Local control of mRNA translation modulates neuronal development, synaptic plasticity, and memory formation. A poorly understood aspect of this control is the role and composition of ribonucleoprotein (RNP) particles that mediate transport and translation of neuronal RNAs. Here, we show that staufen- and FMRP-containing RNPs in Drosophila neurons contain proteins also present in somatic "P bodies," including the RNA-degradative enzymes Dcp1p and Xrn1p/Pacman and crucial components of miRNA (argonaute), NMD (Upf1p), and general translational repression (Dhh1p/Me31B) pathways. Drosophila Me31B is shown to participate (1) with an FMRP-associated, P body protein (Scd6p/trailer hitch) in FMRP-driven, argonaute-dependent translational repression in developing eye imaginal discs; (2) in dendritic elaboration of larval sensory neurons; and (3) in bantam miRNA-mediated translational repression in wing imaginal discs. These results argue for a conserved mechanism of translational control critical to neuronal function and open up new experimental avenues for understanding the regulation of mRNA function within neurons.

Taufliege Me31B erscheint (1) mit einer FMRP-assoziierte, P K&ouml;rper Protein (Scd6p/Anh&auml;ngerkupplung) in FMRP getrieben, Argonaut-abh&auml;ngige translational Unterdr&uuml;ckung im dritten Auge so Scheiben zu beteiligen; (2) in dendritischen Ausarbeitung larvalen sensorische Neuronen; und (3) in bantam MiRNA-vermittelten translational Unterdr&uuml;ckung in Fl&uuml;gel so Scheiben. Diese Ergebnisse sprechen f&uuml;r eine konservierte Mechanismus der translatorischen Kontrolle neuronale Funktion unbedingt und er&ouml;ffnen neue experimentelle M&ouml;glichkeiten f&uuml;r die Regulierung der mRNA-Funktion innerhalb der Neuronen verstehen.

The Fragile-X Mental Retardation protein (FMRP/dFMR1) is one of the best-studied neuronal translational control molecules and here we describe the identification and early characterization of proteins likely to function in the dFMR1 pathway. Induction of the dFMR1 in sevenless-expressing cells of the Drosophila eye causes a disorganized (rough) eye through a mechanism that requires residues necessary for dFMR1/FMRP's translational repressor function. Several mutations in dco, orb2, pAbp, rm62, and smD3 genes dominantly suppress the sev-dfmr1 rough-eye phenotype, suggesting that they are required for dFMR1-mediated processes. The encoded proteins localize to dFMR1-containing neuronal mRNPs in neurites of cultured neurons, and/or have an effect on dendritic branching predicted for bona fide neuronal translational repressors. Genetic mosaic analyses indicate that dco, orb2, rm62, smD3, and dfmr1 are dispensable for translational repression of hid, a microRNA target gene, known to be repressed in wing discs by the bantam miRNA. Despite the frequency of seizure disorders in the human population, the genetic and physiological basis for these defects has been difficult to resolve. Although many genetic contributions to seizure susceptibility have been identified, these involve disparate biological processes, many of which are not neural specific. The large number and heterogeneous nature of the genes involved makes it difficult to understand the complex factors underlying the etiology of seizure disorders. Examining the effect known genetic mutations have on seizure susceptibility is one approach that may prove fruitful. This approach may be helpful in both understanding how different physiological processes affect seizure susceptibility and identifying novel therapeutic treatments. We review here factors contributing to seizure susceptibility in Drosophila, a genetically tractable system that provides a model for human seizure disorders. Seizure-like neuronal activities and behaviors in the fruit fly are described, as well as a set of mutations that exhibit features resembling some human epilepsies and render the fly sensitive to seizures. Especially interesting are descriptions of a novel class of mutations that are second-site mutations that act as seizure suppressors. These mutations revert epilepsy phenotypes back to the wild-type range of seizure susceptibility. The genes responsible for seizure suppression are cloned with the goal of identifying targets for lead compounds that may be developed into new antiepileptic drugs.

A pesar de la frecuencia de los trastornos convulsivos en la poblaci&#xF3;n humana, las bases gen&#xE9;ticas y fisiol&#xF3;gicas de estos defectos ha sido dif&#xED;cil de resolver. Aunque muchas contribuciones gen&#xE9;ticas a la susceptibilidad a las crisis han sido identificados, &#xE9;stos implican diferentes procesos biol&#xF3;gicos, muchos de los cuales no son espec&#xED;ficos de los nervios. El gran n&#xFA;mero y la naturaleza heterog&#xE9;nea de los genes implicados hace que sea dif&#xED;cil de comprender los complejos factores que subyacen a la etiolog&#xED;a de los trastornos convulsivos. Al examinar el efecto que se conoce mutaciones gen&#xE9;ticas en la susceptibilidad a convulsiones es un enfoque que puede resultar fruct&#xED;fera. Este enfoque puede ser &#xFA;til tanto en la comprensi&#xF3;n de c&#xF3;mo los diferentes procesos fisiol&#xF3;gicos afectan susceptibilidad a las crisis y la identificaci&#xF3;n de nuevos tratamientos terap&#xE9;uticos. Aqu&#xED; revisamos los factores que contribuyen a la susceptibilidad a las crisis en la Drosophila, un sistema manejable de forma gen&#xE9;tica que proporciona un modelo para los trastornos convulsivos humanos. Convulsiones: al igual que las actividades neuronales y las conductas en la mosca de la fruta se describen, as&#xED; como un conjunto de mutaciones que presentan caracter&#xED;sticas parecidas a algunas epilepsias humanos y hacer que la mosca sensible a las convulsiones. Especialmente interesantes son las descripciones de una nueva clase de mutaciones que son segundo sitio mutaciones que act&#xFA;an como supresores convulsivos. Estas mutaciones revertir fenotipos epilepsia de nuevo a la gama de tipo salvaje de la susceptibilidad a las crisis. Los genes responsables de la supresi&#xF3;n de crisis son clonadas con el objetivo de identificar los objetivos para los compuestos de plomo que se puedan desarrollar en los nuevos f&#xE1;rmacos antiepil&#xE9;pticos.
