Biotechnology needs to explore the capacity of different organisms to overproduce proteins of interest at low cost. In this paper, we show that Streptomyces lividans is a suitable host for the expression of Thermus thermophilus genes and report the overproduction of the corresponding proteins. This capacity was corroborated after cloning the genes corresponding to an alkaline phosphatase (a periplasmic enzyme in T. thermophilus) and that corresponding to a beta-glycosidase (an intracellular enzyme) in Escherichia coli and in S. lividans. Comparison of the production in both hosts revealed that the expression of active protein achieved in S. lividans was much higher than in E. coli, especially in the case of the periplasmic enzyme. In fact, the native signal peptide of the T. thermophilus phosphatase was functional in S. lividans, being processed at the same peptide bond in both organisms, allowing the overproduction and secretion of this protein to the S. lividans culture supernatant. As in E. coli, the thermostability of the expressed proteins allowed a huge purification factor upon thermal denaturation and precipitation of the host proteins. We conclude that S. lividans is a very efficient and industry-friendly host for the expression of thermophilic proteins from Thermus spp.

High-level overproduction of Thermus enzymes in Streptomyces lividans.

Biotechnologie doit explorer la capacit&eacute; des diff&eacute;rents organismes de surproduction de prot&eacute;ines d&#39;int&eacute;r&ecirc;t &agrave; faible co&ucirc;t. Dans cet article, nous montrons que les Streptomyces lividans est un h&ocirc;te appropri&eacute; pour l&#39;expression des g&egrave;nes de Thermus thermophilus et rapport la surproduction de prot&eacute;ines correspondantes. Cette capacit&eacute; a &eacute;t&eacute; confirm&eacute;e apr&egrave;s le clonage des g&egrave;nes correspondant &agrave; une phosphatase alcaline (une enzyme p&eacute;riplasmique chez T. thermophilus) et celle qui correspond &agrave; une b&ecirc;ta-glycosidase (une enzyme intracellulaire) chez Escherichia coli et de S. lividans. Comparaison de la production dans les deux h&ocirc;tes ont r&eacute;v&eacute;l&eacute; que l&#39;expression de la prot&eacute;ine active atteint chez S. lividans &eacute;tait beaucoup plus &eacute;lev&eacute;e que chez e. coli, surtout dans le cas de l&#39;enzyme p&eacute;riplasmique. En fait, le peptide signal natif de la T. thermophilus phosphatase fonctionnait dans S. lividans, trait&eacute;s &agrave; la m&ecirc;me liaison peptidique chez les deux organismes, permettant &agrave; la surproduction et la s&eacute;cr&eacute;tion de cette prot&eacute;ine pour le surnageant de culture de S. lividans. Comme e. coli, la thermostabilit&eacute; des prot&eacute;ines exprim&eacute;es a permis un facteur &eacute;norme de purification &agrave; la d&eacute;naturation thermique et la pr&eacute;cipitation des prot&eacute;ines de l&#39;h&ocirc;te. Nous concluons que le S. lividans est un h&ocirc;te tr&egrave;s efficace et respectueux de l&#39;industrie pour l&#39;expression des prot&eacute;ines thermophiles de Thermus spp.

Surproduction de haut niveau des enzymes de Thermus chez Streptomyces lividans.

Biotechnologie muss die F&auml;higkeit der verschiedenen Organismen, &Uuml;berproduktion der Proteine des Interesses zu niedrigen Kosten zu pr&uuml;fen. In diesem Beitrag zeigen wir, dass Streptomyces Lividans eine geeignete Host f&uuml;r den Ausdruck von Thermus Thermophilus Gene und Bericht der &Uuml;berproduktion der entsprechenden Proteine ist. Diese Kapazit&auml;t wurde nach dem Klonen die Gene, die eine alkalische Phosphatase (ein periplasmatischen Enzym in T. Thermophilus) und entsprechend eine Beta-Glycosidaseinhibitoren (intrazellul&auml;re Enzym) in Escherichia coli und S. Lividans best&auml;tigt. Vergleich der Produktion in beide Hosts ergab, dass der Ausdruck des aktiven Proteins erreicht in S. Lividans wesentlich h&ouml;her als in E. Coli, insbesondere bei den periplasmatischen Enzym. In der Tat war das native Signal-Peptid von T.-Thermophilus-Phosphatase funktionell in S. Lividans, bei der gleichen Peptidbindung in beiden Organismen, so dass die &Uuml;berproduktion und Sekretion dieses Proteins auf den S. Lividans Kultur &uuml;berstand verarbeitet werden. In E. Coli erlaubt die Thermostabilit&auml;t der exprimierten Proteine einen gro&szlig;e Reinigung-Faktor auf thermische Denaturierung und Niederschlag f&uuml;r die Host-Proteine. Wir feststellen, dass S. Lividans sehr effizient und Industrie-freundliche Host f&uuml;r den Ausdruck der thermophile Proteine aus Thermus spp.

Allgemeine &Uuml;berproduktion von Thermus Enzyme in Streptomyces Lividans.

Biotecnologia ha bisogno di esplorare la capacit&agrave; degli organismi differenti di sovrapproduzione di proteine di interesse a basso costo. In questa carta, mostriamo che Streptomyces lividans &egrave; un host adatto per l&#39;espressione dei geni di Thermus thermophilus e relazione la sovrapproduzione di proteine corrispondente. Questa capacit&agrave; &egrave; stata confermata dopo la clonazione dei geni corrispondenti a una fosfatasi alcalina (un enzima periplasmico in T. thermophilus) e quello corrispondente a un beta-glicosidasico (un enzima intracellulare) in Escherichia coli ed in S. lividans. Confronto della produzione in entrambi gli host ha rivelato che l&#39;espressione della proteina attiva realizzata in S. lividans era molto superiore in e. coli, soprattutto nel caso dell&#39;enzima periplasmico. Infatti, il peptide segnale nativo della fosfatasi thermophilus T. era funzionale in S. lividans, in corso di elaborazione presso lo stesso legame peptidico in entrambi gli organismi, consentendo la sovrapproduzione e la secrezione di questa proteina per il surnatante di cultura S. lividans. Come e. coli, la termostabilit&agrave; delle proteine espresse ha permesso un fattore enorme purificazione dopo denaturazione termica e precipitazione delle proteine ospite. Concludiamo che S. lividans &egrave; un ospite molto efficiente e industria-friendly per l&#39;espressione delle proteine termofili da Thermus spp.

Biotecnolog&iacute;a necesita explorar la capacidad de diversos organismos que sobreproducen unas con otras prote&iacute;nas de inter&eacute;s a bajo costo. En este trabajo mostramos que el Streptomyces lividans es un hu&eacute;sped adecuado para la expresi&oacute;n de genes de Thermus thermophilus e informe la sobreproducci&oacute;n de las prote&iacute;nas correspondientes. Esta capacidad se corrobor&oacute; despu&eacute;s de clonaci&oacute;n de los genes correspondientes a una fosfatasa alcalina (una enzima periplasmic en T. thermophilus) y el correspondiente a un beta-glucosidasa (una enzima intracelular) en Escherichia coli y en S. lividans. Comparaci&oacute;n de la producci&oacute;n en ambas m&aacute;quinas revel&oacute; que la expresi&oacute;n de la prote&iacute;na activa en S. lividans era mucho m&aacute;s alta que en e. coli, especialmente en el caso de la enzima periplasmic. De hecho, el p&eacute;ptido se&ntilde;al nativa de la fosfatasa de T. thermophilus era funcional en S. lividans, siendo procesados en el mismo enlace pept&iacute;dico en ambos organismos, permitiendo la sobreproducci&oacute;n y la secreci&oacute;n de esta prote&iacute;na en el sobrenadante de cultivo de S. lividans. Como en e. coli, la termoestabilidad de las prote&iacute;nas expresadas permiti&oacute; un factor enorme purificaci&oacute;n desnaturalizaci&oacute;n t&eacute;rmica y precipitaci&oacute;n de las prote&iacute;nas del hu&eacute;sped. Concluimos que S. lividans es un anfitri&oacute;n muy eficiente y respetuoso con la industria para la expresi&oacute;n de prote&iacute;nas term&oacute;filas de Thermus spp.

PstS is a phosphate-binding lipoprotein that is part of the high-affinity phosphate transport system. Streptomyces lividans accumulates high amounts of the PstS protein in the supernatant of liquid cultures grown in the presence of different carbon sources, such as fructose or mannose, but not in the presence of glucose or in basal complex medium.

The abundance of two-component systems (TCSs) in Streptomyces coelicolor A3(2) genome indicates their importance in the physiology of this soil bacteria. Currently, several TCSs have been related to antibiotic regulation, and the purpose in this study was the characterization of five TCSs, selected by sequence homology with the well-known absA1A2 system, that could also be associated with this important process. Null mutants of the five TCSs were obtained and two mutants (Î”SCO1744/1745 and Î”SCO4596/4597/4598) showed significant differences in both antibiotic production and morphological differentiation, and have been renamed as abr (antibiotic regulator). No detectable changes in antibiotic production were found in the mutants in the systems that include the ORFs SCO3638/3639, SCO3640/3641 and SCO2165/2166 in any of the culture conditions assayed. The system SCO1744/1745 (AbrA1/A2) was involved in negative regulation of antibiotic production, and acted also as a negative regulator of the morphological differentiation. By contrast, the system SCO4596/4597/4598 (AbrC1/C2/C3), composed of two histidine kinases and one response regulator, had positive effects on both morphological development and antibiotic production. Microarray analyses of the Î”abrC1/C2/C3 and wild-type transcriptomes revealed downregulation of actII-ORF4 and cdaR genes, the actinorhodin and calcium-dependent antibiotic pathway-specific regulators respectively. These results demonstrated the involvement of these new two-component systems in antibiotic production and morphological differentiation by different approaches. One is a pleiotropic negative regulator: abrA1/A2. The other one is a positive regulator composed of three elements, two histidine kinases and one response regulator: abrC1/C2/C3.

L&#39;abondance des deux composantes de syst&egrave;mes (TCSs) dans le g&eacute;nome de Streptomyces coelicolor A3 (2) indique leur importance dans la physiologie de cette bact&eacute;rie de sol. Actuellement, plusieurs TCSs ont &eacute;t&eacute; li&eacute;es &agrave; la r&eacute;glementation aux antibiotiques, et le but de cette &eacute;tude portait sur la caract&eacute;risation des cinq TCSs, s&eacute;lectionn&eacute; par homologie avec le syst&egrave;me d&#39;absA1A2 bien connu, qui pourrait &eacute;galement &ecirc;tre associ&eacute;e &agrave; ce processus important. Die F&uuml;lle der zwei-Komponenten-Systeme (TCSs) in Streptomyces Coelicolor A3(2) Genom zeigt ihre Bedeutung in der Physiologie von diesem Bodenbakterien. Derzeit mehrere TCSs antibiotische Verordnung verwandt, und der Zweck dieser Studie war die Charakterisierung der f&uuml;nf TCSs, ausgew&auml;hlt von Sequenz-Homologie mit den bekannten absA1A2-System, das auch diesen wichtigen Prozess zugeordnet werden konnte. NULL Mutanten von den f&uuml;nf TCSs wurden erzielt und zwei Mutanten (&Delta;SCO1744/1745 und &Delta;SCO4596/4597/4598) zeigten signifikante Unterschiede in der antibiotischen Produktion und morphologischen Differenzierung und als Abr (antibiotische Regler) umbenannt worden. Keine nachweisbaren &Auml;nderungen in der antibiotischen Produktion wurden in die Mutanten in den Systemen gefunden, die den ORFs SCO3638/3639, SCO3640/3641 und SCO2165/2166 in den Kultur-Bedingungen gepr&uuml;ft. Einer ist einen pleiotropen negativen Regulator: abrA1/A2. Das andere ist ein positiver Regulator bestehend aus drei Elementen, zwei Histidin-Kinasen und eine Antwort-Regler: abrC1/C2/C3.

Attualmente, TCSs diversi hanno stato relative al regolamento agli antibiotici, e lo scopo di questo studio &egrave; stata la caratterizzazione dei cinque TCSs, selezionati da omologia di sequenza con il sistema ben noto absA1A2, che potrebbe anche essere associato con questo importante processo. Analisi di Microarray della transcriptomes &Delta;abrC1/C2/C3 e selvaggio-tipo rivelato rispettivamente downregulation dei geni actII-ORF4 e cdaR, il actinorhodin e regolatori specifici pathway antibiotici calcio-dipendente. Questi risultati hanno dimostrato il coinvolgimento di questi nuovi sistemi bicomponenti in produzione antibiotica e differenziazione morfologica di approcci diversi. La abundancia de sistemas de dos componentes (TCSs) en el genoma de Streptomyces coelicolor de los A3(2) indica su importancia en la fisiolog&iacute;a de esta bacteria del suelo. Actualmente, varios TCSs han estado relacionados con regulaci&oacute;n de antibi&oacute;tico, y el prop&oacute;sito de este estudio fue la caracterizaci&oacute;n de cinco TCSs, seleccionado por homolog&iacute;a de secuencia con el sistema de absA1A2 bien conocido, que tambi&eacute;n podr&iacute;a estar asociado con este importante proceso. Se obtuvieron mutantes nulos de las cinco TCSs y dos mutantes (&Delta;SCO1744/1745 y &Delta;SCO4596/4597/4598) mostraban diferencias significativas en la producci&oacute;n de antibi&oacute;tico y diferenciaci&oacute;n morfol&oacute;gica y han pasado a llamarse abr (regulador de antibi&oacute;tico). Ning&uacute;n cambio perceptible en la producci&oacute;n de antibi&oacute;tico fueron encontrado en los mutantes en los sistemas que incluyen ORFs SCO3638/3639, SCO3640/3641 y SCO2165/2166 en cualquiera de las condiciones de cultivo analizadas. An&aacute;lisis de microarrays de los transcriptomas &Delta;abrC1/C2/C3 y salvaje-tipo revelaron downregulation de genes actII-ORF4 y &#x130;maj, actinorhodin y dependiente de calcio antibi&oacute;ticos espec&iacute;fico v&iacute;a reguladores respectivamente. Estos resultados demostraron la implicaci&oacute;n de estos nuevos sistemas de dos componentes en la producci&oacute;n de antibi&oacute;tico y diferenciaci&oacute;n morfol&oacute;gica por diferentes enfoques.
