Neddylation promotes the process of ubiquitination, which plays a critical role in the degradation of numerous proteins, including cell cycle and apoptosis regulators. In our previous study, an increase in neddylation was identified in melanoma cell lines. In the present study, the upregulation of neddylation was detected in melanoma tissues which confirmed the results of our previous study on melanoma cell lines. To explore the mechanism by which the process of neddylation was increased, the enzymes that regulate the process were investigated. These neddylation-related regulatory enzymes are potential targets for melanoma therapy. Downregulation of UBA3, a subunit of the E1 enzyme, by RNA interference caused cell cycle arrest at G0G1 in the M14 cell line. In addition, cyclin D expression declined, whereas p27, p21 and bax expression increased. These findings suggest that interfering with the neddylation pathway may decrease the proliferation of melanoma through the modulation of cell cycle regulators and apoptosis promoters.

Expression of neddylation-related proteins in melanoma cell lines and the effect of neddylation on melanoma proliferation.

The present study attempted to analyze the clinical characteristics and pathogenesis of Kawasaki disease (KD) in children with hyperbilirubinemia.A total of 390 children with KD hospitalized in our hospital from September 2018 to July 2019 were selected and divided into control (270 cases) and hyperbilirubinemia (120 cases) groups based on the total, direct, and indirect bilirubin values after admission. Clinical data of the inflammatory index and fever process of the 2 groups were analyzed and compared.The difference in sex and age between the 2 groups was statistically nonsignificant (P > .05). In the hyperbilirubinemia group, the white blood cell count, C-reactive protein, hemoglobin, platelet count, erythrocyte sedimentation rate, alanine aminotransferase, aspartate aminotransferase, albumin, and routine urine leucocyte; and incidence of coronary artery expansion, heart injury, and unreactive gamma globulin treatment were higher than those in the control group and the differences were statistically significant (P < .05). In the hyperbilirubinemia group, the mean fever duration before admission was shorter than that in the control group, whereas the fever duration after gamma globulin treatment was longer than that in the control group; these differences were statistically significant (P < .05).Hyperbilirubinemia incidence in children with KD was approximately 30.77% (120 cases), of which increased direct bilirubin was observed in 70.83% (85 cases) and increased indirect bilirubin in 29.17% (35 cases). Children with KD combined with hyperbilirubinemia exhibited a strong inflammatory reaction, which may be due to liver damage or biliary block.

Analysis of hyperbilirubinemia in patients with Kawasaki disease.

It has been reported that ubiquitin C-terminal hydrolase-L3 (UCHL3) plays an important role in cancer development; however, the role of UCHL3 in melanoma remains unclear. The present study aimed to investigate the role of ubiquitin C-terminal hydrolase-L3 (UCHL3) and determine its underlying molecular mechanisms in melanoma occurrence and development using  studies. Reverse transcription-quantitative PCR analysis was performed to detect UCHL3 mRNA expression. The MTT assay was performed to assess cell proliferation. Cell apoptosis was analyzed via flow cytometry and the TUNEL assay. Cell ultrastructure was observed via transmission electron microscopy. LC3B protein expression was detected via cellular immunofluorescence, while neural precursor cell-expressed developmentally downregulated protein 8 (NEDD8) and LC3 protein expression levels, and NEDD8 ubiquitination were detected via western blot analysis. The results demonstrated that transfection with small interfering (si)RNA-UCHL3 significantly suppressed cell proliferation, whereas apoptosis was significantly enhanced, as well as autophagy, autophagosome formation and LC3B protein expression. In addition, NEDD8 protein expression and autophagosome numbers significantly decreased, while the LC3II/LC3I ratio significantly increased. NEDD8 knockdown via transfection with si-NEDD8 had similar effects to si-UCHL3, as well as si-UCHL3+ si-NEDD8. Taken together, the results of the present study suggest that UCHL3 knockdown decreases melanoma cell proliferation by increasing cell autophagy through regulating NEDD8 expression and autophagosome numbers.

UCHL3 plays an important role in the occurrence and development of melanoma.

The present study aimed to discuss the effects and relative mechanisms of NEDD4 E3 ubiquitin protein ligase (NEDD4) in cutaneous melanoma (CMM) occurrence and development. Clinical cancer and adjacent normal tissues samples were collected to analyze pathological changes and protein expression of NEDD4. Moreover, small interfering (si)RNA was used to knockdown NEDD4 expression in SK-MEL-2 and Malme-3M cells. Cellular proliferation, apoptosis, invasiveness and migration were examined using colony formation, flow cytometric, Transwell and wound-healing assays, respectively. In addition, the relative mRNA and protein expression levels of NEDD4, notch receptor 1 (Notch1) and PTEN were evaluated via reverse transcription-quantitative (RT-q) PCR and western blotting. It was found that NEDD4 mRNA and protein expression were significantly upregulated (both P<0.01). Following NEDD4-knockdown, colony number was significantly decreased, while the apoptotic rate was significantly increased, the invasive cell number was significantly inhibited and the wound-healing capacity was significantly decreased. Following si-NEDD4 transfection, RT-qPCR and western blotting revealed that NEDD4 and Notch1 mRNA and protein expression levels were significantly downregulated, while those of PTEN were significantly upregulated in the SK-MEL-2 and Malme-3M cell lines. Collectively, the current results suggest that NEDD4-knockdown effectively suppressed CMM biological activity by regulating the Notch1/PTEN pathway .

Two novel mutations in LSS gene associated with hypotrichosis simplex in a Chinese family.

Glioblastomas (GBM), the most common primary brain tumor, lack accurate prognostic markers and have a poor prognosis. Our study was designed to identify effective biomarkers for GBM prognosis analysis and development of precise treatments. Differentially expressed genes (DEGs) between GBM patients and controls were analyzed from the Xena database and GEPIA. Based on the screened DEGs, univariate COX and LASSO regression analysis were performed to identify the most relevant genes associated with GBM prognosis. Genes highly expressed in GBM patients were selected to construct receiver operating characteristic analysis and enrichment analysis was constructed on groups of high and low expression of adipocyte enhancer-binding protein 1 (AEBP1). CIBERSORT, ssGSEA and ESTIMATE were used to perform immune infiltration analysis. About 3297 DEGs were identified using data from Xena database; 8 prognostic genes were identified. AEBP1, which plays a role in neuronal differentiation and development, was positively correlated in GBMs with immune infiltration; its high expression in cancer patients is associated with short overall survival and advanced tumor staging. This study suggests that AEBP1 could serve as a prognostic marker for GBMs and that patients with high expression may have a better response to immunotherapy.

Clinical significance and potential mechanism of AEBP1 in glioblastoma.

: To explore the clinical value of miR-193a-5p in neonatal acute respiratory distress syndrome (ARDS) and its role in ARDS cell model . : RT-qPCR was utilized to detect miR-193a-5p level. Correlation analysis was implemented to assess the correlation between miR-193a-5p and clinical indicators (IL-6, IL-1β, TNF-α, LUS). Human lung epithelial cells induced by LPS were used to construct ARDS cell model. The effects of miR-193a-5p on cell viability, apoptosis and inflammation were evaluated by CCK-8, flow cytometry and ELISA. The target gene of miR-193a-5p was predicted and verified by StarBaseV2.0 and luciferase reporter gene, respectively. : MiR-193a-5p level in the ARDS group was down-regulated. MiR-193a-5p levels were negatively correlated with clinical indicators.  studies revealed that up-regulation of miR-193a-5p significantly improved LPS-induced apoptosis, inflammation and viability inhibition. : The expression of miR-193a-5p was decreased in neonatal ARDS, it is negatively correlated with the pro-inflammatory factors levels.

Clinical Diagnostic Value of miR-193a-5p in Neonatal Acute Respiratory Distress Syndrome and Analysis of Its Effect on Human Lung Epithelial Cells.

Effect of pulmonary rehabilitation nursing approach with integrated traditional Chinese and western medicine in children with Mycoplasma Pneumonia after Bronchoalveolar Lavage.

 Stapokibart is a novel anti-interleukin-4 receptor α subunit monoclonal antibody approved for moderate-to-severe atopic dermatitis (AD) in adults.  To evaluate the feasibility of extending dosing interval of stapokibart based on efficacy response in AD.  In this ongoing, multicenter, open-label, single-arm trial (NCT06116565), stapokibart 300 mg was administered subcutaneously every 2 weeks (Q2W) for 12 weeks, followed by dosing interval adjustments at weeks 12 and 36. At week 12, nonresponders (failed to achieve ≥50% improvement from baseline in Eczema Area and Severity Index [EASI-50]) were withdrawn, fast responders (achieved ≥75% improvement from baseline in EASI score [EASI-75] or Investigator's Global Assessment score of 0/1 [IGA 0/1]) were extended to every 4 weeks (Q4W), and slow responders (achieved EASI-50 but not EASI-75 or IGA 0/1) continued Q2W dosing. At week 36, fast responders achieving ≥90% improvement from baseline in EASI score (EASI-90) or IGA 0/1 were further extended to every 8 weeks (Q8W) (Q2W-Q4W-Q8W group), slow responders with EASI-90 or IGA 0/1 were extended to Q4W (Q2W-Q2W-Q4W group), and others maintained their prior dosing through week 52 (Q2W-Q4W-Q4W and Q2W-Q2W-Q2W groups).  As of April 7, 2025, 256 and 200 patients completed 12 and 36 weeks of treatment, respectively. In the Q2W-Q4W-Q8W group, 93.0%, 73.7%, and 70.7% of patients sustained EASI-75, IGA 0/1, and ≥4-point reduction in weekly average of daily Peak Pruritus Numerical Rating Scale at week 52, versus 99.2%, 90.1%, and 71.1% at week 36, and 98.3%, 54.5%, and 58.3% at week 12. These response rates in the Q2W-Q4W-Q4W group were 96.2%, 23.1%, and 38.5% at week 12, versus 81.8%, 63.6%, and 72.7% at week 52. In the Q2W-Q2W-Q4W group, these responses were achieved in 95.8%, 83.3%, and 77.3% of patients at week 36, increasing to 100%, 80.0%, and 80.0% at week 52. In the Q2W-Q2W-Q2W group, 84.6%, 23.1%, and 71.4% of patients progressively achieved these responses at week 52 with continued standard dosing. The overall incidence of treatment-emergent adverse events was 78.6%, mostly mild or moderate.  Extending dosing interval of stapokibart based on efficacy response demonstrated sustained efficacy and favorable safety in AD.

There is an unmet treatment need and a lack of efficacy evidence for Huoxiang Zhengqi (HXZQ) oral liquid for atopic dermatitis (AD).

Chinese herbal formula Huoxiang Zhengqi Qral liquid for atopic dermatitis: The CHARM randomized controlled trial.

This study was conducted aimed at investigating the effectiveness of combined laser and topical tranexamic acid in the treatment of melasma. A total of 160 melasma patients treated in the dermatology department at the author's hospital from May 2021 to October 2022 were randomly assigned to a control group (laser treatment only, n = 80) and an observation group (combined laser and topical tranexamic acid, n = 80). General patient information was collected, and clinical analysis of melasma area and depth scores was performed before and after treatment. After treatment, the observation group showed significantly smaller melasma area and depth compared to the control group (P < 0.05). Skin quality scores post-treatment were higher in the observation group (P < 0.05). Upon re-examination at the 6th and 12th month, the observation group exhibited smaller melasma areas than the control group (P < 0.05), indicating superior long-term control. The observation group had a lower incidence of side effects (P < 0.05), higher rates of both very satisfied and totally satisfied patients (P < 0.05), and lower dissatisfaction rates (P < 0.05) than the control group. Combined laser and topical tranexamic acid treatment is significantly more effective than laser treatment alone in reducing melasma area and depth, minimizing side effects, and improving patient satisfaction.
