Wachstumsfaktoren und Wachstumsfaktor-Rezeptoren. Durch solche Interaktionen spielen CS/DS Ketten wichtige Rollen in Neurobiochemical Prozesse, Bindegewebe-Hom&ouml;ostase, Gerinnung und Zelle Wachstum Verordnung. Ausdruck der DS hat beobachtet worden, um in Krebsgewebe relativ zu Steuerelementen zu erh&ouml;hen. In fr&uuml;heren Studien wurde MS(2) verwendet, um die Typen CS/DS-Isomere in biologischen Proben vorhanden zu vergleichen. Die Ergebnisse gezeigt, dass Produkt Ion H&auml;ufigkeiten die Typen CS/DS Wiederholungen anwesend reflektieren und quantitativ eingesetzt werden. Condroitina/dermatan sulfato (CS/DS) es un glicosaminoglicano (GAG) que se encuentra en abundancia en matrices extracelulares. En tejido conectivo, proteoglicanos CS/DS un papel estructural en el mantenimiento de viscoelasticidad a trav&eacute;s de la gran cantidad de grupos sulfato inmovilizados en cadenas de CS/DS. Cadenas de CS/DS tambi&eacute;n obligar a familias de prote&iacute;nas como factores de crecimiento y receptores de factor de crecimiento. A trav&eacute;s de estas interacciones, cadenas de CS/DS juegan papeles importantes en los procesos de neurobiochemical, homeostasis del tejido conectivo, coagulaci&oacute;n y regulaci&oacute;n del crecimiento celular. Expresi&oacute;n de DS se ha observado que el aumento de tejido canceroso en relaci&oacute;n con los controles. En estudios anteriores, el MS(2) fue utilizado para comparar los tipos de is&oacute;meros de CS/DS presentes en muestras biol&oacute;gicas. Los resultados demostraron que producto ion abundancia refleja los tipos de repeticiones de CS/DS presentes y puede ser utilizada cuantitativamente. No estaba claro, sin embargo, para que de la CS/DS repite que la abundancia de los iones del producto era sensible.
